<P>Prevention and restoration of hepatic fibrosis from chronic liver injury is essential for the treatment of patients with chronic liver diseases. Vitamin C is known to have hepatoprotective effects, but their underlying mechanisms are unclear, especially those associated with hepatic fibrosis. Here, we analyzed the impact of vitamin Con bile acid induced hepatocyte apoptosis in vitro and lithocholic acid (LCA) induced liver injury in vitamin C-insufficient Gulo(-/-) mice, which cannot synthesize vitamin C similarly to humans. When Huh BAT cells were treated with bile acid, apoptosis was induced by endoplasmic retiiculum stress related JNK activation but vitamin C attenuated bile acid induced hepatocyte apoprosis in vitro. In our in vivo experiments. LCA feeding increased plasma marker of cholestasis and resulted in more extensive liver damage and hepatic fibrosis by more prominent apoptotic cell death and recruiting more intrahepatic inflammatory CD11b(+) cells in the liver of vitamin C-insufficient Gulo(-/-) mice compared to wild type mice which have minimal hepatic fibrosis. However, when vitamin C was supplemented to vitamin C-insufficient Gulo(-/-) mice, hepatic fibrosis was significantly attenuated in the liver of vitamin C-sufficient Gulo(-/-) mice like in wild type mice and this hepatoprotective effect of vitamin C was thought to be associated with both decreased hepatic apoptosis and necrosis. 한국산 겨우살이 렉틴(KML-C)에 대한 단일크론항체의 생산과 특징

We have reported that water-extracted Korean mistletoe(KM-110) had various biological activities such as antitumor and immunomodulatory activity and the lectin fraction(KML-C) of the extract was one of the major factors related to its biological functions. In this paper, we produced murine monoclonal antibody(mAb) against KML-C. The mAbs obtained were largely classified into two groups according to specificity to KML-C and ML-I, a lectin from European mistletoe. One group mABs(9H7-D10 and 3C2-1H4) strongly reacted with KML-C, but not ML-I. In contrast, another group mAbs(8B11-2C5, 8E12-3E9 and 5E10-F1) reacted with both KML-C and ML-1. The subisotypes of these mAbs were shown to be IgG1(9H7-1D10, 3C2-1H4 and 8B11-2C5) or IgM(8E12-3E9 and 5E10-F1). To develop an assay system for determination of the amonunt of KML-C, we established the sandwich ELISA(enzyme-linked immunosorbent assay) method using these mAbs and horse radish peroxidase(HRP) labelled mAbs.In various combinations of the mAbs for coated antibody and detection antibody, the sandwich ELISA quantitatively detected KML-C,showing the detection limit ranging from 7-5,000ng/ml. Especially, reproducibility(C.V.) of the sandwich ELISA, in which 8E12-3E9 was used for coating antibody and 8B11-2C5-HRP for detection antibody, was 4.59-5.83 in intra assay, and 3.9-9.4 in inter assay.

<P>A novel Gram-negative, aerobic, motile, short rod-shaped bacterium, designated J64T, was isolated from black sand collected from Soesoggak, Jeju Island, Korea. Cells grew at 4-37 degrees C, at pH 5.5-10.0 and with 0-10 % NaCl. The strain was found to be oxidase- and catalase-positive. Phylogenetic analyses showed that strain J64T belongs to the genus Pseudomonas, forming a monophyletic group with Pseudomonas pachastrellae, Pseudomonas pertucinogena and 'Pseudomonas denitrificans'. The 16S rRNA gene sequence similarity between strain J64T and type strains of all Pseudomonas species with validly published names was below 96.6 %. Low levels of DNA-DNA relatedness were found with respect to type strains of P. pachastrellae and P. pertucinogena, supporting the classification of strain J64T within a novel species of the genus Pseudomonas. Strain J64T contained C(18 : 1)omega7c (37.2 %), C(16 : 0) (20.4 %), summed feature 3 (17.4 %; comprising iso-C(15 : 0) 2-OH and/or C(16 : 1)omega7c) and C(12 : 0) (7.6 %) as major cellular fatty acids. On the basis of the phenotypic and phylogenetic data, strain J64T represents a novel species of the genus Pseudomonas, for which the name Pseudomonas sabulinigri sp. nov. is proposed. The type strain is J64T (=KCTC 22137T =JCM 14963T).</P>

Background: Protection of distal embolization by balloon occlusion and thrombus aspiration has not improved microvascular circulation nor decreased myocardial injury during primary percutaneous intervention (PCI) for ST-elevation myocardial infarction (STEMI) in randomized trials. In a prospective randomized trial, we investigated the mechanism of the poor effect of distal protection and thrombus aspiration (DP-TA) in 126 patients with STEMI. Methods: Patients with first-diagnosed STEMI were randomly assigned to DP-TA pretreatment or conventional PCI (c-PCI). Primary endpoint was reduced left ventricular end-diastolic volume (LVEDV) measured by MRI at post-PCI and 6months after PCI. Secondary end points were infarct ratio (infarct size to entire LV size) by delayed enhancement (DE), area at risk (AAR) ratio (AAR to entire LV size) by T2 high signal, microvascular occlusion index (MVO) ratio (MVO to entire LV size) by DE, and myocardial salvage index (MSI: (AAR-infarct size)*100/AAR) using cardiac magnetic resonance imaging (MRI) within 3days after PCI. Results: Baseline characteristics of the patients including cardiovascular risk factors and lesion characteristics were similar between the two groups. DT-PA failed to improve LV remodeling at 6months (LVEDV 140+/-39 vs 133+/-37 in c-PCI group, p=0.418). Infarct ratio, AAR ratio and MSI were not statistically different between DP-TA group and c-PCI group. However, MVO ratio was significantly larger in DP-TA group than in c-PCI group (2.4+/-2.7 vs 1.1+/-1.9, p=0.045). Conclusion: DP-TA was potentially hazardous in primary PCI for STEMI by increasing MVO. DP-TA should not be used in STEMI.

Multiprocessor system이 널리 보급되고 사용됨에 따라 concurrent programming은 더욱 더 중요한 feature가 되어가고 있다. 기존의 C 언어는 concurrent programming을 위한 feature들을 가지고 있지 못하나, 본 논문에서는 concurrent processing이 가능한 Concurrent-C 언어를 설계, 구현하였다. Concurrent feature들을 첨가하는 방법으로는 여러 종류의 runtime library routine들을 제공하여 C program 내에서 이 routine들을 call하는 방식을 사용하였다. Concurrent-C는 UNIX 환경하에서 구현되었으며, 실제로 C compiler를 제공하는 어떠한 OS 상에서도 host machine의 종류에 관계없이 구현될 수 있다.
Identification of Novel SNPs in Bovine Insulin-like Growth Factor Binding Protein-3 (IGFBP3) Gene

The insulin-like growth factors (IGFs), their receptors, and their binding proteins play key roles in regulating cell proliferation and apoptosis. Insulin-like growth factor binding protein-3 (IGFBP3, OMIM #146732) is one of the proteins that bind to the IGFs. IGFBP3 is a modulator of IGF bioactivity, and direct growth inhibitor in the extravascular tissue compartment. We identified twenty-two novel single nucleotide polymorphisms (SNPs) in IGFBP3 gene in Korean cattle (Hanwoo, Bos taurus coreanae) by direct sequencing of full gene including -1,500 bp promoter region. Among the identified SNPs, five common SNPs were screened in 650 Korean cattle; one SNP in promoter (IGFBP3 G-854C), one in 5'UTR region (IGFBP3 G-100A), two in intron 1 (IGFBP3 G+421T, IGFBP3 T+1636A), and one in intron 2 (IGFBP3 C+3863A). The frequencies of each SNP were 0.357 (IGFBP3 G-854C), 0.472 (IGFBP3 G-100A), 0.418 (IGFBP3 G+421T), 0.363 (IGFBP3 T+1636A) and 0.226 (IGFBP3 C+3863A), respectively. Haplotypes and their frequencies were estimated by EM algorithm. Six haplotypes were constructed with five SNPs and linkage disequilibrium coefficients (|D'|) between SNP pairs were also calculated. The information on SNPs and haplotypes in IGFBP3 gene could be useful for genetic studies of this gene.
