      • 결핵균 감염에 의한 대식세포에서의 TNFα 유전자 발현 증가

        Mycobactetium tuberculosis is capable of growing and survival dormant for a long time within macrophages. This might be due to a balance between antimicrobial activity of host cell and pathogencity of M. tuberculosis. This study examined gene regulation in human monocyte derived THP-1 cell infected with mycobacteria. Gene expression was evaluated by differential display RT-PCR and was compared to uninfected cells. Results were confirmed by real time quantitative RT-PCR(TaqMan). Among many up or down-regulated clones, 25 colnes were sequenced and compared with known genes on GenBank. One of over-expressed clones from THP-1 cells infected with mycobacteria was identical to human tumor necrosis factorα(TNFα). In particular, the expression of TNFα was highly over-expressed in THP-1 cells infected with M. tuberculosis H_(37)Ra(avirulent) than H_(37)Rv(virulent). This may in fact be a mycobactetial virulence tactic.

        The purpose of this study is to find a Mycobacterium tuberculosis specific protein for a new diagnositic method of tuberculosis. We found a protein reacting with Kris antiserum(goat antiserum against Mycobacterium intracellulare) in culture filtrate of Mycobacterium avium LR114F by Western blot. The N-terminal amino acid sequence of this if compared with that of Mycobacterium tuberculosis using TIGR Database. It is cloned, expressed in E. coli. Recombinant TB-14 protein of M. tuberculosis is composed of 148 amino acid including 30 amino acid of signal peptide and it showed 78% of homology with M. avium. These results suggest that recombinant TB-14 protein may be useful for the devolpment of new diagnostic method for tuberculosis.

        PMMA를 이용한 다공질 β-TCP 골충진제 제조 및 생체적합성 평가

        Porous ${\beta}-tricalcium$ phosphate $({\beta}-TCP)$ bioceramic was fabricated by pressureless sintering using commercial HAp and different volume percentages of PMMA powders (30-60 vol.%). The range of spherical pore size was about $200-250\;{\mu}m$ in diameter. By increasing the PMMA content, the number of pores and their morphology were dramatically changed as well as decreased the material properties. In case of using 60 vol.% PMMA content, network-type pores were found, due to the necking of the PMMA powders. The values of relative density, elastic modulus, bending strength and hardness of the 60 vol.% PMMA content sample, sintered at $1500^{\circ}C$, were about 46%, 22.2 GPa, 5MPa and 182 Hv respectively. Human osteoblast-like MG-63 cells and osteoclast-like Raw 264.7 cells were well grown and fully covered all of the porous ${\beta}-TCP$ bodies sintered at $1500^{\circ}C$.

        차가버섯의 항결핵 활성에 대한 연구

        Inonotus obliquus is a traditional medicine mushroom that was developed from traditional medicine originating in ancient. It has been applied for cancer or immunotherapy,but its effect on pulmonary tuberculosis is not reported. Therefore, we measured the pulmonary tuberculosis therapeutic effect of methyl alcohol extract from MGIT 960 system with fluorescent indicator. Inonotus obliquus extract showed 14 day more inhibitory activity than the positive control. In addition, the anti-pulmonary tuberculosis activity of Inonotus obliquus was 50㎛. These results suggest that Inonotus obliquus methyl alcohol extracts could contribute to inhibition of pulmonary tuberculosis.

      • 생체세라믹 분말이 뼈모세포의 증식에 미치는 영향

        The important cause of re-operation in patients who received total hip arthroplasty is wear-debries-mediated osteolysis in bone-implant interface. We observed the effects of Al_(2)O_(3) and Ag-coated alumina (Al_(2)O_(3)-Ag) particles on the growth and morphologic changes of osteoblast. The results were compared with that of osteoblast cultured on Al_(2)O_(3), HAp-coated Al_(2)O_(3) and HAp-Ag composite Al_(2)O_(3) thin films. According to the effects of the particles, the cell number was decreased and the morphology of osteoblast was changed considerably compared with control. However, the proliferation of osteoblast cultured on HAp and HAp-Ag thin films were increased than control. These results suggest that the shape (particles) of material as well as characteristics of material itself may give a significant effect on the behavior of cell.

      • 대식세포내 결핵균 증식에 미치는 Prothymosin alpha Antisense Oligonucleotide의 영향

        Mycobactetium tuberculosis remains one of the world's most important pathogens. It is able to grow and survive within macrophage. I recently have identified the up-regulation of prothymosin α in THP-1 cells infected with Mycobacterium tuberculosis H_(37)Rv(virulent strain) using the differential display reverse transcriptase polymerase chain reaction(DD RT-PCR) and nothern blot. The purpose of this study was to identify the effect of prothymosin α antisense oligonucleotide in multiplication of Mycobacterium tuberculosis in macrophages. I have confirmed the over-expression of prothymosin α in THP-1 cells infected with M. Tuberculosis H_(37)Rv 48 hours postinfection. Also I found that the prothymosin α antisense oligonucleotide induced the replication of mycobacteria in macrophages infected with M. tuberculosis H_(37)Rv. This result suggests that the over-expression of prothymosin α protein is induced by infection of a virulent mycobacteria and it might be inhibit the replication of mycobacteria in macrophages.

        방전플라즈마소결법에 의해 제조된 저탄성 타이타늄 다공질체의 생체적합성 평가

        Porous Ti compacts were fabricated by spark plasma sintering (SPS) method and their in vitro and in vivo biocompatibilities were investigated. Alkaline phosphatase (ALP) activity representing the activity of osteoblast was increased when osteoblast-like MG-63 cells were cultured on the Ti powder surface. Some genes related to cell growth were over-expressed through microarray analysis. The porous Ti compact with 32.2% of porosity was implanted in the subcutaneous tissue of rats to confirm in vivo cytotoxicity. 12 weeks post-operation, outer surface and inside the porous body was fully filled with fibrous tissue and the formation of new blood vessels were observed. No inflammatory response was confirmed. To investigate the osteoinduction, porous Ti compact was implanted in the femur of NZW rabbits for 4 months. Active in-growth of new bone from the surrounded compact bone was observed around the porous body. From the results, The porous Ti compacts fabricated by spark plasma sintering might be available for the application of the stem part of artificial hip joint.

        마이크로웨이브법에 의해 제조된 HAp 및 BCP 분말이 뼈모세포 및 파골세포의 활성에 미치는 영향

        Hydroxyapatite (HAp) and biphasic calcium phosphate (BCP) nano powders were synthesized using the microwave-assisted synthesis process dependent on pH and microwave irradiation time. The average size of a powder was less than 100 nm in diameter. Through in-vitro cytotoxicity tests by an extract dilution method, the HAp and BCP nano powders have shown to be cytocompatible for L-929 fibroblast cells, osteoblastlike MG-63 cells and osteoclast-like Raw 264.7 cells. The activation of osteoblast was estimated by alkaline phosphatase (ALP) activity. When the HAp and BCP were treated to MG-63 cells, alkaline phosphatase activities increased on day 3, compared with those of the untreated cells. Also, the collagen fibers increased when the HAp and BCP powders suspension were treated to MG-63 cells, compared to those of the untreated cells. Quantitative alizarin red S mineralization assays showed a trend toward increasing mineralization in osteoblast cultured with powder suspension. In conclusion, hydroxyapatite and biphasic calcium phosphate appeared to be a bone graft substitute material with optimal biocompatibility and could be further applied to clinical use as an artificial bone graft substitute.

        Cellular Toxicity of Surfactants Used as Herbicide Additives

        The cellular toxicities of surfactants, a solvent, and an antifreeze that are included in herbicide formulations were assessed by measuring their effects on membrane integrity,metabolic activity, mitochondrial activity, and total protein synthesis rate in a cell culture. Polyethylene glycol, propylene glycol, and monoethylene glycol exhibited no cellular toxicity even at a high concentration of 100 mM. Sodium lauryl ether sulfate and polyoxyethylene lauryl ether significantly damaged the membrane, disturbed cellular metabolic activity, and decreased mitochondrial activity and the protein synthesis rate;however, their toxicity was far below those of the severely toxic chemicals at comparable concentrations. The severely toxic category included polyoxypropylene glycol block copolymer, polyoxyethylene tallow amine, and polyoxyethylene lauryl amine ether. These surfactants were cytotoxic between 3.125 μM and 100 μM in a dose-dependent manner. However, the toxicity graph of concentration vs toxicity had a point of inflection at 25 μM. The slope of the toxicity graph was gentle when the concentration was below 25 μM and steep when the concentration was greater than 25 μM. In conclusion, our results suggest that the toxicity of surfactants be taken care of pertinent treatment of acute herbicide intoxication.

      • Vibrio cholerae균 분리를 위한 새로운 분별배지

        The purpose of this experiment is to develop a new differential medium for Vibrio cholera. The TCBS agar medium is used commonly for the identification of vibrio species, but it contains some inhibitors against other bacteria to increase the selectivity for vibrio species. So we developed the BYS agar medium as a new differential medium for V. cholera. We compared the colony forming unit(CFU) between BYS and TCBS using streak culture method and micro-pour plate method(100 ㎕). The CFU count was more than thirty times higher in BYS agar medium than in TCBS in both methods. These results suggest that BYS agar medium is a differential medium better than TCBS agar medium for the identification of V. cholera.
