      • ISOLATION AND CHARACTERIZATION OF MOLECULAR MARKER FOR SEVERAL RACES OF PYRICULARIA ORYZAE USING RANDOM AMPLIFIED POLYMORPHIC DNA

        Primer 155(GCTGGCGCTG) was the most useful primer with distinctive polymorphic DNA pattern to distinguish between KI races and KJ races. Primer 173(CAGGCGGCGT) showed polymorphic DNA pattern to distinguish isolates of KI-100 race. The specific DNA fragment of 0.6Kb, generated from 88-62 of KJ-201 race by RAPD with primer 412)TGCGCCGCTG), was promising as a molecular marker to identify the isolate.

      • 김치에서 분리한 Lactobacillus sp. JC-7과 Lactobacillus acidophilus 88간의 Electrofusion 최적조건 설정

        김치 발효 숙성기간을 연장하고 신선도를 오랫동안 유지 할 수 있는 김치발효 starter의 개발을 목적으로 최적숙성기에 있는 김치로부터 bacteriocin생성능이 없는 유산균을 분리하여 동정하였으며, bacteriocin생성능이 우수한 L.acidophilus 88을 융합시키기 위해 electrofusion에 대한 최적 조건을 검토하였다. 분리균주는 Lactobacillus속으로 동정되어 Lactobacillus sp. JC-7이라 명명하였다. Electrofusion에 의해 생성된 융합주를 식별하기 위해 streptomycin(2.5㎎/ml)에 내성을 나타내는 Lactobacillus sp. JC-7변이주와 kanamycin(600㎍/ml)에 내성을 나타내는 L.acidophilus 88변이주를 분리하였다. Electrofusion을 100V/㎝, 120msec(72ohms,1670 capacitance) 에서 수행했을 때 융합효율이 가장 양호하였으며 전기장의 세기와 시간이 중가할수록 융합 효율이 현저히 감소하였다. 2가 양이온은 농도가 중가할수록 대체적으로 융합효율을 감소시키는 경향을 나타내었으나 1mM MGCl_2에서는 대조군에 비해 융합효율이 약간 증가하였다. PEG매개에 의한 융합법의 융합효율을 비교한 결과, 융합효율은 chemical fusion<electrofusion<electrofusion+20% PEG순을 나타내었다. Evaluation of Optimum Conditions for the Electrofusion between Lactobacillus sp. JC-7 Isolated from Kimchi and Lactobacillus acidophilus 88. Young-Bae Jo, Hyun-Jung Choi, Hyung-Suk Baik and Hong-Ki Jun*. Department of Microbiology, Pusan National University, Pusan 609-735,Korea-A lactic acid bacterium was isolated from kimchi. The isolated strain was identified as the genus Lactobacillus thrugh its morphological characteristics and named as Lactobacillus sp. JC-7. The optimum conditions for the electrofusion between streptomycin(2.5㎎/ml)resistant mutant of Lactobacillus acidophilus 88 and kanamycin(600㎍/ml) resistant mutant Lactobacillus sp. JC-7 were evaluated. The highest number of fusants were obtained at a capacitance value of 120msec(1670㎌), a field strength of 100V/㎝,and a pulse controller setting of 72Ω. The potimum pH of elecroporation buffer was 7.5 and the concentration of divalent cation was 1mM MG^2+. Electrofusants were efficiently obtained by addition 20% polyethylene glycol to electroporation buffer. The yield of fusion was better than that of using polyethylene glycol mediated chemical induction.

        수출입 컨테이너 장치장 배정을 위한 소프트웨어의 개발

        The Pusan Container Terminal faces a rapid increase in berthing time of container ships as well as in waiting time of external trucks, which is due to an absolute lack of yard space. This research is focused on the development of a decision support system for the planning of the container terminal yard assignment so that the yard space would be utilized most effectively. Efforts should be given to the reasonable assignment of the yard storage and the dynamic adaptation to the ever changing environment. The software introduced here is based o the know-how of the field exports and its framework takes the approach of the hierarchical decision making.

        Interrelation of Cyclin D1, Cyclin E, and p27Kip1 Expression on Tissue Arrays of Breast Cancer

        Purpose: To evaluate the clinical impact of the alteredexpression of cell cycle regulators in stage I and II breastcancers.Materials and Methods: The interaction between cyclinD1/E and p27Kip1 expressions were analyzed usingtissue microarray (TMA) technology in 133 breast cancers.Data from the immunohistochemical assays of 3molecules were merged, and analyzed, with a Ki67labeling index of the same tumors.Results: Cyclin D1 was expressed in 72 breast carcinomas(54.1%) and cyclin E in 60 (45.1%) out of the 133breast carcinomas. Expressions of cyclin D1 and cyclinE were inversely related to each other, and significantlyassociated with the estrogen receptor (ER) expressionand differentiation of the breast carcinoma. The expressionof cyclin E was significantly decreased in tumorsexpressing cyclin D1 (p=0.022). There was a trend forcyclin D1 expression to increase in tumors expressingp27Kip1 (p=0.053), but the expression of cyclin E didnot correlate with p27Kip1 expression. The Ki67 labelingindex was markedly increased in tumors expressing cyclinE, whereas it was significantly decreased in the cyclin D1or p27Kip1 expressing-tumors. From survival analysis,cyclin E expression was the only significant variable forthe prediction of poor survival.Conclusion: The abnormal expressions of cell cycleregulatory molecules are prevalent, and interrelated witheach other in breast cancer. Integration of TMA technologyallowed a high-throughput analysis for correlating molecularthe in situ findings, with the clinico-pathologicinformation. Among the three molecules studied, thecyclin E had a prognostic implication for stage I and IIbreast cancer. (Cancer Res Treat. 2002;34:388-393)

        Expression Profiles of Retinoblastoma Protein and p27Kip1 According to HER2 Amplification Status in Primary Breast Cancer

        Purpose: Human epidermal growth factor receptor-2 (HER-2)/neu amplification affects the cell proliferation through the modulation of multiple G1 cell cycle regulators in breast tumor cells. We performed this study to investigate whether retinoblastoma protein (pRB) and p27Kip1 were differently expressed according to the HER2 amplification status in human breast cancer. Methods: HER2 amplification was assayed by fluorescence in situ hybridization and the expression of cell cycle regulators were assayed by immunohistochemistry on 153 consecutive invasive breast cancers. The proliferative activity of breast cancer was analyzed according to the HER2 amplification and cell cycle protein expression status. Results: HER2 amplification was observed in 39 (25.5%) of 153 breast cancers. In the HER2 amplified breast cancers, the pRB expression was significantly increased (p=0.011) whereas there was no significant relationship between HER2 amplification and p27Kip1 expression. There was an inverse correlation between pRB expression and Ki-67 labeling index in the HER2 amplified breast cancers (p=0.036). In contrast, Ki67 labeling index was significantly decreased as p27Kip1 expression increased in HER2 non-amplified breast cancers (p=0.028). In HER2 non-amplified breast cancers, we could not observe any association between the pRB expression and Ki67 labeling index. Conclusion: The proliferation of the breast cancers was associated with pRB expression in HER2 amplified tumors whereas it was associated with p27Kip1 expression in HER2 non-amplified tumors. The results of the current study indicate that the cell proliferative activity of the breast cancer is under different growth signal pathways according to HER2 amplification status.

      • 화재 발생시 연기 거동과 온도분포에 대한 수치해석적 연구

        본 연구는 세 가지 유형의 아트리움 공간에 대해서 Zone 모델과 Field 모델을 비교하였으며, Zone 모델로는 FIRST, CFAST, NIST에서 개발된 CCFM.VENTS 그리고 CSIRO에서 개발된 NBTC 1-room 모델을 사용하였고 Field 모델로는 Chow에 의해 개발된 화재모델과 본 연구에서 개발된 SMEP을 사용하였다. 두 모델들에 대한 비교는 서로 유사한 결과를 보였으며, 특히 PISO 알고리즘을 사용한 SMEP의 경우가 SIMPLE 알고리즘 또는 SIMPLER 알고리즘이 적용된 field 모델에 비해 더 빠른 계산시간을 보여 주었다. In this paper, the smoke filling process for the three types of atrium spaces are simulated using the two types of deterministic fire model; zone models and field models. The zone models used are the FIRST, CFAST, and CCFM.VENTS models developed at the Building and Fire Research Laboratories, NIST, USA and the NBTC one-room model of FIRECALC delveloped at CSIRO, Australia. The field models used are the fire field model developed by W. K. Chow and a self-developed SMEP(Smoke Movement Estimating Program) based on computational fluid dynamics theories. The results predicted by the two approaches are very similar. The field model using SIMPLE algorithm or SIMPLER algorithm requires much more computing time compared with the use of SMEP using PISO algorithm.

      • 성인의 비만 및 체지방 분포에 따른 건강관련 체력의 비교

        This study investigated the comparison of the health-related physical fitness following to obesity and body fat distribution. The subjects were volunteered by the 20-50 aged male and female adult persons, and obese and non-obese groups were divided by BMI, and abdominal and gluteal-femoral obesity groups were divided by waist circumferences(WC). Grip muscular strength, back muscular strength, sit-up, sit & reach, and PEI of Harvard step test were compared following to the differences of %fat, BMI, and WC. The increasing level of %fat, BMI, and WC influenced the negative depression of health-related physical fitness. Especially, obesity and abdominal obesity could be arose a significant depression of cardiopulmonary function, and male group showed the significant decreasing of health-related physical fitness above WC threshold. Muscular strength per 1㎏ of body weight was reversely correlated with the percentage of body fat, so it was the need of the estimation of relative muscular strength per body weight for the precise comparison of muscular strength following to obesity and body fat distribution.

      • 열차내의 3차원 열 ·유동 특성에 대한 수치 해석적 연구

        In an attempt to design a high efficiency heating and ventilation system for passenger train, a computerized simulation has been conducted for the inlet and outlet ventilation system, and the heater installed under seats, based on the design criteria at which 60% of conditioned air is exhausted and rest of them are refreshed continuously, in a empty car. Thermal distribution and the flow analysis in a car have been investigated utilizing a commercial code(Star-CD), and the result reveals that the air distribution is shown to be a comples three dimensional flow characteristic in case of only the air conditioner operation in summer, and it turns out to be a steady state in the existence of temperature field.
