        白급이 멜라닌 형성 억제에 미치는 영향

        Recently many efforts were focused to understand the mechanical insights of melanogenesis to develop the agents for hyper-pigmentation and hypo-pigmentation. In the melanin biosynthetic pathway, tyrosinase is the rate limiting enzyme, and ${\alpha}$-melanocyte stimulating hormone(MSH) or cAMP-elevating agents stimulate melanogenesis and enhance the melanin synthesis and the tyrosinase activity. The author has analyzed the effects of Rhizoma Bletillae on the basal melanogenic activities of B16 mouse melanoma cells. Rhizoma Bletillae alone markedly suppressed melanin content and tyrosinase activity in a dose-dependent manner. Pretreatment of the cells with Rhizoma Bletillae. The decrease in the tyrosinase activity was paralled by a decrease in the abundance of tyrosinase protein and tyrosinase promoter activity. These results suggest that Rhizoma Bletillae inhibits melanogenesis of B16 melanoma cells via suppression of tyrosinase activity.

        Research articles : Inhibitory Effects of Ethanol Extracts from Nuruk on Oxidative Stress, Melanogenesis, and Photo-Aging

        Nuruk contributes to the unique characteristics of Korean alcoholic beverages. In this study, the effects of nuruk extracts (NE) on anti-oxidant characters, melanogenesis, and anti-photoaging activity were investigated. NEs were obtained from the 70% ethanol extracts of six types of nuruk, which have been used in brewing of fermented alcohol beverages in Korea. First, various antioxidant characteristics were identified in terms of 2, 2`-azino-bis (3-ethylbenzthiozoline-6-sulphonic acid) (ABTS) radical scavenging activity, superoxide dismutase (SOD) expression, and inhibition of xanthine oxidase activity. NE#4 exhibited potent ABTS radical scavenging activity (IC50 = 19.51ug/mL). Compared with NE#4, relatively lower levels of activity were observed for NE#3 and NE#6, with IC50 values of 90.99 and 76.88ug/mL, respectively. According to results of western blot analysis for determination of SOD expression in H2O2-treated HepG2 cells, NE#5 and NE#6 induced a dramatic increase in the expression ratio of SOD, compared to the group treated with H2O2 only. Activity of xanthine oxidase, which converts xanthine into uric acid, generating superoxide ions, was inhibited by NE#4 and NE#6 in a dose-dependent manner. NE#4 induced significant inhibition of mushroom tyrosinase activity. A reduction in cellular melanin contents of 80% was observed in B16F1 melanocytes treated with NE#5 and NE#6; these effects were similar to those of arbutin at 100 uM. In addition, gelatin zymography and reverse transcription-PCR analysis were performed for assessment of anti-photoaging activity of Nuruk. Treatment with NE#6 resulted in dramatically inhibited activities of matrix metalloproteinase (MMP)-2/ -9, suppressed expression of MMP-1, and increased expression of type-1 procollagen. Results of gelatin zymography for NE#4 and NE#5 were similar, to a slightly lesser degree. These results suggest the potential of NE#4 and NE#6 as natural ingredients for use in functional foods and cosmetics.

      • Partially purified <i>Curcuma longa</i> inhibits alpha-melanocyte-stimulating hormone-stimulated melanogenesis through extracellular signal-regulated kinase or Akt activation-mediated signalling in B16F10 cells

        <P>Abstract: </P><P>Bioassay-guided fractionation of <I>Curcuma longa</I> by solvent partitioning and purification with octadecylsilane open column chromatography yielded a partial purification. The active 80% methanol chromatographic fraction from the ethyl acetate layer [partial purification from <I>C. longa</I> (PPC)] was used to investigate the alpha-melanocyte-stimulating hormone (&agr;-MSH)-stimulated melanogenesis signal pathway in B16F10 cells. In cells stimulated &agr;-MSH, PPC inhibited cellular melanin contents, tyrosinase activity and expression of melanogenesis-related proteins including microphthalmia-associated transcription factor (MITF), tyrosinase and tyrosinase-related proteins (TRP). Melanogenesis-regulating signalling such as mitogen-activated protein kinase (MEK)/extracellular signal-regulated kinase (ERK) and phosphatidylinositol 3-kinase (PI3K)/Akt was activated by PPC in &agr;-MSH-stimulated B16F10 cells. The suppressive activity of PPC on &agr;-MSH-induced melanogenesis was abrogated by selective inhibition of MEK/ERK (PD98059) and PI3K (LY294002). MEK/ERK or Akt activation by PPC may contribute to reduced melanin synthesis via MITF and its downstream signal pathway including tyrosinase and TRPs in &agr;-MSH-induced melanogenesis.</P>

        Anti-melanogenesis activity of Ecklonia cava extract cultured in tanks with magma seawater of Jeju Island

        Ecklonia cava is popular in Korea as a marine functional materials. E. cava is generally collected and used on thecoast of Jeju Island. However, the continuous use of collected natural E. cava may be limited because difficult to securethroughout the year and may be exposed to environmental pollution. Jeju magma seawater (MSW) was known to besignificant advantages such as safety, cleanness, stability, and functional improvement. Attempts have been reportedon application of MSW to the culturing of macro- and microalgae and showed improved results. Thus, the objectiveof the present study was to explore the anti-melanogenesis activity of brown seaweed E. cava (E. cava cultured withMSW [MSWE]) extract cultured in tanks with MSW of Jeju Island to evaluate the possibility of cosmeceutical industrialapplication. MSWE extract showed the higher polyphenolic and dieckol contents than natural E. cava (NE) extract. Anti-melanogenesis activity of MSWE extract and NE extract are tested and compared using tyrosinase and dihydroxyphenylalanine(DOPA) oxidation inhibition assay. MSWE extracts evidenced more effective tyrosinase and DOPA oxidationinhibition activity than that of the NE extracts and the commercial whitening agent, arbutin. MSWE extracts alsomarkedly inhibited melanin synthesis and decreased the expression of melanogenesis-related protein in α-melanocytestimulating hormone-stimulated B16F10 melanoma cells without cytotoxicity. These results suggest that MSW cultivationprocess would be more effective in releasing bioactive compounds with whitening effect from seaweed such as E. cava at an industrial scale.

        Melanogenesis Inhibition Effect of Sargassum thunbergii

        Whitening activity of Sargassum thunbergii (S. thunbergii) collected from cost of Sacheonjin beach, Gangneung, South Korea were determined by measuring tyrosinase inhibitory and melanin formation inhibitory effects. Among them, EtOAc fraction of S. thunbergii extract exhibited strong tyrosinase inhibitory and melanin formation inhibitory activities. The EtOAc fraction was separated by Sephadex LH-20, solid phase extraction column, thin layer chromatography and high performance liquid chromatography. Finally, purified fraction, F6-S-F1 evidenced more potent tyrosinase inhibitory effect with an IC50 value of 55 μg/mL than ascorbic acid (IC50 value = 1.75 mg/mL) as a positive control. Lineweaver-Burk plots suggest that F6-S-F1 acts as a noncompetitive inhibitor against tyrosinase. Melanin formation inhibitory effect of F6-S-F1 was evaluated in B16F10 melanoma cells and F6-S-F1 suppressed both transcriptional and translational expression of tyrosinase, tyrosinase-related protein (TRP)-1 and -2, and microphthalmia-associated transcription factor (MITF). Our studies support that F6-S-F1 might prove useful as a novel inhibitor of melanogenesis for skin-whitening material.

        Melanogenesis-Promoting Effects of Rhynchosia nulubilis and Rhynchosia volubilis Ethanol Extracts in Melan-a Cells

        We evaluated the antioxidant activity and melanogenic effects of black soybean ethanol extracts, including Rhynchosia nulubilis bean ethanol extract (RNBEE), R. nulubilis leaf ethanol extract (RNLEE), R. volubilis bean ethanol extract (RVBEE), and R. volubilis leaf ethanol extract (RVLEE). The total polyphenol contents of RNBEE, RNLEE, RVBEE, and RVLEE were 16.0, 57.7, 365.9, and 260.1 mg/g, respectively. The total flavonoid contents of RNBEE, RNLEE, RVBEE, and RVLEE were 40.4, 91.7, 84.7, and 216.5 mg/g, respectively. The electron-donating abilities of RNBEE, RNLEE, RVBEE, and RVLEE at 1,000 μg/ mL were 32.4%, 12.7%, 83.5%, and 84.5%, respectively. RNBEE, RNLEE, RVBEE, and RVLEE at 50 μg/ mL significantly increased (p < 0.01) melanin contents by 30.4%, 32.1%, 35.5%, and 37.4%, respectively, compared to that of the control. RNBEE, RNLEE, RVBEE, and RVLEE at 50 μg/mL significantly increased (p < 0.01) intracellular tyrosinase activity by 18.4%, 21.8%, 21.5%, and 21.1%, respectively, compared to that of the control. These results demonstrated that black soybean ethanol extracts promote melanogenesis in melan-a cells. Among the black soybean ethanol extracts, R. volubilis was found to be more effective than R. nulubilis, and leaf extract was found to be more effective than bean extract. The potential mechanism underlying the hyperpigmentation effects of black soybeans is the promotion of tyrosinase activity.

        To evaluate the antioxidative and whitening effects of Houttuynia cordata Thunb (HCT) extract on lead acetate (LA), hair dye component, cell viability was measured by colorimetric assay after NIH3T3 fibroblasts were cultured in media containing various concentrations of LA. And also, the effect of antioxidant, vitamin E on LA-induced cytotoxicity was analysed. For the protective effect of HCT extract on LA-induced cytotoxicity, NIH3T3 fibroblasts were pretreated with 110 or 130 μg/mL of HCT extract for 2 h before the treatment of LA. And also, the antioxidative and melanogenic effects of HCT extract were assessed by superoxide dismutase (SOD)-like activity, total amount of melanin and tyrosinase activity. In this study, LA significantly decreased cell viability in dose-dependent manner. And the XTT50 value was calculated at 48.3 μM of LA. In the effect of antioxidant, vitamin E, it effectively prevented LA-induced cytotoxicity by the significant increase of cell viability. In the protective effect of HCT extract on LA-induced cytotoxicity, HCT extract significantly increased cell viability which was decreased by the cytotoxicity induced by LA, and also it showed the antioxidative effect by SOD-like activity. In the melanogenesis, HCT extract showed the decreased of amount of melanin and tyrosinase activity. From these results, it is suggested that the cytotoxicity of LA is involved in oxidative stress, and HCT extract effectively prevented the cytotoxicity and the melanogenesis induced by LA.

        느릅나무의 에틸 아세테이트 추출물에 의한 Melanin생성 효과

        Melanogenesis is a physiological process resulted in the synthesis of melanin pigments, which has a role in protecting skin from the damaging effect of ultra-violet (UV)radiation. The main aim of the present study was to examine the effect of Ulmus davidiana var. japonica (UL) on melanogenesis. Cells were cultured in the presence of various concentrations of Ulmus davidiana var. japonica For 48 h, and there were estimated total melanin contents as a final product and activity tyrosinase, a key enzyme, in melanogenesis. Among the four solvent extracts tested, EtOAc extract mostly increased tyrosinase activity And EtOAc extract increased the melanin contents and tyrosinase activity in a dose-dependent manner, Especially it was observed that 100mcg/ml/ EtOAc extract promotes melanin secretion in B16/F10 melanoma cells by 140% at 48 h treatment and activity of increased by 180% in the presence of same concentration. In conclusion, as for EtOAc extract treatment, there was no effect on the viability of B16/F10 cell, only to stimulate melanization.
