        N-Acetyl-L-Cysteine에 의한 생쥐 골수유래 가지세포의 기능적 활성화 저해

        N-acetyl-L-cysteine (NAC)은 thiol기를 포함하는 화합물로서, glutathione (GSH)의 전구체로 작용하여 포유류 세포 내에서 항산화제로 작용한다. 또한 항염기능이 있으며 호산구나 B세포, 가지세포 (dendritic cell, DC)와 같은 면역세포 들에 여러 가지 영향을 미치는 것으로 알려져 있다. 특히 가지세포에 작용하여 활성화를 억제하거나 가지세포에 의한 Th2 반응 유도에 관여한다고 알려져 있다. 그러나 이들 연구는 세부적인 사항에 있어서 그 결과가 서로 상치하는바가 많으며, 또한 조절T세포의 관점에서는 연구된 바가 없다. 따라서 본 연구에서는 NAC 처리가 가지세포 활성화에 미치는 영향을 재확인하였고, NAC 처리된 가지세포의 T세포 활성 능력 저하, 또는 Th2 반응 유도 여부를 알아보았다. 활성화 시 가지세포에서 증가하는 활성산소기 (reactive oxygen species)는 NAC 처리로 낮아져서, NAC이 가지세포에 항산화작용을 나타냄을 확인하였다. NAC 처리로 가지세포에서 보조자극인자인 CD40과 CD86의 발현이 저해되었으며, 활성화 시 정상적으로 낮아지는 포식기능은 처리된 NAC의 농도에 비례하게 보존되었다. 활성화 시 분비되는 IL-6, IL-10, IL-12는 모두 감소하였다. 이러한 NAC-DC와 함께 배양한 T세포의 증식이나 Th1 cytokine인 IFN-γ, Th2 cytokine인 IL-5의 분비가 모두 저하되어 Th1/Th2의 편중 없이 가지세포의 T세포 자극능력이 전반적으로 감소하였음을 나타내었다. 또한 T세포 배양액에서 IL-10과 TGF-β의 농도 역시 NAC-DC로 자극된 경우에 현저히 줄어서, NAC-DC에 의한 T세포 증식 감소 등은 조절T세포 유도에 의한 것이 아니라 T세포 무반응이 유도된 때문임을 나타내 주었다. N-acetyl-L-cysteine (NAC) is a thiol-containing compound and acts as a precursor for glutathione (GSH). It behaves as an antioxidant in mammalian cells and also exerts anti-inflammatory effects. NAC is also known to affect several immune cells including eosinophils, B cells, T cells, and dendritic cells (DC) in many aspects. Even though it has been reported that NAC inhibits DC activation and shifts the immune response to Th2, these studies exhibit some contradictory results in detail and do not give any information with respect to the induction of regulatory T cells. In this study, we re-analyzed the effects of NAC on DC during their activation. We also evaluated whether it induced T cell anergy, Th1/Th2 shift, or regulatory T cells. NAC suppressed the elevation of intracellular reactive oxygen species during DC activation. In parallel, it down-regulated surface expression of CD40 and CD86, suppressed the decrease of phagocytic function, lowered the secretion of cytokines such as IL-6, IL-10, and IL-12. All these effects showed dose-dependency. Thus, it seems likely that NAC inhibited DC activation with regard to their phenotype and cytokine secretion. When we evaluated the T cell-stimulating capacity of these NAC-DC, T cell proliferation and secretion of both Th1 (IFN-γ) and Th2 cytokine (IL-5) were decreased. This implies that the T cell-stimulating activity of NAC-DC decreased without any shift to Th1 or Th2 cytokine (IL-5). The secretion of IL-10 and TGF-β in the supernatants were also decreased, which suggests that the decrease of T cell proliferation and cytokine secretion is due to the induction of T cell anergy, rather than regulatory T cells.

        Effect of Feed Selenium-lysine Supplementation on Milk Compositions and Serum Biochemical Indices in Saanen Dairy Goats

        An experiment was carried out to assess the effect of feed selenium-lysine (Se-Lys) supplementation on milk compositions and serum biochemical indices in Saanen dairy goats in Korea. A total of twelve 36 months old Saanen lactating dairy goats (47±6.21 kg) fed the similar dry matter intake twice a day at 2% of BW (DMI) (10.9% moisture of concentrate and 19% moisture of roughage), milk yield (2.5 kg/d) and parity (2) were randomly selected and subjected for the present study, divided into two groups with six goats in each group. The goats in the control group received rice hulls (10 g/ day) only, and did not receive Se-Lys; goats in the treatment group were fed 0.06 g of Se-Lys with 10 g of rice hulls every day before feeding roughage for six weeks. The milk sample was collected every week, and its compositions were analyzed. The results of the present study showed that there is no significantly increased milk production in Se-Lys treated group goats when compared with control group goats. But, Se-Lys treatment significantly increased the milk protein content (3.98±0.16%), fat (3.72±0.27%), lactose (4.07±0.14%), total solids (12.51±0.28%) and urea (14.42±1.45 mg/dl) content as compared to the control group goats (p<0.05). The somatic cell counts (207,740±28.81 cells/ml) were significantly lower in the Se-Lys treated group than in the control group (p<0.05). Also, the results of the current study showed that supplementation of Se-Lys were significantly decreased the blood biochemical indices of IL-6 (34.34±6.04 pg/ml), TNT-α (0.56±0.22 ng/ml), MDA (5.07±1.03 ng/ml), GPx-1 (9.07±5.17 ng/ml), sCD4 (2.64±1.02 ng/ml) and sCD8 (5.08±2.08 ng/ml) level when compared with without addition of Se-Lys group dairy goats (p<0.05). On the other hand, the selenoprotein P (1,580.18±127.62 ng/ml) level was significantly higher in Se-Lys supplemented group than in the control group (p<0.05). Based on the study results, it was concluded that feed Se-Lys supplementation may improve milk yield with positively improved protein, fat, lactose, total solids, urea content, and biochemical indices without negative effects on milk production traits.

        Potential of Lilium lancifolium in Alleviating Pain and Inflammation in a Rat Model of Monosodium Iodoacetate-Induced Osteoarthritis

        Lilium lancifolium (LL) is widely cultivated in East Asia and used to attenuate airway diseases. Our current study aimed to investigate the therapeutic effect of LL on pain level and inflammatory response in a rat model of monosodium iodoacetate (MIA)-induced osteoarthritis (OA). We first examined the effect of LL on inflammatory cytokines and inflammatory mediators in IL-1β-treated HTB-94 cells. The LL extract was found to significantly inhibit the levels of Interleukin-6 (IL-6), Interleukin-8 (IL-8), and prostaglandin-E2 (PGE-2) in Interleukin-1 β (IL-1β)-stimulated HTB-94 cells in a dose-dependent manner. Moreover, chronic oral administration of LL effectively restored the weight-bearing distribution in the rat model of MIA-induced OA. In addition, administration of LL inhibited inflammatory cytokines and inflammatory mediators, such as C-reactive protein (CRP), IL-6, leukotriene B4 (LTB-4), PGE-2, and matrix metalloproteinase-9 (MMP-9). Our findings collectively suggested LL as one of the potential therapeutic agents for OA, owing to its properties of reducing pain and inflammatory responses.

      • Thymol attenuates the worsening of atopic dermatitis induced by <i>Staphylococcus aureus</i> membrane vesicles

        <P><B>Abstract</B></P> <P> <I>Staphylococcus aureus</I> membrane vesicles (MVs) aggravate atopic dermatitis (AD) through the delivery of bacterial effector molecules to host cells and the stimulation of inflammatory responses. This study investigated the inhibitory effect of thymol, a phenolic monoterpene found in essential oils derived from plants, on the worsening of AD induced by <I>S. aureus</I> MVs both <I>in vitro</I> and <I>in vivo</I>. The sub-minimal inhibitory concentrations of thymol disrupted <I>S. aureus</I> MVs. Intact <I>S. aureus</I> MVs induced the expression of pro-inflammatory cytokine (interleukin (IL)-1β, IL-6, and tumor necrosis factor-α) and chemokine (IL-8 and monocyte chemoattractant protein-1) genes in cultured keratinocytes, whereas thymol-treated <I>S. aureus</I> MVs did not stimulate the expression of these genes. Topical application of thymol-treated <I>S. aureus</I> MVs or treatment with thymol after intact <I>S. aureus</I> MVs to AD-like skin lesions diminished the pathology of AD. This included decreases in epidermal/dermal thickness and infiltration of eosinophils/mast cells, and inhibited expression of pro-inflammatory cytokine and chemokine genes in mouse AD model. Moreover, thymol significantly suppressed the Th1, Th2, and Th17-mediated inflammatory responses in AD-like skin lesions induced by <I>S. aureus</I> MVs, and reduced the serum levels of immunoglobulin (Ig) G2a, mite-specific IgE, and total IgE. In summary, thymol disrupts <I>S. aureus</I> MVs and suppresses inflammatory responses in AD-like skin lesions aggravated by <I>S. aureus</I> MVs.         LPS로 유도한 대식세포의 염증반응에서 우슬의 항염증 효과

        Objectives: Achyranthes japonica (AJ ) has been used as an anti-bacterial and anti-inflammatory agent. However, it is unclear that AJ could show the anti-inflammatory effects in macrophages. In this experiment, we studied whether AJ could inhibit the inflammatory responses in macrophages. Methods: To measure out the cytotoxicity of AJ, we performed the MTT assay. We evaluated the nitric oxide (NO) production, and cytokine production such as interleukin (IL)-1b, IL-6 and tumor necrosis factor (TNF)-a. We also investigated the cellular mechanims such as mitogen activated protein kinases (MAPK)s and nuclear factor kappa B (NF-kB). Results: AJ inhibited lipopolysaccharide (LPS)-induced NO production. AJ also inhibited production levels of IL-1b, IL-6 and TNF-a in LPS-stimulated macrophage. Finally, western blot analysis showed that AJ treatment inhibited the activation of p38 but not of extracellular signal-regulated kinase, c-jun NH2-terminal kinase and NF-kB. Conclusions: These results showed that AJ down-regulated the inflammatory response via p38 in macrophages, which suggest that AJ could be a candidate on treating inflammatory diseases.

        The Lipopolysaccharide from Porphyromonas gingivalis Induces Vascular Permeability

        Porphyromonas gingivalis, one of the major periodontal pathogens, is implicated in the initiation and progression of periodontal disease. The initial stages of periodontal inflammation are accompanied by vascular hyperpermeability. In our present study, we report that the P. gingivalis lipopolysaccharide (LPS) increases the mRNA expression of interleukin-8 (IL-8), a major inducer of vascular permeability, in vascular endothelial cells. P. gingivalis LPS also stimulated the induction of IL-8 secretion in endothelial cells. The P. gingivalis LPS-induced expression of IL-8 was primarily modulated by nuclear factor-κB (NF-κB). P. gingivalis LPS significantly enhanced the vascular permeability both in vitro and in vivo, and a blockade of the IL-8 receptor decreased the P. gingivalis LPS-induced vascular permeability. Taken together, these results suggest that P. gingivalis LPS increases vascular permeability through the NF-κBdependent production of IL-8 in vascular endothelial cells.

      • 공수도 교육의 가치에 관한 연구

        본 연구의 목적은 공수도 철학과 자도자의 신념이나 내용을 다루는 철학적 과정을 고찰함으로서 공수도 교육에 대한 근본적인 의미나 과제를 명확히 파악할 뿐만 아니라 공수도의 교육방향을 정립하였다. 첫째, 공수도의 교육학적 가치이다. 자기교욱에 중점을 둔 교육적 효과 및 가치를 가지며, 지속적이고 반복적인 학습으로만 가능하며 자신의 용기에 의한 자발적인 학습이 필요하기 때문에 학습의 가치를 가진다. 둘째, 공수도의 심리학적 가치이다. 용기, 예의, 겸손 그리고 절제라는 특징들을 수양하는 것이다. 셋째, 공수도의 사회학적 가치이다. 공수도 수련과정 속에서 만날 수 있는 사범, 선후배, 동료 간의 밀접한 인간관계는 개인의 사회성의 향상에 기여하게 되고, 집단의 목적을 원활히 하는 친화성을 강화시킨다. 넷째, 공수도의 운동학적 가치이다. 평형성을 비롯한 근력, 근지구력, 민첩성, 순발력, 유연성 등의 제반 기능을 배양하고 신체의 조직력과 균형있는 신체발달을 유도한다. 이러한 공수도 교육을 통하여 사회적으로 요구하는 조화로운 인간육성이 가능하며, 신체적으로도 균형있는 발육발달뿐만 아니라 체력유지 및 노화현상을 지연시킴으로서 신체의 기능유지가 가능하다. 이처럼 우리 삶에 있어서 공수도는 교육적 의미를 가지고 그 역할과 기능을 수행하고 있으며 우리를 인간다운 인간으로 만드는데 이의 교육이 필요로 할 것이다.

        국가직업능력표준 실용화를 위한 제도화 방안

        이 연구의 목적은 일-교육훈련-자격이 연계될 수 있도록 노동부의 국가직업능력표준을 국가차원에서 개발·운영·관리될 수 있는 제도화 방안을 마련하는데 있었다. 연구목적 달성을 위해 문헌고찰 및 내용분석, 포커스그룹인터뷰를 실시하였고, 연구결과의 타당성 확보를 위해 전문가협의회 및 공청회를 실시하였다. 국가직업능력표준 제도화는 표준의 실용화를 제고할 수 있도록 관련용어 정의, 총괄기구, 표준개발 및 승인, 표준 활용에 관한 인프라 구축에 필요한 사항이 규정되도록 관계 법령을 정비하는 것이다. 표준 제도화를 위해 다음과 같은 사항이 관계 법령에 명시되도록 해야 한다. ① 표준 제도화를 위해 직업, 직업능력, 직무, 표준 등을 관련법에 정의해야 한다. ② 국가직업능력표준 관련 부처 및 다양한 이해당사자의 참여를 기반으로 한 국가직업능력표준 사업 전담 조직을 설치하고, 표준사업에 대한 자문과 심의를 담당할 수 있는 위원회를 구축한다. ③ 다양한 산업분야 단체가 표준개발의 주체가 될 수 있도록 지원하고, 표준의 수준 설정을 통해 직종 내 수직적 경력개발을 촉진하며, 개발된 표준에 대한 지속적인 관리와 현장적합성을 승인하는 체제를 구축해야 한다. ④ 국가직업능력표준은 산업현장에서는 인력을 채용하고 관리하는 기준으로, 교육훈련에서는 교육과정을 개발하고 교과목 및 교재를 개발하며 현 교육과정을 평가하는 기준으로, 자격체계와는 표준의 자격화, 새로운 자격설계, 자격의 출제기준으로 활용될 수 있도록 지원해야 한다. 이를 위해 국가기술자격법, 근로자직업능력개발법, 산업인력공단법 등 관계 법령을 개정하거나 국가직업능력표준에 관한 특별법을 제정할 필요가 있다. 한편, 국가직업능력표준 제도화 단계에 따른 구체적인 추진과제와 이를 실현하기 위한 정책적 제언을 제시하였다. The purpose of this study was to develop the institutionalization that NOS(National Occupational Standard) was developed, operated and managed for linking with work, training and qualification. To achieve the purpose, literature reviews, contents analysis and focus group interview were conducted, and experts council and a public hearing were executed. The institutionalization is the legislation consolidation to regulate details related infra construction as follows for improving utilization of NOS. ① It should be defined occupation, occupational competency, job, standards and so forth on the related laws. ② A organization which take exclusive charge with and a council which consult and deliberate on NOS projects should be established through the cooperation between stakeholder and government organization. ③ It should be supported that various industrial parties including SHRDC are the main body for development of NOS, promoted vertical career development by occupation through establishing level of NOS, and established system that manage continually and approve practical application of NOS. ④ It should be supported that NOS is used as a criterion for recruiting and managing workforce, a criterion for developing and evaluating curriculum and educational materials in training, having qualification for itself, designing for new qualification, and a criterion for setting questions for a qualifying examination. To improve utilization of NOS, It need to be consolidated the related laws including National Technical Qualifications Act and Workers' Vocational Competency Development Act or to be enacted a special law on NOS. Moreover, this study suggested action plan according to the stages of institutionalization, and political proposal for effective actualization of action plan.
