        Adiramycin이 C6 glioma 세포에 미치는 영향에 대한 원지의 효과

        To elucidate the cytotoxic effect of adriamycin on C6 glioma cells, we examined the neurotoxicity induced by adriamycin by MTX assay when C6 glioma cells were cultured in the media containing various concentrations of adriamycin for 72 hours. In addition, neuroprotective effects of herb extract such Polygalae Radix(PR), on adriamycin-induced neurotoxicity in cultured C6 glioma cells were evaluated after C6 glioma cells were preincubated with various concentrations of herb extract, PR for 2 hours before 6uM adriamycin for 72 hours. Adriamycin decreased remarkably cell viability in dose-and time-dependent manner in these cultures, and also herb extract, PR increased cell viability of C6 glioma cells damaged by adriamycin. From these results, it is suggested that adriamycin was highly toxic in cultured C6 glioma cells, and PR was effective in blocking the neurotoxicity induced by adriamycin in these cultures.

        芝山先生이 주창한 膽 膀胱 용어에 대한 연구

        In terms of Yin and Yang theory, Dam represents excessive Yang and deficient Yin, and Bangkwang means excessive Yin and deficient Yang. In terms of Ki and Hyung, Dam is vigorous Ki and flabby Hyung, and Bangwang is strong Hyung and weak Ki. In terms of the balance of Ki and Blood, Dam is more Ki and less Blood and Bangkwang is the opposite. In terms of meridians concerned, Dam belongs to Joksoyangdam meridian(足少陽膽經) and Bangkwang belongs to Joktayangbangwang meridian(足太陽膀胱經). Mr. Jeesan saw that the person having vigorous Ki and flabby Hyung tends to be dynamic and the person having strong Hyung and weak Ki tends to be static and designated both as Dam and Bangkwang, respectively. Meridians are the route of Ki and Blood in the body and since their titles are thought to imply basic oriental medical theories, they are better choices to be comprehensive and systematic than other terms. In the 12 meridians, the 6 foot meridians are more important than the other hand meridians because human beings adapt themselves to the earth. Among the 6 foot meridians, the 3 Yang foot meridians are more representative of the body since they cover more of the body. Joksoyangdam meridian(足少陽膽經) is related to more Ki and less Blood and thus is a relevant term to describe Dam. Likewise Joktaeyangbangkwang meridian(足太陽膀胱經) is related to more Blood and less Ki and thus can correspond to the term 'Bangkwang'. In addition, the Dam organ and Dam meridians meet the condition of excessive Yang and deficient Yin, vigorous Ki and flabby Hyung, more Ki and less Blood, on the other hand, the BangKwang organ and Bangkwang meridians meet the condition of excessive Yin and deficient Yang, strong Hyung and weak Ki, more Blood and less Ki. Thus the terms of Dam and Bangkwang satisfy the basic concepts Mr. Jeesan established.

        生體 氣에 대한 波動共鳴的인 연구

        The present condition of study on the wave-resonance for the bio-KI is observed. And the probroms of reception attendant on the stimulation of wave is observed centering around the sensory organ. The results is as follows: In the wave-resonant stand point, the tendency of studying on KI is showed in the several field all over the world. Because it is originated radionics, the wave-resonant tools of MRA insistenting minute-magnetism-resonance-apparatus need the more severe data in the side of electric circuit. The wave resonance apparatus according to the frequency occurance transmits low-frequency's vibration ratio to the electric stimulating aspect. The wave-water is considered on the application of wave-resonance transcription on the water, and needs the comprehension of torsion-field level. The wave-stimulation of the bio-KI and the reception of the sensory-organ is observed the connection of the sensory-organ and it's corresponding wave. The informations recorded in the wave are distinguished patternly. And the several shaping waves transmit the informations each other through the resonnance. The wave theory is explained the LEE(理) as the wave-pattern and the KI(氣) as the revelation of the patterned-wave in the LEE-KI theory(理氣論), moreover the SHIN(神) as the information of the wave-pattern and the KI as the energy of the wave and the JEUNG(精) as the material of the wave in the JEUNG-KI-SHIN theory(精氣神論). In this point, the study on the wave-resonance of bio-KI is thought that it is significantly in the study method for oriental medicine. The sum of the wave makes the moving body and forms the universe. It means that the several wave patterns gather and form new field. The pattern is a kind of the information and the information is not materially. The information of wave-pattern is the arrangement and the combination of the material source.

        Clinical Review on Complications of Stroke Patients Admitted Oriental Medical Hospital

        Background and Purpose : In the acute stage of stroke, many medical and neurological problems complicate and the clinical course of patients, according to previous reports. Although some of them may be predicted, few data exist about them. So we were to investigate the characteristics of complications in hospitalized patents due to stroke. Methods : We retrospectively examined the clinical notes of patients admitted in Won Kwang oriental medical hospital after stroke. Two observers inspected the clinical notes using predefined diagnostic criteria and recorded-the type, timing, and frequency of complications that occurred during hospitalization. Results : Total 78 subjects were included in this study. Medical complications(88%) were more frequently complicated than neurological ones(65%). The most common medical complication was constipation(29%) followed by fever(28.2%) and overflow incontinence associated with dysuria(28%). The most common neurological complication was dysphagia(23.7%) and the second was agitation or lnsomnia(36%) and the third was headache(21%). Conclusion : Complications after acute stroke were commonly observed. And There were more frequent medical complications than neurological ones. So we should have much knowledge about medical complications and treat them actively. The differences between our study and previous studies are attributable to the different methods including patent selection and diagnostic criteria.

        The Effects of Acupuncture(ST36. LI4) on the Colonic Transit Time in Chronic Constipation Patients

        Background and Purpose : We were to investigate whether simple and electric acupuncture can affect the colonic transit time in both normal persons and chronic constipation patients. Methods : Twenty one volunteers were divided into two groups; first, normal control group(N=12) who had normal defecation habits, second, chronic constipation group(N=13). Before acupuncture, colonic transit time was checked using radio-opaque markers. Then simple acupuncture was done at four acupoints(both ST36, L14) and maintained for 15 minutes during 4 days. Electric acupuncture was done using same methods except for applying 2Hz electrical stimulation. Result : In the normal group, after simple acupuncture, each transit time in the total, right, left was not changed statistic significance compared to pre-acupuncture(P>0.05), but which of rectosigmoid colon shortened statistic significance(P<0.05). After Electric acupuncture, transit time of right colon was shortened(P<0.05), and extended(P>0.05) in rectosigmoid colon compared to pre-acupuncture and simple acupuncture. In constipation group, after simple acupuncture, only rectosigmoid colonic transit time shortened statistic significance compared to pre-acupuncture(P<0.05). After electric acupuncture, also the transit time of rectosigmoid colon was shortened statistic significance compared to pre-acupuncture(P<0.05), but not to simple acupuncture(P>0.05). Conclusion : In normal persons without constipation, acupuncture affect the colonic transit time differently to the methods of it. In chronic constipation group, simple and electric acupuncture only reduces the rectosigmoid colonic transit time statistically significant(P<0.05).

        CD Gene Microarray Profiles of Bambusae Caulis in Liquamen in Human Mast Cell

        Bambusae Caulis in Uquamen(BCL) has been used to relieve the cough and asthma, and remove the phlegm in traditional Oriental medicine. In recent years, it was studied for its antiinflammatory, antiallergenic, immune-modulating, and anticarcinogenic capabilities. This experiment was performed to evaluate the microarray profiles of CD genes in human mast cells before and after SCL treatment. The results are as follows: The expression of 51 of the genes studied was up-regulated in the BCL-treated group; they include the genes coding L apoferritin, beta-2-microglobulin, ferritin light polypeptide, CD63, monocyte chemotactic and activating fact, heme oxygenase 1, CD140a, integrin alpha M, colony stimulating factor 2 receptor, eukaryotic translation elongation factor, CD37, interleukin 18, NADH dehydrogenase 1 beta, CD48, 5-lipoxygenase activating protein, interleukin 4, ribosomal protein L5, GABA(A) receptor-associated protein, beta-tubulin, integrin beta 1, CD162, CD32, lymphotoxin beta, alpha-tublin, integrin alpha L, CD2, CD151, CD331, 90 kDa heat shock protein, CD59, CD3Z, microsomal glutathione S-transferase 2, CD33, CD162R, cyclophilinA, CD84, interleukin 9 receptor, interleukin 11, CD117, CD39-like 2, and so forth. The expression of 7 of the genes studied was down-regulated in the BCL-treated group; they include the genes coding CD77, CD238, SCF, CD160, CD231, CD24, and CD130. Consequently, the treatment of BCL on the human mast cells increased the expression of 51 genes and decreased the expression of 7 genes. These data would provide a fundamental basis to the traditional applications of Bambusae Caulis in Liquamen.

        夏枯草 ethyl acetate분획의 U937세포에 대한 세포고사 유도효과

        Prunellae Spica is a flower petal of Prunella vulgaris var. lilacina used for treatment of lymphoma, breast cancer, hepatitis and pathological fluid related diseases in oriental medicine. We tried to evaluate the mechanism of Prunellae Spica in the treatment of cancer. The ethyl acetate layer of Prunellae Spica showed a good cytotoxicity on U937 cells with IC50 of 8 ug/ml. It induced apoptosis in U937 dose-dependently by cell cycle analysis following PI staining. We also confirmed it induced DNA fragmentation in U937 cells from the concentration of 10 ug/ml. From western blot assay we observed@ the ethyl acetate layer of Prunellae Spica downregulated procaspase-3 and cleaved PARP in a dose dependent manner, whereas it didn't affect bax and bcl-2. Taken together, these results indicate the ethyl acetate layer of Prunellae Spica can induce apoptosis in U937 cells suggesting it can be potently applied to cancer.

        Effect of Hwao-tang on Superoxide Generation and Neutrophil Functions

        We investigated that Hwao-tang had various effects on stimulus-induced superoxide generation in human neutrophils. Hwao-tang significantly inhibited N-formyl-methionyl-leucyl-phenylalanine-induced superoxide generation in a concentration-dependent manner, but not that induced by arachidonic acid. Also, Hwao-tang significantly reduced mouse paw oedema induced by carrageenan. The results suggest that protein tyrosine -kinase participates in fMLP-mediated superoxide generation by Hwao-tang-treated human neutrophils. Also, the results indicate that Hwao-tang exerts ant-inflammatory effects related to the inhibition of neutrophil functions and of NO and prostaglandin E2 production, which could be due to a decreased expression of iNOS and COX-2.

        CCl_4로 誘發된 白鼠의 肝損傷에 대한 葛根의 肝保護作用 연구

        In the present study, we investigated the protective effect of the Puerarie Radix water extract (PRE) against CCI_4-induced hepatotoxicity and the mechanism underlying these protective effects in the rats. The pretreatment of PRE has shown to possess a significant protective effect by lowering the serum alanine and aspartate aminoteansferase (AST and ALT) and alkaline phosphatase (ALP). This hepatoprotective action was confirmed by histological observation. In addition, the pretreatment of PRE prevented the elevation of hepatic malondialdehyde (MDA) formation and the depletion of reduced glutathione (GSH) content and catalase activity in the liver of CCI_4-injected rats. The PRE also displayed hydroxide radical scavenging activity in a dose-dependent manner (IC50 = 83.6 ㎍/ml), as assayed by electron spin resonance (ESR) spin-trapping technique. Moreover, the expression of cytochrome P450 2E1 (CYP2E1) mRNA, as measured by reverse transcriptase-polymerase chain reaction (RT-PCR), was significantly decreased in the liver of PRE-pretreated rats when compared with that in the liver of control group. Based on these results, it was suggested that the hepatoprotective effects of the PRE may be related to anfioxidant effects and regulation of CYP2E1 gene expression

        Cisplatin의 신장독성에 대한 영지추출물 복합제제의 보호효과

        In this paper, the effect of Ganopoly(extracts of Ganoderma lucidum) and Ganopoly/C^+(70% Ganopoly + 30% chitosan) on cisplastin-induced nephrotoxicity was investigated in Sprague-Dawley rats. A single dose of dispiastin(5 ㎎/kg) was administered intraperitoneally after pretreatment of saline, Ganopoly and Ganopoly/C^+ for 7 days. The nephrotoxicity and renal function were manifestated by the changes of body weight, blood pressure, biochemical changes and solute in urine and plasma. After the treatment of CDDP(cis-dichlorodiamineplatinum), a significant elevation of kidney weight, serum urea, cretinine, urine volume for 24 hours, urine magnesium, and a severe or significant decrease in body weight, blood pressure, creatinine clearance, urine osmolarity, serum albumin, etc. The nephrotoxicity was further confirmed by a significant decrease in glutathione S-transferase(GSH) in urine and kidney homogenate, GSH, glutathione peroxidase(GSH-Px) and catalase in kidney tissue. And also the lipid peroxidation was significantly increased in kidney homogenate. These signs of nephrotoxicity was ameliorated by the pretreatment and consecutive administration of Ganopoly and Ganopoly/C^+ for 14 days after the i.p. injection of CDDP on 7th day after pretreatment of Ganopoly and Ganopoly/C^+. The amelioration of nephrotoxicity was evidenced by significant reduction in serum urea and creatinine concentration, and improvement of other index of renal function. And The activity of antioxidant enzymes were partially recovered in kidney tissue of rats treated by CDDP and the administration of Ganopoly and Ganopoly/C^+. These results indicate the cispastin induced nephrotoxicity is due to an impairment of tubular reabsorption systems enhanced by necrosis of proximal tubule, and the Ganopoly and Ganopoly/C^+ has a partial protective effect on nephrotoxicity induced by CDDP. The polysacchride of Ganoderma lucidum may improve the therapeutic index of nephrotoxicity induced by CDDP. However, it is needed to elucidate the mechanism for confirming the therapeutic effect.
