        사향의 크로마토그램 패턴 분석을 통한 품질비교 연구

        Objectives : This study is to check the quality of Moschus products in Korean markets using a chromatographic analysis. Methods : We collected musk products, two genuine products from Russia (A, B), two authentic products from Hong Kong (C) and Korea (D), the Ministry of Food and Drug Safety (MFDS) standard (E), and two fire ants products (E, F). Results : For identification, TLC analysis of 60% ethanol extracts of each product showed that A, B, C, and E have the suitable patterns before color development at 365 nm UV light for the MFDS regulation in Korea. A clear red spot was observed from the E at Rf 0.6 with 365 nm UV light after color development, but this spot was not found in A and B. For the purity test, a distinct violet spot with Rf 0.87 was observed from the A and B in TLC analysis of methanol extracts of each product at white light, however, this spot was not matched with the impurities of E and F. In HPLC-UV pattern analysis, a similar peak pattern was shown in A, B and E, and similar peaks were observed C, although the similarity was weaker than that of A, B, and E. The F and G showed different peak patterns compared with the peak patterns of other samples. Conclusions : Hence, it is considered that the test methods need flexibility in application for identification and purity test depending on the type of sample.

      • 다중요인분석을 이용한 전탕용수에 따른 신이 성분의 추출효과 비교

        Abstract – The composition of minerals and anions in six types of decoction water (S1 to S6) were analyzed by inductively coupled plasma-atomic emission spectomerty (ICP-AES) and inductively coupled plasma-mass spectromerty (ICP-MS), respectively. Extraction efficiency of active compound of Magnoliae Flos depending on decoction water was determined by analyzing seven major peaks using HPLC and LC-MS. In results, S5 contained highest Ca, Mg and SO4 content and S4 showed the highest Cl and NO3 content than other waters. Using multiple factor analysis, it was proved that the variation of the area for peak1 and peak7 of Magnoliae Flos extract related to K, Si and Na, and peak5 linked to SO4, Cl and NO3. Considering peak5 which is highly variable depending on the type of decoction water, the area of peak5 decocted with S4, S5 and S6 were higher than those decocted with S1, S2 and S3. In conclusion, the content of compounds in Magnoliae Flos extracts was different depending on decoction water, indicating that decoction water affected the extraction efficiency and the bioactivity of Magnoliae Flos extract.

      • 시트르산처리와 열처리가 인삼 잎 중 진세노사이드의 함량 변화에 미치는 영향

        The rare ginsenosides are seldom found in nature. However, they are recognized as showing higher biological activities than other ginsenosides. There are many reports about increasing the contents of rare ginsenosides in ginseng root using chemical, physical or biological methods. Some of the methods were very successful and contributed to increase the quality of the ginseng products. We tried to increase the rare ginsenosides in ginseng leaves with citric acid or heat treatment. In this way, the industrial value of ginseng leaves could be highly increased. The ginsenosides in untreated and treated ginseng leaves were identified and determined using high performance liquid chromatography with ultra- violet detection and mass spectrometry. The results showed that the contents of rare ginsenosides in- creased after heat treatment or citric acid treatment, however heat treatment was more effective. Furthermore, when the ginseng leaves were treated with 1 mol/L citric acid together with 3 hours of heat treatment, the contents of eight rare ginsenosides of Rg6, F2, F4, Rk3, Rg3(S), Rg3(R), Rk1, Rg5 were increased to 0.64%, 1.26%, 0.54%, 1.11%, 0.73%, 0.73%, 0.23%, 0.58% and 1.14%, respectively. This study suggested that the combined treatment of citric acid and heat treatment could be applied for the process of ginseng leaves with enhanced biological activities through increased contents of rare ginsenosides.

        육미지황탕가미방에 의한 흰쥐 기억력 향상과 관련된 Hippocampus 부위의 특이 유전자 발현에 대한 연구

        The herbal extract(YMT_02) is a modified extracts from Yukmijihwang-tang(YMJ) to promote memory-enhancing. The YMJ extracts has been widely used as replenishing yin and tonifying the kidneys herbal medicine for hundred years ia Asian countries. The purpose of this study is to: 1) quantitatively evaluate the memory-enhancing effect of YMT_02 by passive avoidance test, 2) statistical evaluation of candidate gene expression (pentraxin, PEP-19, transthyretin) in rat hippocampus. The hippocampi of YMT_02 and control group were dissected and mRNA was further purified. After synthesizing cDNA using oligo-dT primer, the cDNA were applied to Real Time PCR. The results were as follows : 1) passive avoidance test showed enhancing memory retentin by YMT_02 treatment, 2) expression of pentraxin, that accelerate degenerating of neuronal cell, was significantly decreased, 3) the mRNA of genes that has been known to be associated with protecting neuronal cell degeneration, such as PEP-19 and transthyretin, were significantly increased upon YMT_02 treatment. From above results, the administration of YMT_02 which tonify the function of Kidneys could enhance the ability of memory and learning. In addition, the administration of YMT_02 enhance memory retention through modulating particular gene (pentraxin, PEP-19, transthyretin) expressions in hippocampu

        牡丹皮의 PC12 cell 항산화 효과와 관련 HO, MIF, COMT 유전자 발현에 미치는 영향

        Oriental medicine explains aging as the weakening of Kidney-Qi, and Kidney-strengthening herbal medicines such as Yukmijihwang-tang have been studied for anti-aging effects. In Western Medicine, the hypothesis that reactive oxidant species(ROS) contribute to the aging process is generally accepted. It has been reported that Moutan Cortex Radicis extract (MCR) was the most effective constituent of Yukmijihwang-tang in decreasing ROS production in oxidative-stressed cells. The purpose of this study is to confirm the anti-oxidant effect of MCR on PC12 cells, the expression of Heme oxygenase (HO), Macrophage migration inhibitory factor (MIF), Catechol-0-methyltransferase (COMT) using real time RT PCR. PC12 cells were treated without or with hydrogen peroxide in the presence or absence of MCR using MTS assay. Hydrogen peroxide decreased the viability of PC12 cells by 53% and MCR did not influence that of stressed PC 12 cells irrespective of dose or incubation period. However, MCR showed an inhibitory effect on production of ROS in stressed cells, both dose and incubation time dependently. In particular, 1 mg/ml of MCR for 24 h culture almost returned to normal level. In the quantiation of anti-aging related gene expression, MCR at 1 mg/ml increased the expression of HO by 370%, MlF by 180% and COMT by 280% through real time RT PCR. In conclusion, MCR treatment protected PC12 cells from hydrogen peroxide and decreased ROS production and enhanced anti-oxidative gene expression such as HO, COMT and MlF, which suggests that MCR is involved in controlling anti-aging of nerve cells through elimination of cytotoxic stimuli..
