        Mutation in the GA3ox gene governs short-internode characteristic in a korean cucumber inbred line

        Here, we identifi ed a new natural mutant, si-2 , from a Korean cucumber inbred line. The genes responsible for dwarf (DW) and/or short internode (SI) in cucurbits were selected and used to identify the gene(s) controlling the si-2 phenotype. We evaluated the transcript levels of the candidate genes between wild type (WT) and si-2 mutant and identifi ed sequence polymorphism from the diff erentially expressed genes between the WT and si-2 mutants. Finally, we identifi ed that CsGy7G019320 encodes GA3ox (gibberellin 3-beta-dioxygenase 1-like protein) with an SNP (single nucleotide polymor- phism; A ◊ G) in an intron and a 20 bp deletion in the si-2 mutant. This fact strongly speculates that the GA3ox- mediated gibberellic acid (GA) signaling pathway could be involved in the si-2 mutant. It is well known that GAs have a signifi cant impact on plant growth and development. In the si-2 mutant, a 20 bp deletion in the promoter may abolished the transcript levels of CsGy7G019320. Promoter analysis indicated that the 20 bp deleted promoter sequence of CsGy7G019320 may be considered as the AT-hook protein binding site. Although the involvement of the genes encoding GA3ox and AT-hook protein in GA signaling has been well identifi ed, a 20 bp deletion in the promoter sequence of GA3ox in cucumber has not been revealed in other studies. Our data suggests that a mutation in the promoter sequence of CsGy7G019320 caused the si-2 mutant phenotype by suppression of GA3ox expression, possibly leading to decreased GA content in cucumber.

        지베렐린 처리가 땅콩 鍾間 交雜 親和性에 미치는 影響

        Interspecific crosses between three peanut cultivars(Arachis hypogaea) having different plant type, and four wild species, Arachis monticola, Arachis correntina, Arachis applisippla, Arachis rigonii were made by gibberellin(GA₃) treatment and the GA₃ effect on the interspecific crossability and growth of pollen tube in pistil was investigated. Pollen germination on the stigma and penetration of the pollen tubes into the style were increased by GA₃ treatment on the flower base. The GA₃ treatment increased pegging and pod formation up to 47% and 1.6% higher than those of control, respectively. Interspecific-crossability was varied with the combination of cultivar and wild species. Combination of Shipung-type variety Daekwangtangkong×wild species and Spanish-type variety Jinpungtangkong×wild species showed about 2% higher crossability than those of Virginia type variety Namdaetangkong×wild species. Arachis monticola among wild species showed the highest crossability with peanut cultivars, while Arachis correntina did the lowest. This result suggested that interspecific crossbility become higher in the cross between autoplyploids rather than those between allopolyploids.

        A novel gibberellin 2-oxidase gene CaGA2ox1 in pepper is specifically induced by incompatible plant pathogens

        Phytohormone balance is increasingly recognized as central to the outcome of plant–pathogen interactions. Differential screening for genes induced by a non-host pathogen in pepper plants (Capsicum annuum) identified a putative gibberellin 2-oxidase gene, CaGA2ox1. Analysis of the deduced amino acid sequence of CaGA2ox1 showed 53and 50 % amino acid identity to Pisum sativum PsGA2ox2and Arabidopsis AtGA2ox6, respectively. Expression in pepper plants of CaGA2ox1 was preferentially increased in response to non-host pathogen inoculation and during the host resistance response. CaGA2ox1 expression increased following treatment with salicylic acid and ethephon (albeit with different induction patterns), but remained unchanged following treatment with methyl jasmonate and abscisic acid. The gene product of CaGA2ox1 is predicted to catalyze the metabolism of GA4, and does so in recombinant E. coli extracts. Further PEG-mediated transient expression studies showed that CaGA2ox1 fused with soluble modified green fluorescent protein localized to the cytosol in chili pepper protoplasts. Interestingly, the transcript level of CaGA2ox1was not affected by treatments of either pepper with bioactive GA4?7 or paclobutrazol, an inhibitor of GA biosynthesis. Taken together, these results provide the first evidence that a GA 2-oxidase, which is important in GA metabolism,may also play a role in plant defense signaling and plant–microbe interactions.

        Characterization of Gibberellin Biosynthetic Gene Cluster from Fusarium proliferatum

        Gibberellins (GAs) are a group of phytohormones that control many developmental processes in higher plants. We report the cloning and expression pattern of gibberellin biosynthesis genes from a new GA-producing fungus, Fusarium proliferatum (strain KGL0401). These genes sequences are deposited in the National Center for Biotechnology Information (NCBI) under accession numbers EF119831, EF119832, DQ313173, DQ313174, DQ313175, DQ313176, and DQ313177. The expression level of these genes was maximal at a 0.5 M : 0.17 M carbon : nitrogen ratio, and minimal at a 0.25 M : 0.47 M carbon : nitrogen ratio.

        사과 종자 발아에 미치는 기계적 상처, 자엽제거 및 영양분의 영향

        When dehulled apple seeds (Malus pilmila var. domestica) were induced to germinate, the rate of germination, which was increased by piercing both the radicle and cotyledon, was related to the site of injury. Also, the germination rates of an embryo with ¾ cotyledon (¾ CE), embryo with ½ cotyledon (½ CE), embryo with ¼ cotyledon (¼ CE), and embryo without cotyledon (NCE) gradually decreased compared to an embryo with a normal cotyledon, but it significantly recovered on the agar media containing MS solution and/or 80 mg/L gibberellin (GA<sub>3</sub>) as nutrition. The results show that the germination of the embryo was inhibited by the partial removal of cotyledon and also demonstrated that the rate of germination could be significantly increased by adding GA<sub>3</sub> (80 mg/L) and/or MS medium. 유근과 자엽 부위에 기계적 상처를 준 사과종자의 발아율을 측정하였을 때 상처를 받지 않은 종자에 비하여 발아율이 증가되었으며, 상처부위와 밀접한 관계가 있는 것으로 나타났다. 또한 ¾자엽 부위를 갖는 배(¾CE), ½자엽 부위를 갖는 배(½CE), ¼자엽 부위를 갖는 배(¼CE) 그리고 자엽을 완전히 제거한 배(NCE)의 발아율은 정상적인 자엽을 갖는 배에 비해 감소하였으며, 감소된 발아율은 영양분으로서 MS용액과 적정 농도의 호르몬, 즉 GA<sub>3</sub>가 단독 혹은 혼용 첨가된 한천배지에서 증가되는 것으로 나타났다. 이러한 결과는 자엽 제거에 의해 사과의 발아율이 감소되며, 그 감소된 발아율은 GA<sub>3</sub> (80 mg/L)와MS용액에 의해 상당히 회복됨을 보여주었다.

        사과의 접합자배 encapsulation에 의한 인공종자 개발

        To study for germination rate of apple artificial seeds encapsulated with 2% alginic acids, the rates of germination for artificial seeds of various embryos with partial elimination of cotyledon measured by adding 80 ㎎/L gibberellic acid or/and nutritional media. The germination rate of artificial seeds produced embryo with 3/4 cotyledon (3/4 CE), embryo with 1/2 cotyledon (1/2 CE), embryo with 1/4 cotyledon (1/4 CE), and embryo without cotyledon (NCE) were gradually decreased with compare to normal embryo, but it’s considerably recovered on the alginic acid containing MS solution or/and 80 ㎎/L GA3. In special, the low frequency (55.81%) of germination for artificial seeds of 1/2 cotyledon embryo with alginic acid was increased by treatment with combination of alginic acid, 3% sucrose, and 80 ㎎/L gibberellic acid (98.74%). In case of embryo without cotyledon, the low frequency of embryo without cotyledon (24.44%) in alginic acid almost recovered to 82.45% in alginic acid with MS solution and 80 ㎎/L gibberellic acid. The results shown that the germination of embryo was decreased by the removal of cotyledon, and demonstrated that the low rate of germination could be considerably increased by adding GA3 (80 ㎎/L) or/and nutrition.

        Identification and investigation of Zoysiagrass DELLA proteins orthologous to that of maize DWARF8 gene

        Warm-season turfgrasses like zoysia grass (Zoysia spp.) are gaining traction because of their cold and salt tolerance, hardiness, shade resistance, and athletic field suitability. Compared to other turfgrass species, research into the zoysia grass genome is lagging behind. This study focused on gibberellic acid (GA), a plant hormone critical to plant height and development. Plants with disrupted GA signaling often exhibit stunted growth and flowering. DELLA proteins repress this pathway; however, GA triggers their degradation and promotes internode elongation. We used in -silico analysis to identify the DELLA genes, similar to the known maize DWARF8 gene, in the three zoysia grasses. Our analysis revealed that two of these DELLA proteins originated from Zoysia japonica, four from Zoysia matrella, and two from Zoysia pacifica. A comparison of the predicted DELLA proteins from zoysia grass with the maize DWARF8 DELLA protein model revealed a high degree of sequence similarity. We identified eight putative DELLA protein sequences with high similarity (approximately 91%) to that of the maize DWARF8 model. The aim of this study was to understand the functional significance of genetic variations within these DELLA genes across three Zoysia species using comparative genomics approaches, providing insight into the species' plant height differences.
