Neutrophils are also known to acquire the character-istics of dendritic cells (DCs) under the appropriate conditions. In this study, neutrophils were cultivated in vitro in the presence or absence of compounds mod-expression profile of the DC markers. Higher MHC-I, CD80, CD86, CD83, and CD40 expression levels were detected on the surface of the cultured neutrophils for 24 h than on the freshly isolated cells. The annexin V-positive cells showed a higher expression level of the DC markers than the annexin V-negative cells. The population of neutrophils double stained with anexin V and the DC markers increased after being incubated with agonistic anti-Fas Ab. LPS, the anti-apoptotic compound, decreased the CD86 and MHC-I ex-pression levels but 50-60% of the DC marker-positive cells were detected in the annexin V-positive cells. In contrast, CD80, CD86, CD83, and HLA-DR mRNA levels increased in the GM-CSF-treated neutrophils but not in the anti-Fas Ab-treated neutrophils. T cell pro-liferation was inhibited by co-culturing them with an-ti-Fas Ab- or LPS-treated neutrophils at a high neu-ated T cell proliferation was increased by the LPS-treat-ed neutrophils but decreased by the anti-Fas Ab-treat-ed neutrophils. There was a lower level of interferon-γ production in the T cells co-cultured with anti-Fas Ab-treated neutrophils than with the LPS-treated neutrophils. This suggests that apoptotic neutrophils express DC markers on their surface and the difer-ential expression of DC markers might have a detri-detrimental effect on the immune reaction.
Interleukin-10-Producing B Cells Help Suppress Ovariectomy-Mediated Osteoporosis

Osteoporosis is prevalent in elderly women and it may cause dental implant failure. In particular, estrogen deficiency in postmenopausal women leads to higher rates of osteoporosis prevalence. Immune cell-mediated effects involving the development of osteoporosis have been studied previously; however, the role of IL-10-producing regulatory B (B10) cells in osteoporosis is largely unclear. Here, we examined the role of B10 cells in osteoporosis. C57BL/6 mice were subjected to ovariectomy (OVX). Fifteen weeks after OVX surgery, the first molar of the right maxillary was extracted, and twenty-four weeks after OVX surgery, serous progression of osteoporosis was observed in the alveolar bone. Moreover, the proportion of CD19+CD5+CD1dhigh regulatory B cells, B10, and CD4+CD25+FoxP3+ regulatory T cells from the spleen of OVX mice decreased during the progression of osteoporosis, compared to controls. In contrast to regulatory cells, IL-17-producing Th (Th17) cell levels were increased in OVX mice. Adoptive transfer of B10 cells to OVX mice led to a decrease in Th17 cell abundance and inhibited the development of osteoporosis in the alveolar bone from OVX mice. Thus, our results suggest that B10 cells may help suppress osteoporosis development.

Rylene dyes exhibit superior photophysical properties including outstanding chemical and photochemi-cal stabilities as well as highfluorescence quantum yields (FQYs). However, the rylene chromophoreshave some limitations, such as high hydrophobicity and low solubility in aqueous solution, as afluorescent tag for biological system. Herein, we present a general method to load the rylene dyes intotriblock copolymer nanoparticles (NPs) and stabilize the core. The rylene dyes in confined hydrophobiccore fairly preserve their FQYs in aqueous environment. Lumogen probe nanoparticles (PNPs) wereprepared with four different commercial rylenes (Lumogen Violet, Yellow, Orange and Red) in thespectral region of visible and infrared (400–750 nm) and their photophysical properties werecharacterized. They exhibited enhanced photostability compared to commercialfluorescent dyes. Alsothe internalization of Lumogen PNPs in HeLa cell was confirmed by cell uptake test. These results indicatethe Lumogen PNPs can be widely applicable influorescence detection and imaging such asflowcytometry andfluorescence microscopy.
