        Triptolide suppresses interleukin-1beta-induced human beta-defensin-2 mRNA expression through inhibition of transcriptional activation of NF-kappaB in A549 cells.

        <P>The immunosuppressive effect of triptolide has been associated with suppression of T-cell activation. However, the immunosuppressive effects of triptolide on innate immunity in the epithelial barrier remain to be elucidated. Human beta-defensin (HBD)-2 is an inducible antimicrobial peptide and plays an important role in the innate immunity. We have previously demonstrated that IL-1beta induced HBD-2 mRNA expression in A549 cells through activation of nuclear factor-kappaB (NF-kappaB) transcriptional factor as well as p38 mitogen-activated protein kinase (MAPK), c-Jun N-terminal kinase (JNK), or phosphatidylinositol-3-kinase (PI3K). In this study, we investigated effects of triptolide on IL-1beta-induced HBD-2 mRNA expression in A549 cells. Triptolide inhibited IL-1beta-induced HBD-2 mRNA expression in a dose-dependent manner. Addition of triptolide did not suppress activation of p38 MAPK, JNK, or PI3K in response to IL-1beta. Triptolide inhibited IL-1beta-induced MAPK phosphatase-1 expression at the transcriptional level and resulted in sustained phosphorylation of JNK or p38 MAPK, explaining the little effect of triptolide on IL-1beta-induced phosphorylation of these kinases. Although triptolide partially suppressed IL-1beta-mediated degradation of IkappaB-alpha and nuclear translocation of p65 NF-kappaB, triptolide potently inhibited NF-kappaB promoter-driven luciferase activity in A549 cells. These results collectively suggest that the inhibitory effect of triptolide on IL-1beta-induced HBD-2 mRNA expression in A549 cells seems to be at least in part mediated through nuclear inhibition of NF-kappaB transcriptional activity, but not inhibition of p38 MAPK, JNK, or PI3K. This inhibition may explain the ability of triptolide to diminish innate immune response.</P>

      • Interleukin-1B(1L-1B) polymorphisms and gastric mucosal levels of IL-Iβ cytokine in Korean patients with gastric cancer

        Interleukin-1B and IL-1 receptor antagonist gene polymorphisms are associated with an increased risk of gastric cancer (GC) in Caucasian populations. However, recent studies could not find any association between IL-1B-511T polymorphism and the risk of GC in Asians. We tested for an association between IL-1 loci polymorphisms with increased gastric mucosal levels of IL-1β and an increased risk of developing GC in a Korean population. Polymorphisms of IL-1A-889, IL-1B-31, IL-1B-511 and IL-1RN were genotyped in 434 controls and 234 patients with GC. Mucosal IL-1β cytokine was measured using an ELISA. The frequencies of IL-1A, IL-1B-511, IL-1B-31 and IL-1RN were not statistically different between controls and all patients with GC. After subclassification of GC, only patients with intestinal-type GC showed a higher frequency of IL-1B-31T homozygotes (OR = 2.2; 95% CI = 1.1-4.3) compared with controls. Risk was also significantly increased in these patients for IL-1B-31T homozygotes compared with patients with diffuse-type GC (OR = 3.4; 95% CI = 1.5-7.7). As in Caucasian populations, linkage disequilibrium between IL-1B-31 and IL-1B-511 was nearly complete, but the pattern of haplotype related to the risk of GC (IL-1B-31T/IL-1B-511C) was opposite (IL-1B-511T/IL-1B-31C). Mucosal IL-1β levels in H. pylori-infected GC patients were higher in patients homozygous for IL-1B-31T compared with IL-1B-31C/T and IL-1B-31C/C. Thus, the combined effects of H. pylori infection and IL-1B-31T/IL-1B-511C polymorphisms with enhanced mucosal IL-1β production contributed to the development of intestinal-type GC in this Korean population.

      • 레토르트 파우치 포장을 이용한 칡즙의 저장 중 항산화 활성의 변화

        The changes of antioxidant activities during storage of arrowroot juice by retort pouch packaging (AJRP) were investigated. The antioxidant activities of arrowroot juice such as polyphenol content (TPC), DPPH and ABTS radical scavenging activities (DPPHRS and ABTSRS) were analyzed during storage at 4oC and 25oC for 30 days. 0 day, TPC, DPPHRS and ABTSRS were 7.8 mg gallic acid equivalent( GAE)/mL juice, 3.7 mg ascorbic acid eqivalent(AE)/mL juice, and 11.5 mg ascorbic acid equivalent/ mL juice, respectively. In the storage for 30 days, TPC, DPPHRS and ABTSRS of PET+CPP packaging arrowroot juice (control) showed 7.6 mg GAE/mL juice, 3.5 mg/ AE/mL juice, 11.3 mg AE/mL juice at 4oC, respectively and 7.4 mg GAE/mL, 3.3 mg/ AE/mL, 10.6 mg AE/mL at 25oC, respectively. Whereas all antioxidant activities such as TPC, DPPHRS and ABTSRS of AJRP were consistently maintained. In conclusion, the storage of arrowroot juice with retort pouch film packaging was effective for maintaining antioxidant activities at various temperatures.

        Characteristics of Korean ginseng varieties of Gumpoong, Sunun, Sunpoong, Sunone, Cheongsun, and Sunhyang

        Background: Ginseng (Panax ginseng Meyer) is an important medicinal herbs in Asia. However, ginsengvarieties are less developed. Method: To developed ginseng varieties, a pure line selection method was applied in this study. Results: Gumpoong was testing of 4-yr-old specimens in 2002, the proportions of the below-groundroots that were rusty colored for Gumpoong was 1.29 in Daejeon and 1.45 in Eumseong, whereas theproportions for its yellow berry variant were 2.60 and 2.45 in the two regions, respectively. Thus theGumpoong was resistant to root rust. Sunpoong has a high yielding property. Its average root weight is70.6 g for 6-yr-old roots. Its yield is 2.9 kg/1.62m2 and the rate of heaven- and earth-grade product is20.9%, which is very high compared to 9.4% for Yunpoong. Sunone is resistance to root rot and thesurvival rate of 4-yr-old roots was 44.4% in 1997, whereas that of the violet-stem variant landrace was21.7%. Sunhyang has content of arginyl-fructosyl-glucose (AFG), which produces the unique scent of redginseng, is 95.1 mmol/g and greater than the 30.8 mmol/g of Chunpoong in 6-yr-old plants. Sunun andCheongsun are being nurtured to protect genetic resources. Conclusion: Developed ginsneg varieties will be used as the basis for the protection of genetic resourcesand breeding. Jang-Ho Lee

        Sol-Gel 법에 의한 친수성 무기-유기 혼성 코팅 용액의 제조

        소수성 고분자 필름의 무적성을 향상시키기 위하여 우수한 친수성을 보이는 무기-유기 혼성 코팅용액을 Sol-Gel법으로 합성하였다. 코팅용액은 무기물인 콜로이드 실리카 용액(Ludox)에 유기 조성을 함유한 화합물인 APS(3-aminopropyl triethoxysilane)를 첨가하여 제조하였다. 실란 커플링제인 APS는 콜로이드 실리카와 주변의 매트릭스인 고분자 필름(PET)에 각각 강한 결합을 형성하여 두 개의 서로 다른 물질을 강하게 연결하는 결합제 역할을 한다. 산성과 중성인 조건에서(pH 2,7) APS가 첨가되어 제조된 경우에는 순간적으로 용액은 겔이 되어 고분자 필름 위에 코팅 할 수 없었다. 반면 염기성 조건에서(pH 9, 11) 만들어진 혼성 코팅용액으로 고분자필름 위에 코팅한 경우에는 코팅 필름의 표면이 거의 균열이 없는 미세 구조를 보이며, 낫은 접촉각을 보여 우수한 친수성을 나타내었다. In order to improve the anti-fogging property of hydrophobic polymer films, inorganic-organic hybrid coating solutions with a good hydrophilic property were synthesized by the sol-gel method. The coating solutions were prepared by adding 3-aminopropyl triethoxysilane (APS) to a colloidal silica solution(Ludox). APS as a silane coupling agent forms strong bonds to the colloidal silica and surrounding polymer matrix (PET), and links two different materials together. Solutions prepared by the addition of APS at acidic and neutral conditions (pH 2.7) resulted in gels which can not be used as coating solutions. On the other hand, those at basic conditions (pH 9, 11) resulted in coatings that were less prone to cracking and showed a good hydrophilic property.

        주축 변위 센서를 이용한 절삭력 측정에 관한 연구

        A cylindrical capacitance-type spindle displacement sensor was developed and its effectiveness as a system to monitor cutting forces during hard turning was tested in this research. The sensor was installed between the face of spindle cover and the chucking element, and measured pure radial motion of the spindle under the condition with presence of roundness error at measured surface. To prove the effectiveness of the developed system, hard turning tests using ceramic inserts and tool steel as workpiece were hardened up to 65 Rc. The variations of pure radial motion of the spindle were measured during the cutting tests. The signals from the sensor showed the same pattern of cutting force variations from the tool dynamometer due to the progress of tool wear. As the flank wear of the ceramic tool increased, both static component of cutting forces and the amount of center shift of spindle orbit increased. Results from the research showed that the developed sensor could be utilized as an effective and cheap on-line sensing device to monitor cutting conditions and tool performance in the un-manned machining center.

        한국 재래산양에서 Bovine Viral Diarrhea, Bovine Herpesvirus-1, Parainfluenza Type-3 바이러스에 대한 혈청학적 연구

        The 948 goat sera were serologically tested for ten pathogens including bovine viral diarrhea (BVD), bovine herpes virus type-1 (BHV-1) and parainfluenza virus type-3 (PI-3). By viral neutralizaion test, three pathogens including BVD virus, BHV-1, PI-3 were applied for screening each neutralizing antibody and its titration. Among 948 sera, there were 215 seropositives (22.7%) against BVD virus, 25 (2.6%) against BHV-1 virus, and 161 (17.0%) against PI-3 virus, respectively. In the seropositive cases to BVD virus, provincial prevalence of them were recorded as Gyeongnam(30.8%) for the highest one, while as Jeonbuk (15.8%) and Gyeongbuk(l6.7%) relatively for the lower ones. Among them, higher than 1:8 in antibody titration was classified as 74 cases (34.4%) which were suspected the cycle of its infection in their herds. Among the seropositives to BHV-1, it was relatively lower prevalence rate ranging from 1.7% to 6.7% that those of BVD virus and PI-3, but more importantly they were prevailing across the country. Most of the seropositive cases (42.9%) to PI-3 were classified as 1:4-1:16 in titration, but also distributed all around the country. The prevalence in goats varied according to areas (15.8 to 30.8%). Related to the provincial prevalence of three viral agents in Korean goats, it seemed that three viral pathogens would be directly or indirectly involving in various disorders as primary or secondary causative agents resulting in decreased productivity.

        Due to the rising cost of energy, new separation processes based on extraction are becoming more attractive than before. Thus, the need for calculating and predicting liquid-liquid equilibria(LLE) compositions has very much increased. The purpose of this study is to determine and describe LLE data for the quaternary system containing acetic acid aqueous solution and solvent at 25℃. We investigated that the selection of the optimum solvent to separate acetic acid from aqueous solution and compared with ternary and quaternary LLE including binary solvent also.

        봉독(蜂毒)의 연구 경향에 대한 분석 -국내 학위논문 중 봉독에 관한 연구를 중심으로-

        Objectives : To research the trend of the study related to bee venom and to establish the hereafter direction for the study on bee venom therapy. Methods : I referred to national assembly library and national library of Korea, with `bee venom`, `apitoxin`, `apitherapy`, `bee sting`. After collecting all the theses about bee venom published by domestic university from 1992 to 2005, I figured out number and percentage of theses according to university, year, major field of study, subject. In case of subject, I classified and analyzed it with anti-inflammation effect;travail effect;immunity relation;safe treatment & prevention of side effect of bee venom. Results : The following results were obtained in this study. 1. In the arrangement by the year, the theses started to publish after 1992, and the number of theses has been increasing rapidly since 2000. And it has been decreasing since gaining the summit at 2003. 2. Classified by university, Kyung-Hee University published the most theses, and other theses were published by Kyung-Won University, Dong-Guk University, Kyoung-San University, Seoul National University order. 3. Classified by the major field of study, number of theses was the most by 69 in oriental medicine, next, it was 8, 2 in veterinary science and medicine. 4. Analyzed by the subject, numbers of theses that is related to anti-inflammation effect are 39(50%), numbers of theses related to travail effect are 15(18%), numbers of theses related to immunity are 19(23%), numbers of theses related to safty are 7(8%). 5. Theses that is related to anti-inflammation effect ; travail effect of bee venom shaded to mechanism study inclination after 2002 in primitive effect proof study inclination. Theses that is related to immunity of bee venom shaded to anticancer-related research inclination after 2000 in primitive immunity promotion effect study inclination. 6. In the study for safe treatment & prevention of side effect of bee venom, there were reports bee venom secures when it is injected in wide region, by low concentration, in 3 days cycle. 7. Theses related to bee venom are for the most part experiment theses by 76. Conclusions : As see in above result, safety of bee venom was proved, I think that the more clinical access by using bee venom should be achieved because objective, scientific basis was readied for effect of anti-inflammation, travail, immunity promotion, anticancer etc.

        Ion exchange performance to remove nitrate in water was studied using commercially available strong base anion exchange resin of Cl^- type in the batch reactors. Anion exchange resin was more effective than activated carbon or zeolite. With large resin amount or high temperature or low initial concentration, nitrate removal characteristics for a typical gel-type resin was increased. The curves showed the generally accepted selectivity sequence as SO₄^2-$gt;NO₃^-$gt;NO₂^-$gt;HCO₃^-.
