      • ‘숙명역사관’뮤지엄 아이덴티티 개선을 위한 마케팅, 디자인 전략 수립 -특별 기획전, 홍보, 기프트 샵을 중심으로-

        본 연구는 디자인과 마케팅이 이분법적으로 나누어질 수 없음을 인식하고, 디자인과 마케팅을 융합하여 스페이스 디자인 마케팅을 통한 뮤지엄아이덴티티를 구축하는 일련의 프로세스를 제안하는데 목적이 있다. 본 연구는 2017년 3월~6월 총 15주간 숙명여자대학교 환경디자인과 ‘브랜드·스페이스·마케팅’수업에 서 진행된 결과물로서 작업물을 정리하였다. 본 연구의 범위는 4개(物, 事, 人, 場)의 주제 중, 장소(場)를 중심으로 ‘숙명역사관’의 그래픽, 제품, 공간을 개발하는 브랜드 매뉴얼과 연관하여 한정한다. 이에 본 연구는 ‘숙명역사관’이 가지고 있는 역사적 컨텐츠와 가치를 통해 현재 상황과 문제점을 분석하였다. 그 결과 ‘숙명역사관’의 다양한 마케팅 전략을 제시하고 이에 따른 디자인 전략과 해결 방안을 통해 디자인과 마케팅의 융합을 꾀하였다. 아울러 SWOT 분석을 통해 로고, 명함부터 새로 기획하는 숙명역사관 기프트샵, 인테리어디자인, 디스플레이, 공간연출과 그 안의 제품디자인에 이르기까지 일관성과 통일성을 추구했다. 끝으로 본 연구는 뮤지엄아이덴티티 구축을 위한 창의적 디자인 프로세스를 고찰하며, 이를 바탕으로 개발 매뉴얼을 총체적으로 정리함에 있어서 뮤지엄 브랜딩의 기초 자료로 활용될 것으로 기대된다.

      • 여수국가산업단지 인근 지역 대기오염물질 농도에 영향을 미치는 요인분석

        Eight monitoring stations in the Yeosu National Industrial Complex will determine the concentration of air pollutants measured every hour in 2019, analyze what factors affect them, and how they correlate with the air pollutant measurements. Six air pollutants (SO2, NO2, O3, CO, PM-10 and PM-2.5) measured at eight monitoring stations around Yeosu National Industrial Complex (Seogang-dong, Wolnae-dong, Munsu-dong, Yeocheon-dong, Deokchung-dong, Hwayang-myeon, Yulchon-myeon, Samil-dong) the measurement data for each hour of the year were downloaded to analyze the concentration by weather condition, time zone, season, separation distance from Yeosu Industrial Complex, and separation distance from Gwangyang Industrial Complex. SO2, NO2, and CO concentrations were significantly higher in winter, significantly lower in summer, and O3, PM-10, and PM-2.5 concentrations were significantly higher in spring. SO2, CO, PM-10, and PM-2.5 concentrations were significantly higher between 10 a.m. and 12 p.m., NO2 concentrations were significantly higher between 7 a.m. and 9 a.m., and O3 concentrations were significantly higher between 13 p.m. and 15 p.m.(p<0.001). The concentration of air pollutants (SO2, NO2, CO, PM-10, PM-2.5) excluding O3 was found to have a negative (-) correlation with weather condition, and in the case of O3, it was found to have a positive (+) correlation with PM-10, PM-2.5, temperature and wind speed. It was confirmed that the concentration of air pollutants emitted from the Yeosu National Industrial Complex differs depending on the season, time and distance to the industrial complex. This study aims to establish measures to minimize health damage to residents living relatively close to the industrial complex and contribute to the establishment of management and response measures according to the properties of substances to reduce the concentration of air pollutants in the future.

        Effect of Korean Red Ginseng extract on colorectal lung metastasis through inhibiting the epithelialemesenchymal transition via transforming growth factor-b1/Smad-signaling-mediated Snail/E-cadherin expression

        Background: In colorectal cancer (CRC), 40e60% of patients develop metastasis. The epithelialemesenchymal transition (EMT) is a pivotal and intricate process that increases the metastatic potential of CRC. The aim of this study was to investigate the effect of Korean Red Ginseng extract (RGE) on colorectal metastasis through inhibition of EMT and the metastatic abilities of CRC cells. Methods: To investigate the effect of RGE on the metastatic phenotypes of CRC cells, CT26 and HT29 cells were evaluated by using an adhesion assay, a wound-healing assay, an invasion assay, zymography, and real-time reverse transcriptionepolymerase chain reaction. Western-blot analysis was conducted to elucidate the molecular mechanisms of RGE, which showed an inhibitory effect on the transforming growth factor-b1 (TGF-b1)-induced EMT in HT29 cells. Additionally, the antimetastatic effect of RGE was evaluated in a mouse model of lung metastasis injected with CT26 cells. Results: RGE decreased the adhesion and migration ability of the CT26 cells and TGF-b1-treated HT29 cells. The invasion ability was also reduced by RGE treatment through the inhibition of matrix metalloproteinase-9 expression and activity. Moreover, RGE suppressed the TGF-b1-induced EMT via TGF-b1/Smad-signaling-mediated Snail/E-cadherin expression in HT29 cells and lung tissue in CT26 tumor-bearing mice. Conclusion: Our results demonstrated that RGE inhibited colorectal lung metastasis through a reduction in metastatic phenotypes, such as migration, invasion, and the EMT of CRC cells.

        Korean Red Ginseng improves atopic dermatitis-like skin lesions by suppressing expression of proinflammatory cytokines and chemokines in vivo and in vitro

        Background: The prevalence of allergic inflammatory diseases such as atopic dermatitis (AD), asthma, and allergic rhinitis worldwide has increased and complete recovery is difficult. Korean Red Ginseng, which is the heat-processed root of Panax ginseng Meyer, is widely and frequently used as a traditional medicine in East Asia. In this study, we investigated whether Korean Red Ginseng water extract (RGE) regulates the expression of proinflammatory cytokines and chemokines via the mitogen-activated protein kinases (MAPKs)/nuclear factor kappa B (NF-kB) pathway in allergic inflammation. Methods: Compound 48/80-induced anaphylactic shock and 1-fluoro-2,4-dinitrobenzene (DNFB)-induced AD-like skin lesion mice models were used to investigate the antiallergic effects of RGE. Human keratinocytes (HaCaT cells) and human mast cells (HMC-1) were also used to clarify the effects of RGE on the expression of proinflammatory cytokines and chemokines. Results: Anaphylactic shock and DNFB-induced AD-like skin lesions were attenuated by RGE administration through reduction of serum immunoglobulin E (IgE) and interleukin (IL)-6 levels in mouse models. RGE also reduced the production of proinflammatory cytokines including IL-1β, IL-6, and IL-8, and expression of chemokines such as IL-8, thymus and activation-regulated chemokine (TARC), and macrophage-derived chemokine (MDC) in HaCaT cells. Additionally, RGE decreased the release of tumor necrosis factor-α (TNF-α), IL-1β, IL-6, and IL-8 as well as expressions of chemokines including macrophage inflammatory protein (MIP)-1α, MIP-1b, regulated on activation, normal T cell expressed and secreted (RANTES), monocyte chemoattractant protein (MCP)-1, and IL-8 in HMC-1 cells. Furthermore, our data demonstrated that these inhibitory effects occurred through blockage of the MAPK and NF-kB pathway. Conclusion: RGE may be a useful therapeutic agent for the treatment of allergic inflammatory diseases such as AD-like dermatitis.

      • Anti-obesity effects of Arctii Fructus (Arctium lappa) in white/brown adipocytes and high-fat diet-induced obese mice

        <P>Arctii Fructus is traditionally used in oriental pharmacies as an anti-inflammatory medicine. Although several studies have shown its anti-inflammatory effects, there have been no reports on its use in obesity related studies. In this study, the anti-obesity effect of Arctii Fructus was investigated in high-fat diet (HFD)-induced obese mice, and the effect was confirmed in white and primary cultured brown adipocytes. Arctii Fructus inhibited weight gain and reduced the mass of white adipose tissue in HFD-induced obese mice. Serum levels of triglyceride and LDL-cholesterol were reduced, and HDL-cholesterol was increased in the Arctii Fructus treated group. In 3T3-L1 cells, a water extract (WAF) and 70% EtOH extract (EtAF) of Arctii Fructus significantly inhibited adipogenesis and suppressed the expression of proliferator-activated receptor gamma and CCAAT/enhancer-binding protein alpha. In particular, EtAF activated the phosphorylation of AMP-activated protein kinase. On the other hand, uncoupling protein 1 and peroxisome proliferator-activated receptor gamma coactivator 1-alpha, known as brown adipocytes specific genes, were increased in primary cultured brown adipocytes by WAF and EtAF. This study shows that Arctii Fructus prevents the development of obesity through the inhibition of white adipocyte differentiation and activation of brown adipocyte differentiation which suggests that Arctii Fructus could be an effective therapeutic for treating or preventing obesity.</P>

      • Up-dating of new dCAPS markers for mapping yield-related traits using MGRIL

        The next-generation sequencing(NGS) technology is being used for more effective genetic mapping. In previous study, we obtained 60x coverage of sequence from Milyang23 and Gihobyeo on average comparing with Nipponbare reference genome. Also, we developed new derived cleaved amplified polymorphic sequence(dCAPS) markers based on the single nucleotide polymorphisms(SNPs) in coding region sequence(CDS) between these varieties. Totally, 1,726,798 SNPs between Milyang23 and Gihobyeo were detected. Among them, 146 SNP were selected for making dCAPS markers and located on genetic map with previously reported 219 PCR-based DNA markers. The map was applied to the detection of quantitative trait loci(QTLs) for stem internode diameters, culm length and panicle length within MGRIL population, and six QTLs with relatively high LOD score were found at three chromosomes; culm length and stem diameter including the first internode diameter, third and fourth internode diameter. This study showed that the NGS allowed the rapid discovery of a large number of SNPs for dCAPS marker. So, we tried to find out more single nucleotide polymorphisms(SNPs) which were located on the whole genome sequence, such as un-translated region(UTR), intron, Inter-region and coding region sequence(CDS) between Milyang23 and Gihobyeo varieties. And we collected phenotypic information about culm length, panicle length, four stem internode diameters and panicle number in rice MGRIL population for QTLs. Furthermore, results of QTL analysis described above will shows relevance of molecular markers in mapping genes for useful breeding.

        Rubrofusarin-6-β-gentiobioside inhibits lipid accumulation and weight gain by regulating AMPK/mTOR signaling

        <P><B>Abstract</B></P> <P><B>Background</B></P> <P>Although rubrofusarin-6-β-gentiobioside (RFG), which is a component of <I>Cassiae tora</I> seed, could likely regulate hyperlipidemia, its anti-obesity effect and related mechanism have not been elucidated.</P> <P><B>Purpose</B></P> <P>The aim of this study was to examine whether RFG can ameliorate obesity and the mechanism of lipid accumulation regulated by RFG.</P> <P><B>Study design</B></P> <P>In <I>in vitro</I> experiments, we confirmed the anti-adipogenic effect of RFG using 3T3-L1 cells and human adipose mesenchymal stem cells (hAMSCs). To confirm the anti-obesity effect, High-Fat Diet (HFD)-induced obese mice were selected as a model.</P> <P><B>Methods</B></P> <P>We investigated anti-adipogenic effects of RFG using MTS assay, Oil Red O Staining, real-time RT-PCR, western blot analysis, and immunofluorescence staining. The anti-obesity effect of RFG was confirmed in HFD-induced mice model using hematoxylin and eosin staining and serum analysis.</P> <P><B>Results</B></P> <P>RFG inhibited lipid accumulation in 3T3-L1 cells and hAMSCs by reducing expression of mammalian targets of rapamycin (mTOR), peroxisome proliferator-activated receptor (PPAR)γ, and CCAAT-enhancer binding protein (C/EBP)α. RFG phosphorylated AMP-activated protein kinase (AMPK) in a liver kinase B (LKB) 1-independent manner. Moreover, the anti-adipogenic effect of RFG was blocked by AMPK inhibitor. These results suggest that RFG inhibits lipid accumulation via AMPK signaling. Furthermore, RFG reduced the body weight, size of epididymal white adipose tissue (eWAT), and fatty liver in the mice. RFG also suppressed levels of adipogenic factors PPARγ, C/EBPα, FAS, LPL, and aP2) by activating AMPK in the eWAT and liver.</P> <P><B>Conclusion</B></P> <P>RFG can ameliorate obesity, and thus, could be used as a therapeutic agent for treating obesity.</P> <P><B>Graphical abstract</B></P> <P>[DISPLAY OMISSION]</P>

        Arctii Fructus Inhibits Colorectal Cancer Cell Proliferation and MMPs Mediated Invasion via AMPK

        <P>Although Arctii Fructus (AF) has been shown to have various pharmacological effects, there have been no studies concerning the inhibitory effects of AF on the metastatic properties of colorectal cancer (CRC). The aim of this study was to investigate whether AF could suppress CRC progression by inhibiting cell growth, epithelial-mesenchymal transition (EMT), migration, and the invasion ability of CRC cells. AF decreased proliferation of CRC cells by inducing cell cycle arrest and apoptosis via extrinsic and intrinsic apoptotic pathways. Regarding metastatic properties, AF inhibited EMT by increasing the expression of the epithelial marker, E-cadherin, and decreasing the expression of the mesenchymal marker, N-cadherin, in CT26 cells. Moreover, AF decreased the migration and invasion of CT26 cells by inhibiting matrix metalloproteinase-2 (MMP-2) and MMP-9 activity. We confirmed that the decreased invasion ability and MMP-9 activity by AF treatment involved AMP-activated protein kinase (AMPK) activation. Collectively, this study demonstrates that AF inhibits the proliferation and metastatic properties of CRC cells.</P>
