8-Hydroxydeoxyguanosine (oh8dG) treatment induced senescence-like changes in KG-1 cells, a human acute myelocytic leukemia cell line. The oh8dG-treated cells stained positive for senescence associated β-galactosidase (SA-β-galactosidase) and had enlarged cell shape, both of which are senescence indexes. The oh8dG-treated cells were also cell growth inhibited and arrested at G1 in the cell cycle. The accumulation of cdk (cyclin dependent kinase) inhibitors, such as p16, p21, and p27, also implies that cellular senescence was induced in oh8dG-treated cells. However, these changes were not accompanied by cell differentiation or telomerase activity. Taken together, we conclude that oh8dG treatment of KG-1 cells induces cellular senescence.

Telomerase activation is detected in most cancerous cells; hence, telomerase is a highly selective target for cancer therapy, which plays an important role in the apoptotic process. We have previously reported that the ginseng saponin metabolite, Compound K (20-O-D-gluco-pyranosyl-20(S)-protopanaxadiol, IH901), inhibits cell proliferation by inducing apoptosis and cell cycle arrest at the G1 phase. The present study investigated the regulation of telomerase activity in Compound K treated U937 cells. Compound K treatment caused a reduction in telomerase activity and down-regulated the human telomerase reverse transcriptase (hTERT) gene, resulting in the decreased expressions of its protein, and of the c-Myc and Sp1 proteins (transcription factors of hTERT). These results indicate that the anticancer activity of Compound K could be mediated by inhibition of the telomerase activity.

Antineoplastic activity against human gastric and colon carcinoma cell lines was measured in 100 extracts from 90 plants using MTT (3-[4,5-dimethyl thiazo-2-yl]-2, 5-diphenyl tetrazolium bromide) method. Four extracts from four plants have been reported to have antineoplastic effect. Extracts from remaining 86 plants failed to show significant cytotoxic effect at the concentration of less than $230\;{\mu}/ml$.

Homogentisic acid was found to scavenge intracellular reactive oxygen species (ROS), and 1,1-diphenyl-2-picrylhydrazyl (DPPH) radicals and thus prevented lipid peroxidation in human fibroblast (WI 38) cells. The radical scavenging activity of homogentisic acid was found to protect WI 38 cells against hydrogen peroxide (H2O2) induced oxidative stress, via the activation of extracellular signal regulated kinase (ERK) protein. Homogentisic acid increased the activity of catalase. Hence, from the present study, it is suggested that homogentisic acid protects WI 38 cells against H2O2 damage by enhancing the intracellular antioxidative activity.

Antineoplastic activity against human gastric and colon carcinoma cell lines was tested in eighty-three species of Korean plants including Korean medicinal plants which have been frequently used in oriental herb prescriptions. The plant materials were extracted with methanol and the cytotoxic activity was tested using a calorimetric tetrazolium assay (MTT assay). Twenty-six plant extracts against gastric carcinoma cell line, eighteen extracts against colon carcinoma cell line and fourteen plant extracts against both carcinoma cell lines showed antineoplastic activity at the concentration of less than $100{\mu}g/ml$. The effective components from four species have been isolated and reported.

Rapid technology scale-down results in significant performance improvement on digital processing speed associated with enhanced power efficiency due to reduced supply voltage. However, relative circuit noise power becomes enlarged. Thus, low noise clock generation circuits are highly required. As a key factor of evaluating the clocking system, analyzing jitter contribution becomes essential. However, the more technology scaling down has been progressed, the more jitter analysis has been complicated. In this paper, we implemented 4 methods to enhance jitter performance of delay-locked-loop (DLL) circuit to support systematic jitter analysis. After applying our proposed DLL design optimization technique targeting for 1GHz clock generation under 1.2V supply voltage, we obtained 63.7% jitter noise reduction in 65nm standard CMOS technology.
