        Expression of DcHsp17.7, a Small Heat Shock Protein Gene in Carrot (Daucus carota L.)

        We examined the expression pattern of the small heat shock protein (sHSP) gene, DcHsp17.7, under various conditions. In non-stressed leaf tissue, DcHsp17.7 was absent or present at very low levels; however, exposure to an elevated temperature (40℃, 4 hours) led to an accumulation of this protein. Furthermore, after cessation of heat stress, the levels were sustained for two days, but diminished entirely thereafter. The persistence of DcHsp17.7 in post-heat-shocked leaves suggests that this sHSP may play a role during recovery after heat stress. Overnight heat stress (40℃, from 9 pm to 9 am) induced a higher level of DcHsp17.7, compared with that of plants exposed to heat stress during the day (40℃, from 9 am to 9 pm). These observations indicate that DcHsp17.7 gene expression is influenced by circadian rhythm. The levels of the DcHsp17.7 transcripts and their encoded protein increased after exposure to exogenous abscisic acid (ABA) and salicylic acid (SA), suggesting that these plant growth hormones may be involved in the expression of the DcHsp17.7 gene. Additionally, exposure to heavy metals, such as lead (Pb)and arsenic (As), led to increased levels of DcHsp17.7 transcripts and protein.

        국내 약학대학 학생들의 보건의료전문직간 교육에 대한 인식

        Background: Interprofessional education (IPE) is important for collaboration between professionals in the team-based practices of the healthcare field. Objectives: This study aimed to examine pharmacy students across in Korea on the experience of IPE and the perceptions of the importance of interprofessional collaboration (IPC) and the need of IPE. Methods: A cross-sectional survey using a 20 questionnaire to pharmacy students nationwide was conducted from March to May 2019. Results: A total of 555 students from 32 pharmacy schools participated. They recognized that the importance of close pharmacist-medical doctor collaboration was an average of 5.38 points (out of 6 points), but the current level of pharmacist-to-medical doctor collaboration was an average of 2.51 points (out of 6 points), and the reasons for the insufficient IPC were the lack of an environment encouraging mutual cooperation (79.5%) and the psychological distance to the other professional (35.3%). They perceived the necessity of IPE between pharmacistmedical doctor was 4.95 points (out of 6 points), with the curriculum including how to cooperate with medical doctors (78.2%), communication skills (51.0%), and understanding of medical doctors’ functions (44.5%), etc. Only 52 respondents (9.4%) had experience in IPE. Respondents who recognized the importance of IPC (≥5 points) showed 4.44-fold higher agreement on the need for IPE than those who did not (≤4 points) (OR 4.44, 95% CI 2.56, 7.68). Conclusions: Further attention and discussion to add IPE program in the pharmacy school curriculum is necessary in order to cultivate pharmacists with sufficient professional collaboration capabilities.

        Rad50 mediates DNA demethylation to establish pluripotent reprogramming

        DNA demethylation is characterized by the loss of methyl groups from 5-methylcytosine, and this activity is involved in various biological processes in mammalian cell development and differentiation. In particular, dynamic DNA demethylation in the process of somatic cell reprogramming is required for successful iPSC generation. In the present study, we reported the role of Rad50 in the DNA demethylation process during somatic cell reprogramming. We found that Rad50 was highly expressed in pluripotent stem cells and that Rad50 regulated global DNA demethylation levels. Importantly, the overexpression of Rad50 resulted in the enhanced efficiency of iPSC generation via increased DNA demethylation, whereas Rad50 knockdown led to DNA hypermethylation, which suppressed somatic cell reprogramming into iPSCs. Moreover, we found that Rad50 associated with Tet1 to facilitate the DNA demethylation process in pluripotent reprogramming. Therefore, our findings highlight the novel role of Rad50 in the DNA demethylation process during somatic cell reprogramming.

        블렌디드 러닝(Blended Learning)을 활용한 생태시민성 함양 환경교육 프로그램 개발

        이 연구는 초등학교 5∼6학년을 대상으로 비영역성, 성찰성, 형평성, 실천성을 갖춘 생태시민을 양성하기 위하여 블렌디드 러닝을 활용한 환경교육 프로그램을 개발하였다. 프로그램을 개발하기 위하여 2015 개정교육과정 초등학교 5∼6학년(군) 성취기준에서 생태시민성 함양 관련 환경교육 내용이 어떻게 반영되어 있는지를 확인하고, 시사점을 도출하였다. 이를 바탕으로 프로그램의 개발 방향, 학습 목표 및 주제를 선정하고, 선정된 주제를 바탕으로 블렌디드 러닝을 활용한 환경교육 프로그램 개발과 실행방안을 제시하였다. 이 연구를 통해 얻은 결론은 다음과 같다. 첫째, 생태시민성 함양 관련 환경교육 내용요소를 비영역성, 성찰성, 형평성, 실천성으로 추출하여 교육과정을 분석한 결과, 생태시민성 함양 관련 환경교육 내용이 사회, 도덕, 과학, 실과 순으로 반영되어 있었다. 또한 생태시민성함양 관련 환경교육 내용요소는 비영역성, 성찰성, 형평성, 실천성 순으로 높게 제시되어 있었다. 둘째, 프로그램 구성 체제는 문제인식→성찰→판단→실천과 같이 4단계로 수업활동을 적용하였으며, 교수·학습 모형은 Large Z형 블렌디드 유형을 따라 수업 차시의 특성과 목적에 알맞게 차시별로 오프라인 수업과 온라인 수업을 배치하고 블렌디드 러닝 교수·학습 전략을 반영하여 교수·학습 과정안을 개발하였다. 셋째, 개발된 프로그램이 창의적 체험활동 시간이나 관련 교과의 재구성을 통해 활용될 수 있도록 하기 위하여 교수·학습 실행방안을 제시하였다. 결론적으로 블렌디드 러닝을 활용한 생태시민성 함양 환경교육 프로그램은 환경문제에 대한 인식과 성찰, 판단, 실천을 통해 생태시민성을 함양시킬 수 있는 도움 자료가 될 것으로 보인다. This study aimed to develop an environmental education program using blended learning to foster ecological citizenship with nonterritoriality, reflexivity, equity, and practicality for elementary school 5th to 6th graders. How the contents of environmental education related to cultivating ecocitizenship were reflected in the 2015 revised curriculum standards for the 5th and 6th grades was confirmed, and implications were derived. The development direction, learning goals, and program topics were selected, and environmental education programs using blended learning were developed and implemented on ecological citizenship. The conclusions obtained from this study are as follows: First, based on an analysis of the environmental education curriculum, the contents related to cultivating ecocitizenship were ranked by society, morality, science, and practical arts. In addition, the elements related to cultivating ecocitizenship were presented in the order of nonterritoriality, reflection, equity, and practicality. Second, the program composition system applied class activities in four stages: problem recognition→introspection→judgment→practice. The teaching and learning model followed the Z-type blended model and developed the teaching and learning process plan by arranging offline and online classes based on the characteristics and purpose of each class and reflecting the blended learning strategy. Third, a teaching and learning implementation plan was proposed to ensure that the program can be used through a creative experience activity time or reorganization of related subjects.

        Platelet Shape Changes and Cytoskeleton Dynamics as Novel Therapeutic Targets for Anti-Thrombotic Drugs

        Platelets play an essential role in hemostasis through aggregation and adhesion to vascular injury sites but their unnecessary activation can often lead to thrombotic diseases. Upon exposure to physical or biochemical stimuli, remarkable platelet shape changes precede aggregation or adhesion. Platelets shape changes facilitate the formation and adhesion of platelet aggregates, but are readily reversible in contrast to the irrevocable characteristics of aggregation and adhesion. In this dynamic phenomenon, complex molecular signaling pathways and a host of diverse cytoskeleton proteins are involved. Platelet shape change is easily primed by diverse pro-thrombotic xenobiotics and stimuli, and its inhibition can modulate thrombosis, which can ultimately contribute to the development or prevention of thrombotic diseases. In this review, we discussed the current knowledge on the mechanisms of platelet shape change and also pathological implications and therapeutic opportunities for regulating the related cytoskeleton dynamics.

        심폐우회술 중 경식도 심장초음파를 통해 발견된 동맥관개존 - 증례 보고 -

        Transesophageal echocardiography is a useful device to evaluate the posterior structure of heart with an advantage of enabling clearer images, as compared to transthoracic echocardiography. With in- traoperative transesophageal echocardiography, we can reconfirm pre-diagnosed lesions, determine the success of the operation, and in particular, diagnose new lesions that are undetected in pre-operative evaluation. In the present case, undiagnosed patent ductus arteriosus was found on intraoperative transesophageal echocardiography during cardiopulmonary bypass. Subsequently, the patent ductus arteriosus was ligated successfully. With transesophageal echo- cardiography, we can diagnose the structural and functional abnormality of heart unidentified in the pre-operative evaluation. Also, transesophageal echocardiography can play the role of a rescuer to solve the problems that occur during cardiopulmonary bypass.

        Crystal Structure of XoLAP, a Leucine Aminopeptidase, from Xanthomonas oryzae pv. oryzae

        Aminopeptidases are metalloproteinases that degrade N-terminal residues from protein and play important roles in cell growth and development by controlling cell homeostasis and protein maturation. We determined the crystal structure of XoLAP, a leucyl aminopeptidase, at 2.6 Å resolution from Xanthomonas oryzae pv. oryzae, causing the destructive rice disease of bacterial blight. It is the first crystal structure of aminopeptidase from phytopathogens as a drug target. XoLAP existed as a hexamer and the monomer structure consisted of an N-terminal cap domain and a C-terminal peptidase domain with two divalent zinc ions. XoLAP structure was compared with BlLAP and EcLAP (EcPepA) structures. Based on the structural comparison, the molecular model of XoLAP in complex with the natural aminopeptidase inhibitor of microginin FR1 was proposed. The model structure will be useful to develop a novel antibacterial drug against Xoo.
