        Proliferation of T lymphocytes has been determined by a threshold of signals generated by the interaction between interleukin-2 (IL-2) and its spedific membrane receptor (IL-2R). Regulation of IL-2R expression on the surface of lymphocytes is important in the generation of cytototocix T cells and lymphokine-activated killer (LAK) cells carrying anti-tumor effector functions in the body. To obtain the ideas in designing in vivo immunotherapy using biological response modifiers, this study examined the effects of the interferon(IFN)s, steroid, or colchicine on the IL-2R expression and proliferation of human peripheral blood lymphocytes(PBL). As incubation time was lengthened up to 72 hours, the number of Tac-positive cells and prolongation potential of human PBL upon stimulation wit IL-2 or mitogens were increased. IFN-α resulted in a significant inhibition of IL-2R expression induced by IL-2 but IFN-Υ had no effect. Colchicine or dexaamethasone also inhibited IL-2R expression and cell proliferation induced by IL-2 or mitogens. These findings suggest that IL-2R expression and proliferation of human PBL are inhibited by IFN-α, colchicine, or dexamethasone.

        척수손상·비손상인의 신체부위별 신체구성 및 골밀도 비교

        This study was designed to compare segmental body composition and bone mineral density in spinal cord-injured(SCIS; n=9, SCIA; n=9) and non-injured control(AB; n=9) subjects. Dual-energy X-ray absorptiometry(DEXA) was used to measure %body fat, body fat mass, lean body mass, bone mineral contents, and bone mineral density in total and various body regions(arms, legs, trunk). The present results showed that body fat mass and %body fat were significantly higher and lean body mass was significantly lower in the SCIS group compared to AB group, while there was no difference between the SCIA and AB group in those of parameters. In BMC, there was no difference among groups and in BMD, SCI group was significantly lower that AB group, but there was no difference between SCIA group and AB group. Based on these results, in can be suggested that long-term regular exercise can reduce the negative changes in body composition, bone mineral density by spinal cord injury.

        유산균들의 콜레스테를 저하성, 내산성, 내담즙성, 항생제 내성 비교

        기능성 요구르트의 제조에 사용되는 유산균은 인체에 유익한 생리활성과 우수한 생존능력을 보유하고 있는 것이 바람직하다. 본 연구에서는 다양한 유산균주의 산과 담즙 및 항생제에 대한 내성을 조사하였으며 in vitro에서 콜레스테롤 저하능을 조사하였다. In vitro에서 콜레스테롤 저하능이 우수한 13균주를 선발하여 이들의 동결건조 분말을 고 콜레스테롤 식이의 실험쥐에 투여하였다. 그 결과 8균주는 비 투여구보다 유의적으로 18.3~27.3%의 콜레스테롤 저하능을 나타냈다(P≤0.05). 이들 중에서 Bifidobacterium infantis AM-220, Lactobacillus AM-245, Streptococcus MA-1의 3균주를 이용하여 요구르트를 제조한 후 동결건조 분말을 이용하여 급여실험을 수행한 결과, 총 콜레스테롤과 LDL 함량이 유의적으로 감소하는 것으로 나타났다. For a probiotic yoghurt it is desirable to utilize lactic acid bacteria with a high survival rate and beneficial function to human beings. We have examined a variety of lactic acid bacteria to assess the acid and bile tolerance and antibiotic resistance. In addition, an in vitro culture experiment was performed to evaluate their ability to reduce cholesterol levels in the growth medium. Thirteen strains were selected from in vitro cholesterol assays and fed to Sprague-Dawley rats with a high-cholesterol diet. Among the 13 strains tested, 8 strains were shown to reduce serum cholesterol levels significantly after 24 days of administration in vivo. Rats were fed lyophilized yoghurt powder fermented with a combination of 3 selected strains: Bifidobacterium infantis AM-220, Lactobacillus AM-245, and Streptococcus MA-1. The levels of total cholesterol and low density lipoprotein were significantly lower (p≤0.05) in rats fed the yoghurt powder compared with control group. These studies suggest that yoghurt fermented with appropriately selected lactic acid bacteria may have a anticholesterolemic effect.

        Public Evacuation Time Estimates within EPZ of Ulchin Site

        원자력발전소 사고시 방사성물질이 발전소외 지역으로 유출될 경우 주민을 안전하게 보호하기 위한 조치의 일환으로 주민소개가 고려된다. 소개시간 산정에 필요한 인자를 도출하고, 각각의 인자에 대해 원전 주변의 현장자료를 토대로 부지주변의 교통환경 여건을 반영하여 울진원전의 방사선비상계획구역 내의 주민전체를 소개시키는데 소요되는 시간을 예측하였다. 평상교통, 여름철 첨두교통, 그리고 겨울철 첨두교통에 대해 각각 주간 및 야간, 평상기상 및 악기상의 경우로 나누어 12가지의 주민소개 시나리오를 설정하였다. 비상계획구역 경계 남단과 북단에서 모든 소개차량(인구)이 비상계획구역을 벗어나는 데 걸리는 시간은 전체적으로는 분 정도로 예측되었다. 소개시간은 밤이 낮보다 약 45분 정도 더 소요되는 것으로 예측되었다. The strong protection method of radiation emergency preparedness is the evacuation when a great deal of radionuclide material is released to environment. Required factors for evacuation time estimate of Ulchin nuclear power plant site were investigated. The traffic capacity and the traffic volume by season were investigated for the traffic analysis and simulation within EPZ of Ulchin site. As a result, the background traffic volume by season were established. The NETSIM code was applied to simulate for 12 scenarios in the event of normal traffic/summer peak traffic/winter peak traffic, daytime/night, and normal weather/adverse weather conditions. The results showed that the evacuation time required for total vehicles to move out from EPZ took generally minutes. The evacuation time took longer about 45 minutes at night than in the daytime, and 45 minutes in adverse weather than normal condition.

        Lung-specific delivery of paclitaxel by chitosan-modified PLGA nanoparticles via transient formation of microaggregates

        <P>Chitosan-modified paclitaxel-loaded poly lactic-co-glycolic acid (PLGA) nanoparticles with a mean diameter of 200–300 nm in distilled water were prepared by a solvent evaporation method. The mean diameter increased dramatically in contact with the mouse (CDF<SUB>1</SUB>) plasma, as a function of chitosan concentration in the modification solution (e.g., 2670.5 nm for 0.7% chitosan-modified nanoparticles, NP<SUB>3</SUB>), but reverted to almost its original size (i.e., 350.7 nm for NP<SUB>3</SUB>) following 5 min of gentle agitation. The zeta potential of PLGA nanoparticles was changed to positive by the chitosan modification. The in vitro uptake into, and cytotoxicity of the nanoparticles against, a lung cancer cell line (A549) were significantly increased by the modification. Most importantly, a lung-specific increase in the distribution index of paclitaxel (i.e., AUC<SUB>lung</SUB>/AUC<SUB>plasma</SUB>) was observed for chitosan-modified nanoparticles (e.g., 99.9 for NP<SUB>3</SUB> vs. 5.4 for Taxol™) when nanoparticles were administered to lung-metastasized mice via the tail vein at a paclitaxel dose of 10 mg/kg. Transient formation of aggregates in the blood stream followed by enhanced trapping in the lung capillaries, and electrical interaction-mediated enhanced uptake across the endothelial cells of the lung tumor capillary appear to be responsible for the lung-tumor-specific distribution of the chitosan modified nanoparticles. © 2008 Wiley-Liss, Inc. and the American Pharmacists Association J Pharm Sci 98:970–984, 2009</P>

        세균독소로 자극시킨 사람 섬유아 세포에서의 Interleukin-6와 Interleukin-8의 생성

        Cytokines are hormone-like proteins which mediate and regulate inflammatory and immune responses. Interleukin-6 (IL-6) is involved in the final differentiation of B cells into antibody-producing cells. Interleukin-8 (IL-8) is a neutrophil chemotactic factor that plays an important role in the recruitment of neutrophil to inflammatory loci. Inflammatory mediators by cells in the gingiva have been implicated in the initiation and progression of periodontitis and oral infection. The purpose of this study was conducted to investigate the effect of lipopolysaccharide (LPS), staphylococcus enterotoxin B (SEB) on production ofIL-6 and IL-8 by human gingival and facial dermal fibroblasts. Primary cultured human gingival and facial dermal fibroblasts were incubated with LPS(0.01, 0.1, 1.0㎍/ml), SEB (0.01, 0.1, 1.0㎍/ml) or LPS (0.1㎍/ml) plus SEB (0.1㎍/ml). Culture supernatants were collected at 24, 48, and 72 hrs and assessed for IL-6 and IL-8 production by enzyme-linked immunosorbent assay. IL-6 production in gingival fibroblasts stimulated with LPS was higher than that with SEB. IL-6 production by double exposure with LPS plus SEB was amplified in comparison with single exposure of LPS or SEB. IL-6 production in facial demal fibroblasts was increased only by stimulation with a high concertration of LPS (1.0㎍/ml). Its production in facial dermal fibroblasts by exposure with SEB was decreased in comparison with control, nontreated cells. Therefore, gingival fibroblasts showed higher sensitivity than facial dermal fibroblasts in response to low concentration of LPS. Also, IL-6 production by double exposure with LPS plus SEB was amplified in comparison with single exposure of LPS or SEB.IL-6 production in facial dermal fibroblasts was increased only by stimulation with a high concentration of LPS(1.0㎍/ml). Its production in facial dermal fibroblasts by exposure with SEB was decreased in comparison with control, nontreated cells. Therefore, gingival fibroblasts showed higher sensitvity than facial dermal fibroblasts in response to low concentration of LPS. Also, IL-6 production by double exposure with LPS plus SEB was amplified in comparison with single exposure of LPS or SEB. IL-8 production in gingival fibroblasts was enhanced greatly only by stimulation of high concentration of LPS (1.0㎍/ml). That by exposure with SEB was increased only in 24 hrs cultivation. IL-8 production by double exposure with LPS plus SEB was amplified in comparison with single exposure of LPS or SEB. IL-8 production in facial dermal fibroblasts was decreased by LPS and increased only in 48 hrs cultivation by SEB. IL-8 production by double exposure with LPS plus SEB was enhanced only in 48 hrs cultivation in comparison with single exposure of LPS or SEB. therefore, IL-6 and IL-8 production were released at various quantities according to bacterial toxin applied and site of fibroblast harvested. These results suggest that gingival fibroblasts may be concerned with IL-6 and IL-8 related inflammatory response more than facial dermal fibroblasts.

        수요의 불확실성과 현금 흐름을 고려한 공급망 관리의 민감도 분석에 관한 연구

        Supply chains faces various uncertainties such as demand, manufacturing and supply uncertainties. In this study, we consider both normal demand variation and sudden change in demand as risk factors in a supply chain. Capacity is adjusted by hiring and firing workforce to mitigate the impact of demand uncertainty in consideration of cash flow. A mixed-integer linear programming model is developed and numerical experiments are conducted to investigate the impact of uncertain demand in terms of inventory level, cash flow, debt ratio, and bankruptcy. The results show that safety stock of a focal company has less effect on cash flow of suppliers in the case of normal demand variation. Also, it is observed that the sudden change in demand worsens cash flow and causes bankruptcy and high inventory level in a supply chain. The ratio of production and fixed cost has a significant impact on the performance of a supply chain.
