Polysaccharide of Atractylodes macrocephala Koidz Enhances Cytokine Secretion by Stimulating the TLR4–MyD88–NF-κB Signaling Pathway in the Mouse Spleen

        Bing-Xin Li,Wan-Yan Li,Yun-Bo Tian,Si-Xuan Guo,Yun-Mao Huang,Dan-Ning Xu,Nan Cao 한국식품영양과학회 2019 Journal of medicinal food Vol.22 No.9

Polysaccharide of Atractylodes macrocephala Koidz (PAMK) has been reported to have beneficial effects on regulation of immune responses in mammals and poultry. Nonetheless, the immunoregulatory mechanism of action of PAMK remains unclear. The Toll-like receptor 4 (TLR4) signaling cascade has been proved as a classic polysaccharide-regulated pathway. The aim of this study was to explore the effects of PAMK on the TLR4 signaling pathway in the regulation of spleen function in mice. Ninety-six 5-week-old BALB/c female mice were randomly allocated into four groups with three replicates per group and eight mice per replicate in a single-factor completely randomized experimental design. The control group was fed a basic diet (PAMK free); the other three groups were fed 100, 200, or 400 mg/kg PAMK for 28 days. The spleen index, concentrations of cytokines, and mRNA and protein expression levels of genes related to TLR4 signaling were determined in spleen tissue. Compared with the control group, the spleen index significantly increased in all treatment groups. Concentrations of interleukin 2 (IL-2), IL-4, interferon γ (IFN-γ), and tumor necrosis factor α (TNF-α) in the medium-PAMK group also increased significantly. PAMK in the medium-PAMK group significantly increased both mRNA and protein expression of TLR4, myeloid differentiation factor 88 (MyD88), TNFR-associated factor 6 (TRAF6), TRAF3, and nuclear factor kappa B (NF-κB) in the spleen. In conclusion, PAMK may increase immune-response capacity of the spleen in mice via TLR4–MyD88–NF-κB signaling.

Expression profiles of circular RNAs in sheep skeletal muscle

        Cao, Yang,You, Shuang,Yao, Yang,Liu, Zhi-Jin,Hazi, Wureli,Li, Cun-Yuan,Zhang, Xiang-Yu,Hou, Xiao-Xu,Wei, Jun-Chang,Li, Xiao-Yue,Wang, Da-Wei,Chen, Chuang-Fu,Zhang, Yun-Feng,Ni, Wei,Hu, Sheng-Wei Asian Australasian Association of Animal Productio 2018 Animal Bioscience Vol.31 No.10

Objective: Circular RNAs (circRNAs) are a newfound class of non-coding RNA in animals and plants. Recent studies have revealed that circRNAs play important roles in cell proliferation, differentiation, autophagy and apoptosis during development. However, there are few reports about muscle development-related circRNAs in livestock. Methods: RNA sequencing analysis was employed to identify and annotate circRNAs from longissimus dorsi of sheep. Reverse transcription followed by real-time quantitative (q) polymerase chain reaction (PCR) analysis verified the presence of these circRNAs. Targetscan7.0 and miRanda were used to analyse the interaction of circRNA-microRNA (miRNA). To investigate the function of circRNAs, an experiment was conducted to perform enrichment analysis hosting genes of circRNAs using gene ontology (GO) and Kyoto encyclopedia of genes and genomes (KEGG) pathways. Results: About 75.5 million sequences were obtained from RNA libraries of sheep skeletal muscle. These sequences were mapped to 729 genes in the sheep reference genome. We identified 886 circRNAs, including numerous circular intronic RNAs and exonic circRNAs. Reverse transcription PCR (RT-PCR) and DNA sequencing analysis confirmed the presence of several circRNAs. Real-Time RT-PCR analysis exhibited resistance of sheep circRNAs to RNase R digestion. We found that many circRNAs interacted with muscle-specific miRNAs involved in growth and development of muscle, especially circ776. The GO and KEGG enrichment analysis showed that hosting genes of circRNAs was involved in muscle cell development and signaling pathway. Conclusion: The study provides comprehensive expression profiles of circRNAs in sheep skeletal muscle. Our study offers a large number of circRNAs to facilitate a better understanding of their roles in muscle growth. Meanwhile, we suggested that circ776 could be analyzed in future study.

Cordycepin inhibits chondrocyte hypertrophy of mesenchymal stem cells through PI3K/Bapx1 and Notch signaling pathway

( Zhen Cao ),( Ce Dou ),( Jianmei Li ),( Xiangyu Tang ),( Junyu Xiang ),( Chunrong Zhao ),( Lingyu Zhu ),( Yun Bai ),( Qiang Xiang ),( Shiwu Dong ) 생화학분자생물학회(구 한국생화학분자생물학회) 2016 BMB Reports Vol.49 No.10

Mesenchymal stem cells (MSCs) are widely used in cartilage tissue engineering to repair articular cartilage defects. However, hypertrophy of chondrocytes derived from MSCs might hinder the stabilization of hyaline cartilage. Thus, it is very important to find a suitable way to maintain the chondrogenic phenotype of chondrocytes. It has been reported that cordycepin has anti-inflammatory and anti-tumor functions. However, the role of cordycepin in chondrocyte hypertrophy remains unclear. Therefore, the objective of this study was to determine the effect of cordycepin on chondrogenesis and chondrocyte hypertrophy in MSCs and ATDC5 cells. Cordycepin upregulated chondrogenic markers including Sox9 and collagen type II while down-regulated hypertrophic markers including Runx2 and collagen type X. Further exploration showed that cordycepin promoted chondrogenesis through inhibiting Nrf2 while activating BMP signaling. Besides, cordycepin suppressed chondrocyte hypertrophy through PI3K/Bapx1 pathway and Notch signaling. Our results indicated cordycepin had the potential to maintain chondrocyte phenotype and reconstruct engineered cartilage. [BMB Reports 2016; 49(10): 548-553]

Cutting fluid corrosion inhibitors from inorganic to organic: Progress and applications

        Haogang Li,Yanbin Zhang,Changhe Li,Zongming Zhou,Xiaolin Nie,Yun Chen,Huajun Cao,Bo Liu,Naiqing Zhang,Zafar Said,Sujan Debnath,Muhammad Jamil,Hafiz Muhammad Ali,Shubham Sharma 한국화학공학회 2022 Korean Journal of Chemical Engineering Vol.39 No.5

Water-based cutting fluid has a broad application area and a hundred year history, but its poor corrosioninhibition and anti-rust ability limit its further promotion. Adding corrosion inhibitors can effectively solve the aboveproblems. However, no review papers are available on cutting fluid corrosion inhibitors, and their mechanism, suitability, and performance influencing factors have not been revealed. This article discusses cutting fluid corrosion inhibitorsto fill the gaps in theoretical research and industrial applications. Inorganic matters are initially used in corrosion inhibitiondue to their strong oxidizing properties. Therefore, the film formation mechanism of inorganic corrosion inhibitoroxide and precipitation film is first analyzed, and the applications in corrosive medium are summarized. Given thatinorganic corrosion inhibitors are not environmentally friendly and expensive, organic corrosion inhibitors are currentlyused as replacement. Thus, the film formation mechanism of different organic corrosion inhibitors adsorptionfilm is analyzed, and their suitability with metals is determined. The influence of molecular structure and temperatureon their corrosion inhibition effect is also studied, and the performance of inorganic and organic corrosion inhibitors iscompared. However, single organic corrosion inhibitors are greatly affected by metal surface state, temperature, andimmersion time. Therefore, the synergistic film formation after the compounding of organic and inorganic corrosioninhibitors is analyzed. In addition, the influence rate of concentration, molecular structure, and temperature on corrosioninhibition performance is revealed, and a matching database of corrosion inhibitor type and metal type in cuttingis established. Finally, in view of the limitations of cutting fluid corrosion inhibitors, the establishment of a moleculardynamics model of corrosion inhibitor failure and accelerates corrosion and the development of general-purpose greenadditives based on the molecular design and physical and chemical analysis of the suitability of corrosion inhibitor andbase liquid are prospected.

Homing and Restorative Effects of Bone Marrow-Derived Mesenchymal Stem Cells on Cisplatin Injured Ovaries in Rats

        Jiabin Liu,Haiying Zhang,Yun Zhang,Nan Li,Yuku Wen,Fanglei Cao,Hao Ai,Xiaoou Xue 한국분자세포생물학회 2014 Molecules and cells Vol.37 No.12

Premature ovarian failure (POF) is a long-term adverse effect of chemotherapy treatment. However, current available treatment regimens are not optimal. Emerging evidence suggests that bone marrow-derived mesenchymal stem cells (BMSCs) could restore the structure and function of injured tissues, but the homing and restorative effects of BMSCs on chemotherapy injured ovaries are still not clear. In this study, we found that granulosa cell (GC) apoptosis induced by cisplatin was reduced when BMSCs were migrated to granulosa cells (GCs) in vitro. Chemo-therapy-induced POF was induced by intraperitoneal injection of cisplatin in rats. BMSCs labeled with enhanced green fluorescent protein (EGFP) were injected into the rats via the tail vein to investigate the homing and distribution of BMSCs in vivo. The number of BMSCs in the ovarian hilum and medulla was greater than in the cortex, but no BMSCs were found in the follicles and corpus lutea. In addition, the BMSCs treatment group’s antral follicle count and estradiol levels increased after 30 days, compared with the POF group. Hence, our study demonstrates that intravenously delivered BMSCs can home to the ovaries, and restore its structure and function in POF model rats.

Preparation and characteristics of carboplatin-Fe@C-loaded chitosan nanoparticles with dual physical drug-loaded mechanisms

        Yue-Hua Guo,Fu-Rong Li,Shi-Yun Bao,Tao Han,Jun-Jian Cao,Han-Xin Zhou 한국물리학회 2007 Current Applied Physics Vol.7 No.s1

The present work is intended to set up the optimal carboplatin-Fe@C-loaded chitosan nanoparticles method and to compare andassess carboplatin-Fe@C-loaded with carboplatin-Fe-loaded chitosan nanoparticles. Both kinds of nanoparticles were prepared by areverse microemulsion method. The carboplatin-Fe@C-loaded chitosan nanoparticles consisted of Fe@C nanopowder with the adsorbeddrug as the magnetic core, chitosan as the matrix and carboplatin as the model drug. The core of the carboplatin-Fe-loaded chitosannanoparticles was pure iron nanopowder, which was unable to adsorb a drug. The characteristics of both kinds of nanoparticles weredetermined and compared. The results showed that both kinds of nanoparticles were spherical in shape with an average size of210 nm ± 26 nm (size range 150300 nm) and a good magnetic responsivity. The drug content of the nanoparticles wasrespectively. The cumulative release percentages of carboplatin-Fe@C-loaded chitosan nanoparticles in vitro in 1d, 2d, 3d, 4d were60%, 74%, 84%, and 92%, respectively, and those of carboplatin-Fe-loaded chitosan nanoparticles in 1d, 2d were 81% and 91%. Thus, the carboplatin-Fe@C-loaded chitosan nanoparticles with dual physical drug-loaded mechanisms (physical encapsulation and adsorp-tion of active carbon) possessed a higher drug content and showed more sustained releasing. The cooperation of multiple mechanismswas a promising feature to improve the properties of nanoparticles.

Shear-Wave Elastography of the Breast: Added Value of a Quality Map in Diagnosis and Prediction of the Biological Characteristics of Breast Cancer

        Xueyi Zheng,Yini Huang,Yubo Liu,Yun Wang,Rushuang Mao,Fei Li,Longhui Cao,Jianhua Zhou 대한영상의학회 2020 Korean Journal of Radiology Vol.21 No.2

Objective: To determine the added value of a shear-wave elastography (SWE) quality map (QM) in the diagnosis of breast lesions and in predicting the biological characteristics of invasive breast cancer. Materials and Methods: Between January 2016 and February 2019, this study included 368 women with 368 pathologically proven breast lesions, which appeared as poor-quality regions in the QM of SWE. To measure shear-wave velocity (SWV), seven regions of interest were placed in each lesion with and without QM guidance. Under QM guidance, poor-quality areas were avoided. Diagnostic performance was calculated for mean SWV (SWVmean), max SWV (SWVmax), and standard deviation (SD) with QM guidance (SWVmean + QM, SWVmax + QM, and SD + QM, respectively) and without QM guidance (SWVmean - QM, SWVmax - QM, and SD - QM, respectively). For invasive cancers, the relationship between SWV findings and biological characteristics was investigated with and without QM guidance. Results: Of the 368 women (mean age, 47 years; SD, 10.8 years) enrolled, 159 had benign breast lesions and 209 had malignant breast lesions. SWVmean + QM (3.6 ± 1.39 m/s) and SD + QM (1.02 ± 0.84) were significantly different from SWVmean - QM (3.29 ± 1.22 m/s) and SD - QM (1.46 ± 1.06), respectively (all p < 0.001). For differential diagnosis of breast lesions, the sensitivity and areas under the receiver operating characteristic curve (AUC) of SWVmean + QM (sensitivity: 89%; AUC: 0.932) were better than those of SWVmean - QM (sensitivity, 84.2%; AUC, 0.912) (all p < 0.05). There was no significant difference in sensitivity and specificity between SD + QM and SD - QM (all p = 1.000). Among the biological characteristics of invasive cancers, lymphovascular involvement, axillary lymph node metastasis, negative estrogen receptor status, negative progesterone receptor status, positive human epidermal growth factor receptor status, and aggressive molecular subtypes showed higher SWVmean + QM (all p < 0.05), while only lymphovascular involvement showed higher SWVmean - QM (p = 0.036). Conclusion: The use of QM in SWE might improve the diagnostic performance for breast lesions and facilitate prediction of the biological characteristics of invasive breast cancers.
