      • Serratia marcescens에서 cAMP receptor protein(CRP)유전자의 클로닝 및 해석

        전사조절인자로서 잘 알려져 있는 cAMP receptor protein(CRP)은 cAMP와 DNA에 결합하는 특별한 활성을 가지고 있으며, cAMP-CRP complex를 형성하여 수많은 유전자의 발현조절에 관여한다. 이러한 측면에서 cAMP-CRP의 조절은 어떤 면에서 총체적 조절체계라고까지 한다. 본 연구는 Serratia 균주에서 crp유전자의 분자적 특성 및 cAMP에 의한 발현조절을 받는 분자기구를 해석하고자 유전자를 클로닝하고 발현을 확인하였다. MacConkey배지에서 maltose를 탄소원으로 충분히 이용하지 못하는 대장균 TP2139(△crp,△lac)를 숙주로 이용하고, 염색체DNA를 library로 작성하여 얻은 형질전환체 약 일만개의 콜로니에서 red colony를 나타내는 5종류 의 양성 클론을 얻었다. 이들 클론을 Southern방법으로 확인한 결과 3kb의 단편을 가진 pCKB12클론이 crp유전자를 coding하고 있음을 확인하였다. glpD-lacZ 융합 plasmid인 pLDC6 의 BamHI부위에 pCKB12의 3kb 단편을 삽입시킨 재조합 plasmid pLDC 6-Scrp를 작성하여, 클로닝된 Serratia 의 crp 유전자가 대장균에서 유전자 전사조절에 미치는 영향을 확인한 결과 cAMP-CRP 복합체 형성에 의한 전사조절 기능이 확인되어졌다. One of the better-characterized transcription factor of E. coli is the cAMP receptor protein(CRP) and the CRP binds cAMP and DNA. The cAMP-CRP complex is involved in regulation of many genes at bacteria. The cAMP-CRP regulatory element represents, in some respects, a global regulatory network. The aim of this work was to study the structure and the mechanisms controlling the expression of CRP in Serratia marcescens. We have been get 5 different clones from Serratia which stimulated the cells to use maltose as a sole carbon source in E. coli TP2139. The crp gene clone, pCKB12, was confirmed by Southern hybridization with E. coli crp gene. The location of the crp gene was determined by constructing subclones carrying various portions of pCKB12. To investigate the potential role of CRP in E. cloi, lacZ fused plasmids were constructed and investigated the β-galactosidase activity of the fused plasmid. The Serratia marcescens cAMP receptor protein can substitute the E. coli CRP in transcriptional activation at the lacZ gene. These results suggest that Serratia marcescens cAMP receptor protein complex functions to regulate several promoters in E. coli.

      • 팥 신장기 과습처리에 의한 생육특성 및 단백질 발현

        Adzuki bean is sensitive to waterlogging stress. The overall study on the waterlogging stress is limited comparing to the study on the drought and any environmental stress. Therefore, the present study was conducted to investigate the changes in growth characteristics and the expression patterns of proteins at the 5-leaf stage periods of adzuki bean. The domestic cultivar, Arari (Miryang No. 8) was used to test the waterlogging stress. In the waterlogging treatment for 3 days, the plant height showed slightly decrease in the treatment at 3 days of waterlogging, but root fresh weight showed significantly changes at 3 days of the waterlogging treatment. Chlorophyll contents showed also significantly different at 3 days of waterlogging treatment compared to control the plants. The waterlogging stress gradually influenced the growth differences between the control and the treatment respectively. More than 350 pro- tein spots were identified on 2-D gels using an image analysis. Moreover, a total of 28 proteins were analyzed using LTQ-FT-ICR MS. Among these 28 proteins, a total of 18 proteins were up-regulated, and 10 proteins were down-regulated under waterlogging treatment. According to biological process, the most of the proteins were found to be involved in carbohydrate metabolism. Regarding the subcellular local- ization, most of the proteins were localized into chloroplasts in 5-leaf stage.

        We have encountered a rare case of symmetrical peripheral gangrene complicating Escherichia coli sepsis in a 47-years-old male. He was successfully treated with antibiotics, anticoagulants, and vasodilator. To our knowledge, this is the first report on symmetrical peripheral gangrene complicating E. coli sepsis in Korea.

        간암 환자에 합병된 Citrobacter braakii 패혈증 1례 보고

        Citrobacter species is a gram-negative bacilli that can cause opportunistic infections in immunocompromised hosts. Citrobacter braakii refers to the genomospecies 6 of the Citrobacter freundii complex. There are no detailed studies on infections caused by this newly identified specific genetic species in Korea. We herein report a case of a patient with hepatocellular carcinoma who, after undergoing transcatheter arterial chemoembolization, developed biloma which later progressed to C.braakii sepsis and did not respond to treatment. To our knowledge, this is the first reported case in Korea on C. braakii infection resulting in septic shock in a patient with malignancy in Korea.

        Digital Imaging Fiber-Optic Trans-Illumination을 이용한 초기 법랑질 우식병소의 조기 진단

        치아 우식증의 발생과 관련된 분야에 대한 연구는 지난 20년간 활발히 진행되어 괄목할만한 발전을 이루었다. 그러나 치아 우식증의 원천적인 예방을 이루기 위해서는 보다 새로운 실험 장비와 기구를 이용한 다각적인 연구가 요구되며, 이러한 흐름에 부응하여 미국의 인디아나 치과대학을 중심으로 초기 법랑질 우식증에 관한 재조명이 집중적으로 이루어지고 있다. 또한 세계적으로 치과계의 지속적인 대민 교육과 홍보 및 불소화 사업 등의 우식 예방에 대한 노력과 구강 보건에 대한 대중의 인식 향상을 통해 치아 우식증이 감소하는 추세에 있으며, 이로 인해 치아 우식증이 기존의 교합면보다 인접면에서 더 많이 발견되는 추세로 변화되고 있다. 치아 우식증의 조기 진단을 목적으로 새로운 진단 장비들이 속속 개발되고 있으며 이미 성능의 우수성이 실험실 연구를 통해 입증된 바 있다. 본 연구의 목적은 초기 인접면 우식증의 진단에 있어 새로 개발된 DIFOTI^(TM) 시스템의 효능을 기존의 방법인 사진 및 교익방사선사진과 비교 평가하고, 임상 적용시의 문제점을 파악하여 차후 DIFOTI^(TM) 시스템 개발에 필요한 개선안을 제시함과 아울러 치아 우식증의 예방 및 불소를 이용한 초기 우식증 재광화 방법에 대한 기초 연구 자료를 마련하고자 하였다. 학동기 연령에 있는 유치 탈락 시기에 근접한 것으로 기대되는 23명의 아동을 대상으로 구강 검진 2회, 구치부 교익 방사선 필름 판독 2회 그리고 전치부 및 구치부 DIFOTI^(TM) 이미지 판독 2회를 실시하고 각 방법에 대한 신뢰도 평가를 시행한 결과 다음과 같은 결론을 얻었다. 1. 구강 검진시 검사자간 신뢰도는 교합면에서 평균 0.8470으로 가장 높았으며, 근심면 평균 0.6430 원심면 평균 0.5727, 설면 평균 0.2807 그리고 협면 평균 0.2339 순으로 나타났다. 구치부에 국한시킨 경우 교합면에서는 평균 0.8577이었으며, 원심면 평균 0.8211, 설면 평균 0.7728, 협면 평균 0.7152, 근심면 평균 0.6782 순으로 나타났다. 2. 구치부 교익 방사선 사진 판독 결과에 대한 검사자간 신뢰도는 교합면 평균 0.8346, 근심면 평균 0.8675, 원심면 평균 0.8482 순으로 나타났다. 3. DIFOTI^(TM) 이미지 판독 결과에 대한 검사자간 신뢰도는 교합면 평균 0.8437, 협면 평균 8379, 근심면 평균 0.8223, 설면 평균 0.7766, 원심면 평균 0.6781 순으로 나타났다. 4. 치아 우식증 진단율을 비교한 결과 교합면, 협면, 설면에서는 DIFOTI^(TM) 이미지 판독이 가장 우수한 것으로 나타났으며(p<0.05), 근심면과 원심면에서는 방사선 판독이 가장 우수한 것으로 나타났다(p<0.05). Over the past 20 years, great strides have been made in research regarding the mechanisms involved in the progression of carious lesions, but new equipment and research tools need to be developed to continue these advancements in caries research. Various methods have been applied to reduce the incidence of carious lesions, which have led to a significant decrease in the number of occlusal caries, but a concurrent increase in the proportion of proximal carious lesions. New diagnostic equipment has been developed to detect early stage carious lesions, and these have demonstrated excellent laboratory results and show promise in clinical applications. The research presented here examines the efficacy of the newly developed DIFOTI^(TM) system in detecting proximal carious lesions compared to traditional intraoral exam and bitewing radiography, possible problems or deficiencies of using the system in clinic, possible improvements that can be made to the system, and the efficacy of detecting early, reversible carious lesions that can be remineralized by preventative fluoride applications. The subject pool consisted of 23 grammer school age patients just prior to entering the mixed dentition phase. Each patient was given a thorough oral examination, radiographic examination consisting of bitewing radiographs of the posterior teeth, and DIFOTIR examination of the anterior and posterior teeth. Each examination was carried out two times by two examiners, and the data were statistically analyzed. The results are as follows: 1. The mean alpha value of reliability test of the visual oral examination was as follows: occlusal surface was 0.8470, mesial surface was 0.6430, distal surface was 0.5727, lingual surface was 0.2807 and distal surface was 0.2339. When the examination was limited to posterior teeth, the mean alpha value was as follows: occlusal surface was 0.8577, distal surface was 0.8211, lingual surface was 0.7728, buccal surface was 0.7152 and mesial surface was 0.6782. 2. The alpha value of reliability test of the radiographic analysis of carious lesions of the occlusal, mesial, and distal surfaces was 0.8500. 3. The alpha value of reliability test of the DIFOTI^(TM) diagnostic analysis of carious lesions of the occlusal, buccal, lingual, mesial, and distal surfaces was determined to be 0.7917. 4. The DIFOTI^(TM) diagnostic system was found to be the most accurate means of detecting occlusal, buccal, and lingual surface carious lesions (p<0.05), while mesial and distal proximal carious lesions were most accurately assessed using bitewing radiography (p<0.05).

      • Cimetidine製劑의 分析方法에 관한 考察

        The results on the analysis of cimetidine tablets and injections by UV determination, colorimetric determination and HPLC determination are as follows; 1. Cimetidine tablet can be easily made by wet granulation method and diluents in core tablets and film coating tablet do not interfere with analysis. 2. Cimetidine injections are made by aseptic manipulation or autoclaving. When they are adjusted approximately to pH 6.0 because they are stable under the strong acidic condition, they have high stability without the performance of complicated aseptic manipulation 3. UV determination shows more stable absorption spectrum by the Me-OH than by the 0.01-HCL as a solvent. It can be considered as a desirable quantitative analysis for cimetidine material. 4. Colorimetric derermination shows much better results by the BCG dye than by BTB dye. BCG dye doesn't be affected by diluents in tablets and preservatives in injections. 5. HPLC determination using the μBondapak C_18 column can be used for the quantitative analysis of cimetidine preparations. It shows the peak of main component in 1 minute after injection of sample. Compared with other analytical methods, HPLC determination is recognized as a desirable analytical method because of its high representation and short time for measuring the content uniformity in cimetidine preparations.

      • 1kW 소형 풍력발전기의 진동 성능 모니터링

        A vibration monitoring is performed on a 1kW class stand alone wind turbine(W/T). When a W/T model is developed, general performance under various wind condition should be verified to introduce the product in the market. Especially, vibration characteristics within operating speed range are very important in the aspect of structural stability as well as generator's electrical efficiency. This paper examines the vibration performance of a home made 1kW W/T. Various data of the W/T model are acquired in real time using a remote vibration monitoring system installed in Daekwanryung test site. Vibration stability of the W/T structure is diagnosed based upon the data and the result is used to estimate the applicability of the W/T model.

        6kW 독립형 풍력발전기의 진동 모니터링 및 분석

        A vibration monitoring system for a small class of wind turbine (W/T) is established and operated. The monitoring system consists of monolithic integrated chip accelerometer for vibration monitoring, anemometers for wind data acquisition and auxiliary sensors for atmospheric data. Using the monitoring system, vibration response of a 6kW W/T generator is investigated. Acceleration data of the W/T tower under various operation condition is acquired in real time using LabVIEW and is remotely transferred from the test site to the laboratory in school by internet. Vibration state of the tower structure is diagnosed within the operating speed range. Resonance frequency range of the test model is investigated with the wind speed data of the test site.
