        Biocatalysis and Bioprocess Engineering : Overproduction of the Escherichia coli Chaperones GroEL-GroES in Rhodococcus ruber Improves the Activity and Stability of Cell Catalysts Harboring a Nitrile Hydratase

        Three combinations of molecular chaperones from Escherichia coli (i.e., DnaK-DnaJ-GrpEGroEL- GroES, GroEL-GroES, and DnaK-DnaJ-GrpE) were overproduced in E. coli BL21, and their in vitro stabilizing effects on a nitrile hydratase (NHase) were assessed. The optimal gene combination, E. coli groEL-groES (ecgroEL-ES), was introduced into Rhodococcus ruber TH3. A novel engineered strain, R. ruber TH3G was constructed with the native NHase gene on its chromosome and the heterologous ecgroEL-ES genes in a shuttle plasmid. In R. ruber TH3G, NHase activity was enhanced 37.3% compared with the control, TH3. The in vivo stabilizing effect of ecGroEL-ES on the NHase was assessed using both acrylamide immersion and heat shock experiments. The inactivation behavior of the in vivo NHase after immersion in a solution of dynamically increased concentrations of acrylamide was particularly evident. When the acrylamide concentration was increased to 500 g/l (50%), the remaining NHase activity in TH3G was 38%, but in TH3, activity was reduced to 10%. Reactivation of the in vivo NHases after varying degrees of inactivation was further assessed. The activity of the reactivated NHase was more than 2-fold greater in TH3G than in TH3. The hydration synthesis of acrylamide catalyzed by the in vivo NHase was performed with continuous acrylonitrile feeding. The final concentration of acrylamide was 640 g/l when catalyzed by TH3G, compared with 490 g/l acrylamide by TH3. This study is the first to show that the chaperones ecGroEL-ES work well in Rhodococcus and simultaneously possess protein-folding assistance functions and the ability to stabilize and reactivate the native NHases.

        Comparison of Liver Transplantation and Liver Resection for Hepatocellular Carcinoma Patients with Portal Vein Tumor Thrombus Type I and Type II

        Purpose: The aim of this study was to compare the efficacy of liver transplantation (LT) and liver resection (LR) for hepatocellularcarcinoma (HCC) patients with portal vein tumor thrombus (PVTT) and to investigate risk factors affecting prognosis. Materials and Methods: A total of 94 HCC patients with PVTT type I (segmental PVTT) and PVTT type II (lobar PVTT) were involvedand divided into LR (n=47) and LT groups (n=47). Recurrence-free survival (RFS) and overall survival (OS) were comparedbefore and after inverse probability of treatment weighting (IPTW). Prognostic factors for RFS and OS were explored. Results: Two treatment groups were well-balanced using IPTW. In the entire cohort, LT provided a better prognosis than LR. Among patients with PVTT type I, RFS was better with LT (p=0.039); OS was not different significantly between LT and LR(p=0.093). In subgroup analysis of PVTT type I patients with α-fetoprotein (AFP) levels >200 ng/mL, LT elicited significantly longermedian RFS (18.0 months vs. 2.1 months, p=0.022) and relatively longer median OS time (23.6 months vs. 9.8 months, p=0.065). Among patients with PVTT type II, no significant differences in RFS and OS were found between LT and LR (p=0.115 and 0.335,respectively). Multivariate analyses showed treatment allocation (LR), tumor size (>5 cm), AFP and aspartate aminotransferase(AST) levels to be risk factors of RFS and treatment allocation (LR), AFP and AST as risk factors for OS. Conclusion: LT appeared to afford a better prognosis for HCC with PVTT type I than LR, especially in patients with AFP levels>200 ng/mL.

        Efficacy and Safety Profile of Combining Sorafenib with Chemotherapy in Patients with HER2-Negative Advanced Breast Cancer: A Meta-analysis

        Purpose: The aim of the study was to evaluate the efficacy andsafety of combining sorafenib with chemotherapy in patients withhuman epidermal growth factor receptor 2 (HER2)-negative advancedbreast cancer. Methods: MEDLINE, EMBASE, CochraneCentral Register of Controlled Trials, American Society for ClinicalOncology abstracts, and European Society for Medical Oncologyabstracts were searched. Randomized clinical trials that comparedthe efficacy and safety of sorafenib plus chemotherapy inpatients with HER2-negative advanced breast cancer with placeboplus chemotherapy were eligible. The endpoints were progression-free survival (PFS), overall survival (OS), time to progression(TTP), duration of response (DOR), overall response rate(ORR), clinical benefits, and adverse effects. The meta-analysiswas performed using Review Manager 5.2.6 (The Nordic CochraneCentre), and the fixed-effect model weighted by the Mantel-Haenszel method was used. When considerable heterogeneitywas found (p<0.1), further analysis (subgroup analysis, sensitivityanalysis, or random-effect model) was performed to identifythe potential cause. The results are expressed as hazard ratios orrisk ratios, with their corresponding 95% confidence intervals. Results: The final analysis included four trials comprising 844 patients. The results revealed longer PFS and TTP, and higher ORRand clinical benefit rates in patients receiving sorafenib combinedwith chemotherapy compared to those receiving chemotherapyand placebo. OS and DOR were similar in the two groups. Meanwhile,the incidence of some adverse effects, including hand-footskin reaction/hand-foot syndrome, diarrhea, rash, and hypertension,were significantly higher in the sorafenib arm. Conclusion:Sorafenib combined with chemotherapy may prolong PFS andTTP. This treatment was associated with manageable toxicities,but frequent dose interruptions and reductions were required.

        Keratin/PEO/hydroxyapatite Nanofiber Membrane with Improved Mechanical Property for Potential Burn Dressing Application

        Keratin, as a promising substitute for tissue engineering due to its excellent biocompatibility and bioactivity, is used to combine one or more other polymers together. However, compound nanofibers with high keratin content (normally>90 wt.%) may result in the poor elongation of nanofiber membranes such as wound dressing. In this work, different ratios of hydroxyapatites (HA) modified by sodium hexametaphosphate were blended with keratin/polyethylene oxide (PEO) spinning solution to produce reinforced keratin blend nanofiber nonwoven membranes as a potential candidate wound dressing. The tensile strength of keratin blend nanofiber membrane with 15 % modified HA addition was two times higher than that without HA. The morphologies and chemical structure of keratin/PEO/HA nanofiber membranes were investigated using SEM, FTIR, and TG. The biocompatibility and the burn repairing performance of keratin/PEO/HA nanofiber mat were also investigated by cell culture and animal burn model. The results showed that the Keratin/PEO/HA nanofiber membranewas beneficial to enhance the proliferation of L929 cell, exhibiting an advantages in reducing inflammatory response in the infective stage and enhancing skin repairing process in the following recover stages. Our data suggested that keratin/PEO/HA nanofiber membrane could serve as a promising burn dressing for treatment of the skin burn.

        Microwave-assisted pyrolysis of phosphoric acid-activated Goldenberry peel powder biochar for enhancing the adsorption of trace beta-lactamase inhibitors

        Novel efficient biochar of Goldenberry peels (GBPMW-H3PO4) was prepared through a microwave-assistedphosphoric acid activation method. It was characterized and used for removing two beta-lactamase inhibitors, sulbactam(SAM, first listed in Japan in 1986) and avibactam (AVI, first listed in the U.S. in 2015), from aqueous solution. Characterization confirmed that GBPMW-H3PO4 displayed a high surface area (720.046m2 g1), more abundant porestructure, smaller particle size, and higher thermal stability. The experimental results showed that the adsorption of thetwo antibiotics was a spontaneous, favorable, and endothermic process, highly dependent on solution pH. A contacttime of 60 min assured equilibrium, and GBPMW-H3PO4 followed pseudo-first-order kinetics (R2=0.9950-0.9977). Furthermore,the adsorption capacities of GBPMW-H3PO4 for SAM and AVI were 211.86 and 198.81mg g1, respectively,and the performance was better than that of unmodified biochar. Microscopically, the main mechanism could beexplained by - electron donor-acceptor interaction, hydrogen bonding interaction, -hydrogen bonding, hydrophobicinteraction, and electrostatic interaction. The study demonstrates that the microwave-assisted H3PO4 activationmethod could produce biochar, and GBPMW-H3PO4 was confirmed to be a low-cost and high-efficiency adsorbent forremoving beta-lactamase inhibitors from medical wastewater.

      • Research about the Evaporative Cooling Sleeve of 3.6 MW Wind Generator Stator

        The evaporative cooling technology used in Wind generator stator has unique advantages. Combined with the structure of motor stator and operating conditions, this report based on the research project for the evaporative cooling sleeve of the 3.6MW wind generator, introduces the material requirements and structural characteristics of the sleeve, simulates on the stress, displacement and stability by finite analysis method, and tests the products experimentally. The research results show that the epoxy resin-glass materials have a higher strength and better insulation properties, but the evaporative cooling of the wind generator stator sleeve, because of its thin-walled, and the external pressure, so it's the less rigid. Should make full use of the motor stator core structure, increase its stiffness and improve the stability of the epoxy resin-glass sleeve, which for thin-walled the epoxy resinglass sleeve on the successful application of wind turbines has played an important role.

        Transcriptional activation of insulin-like growth factor binding protein 6 by 17β-estradiol in SaOS-2 cells

        Osteoblasts can synthesize the insulin-like growth factors (IGFs) and the IGF-binding proteins (IGFBPs), which may either enhance or attenuate IGF-stimulated bone cell proliferation. Since estrogen induced osteoblastic differentiation and proliferation through an estrogen- responsive gene in target cells, we investigated the effects of estrogen on IGFBP-6 expression in the human osteoblastic-like cell line SaOS-2. Expressions of IGFBP-6 protein and mRNA increased 2.8 and 2-fold, respectively, in the presence of 17-β-estradiol (E2) (0.01 to 1 μM) and estrogen receptor (ER) in SaOS-2 cells. On the other hand, E2 induced a 2-fold increase in SaOS-2 cell proliferation. To identify genomic sequences associated with estrogen responsiveness, the 5'-promoter region (-44 to +118) of the IGFBP-6 gene was cloned into a chloramphenicol acetyltransferase (CAT) reporter vector. E2 induced a 3-fold increase in CAT activity in SaOS-2 cells transiently transfected with this construct. Identification of the estrogen- responsive element (ERE) [5'-CCTTCA CCTG-3'] (-9 to +1) in this IGFBP-6 gene promoter region was confirmed using electromobility shift assays and deletion analysis. This functional ERE was important for E2-induced trans-activation of the IGFBP-6 gene. These results demonstrate that E2 exhibits a positive effect on IGFBP-6 gene transcription through estrogen- liganded ER binding to the functional ERE in the IGFBP-6 gene promoter in SaOS-2 cells.       • Benchtop fabrication of three-dimensional reconfigurable microfluidic devices from paper-polymer composite.

        <P>We presented a benchtop technique that can fabricate reconfigurable, three-dimensional (3D) microfluidic devices made from a soft paper-polymer composite. This fabrication approach can produce microchannels at a minimal width of 100 μm and can be used to prototype 3D microfluidic devices by simple bending and stretching. The entire fabrication process can be finished in 2 hours on a laboratory bench without the need for special equipment involved in lithography. Various functional microfluidic devices (e.g., droplet generator and reconfigurable electronic circuit) were prepared using this paper-polymer hybrid microfluidic system. The developed method can be applied in a wide range of standard applications and emerging technologies such as liquid-phase electronics.</P>

        Prevalence, Risk Factors, and Psychological Effects of Primary Nocturnal Enuresis in Chinese Young Adults

        Purpose: This study aimed to investigate the prevalence, risk factors, and effects of primary nocturnal enuresis (PNE) on physical and mental health in young adults in mainland China. Methods: An anonymous questionnaire was used to collect information including the sociodemographic characteristics, history of PNE, family history, daytime voiding symptoms, Pittsburgh Sleep Quality Index (PSQI) scores, Self-Esteem Scale (SES), and Self-Rating Depression Scale (SDS). A total of 22,500 university students from 23 provinces and 368 cities in mainland China were included. Results: In total, 21,082 questionnaires were collected, and 20,345 of them qualified for statistical analysis. The PNE prevalence was 1.17%, and the distribution of monosymptomatic nocturnal enuresis (MNE) and nonmonosymptomatic nocturnal enuresis (NMNE) was 66.1% and 33.9%, respectively. In total, 28% of respondents with PNE reported bedwetting daily, 31.6% between 1 and 7 times weekly, and 40.4% between 1 and 4 times monthly; 80% of PNE cases had no history of treatment. The prevalence of PNE in patients with a family history, frequency, urgency, urinary incontinence, and recurrent urinary tract infections was significantly higher than in those without these conditions (P<0.001). PNE was significantly correlated with the PSQI total score (sleep quality) (P=0.011). The SES score was lower and the SDS was higher (P<0.001) in the PNE group than in those without PNE. Conclusions: In mainland China, the PNE prevalence among young adults was found to be high, and PNE had significant effects on physical and mental health. Risk factors included a family history, daytime voiding symptoms, and lack of treatment.
