      • MiR-130a Overcomes Gefitinib Resistance by Targeting Met in Non-Small Cell Lung Cancer Cell Lines

        Non-small cell lung cancer (NSCLC) is the most common type of lung cancer and the most common cause of lung cancer death. Currently, the epidermal growth factor receptor inhibitor gefitinib is used for its treatment; however, drug resistance is a major obstacle. Expression of Met has been associated with both primary and acquired resistance to gefitinib, but the mechanisms regulating its expression are not fully understood. Recently, miRNAs such as miR-130a have been shown to play a role in gefitinib resistance, but importance in NSCLC and relationships with Met have not been fully explored. Here we show that miR-130a is over-expressed in gefitinibsensitive NSCLC cell lines, but is low in gefitinib-resistant NSCLC cell lines. Moreover, miR-130a expression was negatively correlated with that of Met. Further analysis revealed that over-expression of miR-130a increased cell apoptosis and inhibited proliferation of NSCLC cells treated with gefitinib, whereas lowering the expression of miR-130a decreased cell apoptosis and promoted cell proliferation after treatment with gefitinib in both gefitinib-sensitive and -resistant NSCLC cell lines, suggesting that miR-130a overcomes gefitinib resistance. We also demonstrated that miR-130a binds to the 3'-UTR of Met and significantly suppresses its expression. Finally, our results showed that over-expressing Met could "rescue" the functions of miR-130a regarding cell apoptosis and proliferation after cells are treated with gefitinib. These findings indicate that the miR-130a/Met axis plays an important role in gefitinib resistance in NSCLC. Thus, the miR-130a/Met axis may be an effective therapeutic target in gefitinib-resistant lung cancer patients.

        Simulation of Permeation of Saturated Cement Paste Based on a New Meso-scale Pore Network Model

        This paper presents the simulation of the permeation of saturated cement paste based on a novel pore network model. First, a 2D hydration model of cement particles was developed by extending the work of Zheng et al. 2005 to provide the background for the network construction. Secondly, the establishment of the pore network model and simulation of permeation of saturated cement paste were carried out. The irregular pores between any two hydrated cement particles were linearized with clear distances as the diameters of pores. The straight tubular pores were interconnected with one another to form the network model. During this process, the weighted Voronoi diagram was employed to operate on the graphical expression of the hydrated cement particles. Water permeation in saturated cement paste was simulated to verify the pore network model. Finally, the factors including water-cement ratio, reaction temperature, reaction time and cement particle size that would influence water permeation were numerically investigated.

        Effects of Pressure and Deposition Time on the Characteristics of In2Se3 Films Grown by Magnetron Sputtering

        Crystalline In2Se3 films were fabricated by magnetron sputtering from a sintered In2Se3-compound target and the effects of the deposition parameters, including the working pressure and deposition time, on the phase composition, structure, morphology, and optical properties were clarified. Single-phase κ-In2Se3 was prepared at 4.0 Pa, but γ-In2Se3 was recognized when the working pressure was lower than 4.0 Pa. The optical transmittance of the films decreased to 45% and the optical band gap varied from 2.9 to 2.0 eV with increasing film thickness from 80 to 967 nm. Metal-semiconductor-metal (MSM) photodetectors based on γ-In2Se3 thin films with various thicknesses were also fabricated. The result of photosensitivity research on such MSM photodetectors suggests that it may be impossible to fabricate wide-absorption-range MSM devices by just using a single material (γ-In2Se3) because of spatial potential fluctuations in the layers.

        Expression and Immunogenicity of SARSCoV-2 Virus-Like Particles based on Recombinant Truncated HEV-3 ORF2 Capsid Protein

        COVID-19 is an emerging disease that poses a severe threat to global public health. As such, there is an urgent demand for vaccines against SARS-CoV-2, the virus that causes COVID-19. Here, we describe a virus-like nanoparticle candidate vaccine against SARS-CoV-2 produced by an E. coli expression system. The fusion protein of a truncated ORF2-encoded protein of aa 439~608 (p170) from hepatitis E virus CCJD-517 and the receptor-binding domain of the spike protein from SARSCoV-2 were expressed, purified and characterized. The antigenicity and immunogenicity of p170-RBD were evaluated in vitro and in Kunming mice. Our investigation revealed that p170-RBD selfassembled into approximately 24 nm virus-like particles, which could bind to serum from vaccinated people (p < 0.001) and receptors on cells. Immunization with p170-RBD induced the titer of IgG antibody vaccine increased from 14 days post-immunization and was significantly enhanced after a booster immunization at 28 dpi, ultimately reaching a peak level on 42 dpi with a titer of 4.97 log10. Pseudovirus neutralization tests showed that the candidate vaccine induced a strong neutralizing antibody response in mice. In this research, we demonstrated that p170-RBD possesses strong antigenicity and immunogenicity and could be a potential candidate for use in future SARS-CoV-2 vaccine development.

        Optimization of preparation and properties of Gardenia yellow pigment-loaded alginate beads

        Gardenia yellow pigment (GYP) loaded alginate beads were prepared by the ionic gelation technique, and the preparation parameters were optimized by response surface methodology for high encapsulation efficiency. The optimized parameters were alginate concentration of 3.3%, CaCl2 concentration of 2.4%, and GYP concentration of 3.2mg/mL, under which the encapsulation efficiency was 73.61%. The surface morphology and bead size analysis showed that the GYP-loaded alginate beads had a roughly spherical morphology with a wrinkled surface, and their average diameter was 0.87 mm. In vitro release test revealed that the GYP release had a pH-dependent release profile and a two-step release process. The Rigter-Peppas model was the most proper model to assess the GYP release from alginate beads. The release mechanism of GYP at pH 1.2 and 7.4 was non-Fickian transport and case-II transport, respectively. The 2,2-diphenyl-1-picrylhydrazyl assay indicated that the encapsulated GYP had effectively maintained 82.56% of the antioxidant activity.

        One Metal-Organic Framework Showing Two-Dimensional Sheet Structure: Synthesis, Structure, and Magnetic Property

        Under hydrothermal condition, the reaction of 2,4-dichlorophenoxyacetic acid (2,4-DH) and 1,3-bis(4-pyridyl)propane with MnCl2·2H2O gave a novel metal-organic framework (MOF), [Mn(2,4-D)2 (H2O)2] n (1). This complex was characterized by IR, elemental analysis, powder X-ray diffraction, and thermogravimetry analysis. X-Ray single-crystal diffraction shows that the Mn(II) ions are bidendate bridged by carboxyl groups in the syn–anti mode, giving a two-dimensional (2D) network. Temperature-dependent magnetic studies reveal that there are weak antiferromagnetic exchange interactions between the Mn(II) ions transmitted by carboxyl groups.

        Genetic Overlap in the Quantitative Resistance of Rice at the Seedling and Adult Stages to Xanthomonas oryzae pv. oryzae

        The genetic components responsible for the qualitative and quantitative resistance of rice to three Chinese races (C2, C4, and C5) of Xanthomonas oryzae pv. oryzae (Xoo)were investigated at the seedling and adult stages in two successive years in set of Lemont/Teqing cross introgression lines (ILs) in a Teqing background, to create a complete linkage map using 160 well-distributed SSR markers. Teqing was resistant to C2 and C4, but moderately susceptible to C5,whereas Lemont was susceptible to all three races. Highly significant correlations were detected among the resistance to different races at different developmental stages. A major gene (Xa4), 14 main-effect QTLs (M-QTLs), and 18 epistatic QTLs were identified in the two developmental stages over 2 years,and were largely responsible for the segregation of resistance in the ILs. In 2007, the Lemont alleles at all loci in the seedling stage, except QBbr10 to C4, increased lesion length (LL) or decreased resistance. The Teqing allele at the Xa4 locus acted as a resistance gene against C2 and C4, but acted as a M-QTL when its resistance was overcome by the virulent race C5. MQTLs showed a degree of race specificity and had a cumulative effect on resistance. Most M-QTLs (94%) consistently expressed resistance to the same race at the seedling and adult stages, indicating that a high degree of genetic overlap exists between Xoo resistance at both developmental stages in rice. Among the digenic interactions, most co-introgressed Lemont alleles at the two epistatic loci lead to significantly smaller LL with all three races, compared to other types of interacting alleles at both development stages. The results indicate that a high level of resistance may be achieved by the cumulative effect of multiple M-QTLs, including the residual effects of “defeated” major resistance genes and the epistatic effects of co-introgression from diverse susceptible varieties.

        Design, performance analysis, and testing of a GFRP racetrack ring for a high-temperature superconducting motor with a flexible support rotor

        A conceptual 20 MW high-temperature superconducting (HTS) motor (HTSM) with a novel glass-fiber reinforced plastic (GFRP) racetrack ring in the flexible support rotor was explored in this study. To support the cold mass at operating conditions, the high-strength, lowleakage GFRP racetrack ring was designed as follows. First, conceptual design of the 20 MW HTSM was conducted, and the torque transmitted by the GFRP racetrack rings was obtained through electromagnetic analysis. Second, a GFRP racetrack ring was designed, and its total cross-sectional area was obtained through theoretical analysis. Third, thermal leakage and stress were calculated and examined through theoretical and finite element analyses, respectively. The GFRP racetrack ring with the “D” ring was selected based on the results. The thickness and width of the GFRP ring were set to 16 and 10 mm, respectively, and the thickness of the “D” ring was 3 mm. Then, a full-scaled GFRP ring was fabricated, and its strength and stiffness were tested via room-temperature and low-temperature tensile tests. The average tensile strength and average Young’s modulus of the GFRP ring at room temperature were 1131.5 MPa and 49.9 GPa, respectively. The strength of the GFRP ring at 193.4 K was 1243.9 MPa, and the Young's modulus of the GFRP ring at 141.3 K was 69 GPa. Lastly, a new 200 kW HTS generator was assembled using a scaled rotor with the same structure as the 20 MW HTSM and the original stator of an existing 200 kW generator. The test system was built to evaluate the feasibility of the GFRP ring before manufacturing the 20 MW HTSM. Test results showed that the GFRP ring is feasible, the design method of the GFRP ring is credible. The GFRP ring is suitable for the cold mass support of HTSMs and other low-temperature devices.

        MicroRNAs (miRNAs) are endogenous non-coding genes that participate in post-transcription regulation by either degrading mRNA or blocking its translation. It is considered to be very important in regulating insect development and metamorphosis. Insects are the largest group of animals and are extremely valuable in biological and agriculture research. Insects are also important pests to human health and agriculture, and efforts are necessary protect both humans and plants from disease and damage. Despite their importance, insects lag behind mammals, nematodes, and plants in miRNA research. At present, only 279 insect miRNAs have been identified from Drosophila melanogaster, Anopheles gambiae, Apis mellifera, Bombyx mori, and D. pseudoobscura in miRBase, and most of these miRNAs were computationally predicted without experimental validation. Functional analysis of insect miRNAs has only been conducted in D. melanogaster.

        A Pole Pair Segment of Oil-cooling Air-Core Stator for a 2 MW Direct-Drive High Temperature Superconducting Wind Power Generator

        A 2 MW direct-drive (DD) high temperature superconducting (HTS) wind power generator with HTS wires in the rotor fi eld windings and copper transposed conductor in the stator coils was explored for the wind turbine application in this study. An oil-cooling air-core stator with non-magnetic teeth of the 2 MW DD HTS generator was designed because the high fl ux density generated by the HTS wires wound result in larger dissipation in iron teeth, and the process of that could be summarized as follows. First, a 2 MW DD HTS wind power generator was designed, and the electromagnetic (EM), loss, EM force, and insulation of the DD HTS generator were analyzed and developed, respectively. Second, the thermal and mechanical of the one pole pair oil-cooling air-gap armature was analyzed by the fi nite element analysis. Then, a pole pair segment of the oil-cooling air-core stator with the same structure as the 2 MW generator was designed and manufactured to identify potential challenges, obtain practical knowledge before production, and then reduce the development risk of the 2 MW DD HTS generator. Furthermore, a test system with converter, oil-cooling system, and data acquisition equipment was developed simultaneously to test the temperature distribution of the slotless stator. The performance test results show that the maximum temperature rise of the one pole pair oil-cooling stator under rated conditions is about 81.4 K, which satisfi es the design requirements of the 2 MW DD HTS generator.
