        Enhancing the Enzymatic Activity of the Multifunctional β-Glycosyl Hydrolase (Cel44C-Man26A<sub>P558</sub>) from Paenibacillus polymyxa GS01 Using DNA Shuffling

        본 연구자들은 이전에 cellulase, xyalnase 및 lichenase의 다기능 효소활성을 지니는 절단된 Cel44C-$Man26A_{P558}$의 ${\beta}$-glycosyl hydrolase를 보고하였다. 본 연구에서는 절단된 Cel44C-$Man26A_{P558}$ 효소의 다기능성 ${\beta}$-glycosyl hydrolase 활성을 증가시키기 위해 DNA shuffling을 시도하였다. DNA shuffling에 의해 단일변이(P438A)를 가진 M2Cel44C-$Man26A_{P558}$와 이중변이(A273T 및 P438A)를 가진 M21Cel44C-$Man26A_{P558}$를 얻었다. 이중변이를 가진 M21Cel44C-$Man26A_{P558}$은 단일변이를 가진 M2Cel44C-$Man26A_{P558}$ 보다 효소활성이 낮게 나타났으나, M2Cel44C-$Man26A_{P558}$와 M21Cel44C-$Man26A_{P558}$은 대조구인 Cel44C-$Man26A_{P558}$ 보다 약 1.3에서 2.2배 정도 높은 효소활성을 나타내었다. 특히, 단일변이를 가진 M2Cel44C-$Man26A_{P558}$는 대조구인 Cel44C-$Man26A_{P558}$보다 cellulase, xylanase 및 lichenase 효소활성이 약 1.5에서 2.2배 정도 높게 나타났다. ${\beta}$-Glycosyl hydrolase의 cellulase, linchenase 및 xylanase 최적 효소활성은 각각 pH 7.0, 7.0 및 6.0에서 이었다. 이러한 결과는, 아미노산 잔기인 Ala438이 다기능성 ${\beta}$-glycosyl hydrolase 활성을 증가시키는 중요한 역할을 한다고 추정할 수 있다. We previously reported that the truncated Cel44C-$Man26A_{P558}$ ${\beta}$-glycosyl hydrolase protein exhibits multifunctional activities, including cellulase, xylanase, and lichenase. DNA shuffling of the truncated Cel44C-$Man26A_{P558}$ enzyme was performed to enhance the enzymatic activity of the multifunctional ${\beta}$-glycosyl hydrolase. Two mutant enzymes, M2Cel44C-$Man26A_{P558}$ that carries one mutation (P438A) and M21Cel44C-$Man26A_{P558}$ that carries two mutations (A273T and P438A) were obtained. The enzymatic activity of the M21Cel44C-$Man26A_{P558}$ double mutant was lower than enzymatic activity of the single mutant (M2Cel44C-$Man26A_{P558}$). However, both mutants displayed the enhancements in their enzyme activities that were ${\approx}1.3$- to 2.2-fold higher than the original enzymatic activity in Cel44C-$Man26A_{P558}$. In particular, the mutant M2Cel44C-$Man26A_{P558}$ exhibited an approximate 1.5- to 2.2-fold increase in the cellulase, xylanase, and lichenase activities in comparison with the control (Cel44C-$Man26A_{P558}$). The optimum cellulase, linchenase, and xylanase activities of ${\beta}$-glycosyl hydrolase were observed at pH 7.0, pH 7.0 and pH 6.0, respectively. These results, therefore, suggest that the amino acid residue Ala438 plays important roles in the enhancement of the activity of multifunctional ${\beta}$-glycosyl hydrolase.

      • 알코올 및 베타 카로틴이 혈액 성분에 미치는 영향에 관한 연구

        Human take in many kinds of foods, and also are exposed to various chemicals which may be harmful. Especially, some food and chemicals may give rise to free radicals during metabolism in the tissues. These free radicals, such as superoxide. hydroxy radical and hydrogen peroxide, induce oxidations of the macromolecules, which can cause a variety of damages in the body. But human have anti-oxidative mechanisms well developed against these damages. Anti-oxidants such as vitamines A, C, E, and β-carotene are believed to have a major role in these mechanisms acting as a cofactor of anti-oxidative system. On the basis that alcohol is one of the most common chemicals that induce oxidative damages in liver, this study was carried out to elucidate biochemical changes of blood after alcohol administration as well as to investigate the effects of β-carotene on the alcohol-injected Sprague-Dawley rats. The following results were obtained from this study. At first, the number of WBC after one week were lowest in the alcohol-treated group. But, β-carotene treatment by oral injection was shown to diminish the increasing rate of WBC in the alcohol-treated group. Second, RBC was increased all the time in all groups tested with alcohol and/or β-carotene. It is of interest that there was a sudden increase of RBC in the alcohol-treated group. Third, Hb was increased in the alcohol-treated groups on the second week. On the other hand, the groups of β-carotene treatment showed more higher rate of increase in Ht and MCV. It is of note worthy that these effects were maximized in the group of β-carotene treatment followed by diet with cheese for a week. Finally, alcohol had little or no effect on MCH and MCHC, although β-carotene showed atypical effects. These results made it clear that alcohol can cause damages in liver, resulting in inflammation and increase of RBC. Also β-carotene is believed to protect liver from oxidative effects by alcohol. In according to these results, it is suggested that diet of anti-oxidants such as Vt. A, B, C, E, and β-carotene is strongly recommended to prevent certain diseases caused by oxidative damages.

      • Apolipoprotein A-I에 특이적인 단일클론항체 제조 및 특성에 관한 연구

        본 연구는 HDL을 구성하는 아포단백 A-I에 특이적인 단일클론항체 A-I30 및 A-I4를 제조하여 이의 특성을 분석하였다. 아포단백 A-I을 BALB/c 생쥐에 면역한후 얻어진 비장과 Sp2/O세포주와 융합시켜 얻어진 hybridoma를 생쥐의 복강내에 주입, 복수를 채취하여 Protein A-Sepharose CL4B크로마토그래피를 통해 단일클론항체를 분리하였다. 항체의 특이성은 ELISA 및 Western blotting으로 확인하였으며 ELISA방법으로 얻어진 해리상수(Kd)치는 A-I30 은 2.25 x 10의 마이너스8승, A-I4는2.15 x 10의 마이너스 8승 이었다.

        7-Substituted 2,3-dihydroxyphenazine 5, 10-dioxides were synthesized by the reaction of 1, 2, 4-trihydroxybenzene with 6-substituted benzofuroxan derivatives which had been obtained from aniline derivativies bearing methoxy, methyl, acetyl and nitro group at the para position. 2, 3-Dihydroxphenazine 5, 10-dioxide was also prepared by the reaction of 1, 2, 4-trihydroxybenzene with benzofuroxan. The antimicrobial activities of these phenazine dioxide were investigated in terms of minimum inhibitory concentration by the common twofold dilution technique. It was observed that the antimicrobial activity of the phenazine dioxides bearing electron releasing substituents was stronger than that of those bearing electron withdrawing substitutents. From this result, it was concluded that the antimicrobial activity of phenazine dioxide derivatives has a direct relationship with the electronic effect of the substituents.

      • Hb변이형에 대한 연구 : Exon Ⅱ Xho Ⅱ인지부위의 이상

        The human β-globin gene has been known to be many DNA sequence variation, and some of these could be detectable as variants in the length of DNA fragments produced by a restriction endonuclease digestion. Accordingly, I investigated the genetic variation on human β-globin coding region. At first, I found the abnormal β-chain of hemoglobin in Korean by aga gel, urea-starch gel and Supersepraphore Ⅲ polyacetate membrane electrophoresis. Polymerase chain reaction(PCR) for detection of Exon Ⅰ-Ⅱ, Ⅱ, and Ⅲ in β-globin gene was performed on normal and abnormal β-globin specimens, which were found at Soonchunhyang University Chunan Hospital by agar gel electrophoresis. Comparison study was made among the DNA fragments obtained from the restriction endonudease digests of PCR products. From the analysis of direct restriction enzyme digestion, I revealed a genetic variation in a XhoⅡ recognition site(-GGATCT-) about gene sequences coding β globin.

        In this paper, we designed, implemented and evaluated 32-bit CLA adder with the ENMODL(enhanced NORA Multiple Output Domino Logic) logic style which is new dynamic CMOS logic. The circuit is implemented using 0.8㎛ CMOS double-metal process technology. It operates stably and the addition time is less 3.9 ns. The ENMODL logic style can improve the performance in the high-speed computing circuits depending on the degree of recurrence.

      • 해수에 의한 부식속도 데이터 베이스의 구축과 이용에 관한 연구

        해수분위기에서 부식속도에 관한 데이터 베이스를 구축하였다. 본 데이터 베이스 system 프로그램은 커다랗게 두가지의 기능을 가지고 있는데 한가지는 데이터를 입력하는 기능이며 다른 한가지는 검색기능이다. 각각의 기능들은 몇개의 필드로 구성 되어 있다. 데이터베이스의 주된 필드들은 record, key-no, material type, material common name, environmental condition, corrosion rate등이다. 이중 중요한 연결고리용 필드는 각 데이터 들의 key-no로 사용하였다. We construct the Data-Base about corrosion rate of metal material in the sea water. In Data-Base system program have mainly 2 functions, one is data-input mode, the other is search mode. Each mode enclose some fields. Main fields are compose of record, key-no, material type, material common name, environmental condition, corrosion rate. Relation field is key-no of each data.

        한국산 배의 Polyphenol 화합물군이 쥐의 면역기능에 미치는 영향

        This study was conducted to investigate immunofunctional activity of the polyphenol fractions isolated from Korean pear. In the experimental of Rosette forming cell, the results showed that all the polyphenol fractions enhance the cell count compared with the control group. Especially polyphenol fraction II and III showed highly significant effect on Rosette forming cell, and allergy inhibition. After antigen challenge, histamine content of blood of the polyphenol groups was lowered to near the normal group.

        저품질의 데이터는 잘못된 의사결정을 초래하여 중요한 사업 기회를 놓쳐버리게 하거나 때로는 기업의 존폐 문제에 까지 영향을 미칠 수 있다. 대부분의 기업들은 데이터 품질 향상을 위한 전문 담당자를 배치하지 않고 있으며 신시스템 구축이나 데이터웨어하우스(DW), 고객관계관리(CRM) 구축 시 일시적인 데이터 정제(Data cleansing)를 실시하는 정도에 그치고 있으며, 데이터 품질의 중요성을 인지한 기업이라도 데이터 저품질로 인한 사업기회의 상실과 경제적인 손실이 가시적으로 당장 사업에 타격을 주는 것이 아니기 때문에 데이터 품질 관리에의 투자를 마지막 순위에 두는 경향이 있다. 본 연구는 먼저 데이터 품질 관리에 대한 정의를 정확히 하고 이를 기반으로 하여 국내 데이터 품질 관리의 실태를 파악한 후에 데이터 품질관리의 요소를 크게 데이터 품질 평가, 데이터 품질 관리 방법론, 데이터 품질관리 통제로 나누어 체계적인 데이터 품질 관리 방안을 연구하도록 한다.

      • 분광학적 방법에 의한 이소니트로소벤조일아세톤의 코발트(Ⅲ) 및 팔라듐(Ⅱ) 착물의 구조

        Isonitrosobenzoylacetone is particularly interesting, as the isonitroso group has two potentially coordinating sites which can compete with the carbonyl groups in forming bonds with the metal ions. In this paper tris(isonitrosobenzoylacetonato)cobalt(III) and bis(isonitrosobenzoylacetonato) palladium(II) have been prepared, and their structures have been investigated. Spectroscopic studies lead to the conclusion that the both complexes do not contain an OH group in the chelated five-membered ring structure in which the ligand coordinates to metal through oxygen of the acetyl group and nitrogen of the isonitroso group. The coordination manner of this ligand is similar to that of isonitrosoacetylacetone obtained by Haldar et al.^(3)(4) Very weak intramolecular hydrogen bonding of the free ligand is indicated by nmr and ir spectra.
