        대량재해에 있어 미토콘드리아 DNA의 다형성을 이용한 개인식별

        The mitochondrial DNA(mtDNA) is a small extranuclear DNA molecule that has been sequenced in man. the mtDNA has distinct characteristics over nuclear DNA. First, the evolutionary rate of nucleotide substitution appears to be larger compared with the nuclear DNA. Second, mtDNA is maternally inherited, so the determination of the various haplotype is unequivocal and no recombination has to be involved. Third, there is a 1.1 kb long control region, which shows severe polymorphism. These characteristics have been applied for several evolutionary study. Furthermore, they exist in cytoplasm with numerous copy number and their size, 16,569 bp is small relative to nuclear DNA, so they are more resistant to degradation or can be types in samples with only cytoplasm such as hair shaft without hair root. These presented the possibility of mtDNA as a tool in individual identification, especially when the nuclear DNA be unavailable. Actually, polymorphism in control region has been applied to individual identification for the skeletal remain found in Vietnam war. So we decided to apply the polymorphism of mtDNA in control region for the individual identification in case of mass disaster-"Downfall of Sampoong Department". Human remains from total 27 different individuals and 178 control persons were submitted for the individual identification. As the mtDNA comparison could be done through maternal lineage, 12 individual from 9 families from the control group were ruled out, because these families were consisted of father or were seeking for their father. Sequencing for the mtDNA control region was done using ABI automatic sequencer and DyeDeoxy Terminator Cycle Sequencing Kit. For convenience, sequencing reaction was done for two separate control region, region I from 16016-16401 in Anderson sequence, and regionⅡ from 048-388. First, sequences in regionⅡ of all samples and control persons were compared, and 20 samples were ruled out with no identical sequences in control group. Remaining 7 samples showed identical sequences with some of the control persons in three groups, and these identical pairs underwent mtDNA region I study. After region I study all samples except two dropped out in blood relationship. For the confirmation, STR and VNTR study such as MCT118, apoB, YNZ22, vWF, MBP, D21S11, TC-11 were done for remaining the two samples. The result denied the blood relationship between these two samples and any of the control group.

        한국인에서 Quadruplex PCR을 이용한 8종류 상염색체 STR 유전좌의 다형성 분석

        Two quadruplex PCR reactions were designed for 7 tetrameric (D3S2406, D4S2368, D5S818, D7S821, D9S925, D13S317, D19S253) and one trimeric (D6S1043) short tandem repeats loci to study the allele frequency and the applicability of genetic variation in these loci in forensic case works. For 310 unrelated Koreans DNA was isolated from peripheral blood using phenol/chloroform method. Quadruplex I was consisted of D4S2368, D6S1043, D7S821, D9S925 and quadruplex II D3S2406, D5S818, D13S317, D19S253. The amplified products were analyzed by 5% polyacrylamide gel electrophoresis followed by silver staining. The heterozygosity in each loci ranged 92.91-66.13%, and PD(Power of Discrimination) was above 085 in each loci. Every loci except D6S1043 followed hardy-Weinberg expectation. The cumulative PI was low as 1.65×10-10. Two mutations were noted, one in D19S253 and the other in D9S925 among 234 gametes. With these results above eight STR loci studied here preyed to be highly polymorphic enough to be used in forensic field. This study provides valuable population data in these loci for Korean.

        녹내장 수술 후 안축장의 변화

        Purpose: This study examines the changes in axial length after trabeculectomy and Ahmed glaucoma valve implantation surgery. Methods: In the forty three eyes of the 42 patients who had the uncontrolled glaucoma, 22 eyes of 21 patients having trabeculectomy and 21 eyes of 21 patients having Ahmed glaucoma valve implantation from October 2004 to September 2005 at Kim`s Eye Hospital were analyzed prospectively. These patients had IOP and axial length measurements preoperatively, at 1 week, 1 month, and 3 months after surgery and refractive power measurements preoperatively, at 3 months after surgery. Axial length was measured by IOL Master?. Results: There was a statistically significant reduction in IOP after 3 months of 26.6±11.2 mmHg overall, 22.4±8.4 mmHg following trabeculectomy, and 30.9±12.3 mmHg after Ahmed glaucoma valve implatation (p<0.001). There was a statistically significant reduction in axial length after 3 months of -0.24±0.10 mm overall, -0.21±0.1 mm after trabeculectomy and -0.28±0.1 mm after Ahmed glaucoma valve implantation. Conclusions: There was a statistically significant decrease in axial length after both trabeculectomy and Ahmed glaucoma valve implantation. We must consider the influence of axial length change on refraction in planning combined glaucoma and cataract surgery or future IOL implantation.

        Bypass surgery, as therapy for morbid obesity, was introduced in 1952. Multiple complications such as arthritis and dermatitis as well as liver dysfunction, renal injury, diarrhea, malnutrition and electrolyte imbalance were reported after bypass surgery. Recently, we have experienced a case of bypass arthritis-dermatitis syndrome in a patient after pyloric exclusion and bypass gastrojejunostomy due to traumatic duodenal perforation. He complained arthralgia of left knee, both wrist and both ankle and developed erythematous maculopapular rash over lower extremities. He was successfully treated with NSAIDs and clindamycin.(Korean J Med 59:583-586, 2000)

        보문: 백인의 상처분장과 노인분장에 활용할 수 있는 실리콘의 조색방법

        As special make-up industry develops, it is necessary to obtain the concrete and objective data about the colorant combination criterion and thickness of silicone which can express diverse skin colors and give delicacy and reality to depicting the characters often expressed like wound and old people make-up. For this purpose, primarily, this study measured 10 white people`s skin chromaticity, and so combined the colorants of beige, permanent yellow light, carmine, and cobalt blue, and then determined the colorant combination ratio similar to such skin chromaticity and investigated the chromaticity change of silicone by its thickness. Secondarily, this study fabricated the prosthesis necessary for wound and old people make-up, and so applied such prosthesis to experimental subjects. According to the results of this study, on 10 experimental subjects` foreheads and cheeks, lightness (L*) was 60.4±1.2 and 60.1±0.9. Redness (a*) was 13.4±0.9 and 14.4±0.5. Yellowness (b*) was 12.6±0.8 and 11.7±0.7. Redness was the 22% and 24% of lightness and yellowness was the 21% and 19% of lightness, so redness was higher in the ratio of white people`s skin colors than yellowness. When beige 1.0 g, carmine 0.66 g, and cobalt blue 0.13 g were combined in the colorant combination ratio of pigments, △Eab* was 5.0, which was very similar to skin colors and so was used for the concentration of fabricating silicone pieces. When three colorant-combined pigment 1.0 g was mixed with silicone 100 g and so the silicone piece was fabricated and attached to white people experiment subjects` left cheek and then their chromaticity was compared, ?Eab* was 3.2 for a thickness of 2 mm, △Eab* was 7.8 for a thickness of 4 mm, and △Eab* was 11.1 for a thickness of 6 mm. So, the thicker the silicone piece was, the darker its color was. When the silicone pieces having a thickness of 2 mm were attached to experimental subjects` skins, colors were similar and so showed no difference to the naked eye. Therefore, because silicone thickness influences its chromaticity, it is desirable to fabricate its prosthesis in consideration of such thickness. When such fabricated silicone pieces were applied to wound make-up, there was no sense of difference on attached boundary surfaces, and the skin surfaces swollen owing to wounds and the incised wound were expressed realistically. In old people make-up, the features of aged skins, including the minute wrinkles on the forehead, the drooping under the eye, and the wrinkles around the mouth, were expressed delicately and realistically. Thus, it was confirmed that such silicon pieces could be used very usefully for would make-up and old people make-up. So, when silicone is used in special make-up by combining colorants for realistic and detailed expressions, even if three pigments, that is, beige, carmine, and cobalt blue, are combined properly with the naked eye, the differences of its chromaticity are not shown clearly. In contrast, the thickness of silicon pieces influences greatly their chromaticity. Therefore, it is very desirable to utilize silicon pieces on the spots of special make-up industry in consideration of the importance of their thickness.

        실리콘 고무를 이용한 황인 여성의 안면 상처분장

        The progress of video technology in Korea has increased concern for special make-up. The purpose of this study was to investigate usefulness of silicone as a means of special make-up. The skin color of 8 yellow women was measured at first. Next, one of the women was selected as the model for special make-up and was applied with a mix of 4 pigments to create a tone similar to her skin color. Then, a silicone piece was produced and applied to the model to analyze the difference in tone and outcomes of the special make-up. In the result, there was slight difference in color between the forehead and cheek and the percentage of yellowness/lightness appeared to be higher than that of redness/lightness, showing that yellowness might play a decisive role in deciding the skin color of the yellow people. A comparison between the piece made by mixing silicone with pigments (magenta, cyanide, yellowish-like) and the forehead and cheek of the model revealed a very little difference in lightness and yellowness with Δ*=-0.9, Δ*=2.0, Δ*=-1.7 and b*= -2.9 and a little difference in redness with Δ*=-4.9 and Δ*=-5.2. No difference could be visible to the naked eye in the chromaticity. When the artificial silicone for wounds was applied to the model, realistic cover-up was possible without any show of strangeness even on the skin surface of which looked swollen due to wounds as well as in the edge of attachment thanks to the semitransparent material and optimal mixing ratio of pigments. This result proved that silicone is the better material for special make-up than other ones. In conclusion, it may be said that toning by mixing silicone with pigments makes it possible to express skin color and quality more realistic and delicate, a method that can be better utilized in the industrial area since silicone is easily reproducible because it won`t change depending on body temperature and weather.
