Objectives: The aim was to confirm the stem cell-like properties of the dental pulp stromal cells and to evaluate the morphologic changes during in vitro chondrogenesis. Materials and Methods: Stromal cells were outgrown from the dental pulp tissue of the premolars. Surface markers were investigated and cell proliferation rate was compared to other mesenchymal stem cells. Multipotency of the pulp cells was confirmed by inducing osteogenesis, adipogenesis and chondrogenesis. The morphologic changes in the chondrogenic pellet during the 21 day of induction were evaluated under light microscope and transmission electron microscope. TUNEL assay was used to evaluate apoptosis within the chondrogenic pellets. Results: Pulp cells were CD90, 105 positive and CD31, 34 negative. They showed similar proliferation rate to other stem cells. Pulp cells differentiated to osteogenic, adipogenic and chondrogenic tissues. During chondrogenesis, 3-dimensional pellet was created with multi-layers, hypertrophic chondrocyte-like cells and cartilage-like extracellular matrix. However, cell morphology became irregular and apoptotic cells were increased after 7 day of chondrogenic induction. Conclusions: Pulp cells indicated mesenchymal stem cell-like characteristics. During the in vitro chondrogenesis, cellular activity was superior during the earlier phase (within 7 day) of differentiation.

The purpose of this study was to investigate the relationships among trunk muscle, knee, and ankle strengths using Cybex770, the isokinetic muscle functioning measurement device. 41 K and Y University soccer players who did not have any experience about back pain or spine surgery were recruited as subjects. The results of the study were as follows. 1. Trunk extension was significantly higher than trunk flexion and muscle strength was diminished with increasing load speed. 2. Trunk flexion/extension ratio was well balanced at 30。/sec but this balance was broken with increasing load speed. 3. At 60。/sec of load speed, the knee extension and flexion of the left side were higher than those of the right side. 4. At 60。/sec of load speed, the ipsilateral muscle strengths of the left knee were well balanced but those of the right side were imbalanced. 5. At 60。/sec of load spped, the ratio of bilateral muscle strength of knee was -18.5±32.3% and this implies the possibility of knee injury. 6. Plantalflexion and dorsiflexion of the right ankle were higher than those of the left side, and dorsiflexion/plantalflexion ratio of the left side was higher than that of the right side. 7. The ratio of bilateral muscle strength of ankle at 60。/sec of load speed represented a balanced muscle development but this balance was broken with increasing load speed.8. There were partially significant relationships between trunk muscle and knee strengths. and between knee and ankle strengths. In conclusion, the trunk muscle strength of soccer player might be a very important factor to improve performance. In addition. a training technique which considering strength and speed is needed to develop not static but dynamic strength. This training not only develops well balanced muscle strength but also protects sport injury. Thus, the isokinetic training of trunk could improve the trunk muscle strength. and increase the ability of flexion/extension of knee and ankle.
Antiallergic Activity of Ginseng and its Ginsenosides

In this study, we measured the antiallergic activities of ginsenosides isolated from the root of Panax ginseng (Araliaceae), and of their metabolites, as produced by human intestinal bacteria. Compound K. which was identified as a main metabolite, had the most potent inhibitory activity on β-hexosaminidase release from RBL-2H3 cells and on the PCA reaction. The inhibitory activity of compound K was more potent than that of disodium cromoglycate, one of the commercial anti-allergic drugs. This compound demonstrated a membrane stabilizing action on differential scanning calorimetry. However, compound K did not inhibit the activation of hyaluronidase and did not scavenge active oxygen. These results suggest that the antiallergic action of compound K originates from its cell membrane stabilizing activity and that the ginsenosides of ginseng are prodrugs with extensive antiallergic properties.

To evaluate the antithrombotic activites of puerarin and daidzin from the rhizome of Pueraria lobata, in vitro and ex vivo inhibitory activities of these compounds and their metabolite, daidzein, were mearured. These compounds inhitited ADP- and collagen-induced platelet aggregation. Daidzein was the most potent. However, when puerarin and daidzin were intraperitoneally administered, their antiaggregation activities were weaket than when these compounds were administered orally. When in vivo antithrombotic activities of these compounds against collagen and epinephrine were measured, these compounds showed significant protection from death due to pulmonary thrombosis in mice. To evaluate the antiallergic activity of puerarin, daidzin, and daidzein, their inhibitory effects on the release of β-hexosaminidase from RBL 2H3 cells and on the passive cutaneous anaphylaxis (PCA) reaction in mice were examined. Daidzein enhibited potent inhibitory activity on the β-hexosaminidase release induced by DNP-BSA and potently inhibited the PCA reaction in rats. Daidzein administered intraperitoneally showed the strongest inhibitory activity and significantly inhibited the PCA reaction at doses of 25 and 50 ㎎/㎏ with inhibitory activity of 37 and 73%, respectively. The inhibitory activity of intraperitoneally administered daidzein was stronger than those of intraperitoneally and orally administered puerarin and daidzin. Therefore we believe that puerarin and daidzin in the rhizome of Pueraria lobata are prodrugs, which have abtiallergic and antithrombotic activities, produced by intestinal microflora.

To evaluate the antithrombotic and antiallergic properties of rhaponticin extracted from Rhei Rhizoma, the in vitro and ex vivo inhibitory activities of rhaponticin and its metabolite, rhapontigenin, were measured. These compounds inhibited in vitro ADP- and collagen-induced platelet aggregation. Rhapontigenin was more potent, with $IC_{50}$ values of 4 and $70{\;}{\mu}g/ml$, respectively. In ex vivo ADP- and collagen-induced rat platelet aggregation, these compounds also exhibited a potent inhibitory effect. The antiplatelet aggregation effects of rhaponticin and rhapontigenin were more potent than those of aspirin. Rhapontigenin showed significant protection from death due to pulmonary thrombosis in mice. Rhapontigenin also showed the strongest inhibitory activity against $\beta-hexosaminidase$ release induced by DNP-BSA. These compounds inhibited PCA reaction in mice. Rhapontigenin intraperitoneally administered showed the strongest inhibitory activity and significantly inhibited PCA at doses of 25 and 50 mg/kg, with inhibitory activities of 48 and 85%, respectively. The inhibitory activity of orally administered rhaponticin was stronger than that of intraperitoneally administered rhaponticin. These results suggest that rhaponticin, in the rhizome of Rhei Rhizoma, is a prodrug that has extensive antiallergic and antithrombotic properties.

The assessment of graft quality is very important for successful engraftment after bone marrow (BM) and peripheral blood stem cell transplantation. In addition to CFU-GM colony counting, flow cytometric enumeration of CD34+ cells has been peformed not only to evaluate graft quality but also to monitor the mobilized hematopoietic stem cells (HSCs) in peripheral blood (PB). Optimization of the gate is essential for flow cytometric measurement of CD34+ cells because these cells appear rarely and any procedures to enrich HSCs result in alteration of the cellular composition of graft. We isolated SBA-CD34+ cells from normal human BM mononuclear cells (MNCs) by panning, and analyzed their flow cytometric and microscopic characteristics. Within the MNC gate, the CD34+ cells reacting with FITC-conjugated anti-HPCA-2 monoclonal antibody (FITC-anti-HPCA) was 41.9% in the nonadherent SBA- fraction, 25.3% in the SBA-CD34- and 97.4% in the SBA-CD34+. On a scattergram, the SBA-CD34+ cells had low-to-intermediate right angle and low-to-high forward light scattering properties. And they clustered at the low-to-intermediate right angle and intermediate-to-high forward light scattering area mainly. Microscopically, the majority of SBA-CD34+ cells looked like large Iymphocyte (the Iymphoblastoid cells). By painting the cells which had intermediate-to-high forward light scatter as red color with PAINT-A-GATE software, they were revealed to be CD34+ on a histogram unexceptionally. Among the total events on each scattergram, 8.8% of the nonadherent SBA- fraction, 3.9% of the SBA-CD34- and 4,2.2% of the SBA-CD34+ belonged to the Iymphoblastoid eells expressing CD34 antigens. From these results, we suggest that (1) panning with SBA and anti-CD34 monoclonal antibody is highly efficient for purification of CD34+ cells, and (2) the optimal gate for measuring CD34+ cells should include both high and low-to-inter-mediate forward light scattering area within the low-to-intermediate right angle scattering zone.
