      • 그람음성 세균에 대한 나노실버의 항균효과

        Silver is one of the most universal anti-microbial substances. To improve the anti-microbial activity of silver, we successfully produced nanosilver by nano-technology. We examined the anti-microbial activity of nanosilver for several Gram negative pathogenic bacteria(Escherichia coli, Pseudomonas aeruginosa, Salmonella typhi). Most bacteria were controlled within 1 ppm of nanosilver witin 1 hour. The mechanism of anti-microbial effects was explored by SEM and TEM. The severe morphologic changes, irregular cell wall, increased electron density of cytoplasm, and plasmolysis, were observed. From the result, nanosilver will be available as an antibiotics alternative.

      • RF-magnetron 스퍼터링에 의해 제조된 HAp와 HAp-Ag 복합재료의 미세구조와 In-vitro 연구

        Microstructure and in-vistro study of RF-magnetron sputtered HAp and HAp-Ag composite films on the ZrO_(2) substrate were investigated using XPS, SEM, AFM and HR-TEM techniques. The homogenous and dense films about 0.8 ㎛ in thickness were successfully obtained and they were comprised with nano-crystalline grains about 30 nm in diameter. Micro cracks were not found at the interfaces between HAp and ZrO_(2) substrate and also on theire films. Without the dependency of Ag addition the fracure mode of HAp-Ag films showed a transgranular type. Due to the excellent bioaffinity, osteoblast MG-63 cells wre grown well on the HAp and HAp-Ag films with dense and healthly morphology, although the growth rate of cells on the HAp-Ag film was slower than that of the HAp film.

      • 생체세라믹 분말이 뼈모세포의 증식에 미치는 영향

        The important cause of re-operation in patients who received total hip arthroplasty is wear-debries-mediated osteolysis in bone-implant interface. We observed the effects of Al_(2)O_(3) and Ag-coated alumina (Al_(2)O_(3)-Ag) particles on the growth and morphologic changes of osteoblast. The results were compared with that of osteoblast cultured on Al_(2)O_(3), HAp-coated Al_(2)O_(3) and HAp-Ag composite Al_(2)O_(3) thin films. According to the effects of the particles, the cell number was decreased and the morphology of osteoblast was changed considerably compared with control. However, the proliferation of osteoblast cultured on HAp and HAp-Ag thin films were increased than control. These results suggest that the shape (particles) of material as well as characteristics of material itself may give a significant effect on the behavior of cell.

      • 사람의 혈관 내피세포에 대한 Salmonella 균종의 감염

        In order to elucidate the mechanism of injury to endothelial cell of blood vessels in typhoid fever, Salmonella typhi was inoculated in cultured human endothelial cells. Endothelial cells were prepared from the umbilical vein that was freshly obtained from human umbilical cord by collagenase treatment. After screening of cultured endothelial cells through the immunofluorescent antibody technique, ultrastructural change of cytopathic Aeffect, obtained by lectron microscopy, showed typical progress of pathogenesis caused by S. typhi; adherence, endocytosis, intracellular replication and release of microorganism. These patholgical changes led to cellular disruption and final death of endothelial cells. Endothelial cells of vascular system found to be the site of amplification as well as the target organ of S. typhi, which could play important role in hematogenous spread of this microorganism and also in pathologic sign or symptom in typhoid fever. An experimental infection of S. typhi in cultured human endothelial cells found to be an excellent and valuable virulence assay system. The above ability of S. typhi to proliferate and lead to endothelial cell death in cultured human endothelial cells confirmed to be a factor of endovascular disorder of typhoid fever such as rose spot on abdomen of patients.

      • 연속 다공질 생체세라믹에 대한 생체친화성 연구

        Continuously Al_(2)O_(3) porous bodies were fabricated by multi-extrusion process. Their biocompatibility was evaluated through in vitro and in-vivo study. Behavior of osteoblast adhesion and proliferation grown on Al_(2)O_(3) porous body was observed. Human osteoblast-like MG-63 cells were grown well on top, bottom, and outside as well as inside surface of porous body. Al_(2)O_(3) the ideal pore size was 150-200 um. The 3- dimensional continuously porous Al_(2)O_(3) body was implanted in subcutaneous tissue of rat for in-vivo study. The porous body was covered with well-developed fibrous tissues and showed the formation of new capillaries in it. No inflammatory response around porous body was found. From the results, the bio-ceramic materials used in the research was proved to have excellent biocompatibility.

      • 결핵균 감염에 따른 큰 포식세포의 Ferritin H chain 유전자 발현

        The purpose of this study was to identify macrophage genes regulated by infection of mycobacteria. Mycobacterium tuberculosis H37Rv (ATCC 27294) was provided from Korean National Tuberculosis Association. We have used a differential display RT-PCR to isolated cDNAs corresponding to transcripts that induced in THP-1 cells infected with M. tuberculosis. Among several differentially expressed trascripts, one clone, designated as 1-TC8 was 322 bp and was identical to human ferritin heavy(H) chain gene. Northern blot analysis confiremed that ferritin H chain gene has been markedly over-expressed in monocytic THP-1 cells infected with live Mycobacterium tuberculosis.

      • Vibrio cholerae균 분리를 위한 새로운 분별배지

        The purpose of this experiment is to develop a new differential medium for Vibrio cholera. The TCBS agar medium is used commonly for the identification of vibrio species, but it contains some inhibitors against other bacteria to increase the selectivity for vibrio species. So we developed the BYS agar medium as a new differential medium for V. cholera. We compared the colony forming unit(CFU) between BYS and TCBS using streak culture method and micro-pour plate method(100 ㎕). The CFU count was more than thirty times higher in BYS agar medium than in TCBS in both methods. These results suggest that BYS agar medium is a differential medium better than TCBS agar medium for the identification of V. cholera.

      • 나사산이 있는 관이음의 효과적인 해석모델의 개발

        This paper is concerned with and efficient model for the structural analysis of threaded connections which is used to assemble the tubular connections such as pipes. Advanced mechanics has been used to develop the model which yields acceptably accurate solutions for stresses when compared to other methods such as experimental or finite element analysis procedures. Beam-on-elastic-foundation equations are used to compute stresses due to assembly of the connection while bolt-nut equation and Lame' equation are used for axial loads and pressures. Numerical results show that our model is computationally efficient and valid in terms of stress distribution for make-up. The current model can provide the quick and inexpensive analysis tools to the production engineer with reasonable accuracy.

      • Estradiol에 의한 흰쥐-자궁의 수축 기능과 Polyamine 대사의 변동에 관한 연구

        The polyamines have been postulated to be essential in most processes associated with both hypertrophy and hyperplasia. The effects of estrogen on uterine contractility vary with animal species, and most reports have shown that in vitro contractility of rat uterus is decreased after estrogen treatment, but estrogen increases polyamine contents in castrated rat uterus. This study was carried out to investigate whether the regulation of polyamine metabolism may be instrumental in the contractile response of castrated nonpregnant uterus of female SD rats to electrical field stimulation (40 V, 100 μsec : EFS). Estradiol (E2) increased uterine contents of putrescine (PT), spermidine (SPD), and spermine (SPM). These E2 effects were inhibited by tamoxifen or DFMO, but E2-incuced increase of uterine SPM content was enhanced by aminoguanidine. E2 decreased the amplitude of force developed in the rhythmic spontaneous contraction of rat uterus, but it exaggerated the increase of uterine contraction force in response to EFS. The E2-dependent exaggeration was inhibited by tamoxifen, DFMO, or indomethacin, but was significantly enhanced by aminoguanidine. The synergism between E2 and aminoguanidine on uterine contractility was little affected by indomethacin. These results suggest that E2-dependent increase of uterine contractility may be ascribed partly to increase of uterus polyamines.

        환자 만족도 조사를 통한 QI활동의 효과 평가 : 일개 병원을 중심으로

        Background : Evaluation of patient's satisfaction is one of the most important aspects of quality improvement. If the patient highly satisfies with the medical service provided in the hospital, he/she will be likely to visit the same hospital again. Patient's satisfaction of a particular hospital is directly correlated with hospital profits as well as reputation in the community. For this reason, various kinds of survey measuring satisfaction level have been performed and many kind of QI activities for enhancing the profits as well as reputation in the community. For this reason, various kinds of survey measuring satisfaction level have been performed and many kind of QI activities for enhancing the patient's satisfaction. This study is to find the effect of hospital QI activities on the patient's satisfaction level. Methods : After questionnares were developed, survey of measuring satisfaction level was performed in August. 1998. On the basis of survey results. Ql activities were carried out to attain the target point of 4.0 and subsequent survey was done in November. 1998. Results : With three main principles of "problem solving approach with kindness". "helping patient to participate in medical procedure with sufficient information", and "putting employees into practice of attitude with human respect", the average level of satisfaction was enhanced from 3.45 to 3.55 level. Also kindness level of employees was increased from 3.71 to 3.82. Level of dissatisfaction about insufficient explanation and unkind attitude was dropped from 69% to 48% and from 82% to 46% respectively. Conclusion : With the result of this study overall satisfaction level was enhanced. In order to keep these advantages a operation of Quality Improvement Task Force Team in each subject will be required.
