        Heme oxygenase-1 induced by desoxo-narchinol-A attenuated the severity of acute pancreatitis via blockade of neutrophil infiltration

        <P><B>Abstract</B></P> <P>Heme oxygenase-1 (HO-1) has an anti-inflammatory action in acute pancreatitis (AP). However, its mechanism of action and natural compounds/drugs to induce HO-1 in pancreas are not well understood. In this study, we investigated the regulatory mechanisms of HO-1 during AP using desoxo-narchinol-A (DN), the natural compound inducing HO-1 in the pancreas. Female C57/BL6 Mice were intraperitoneally injected with supramaximal concentrations of cerulein (50 μg/kg) hourly for 6 h to induce AP. DMSO or DN was administered intraperitoneally, then mice were sacrificed 6 h after the final cerulein injection. Administration of DN increased pancreatic HO-1 expression through activation of activating protein-1, mediated by mitogen-activated protein kinases. Furthermore, DN treatment reduced the pancreatic weight-to-body weight ratio as well as production of digestive enzymes and pro-inflammatory cytokines. Inhibition of HO-1 by tin protoporphyrin IX abolished the protective effects of DN on pancreatic damage. Additionally, DN treatment inhibited neutrophil infiltration into the pancreas via regulation of chemokine (C-X-C motif) ligand 2 (CXCL2) by HO-1.       • Genetic Diversity of Korean Rice Breeding Parents as Measured by DNA Fingerprinting with Simple Sequence Repeat (SSR) Markers

        Molecular markers are useful tools for evaluating genetic diversity and determining cultivar identity. Present study was conducted to evaluate the genetic diversity within a diverse collection of rice accessions used for Korean breeding programs. Two hundred eighty-seven rice cultivars, composed of temperate japonica, tropical japonica, indica, and Tongil-type of Korean crossing parents were evaluated by means of 15 simple sequence repeat (SSR) markers. A total of 99 alleles were detected, and the number of alleles per marker ranged from 4 to 11, with an average of 6.6 per locus. Polymorphism information content (PIC) for each of the SSR markers ranged from 0.2924 to 0.8102 with an average of 0.5785. These results, with the result that use of only 15 SSR markers made all rice cultivars examined could be uniquely distinguished, imply the efficiency of SSR markers for analysis of genetic diversity in rice. Cluster analysis was performed on similar coefficient matrics calculated from SSR markers to generate a dendogram in which two major groups corresponding to japonica (Group I) and indica and Tongil type rice (group II) with additional subclasses within both major groups. The narrowness of the Korean breeding germplasm was revealed by the fact that most of the Korean-bred and Japan-bred temperate japonica cultivars were concentrated into only 2 of the sub-group I-1 (143 cultivars) and I-2 (58 cultivars) among six sub-groups in major group of japonica. This is because of the japonica accessions used in this study was a very closely related ones because of frequent sharing of the crossing parents with similar genetic background with synergy effect of the inherited genetic difference between indica and japonica. A rice breeding strategy with the use of molecular markers was discussed for overcoming of genetic vulnerability owing to this genetic narrowness.

        1992년 첫 삽을 뜨면서 시작한 경부고속철도건설사업이 2004년 4월, 1단계 개통을 한데 이어 지난 2010년 11월 완전 개통하였다. 경부고속철도 개통으로 경부축 지역의 2시간대 서비스 제공을 통한 국토의 균형발전 촉진 및 여가·관광사업 활성화, 기술발전, 환경비용 및 에너지 절감 등 긍정적인 효과를 이룬 반면, 잦은 기본계획 변경으로 인한 사업비 및 사업기간의 증가, 현실과 맞지 않은 설치기준으로 인한 과잉설계 문제 등 부정적 효과도 초래했다. 본 연구를 통해 사업추진 단계별 주요 이슈에 대해 객관적인 시각으로 문제점을 분석하고 시사점을 도출함으로써, 현재 추진하고 있는 호남고속철도와 수도권고속철도 사업에서는 시행착오를 줄이고 사업을 효율적으로 추진하는데 활용하고자 한다. Launched in 1992, the Gyeung-Bu High Speed Railway (HSR) was fully opened in November 2010 following the partial opening of the first phase in April 2004. Since the opening it has made tremendous achievements, including but not limited to, technical innovation, balanced regional development, growth of leisure and tourism industries, and savings of environmental costs and energy. On the other hand, it has brought negative effects such as increase in project time and cost by frequent changes of the basic plan and over-design based on unrealistic criteria. In this study we will analyze major issues that occurred at each stage of the Gyeong-Bu HSR project and draw implications. Thus, this study will enhance to efficiently implement the Ho-Nam and Seoul Metropolitan HSR project through minimizing possible trials and errors.

        Morin exerts cytoprotective effects against oxidative stress in C2C12 myoblasts via the upregulation of Nrf2-dependent HO-1 expression and the activation of the ERK pathway

        <P>In the present study, we investigated the cytoprotective efficacy of morin, a natural flavonoid, against oxidative stress and elucidated the underlying mechanisms in C2C12 myoblasts. Our results indicated that morin treatment prior to hydrogen peroxide (H2O2) exposure significantly increased cell viability and prevented the generation of reactive oxygen species. H2O2-induced comet-like DNA formation and gamma H2AX phosphorylation were also markedly suppressed by morin with a parallel inhibition of apoptosis in C2C12 myoblasts, suggesting that morin prevented H2O2-induced cellular DNA damage. Furthermore, morin markedly enhanced the expression of heme oxygenase-1 (HO-1) associated with the induction and phosphorylation of nuclear factor-erythroid 2-related factor 2 (Nrf2) and the inhibition of Kelch-like ECH-associated protein 1 (Keapl) expression. Notably, these events were eliminated by transient transfection with Nrf2-specific small interfering RNA. Additional experiments demonstrated that the activation of the Nrf2/HO-1 pathway by morin was mediated by the extracellular signal-regulated kinase (ERK) signaling cascade. This phenomenon was confirmed with suppressed Nrf2 phosphorylation and consequently diminished HO-1 expression in cells treated with a pharmacological inhibitor of ERK. Collectively, these results demonstrated that morin augments the cellular antioxidant defense capacity through the activation of Nrf2/HO-1 signaling, which involves the activation of the ERK pathway, thereby protecting C2C12 myoblasts from H2O2,-induced oxidative cytotoxicity.</P>

        Protective Effect of Heme Oxygenase-1 on High Glucose-Induced Pancreatic β-Cell Injury

        <P><B>Background</B></P><P>Glucose toxicity that is caused by chronic exposure to a high glucose concentration leads to islet dysfunction and induces apoptosis in pancreatic β-cells. Heme oxygenase-1 (HO-1) has been identified as an anti-apoptotic and cytoprotective gene. The purpose of this study is to investigate whether HO-1 up-regulation when using metalloprotophyrin (cobalt protoporphyrin, CoPP) could protect pancreatic β-cells from high glucose-induced apoptosis.</P><P><B>Methods</B></P><P>Reverse transcription-polymerase chain reaction was performed to analyze the CoPP-induced mRNA expression of HO-1. Cell viability of INS-1 cells cultured in the presence of CoPP was examined by acridine orange/propidium iodide staining. The generation of intracellular reactive oxygen species (ROS) was measured using flow cytometry. Glucose stimulated insulin secretion (GSIS) was determined following incubation with CoPP in different glucose concentrations.</P><P><B>Results</B></P><P>CoPP increased HO-1 mRNA expression in both a dose- and time-dependent manner. Overexpression of HO-1 inhibited caspase-3, and the number of dead cells in the presence of CoPP was significantly decreased when exposed to high glucose conditions (HG). CoPP also decreased the generation of intracellular ROS by 50% during 72 hours of culture with HG. However, decreased GSIS was not recovered even in the presence of CoPP.</P><P><B>Conclusion</B></P><P>Our data suggest that CoPP-induced HO-1 up-regulation results in protection from high glucose-induced apoptosis in INS-1 cells; however, glucose stimulated insulin secretion is not restored.</P>

      • Effect of Temperature Associated with Early Growth Stimulus on Shortening of Heading Dates in Rice

        The heading date is known to be controlled by two kinds of genetic constituent, photosensitivity and basic vegetable phase. For the latter, the effect of temperature in early growth period is critical to determine the shortness of vegetative growth periods in plant s life. A phytotron experiment on 55 rice cultivars, consisting of two ecotypes of rices, indica and japonica, was conducted at high and low temperature treatments at early growth stage to investigate the possible role of plant growth stimulus by high temperature to associate with shortening of heading date. The high temperature during the early growth stage stimulated the rice growth as measured by plant height with much difference of the growth response between indica and japonica. The conclusive finding that these growth stimulus in early growth stage was highly correlated with the acceleration of heading is, more or less, correlated with the heading of the late growth stage although we could not conclude the genes for early plant growth stimulus by high temperature is the same genes as the genes for accelerating of heading in the late growth stage of plants.

      • 위절제술 후 절제연 양성으로 진단된 진행성 위암 환자의 임상적 경과

        Objectives: Many investigators have recommended adequate resection margin and lymphadenectomy for radical curative resection. The aim of this study is to evaluate clinical characteristics of positive resection margin (proximal or distal) of postgastrectomy in advanced gastric cancer. Methods: We studied 17 patients with gastric cancer who were diagnosed positive resection margin by intraoperative frozen biopsy or permanent biopsy report from January 2005 to December 2007, retrospectively. Surgical margin monitored by endoscopy. Results: Distal gastrectomy was performed in 13 patients and total gastrectomy in 4. Gastrectomy with combined resection including splenectomy was performed in 3, distal pancreatectomy in 2, transverse colon segmental resection in 1, and cholecystectomy in 2. Positive Proximal margin was found in 12, positive distal margin in 3, and both in 2. Palliative chemotherapy was performed in 8 patients. Postoperative follow up endoscopy was established in only 8 patients. Malignant results from endoscopic biopsy in gastroenteric or esophagoenteric anastomotic line were proven in 2 patients during follow up. 9 patients were not performed follow-up endoscopy. Among total 17 patients, 2 patients are alive. Fifteen patients died of aggravation of disease in 13 and postoperative complication in 2. Conclusions: Although positive surgical margin in far advanced gastric cancer were found, it can consider that does not further resection to obtain microscopic clear anastomotic margin.

      • 공간구조 분석에 있어서 GIS기술을 이용한 건축물 [근접성]의 정량화 방법에 관한 연구

        In this paper quantitative method about buildings and land use pattern is discussed by distance among near buildings(DNB). We considered the worth of DNB as urban planning method following two points : 1) DNB average and Building area ratio were pretty independent each other. That is to say DNB did not show density but arrangement. 2)DNB would express not just near buildings pattern and small vacant apace but also lot-street patterns. We analyzed following three steps. Firstly, we figured a calculating algorithm, secondly we analyzed characteristics of 4 index about DNB and statistical relations of DNB to building area ratio by each lot unit, thirdly we discussed combination patterns of DNB average and urban space pattern. The aim of this study is to theoretically develop a simple yet efficient system of data generation for urban planning and spatial structure analysis based on the application of GIS technique.

      • 다핵균 및 단핵균으로부터 고protease 생산균주의 Screening

        4 multinucleate and 7 mononuclear fungi were investigated for screening of high protease productive strains. The protease activity of mononuclear strains was generally greater than that of multinucleate strains. For acid proteinase, Aspergillus awamori AOK 1007 and Asp. oryzae shoyu 2 as mononuclear strains were selected in glucose-polypeptnoe medium. For acid carboxypeptidase, two mononuclear strains were screened as Asp. awamori AOK 1007 and Asp. usamii mut. shirousami AOK 1011 in glusose-sodium glutanate medium. In this result, mononuclear fungi appeared higher protease production than that of multinuclear fungi. So, mononuclear fungi may gave many merits to improve protease activity by using of mutation or cell fusion technology.

      • 예도(銳刀)의 미시사적 관점으로 바라본 무예실제에 관한 분석

        예도(銳刀)에 관한 보다 정확한 고찰은 오늘날 전통무예단체들이 『武藝圖譜通志』를 통한 전통무예의 실제를 복원 및 재현함에 있어 크나큰 도움이 될 것이다. 이는 예도(銳刀)가 전통무예사에 대한 단초적 역할을 제공할 수 있는 초석이 될 수 있기 때문이다. 따라서 이 연구에서는 예도의 설(說: 해설(解說))과 기(技: 기예(技藝))의 형식적 분석과 전통무예의 복원에 관한 이론적 배경을 정립하기 위하여 고서와 도록 등의 다양한 자료를 검토‧분석하였는데 이는 다음과 같은 결론을 도출하였다. 첫째, 예도(銳刀)의 종합적 분석을 통한 예도의 형식은 병기도식(兵器圖式)․설(設) 11개․예도보(銳刀譜)․예도총보(銳刀總譜)와 예도총도(銳刀總圖)로 구성되어 있음을 알았다. 둘째, 예도는 본국검과 더불어 조선을 대표하는 한국 고유의 전통검법으로 임진왜란을 계기로 새롭게 발굴 정립되었으며 정조가 재위했던 1800년대까지 약 200년 걸쳐 전승되면서 민족의 재산과 생명을 지켜왔던 국방무예(24반 무예)였다. 셋째, 국난을 극복하고 국토를 수호하려는 선조들의 의지로 완성된 24반 무예 속에 녹아 든 활달한 몸짓 속에는 민족자존의 의지와 불굴에 투혼이 배어 있다. 이와 같이 오늘날 한․중․일의 칼 쓰는 법을 비교해 볼 때, 『武藝圖譜通志』의 예도가 갖는 의미와 가치는 대단히 귀중하다 할 수 있겠다. 이상과 같이 종합해 볼 때, 앞으로 이 한국무예 예도(銳刀)에 관한 심도 있는 연구가 이루어질 때 비로소 예도(銳刀)가 세계적인 보편무예로 발전하는 중요한 기반이 될 것으로 기대하는 바이다.
