Oxidative Stress, Nrf2, and Epigenetic Modification Contribute to Anticancer Drug Resistance

Nuclear factor E2-related factor 2 (Nrf2), a transcription factor, controls the expression of genes encoding cytoprotective proteins, including antioxidant enzymes that combat oxidative and electrophilic stress to maintain redox homeostasis. However, recent studies demonstrated that, in cancer, aberrant activation of Nrf2 by epigenetic alterations promotes high expression of cytoprotective proteins, which can decrease the efficacy of anticancer drugs used for chemotherapy. In this review, we summarize recent findings regarding the relationship between oxidative stress, Nrf2, epigenetic modification, and anticancer drug resistance, which should aid in development of new strategies to improve chemotherapeutic efficacy.

Purpose: The objective of this study was to provide an understanding of the difficulties for facing parents of children with terminal cancer. Method: The design of this study was an inductive and descriptive study. Thirty‐one parents of children with terminal cancer participated in‐depth were interviewed s. C in depth and content analysis was used for analyzing the data. Results: The main categories in the findings were 'difficulties in coping', 'physical and psychological suffering of children', 'suffering of family', 'bereavement with children', 'economic problems', 'incurable situation', 'preparation of death', 'social isolation', 'coping after a child dies', 'spiritual problems', receiving active treatment', 'informing children of their condition', 'a sense of meaninglessness', 'emotional iunstability', and 'giving up hope'. Conclusion: The main point identified from this result in this study was that parents who have a child with terminal cancer don't never give up the hope of recovery for their child even when the child is in by the terminal stage of their children, even though and they are unwillingly to prepare for their child's death. This is a unique characteristic in the attitude of the families' attitude in child hospice care and differs from that found in adult hospice care. This result can be used as an important guide for nurses to in assessing the parents' needs in the terminal care setting.

Purpose: The purpose of this study was to develop a tool to assess the need for child hospice care in families of children with cancer. Method: The research design was a methodological study. The tool was developed in 4 stages : first, preliminary items were developed based on a questionnaire about the needs for child hospice care that was given to 20 families of children with cancer; second, a panel of specialists reduced the number of preliminary items using 3 validity tests for the content; third, final items were selected from the results of a pre-test. Finally, from February to July 2004, reliability and validity were tested with a sample of 104 families who had a child with cancer. Results: The final tool on the need for child hospice care consisted of 22 items and Cronbach's alpha coefficient for internal consistency was .93. Using factor analysis, 5 factors were extracted and these factors explained 69% of the total variance. Conclusion: The instrument, for assessing the need for child hospice care in families of children with cancer, developed in this study was identified as a tool with a high degree of reliability and validity. In this sense, this tool can be effectively utilized for implementing and improving hospice care for children with cancer.

Purpose: Preventing the effects of physical vulnerability is a practical approach to improving older adults’ depressive symptoms. This study aims to examine the relationship between gender differences related to muscle strength and depressive symptoms mediated by perceived stress. Methods: In this cross-sectional study, data from 2,705 older adults (65 years old or older) from the Korea National Health and Nutrition Examination Survey VII 2016 and 2018 were analyzed. The moderated mediation model was developed; the outcome, independent, mediation, and moderated mediation variables in the literature-based research model were depressive symptoms, muscle strength, perceived stress, and gender, respectively. Results: Perceived stress had a mediating effect on the relationship between muscle strength and depressive symptoms. The indirect effect of muscle strength on depressive symptoms mediated by perceived stress was β = -.02 (95% CI:-0.03~-0.01). The moderated mediation model demonstrated that the interaction term of handgrip strength and gender negatively affected perceived stress, which indicated that gender moderated the mediating model of perceived stress on the association of muscle strength and depressive symptoms (β = -.01, p <.05). The conditional indirect effect model was insignificant in the male group (β = -.00, 95% CI:-0.01~0.01) but significant in the female group (β = -.01, 95% CI: -0.02~0.00). Conclusions: Perceived stress mediated the relationship between muscle strength and depressive symptoms. However, the effect differed by gender. A stress-mediated depressive symptoms intervention program for older adults should be developed to consider women’s needs.

We screened the anti-oxidative effect on V79-4 hamster lung fibroblast cells induced by hydrogen peroxide with fifteen herbal extracts. Uncariae rhynchophylla JACKS and Rheum coreanum NAKAI were found to show DPPH radical scavenge activity (25 and 29% compared to control). Rheum coreanum NAKAI and Siegesbeckia orientalis L. were shown to scavenge intracellular reactive oxygen species (57 and 55% compared to control) which is measured by dichlorodihydrofluorescin diacetate method (DCHF-DA). Rheum coreanum NAKAI which showed the most strong intracellular reactive oxygen species scavenging activity had low DPPH radical scavenging activity compared to Uncariae rhynchophylla JACKS.

Purpose: The purpose of this study was to identify the relationships among self-esteem, meaning in life, and spiritual well-being in middle school students. Methods: The research design was a descriptive correlational survey involving 405 middle school students in grades 1 to 3 in S city, K city, and C city. Data were collected from March to May 2008 using questionnaires. Means with standard deviation, t-test, ANOVA, Duncan post-hoc test, and Pearson correlation coefficients were used to analyze the data. Results: According to general characteristics of the students, there were statistically significant differences in self-esteem for gender, grade, and region, in meaning in life, there were significant differences for grade, gender, and region, and in spiritual well-being, there were statistically significant differences for gender, religion, and region. There were significant correlations between self-esteem and meaning in life, between self-esteem and spiritual well-being, and between meaning in life and spiritual well-being. Conclusion: Self-esteem, meaning in life, and spiritual well-being should be considered as important factors for healthy development in the middle school period. These three variables should be considered as the main content for self-development programs for middle school students.

Purpose: To evaluate the effects of a logotherapy education program. Methods: A nonequivalent control group, non-synchronized design was conducted with a convenience sample of 29 children with cancer. The experimental group (n=17) participated in the logotherapy education program which consisted of 5 daily sessions for one week. The control group (n=12) received the usual nursing care. The effects were measured using suffering, adolescent meaning in life (AMIL), and spiritual well-being (SWBS) scales. Results: There were significant differences in suffering (W=153.00, p< .05) and meaning in life (W=78.00, p< .05) between the experimental and control groups. However, there was no significant difference in spiritual well-being (W=136.50, p> .05). Conclusions: Logotherapy was effective in reducing suffering and improving the meaning in life. Logotherapy can be utilized for adolescents with terminal cancer to prevent existential distress and improve their quality of life.

Reactive oxygen species (ROS) are known to cause oxidative modification of DNA, proteins, lipids and small cellular molecules and are associated with tissue damage and are the contributing factors for inflammation, aging, cancer, arteriosclerosis, hypertension and diabetes. We screened the anti-oxidants in V79-4 hamster lung fibroblast cells induced by hydrogen peroxide with eighteen pure compounds isolated from natural products. Allantoin, brassicasterol, and hypaconitine were found to strongly scavenge intracellular reactive oxygen species, which is measured by dichlorodihydrofluorescin diacetate method (DCHF-DA), and 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical.
