      • Lentivirus-mediated shRNA Interference Targeting SLUG Inhibits Lung Cancer Growth and Metastasis

        Objective: Lung cancer is a deadly cancer, whose kills more people worldwide than any other malignancy. SLUG (SNAI2, Snail2) is involved in the epithelial mesenchymal transition in physiological and in pathological contexts and is implicated in the development and progression of lung cancer. Methods: We constructed a lentivirus vector with SLUG shRNA (LV-shSLUG). LV-shSLUG and a control lentivirus were infected into the non-small cell lung cancer cell A549 and real-time PCR, Western blot and IHC were applied to assess expression of the SLUG gene. Cell proliferation and migration were detected using MTT and clony formation methods. Results: Real-time PCR, Western Blot and IHC results confirmed down-regulation of SLUG expression by its shRNA by about 80%~90% at both the mRNA and protein levels. Knockdown of SLUG significantly suppressed lung cancer cell proliferation. Furthermore, knockdown of SLUG significantly inhibited lung cancer cell invasion and metastasis. Finally, knockdown of SLUG induced the down-regulation of Bcl-2 and up-regulation of E-cadherin. Conclusion: These results indicate that SLUG is a newly identified gene associated with lung cancer growth and metastasis. SLUG may serve as a new therapeutic target for the treatment of lung cancer in the future.

        A 2 MW-3.7 GHz lower hybrid current drive (LHCD) system is under development for physicsexperiments on the HL-2A device. The RF Power is generated by four TH2103A klystron amplifiersand propagates in the TE10 mode through WR284 waveguides. The transmission lines with a lengthof 20 m to 30 m are pressurized with 2 bars of nitrogen to decrease the possibility of arcing. Thelauncher, based on the passive-active multi-junction (PAM) concept, has been developed and iscurrently being realized. It was designed for a power spectrum peaked at Nk = 2.75 with goodcoupling properties over a wide range of plasma parameters. The four klystrons are fed by ahigh-voltage power supply (HVPS) based on the pulse step modulation (PSM) concept with a fastswitch-off time of less than ten μs. This system is expected to be in operation within 1 years and willexplore many international thermonuclear experimental reactor (ITER) related LH experiments inthe following years.

        The Gender-Sensitive Social Risk Factors for Internet Addiction in College Undergraduate Students

        Objective The current study aims to explore precipitating and social risk factors for internet addiction (IA) in university undergraduate students, and to provide evidence for interventions and the early prevention of IA in different genders. Methods Four thousand eight hundred and fifty-eight college sophomores completed an online survey on their internet use-related behaviours and social risk factors. Results We found that more male (8.3%) than female students (5.4%) had moderate and severe IA. The main online activity in the moderate and severe IA groups was online gaming in males and online streaming in females. Roommates engaging in similar internetbased entertainment was a risk factor of IA only for males, while not being in a romantic relationship was a risk factor of IA for females only. Infatuation with the internet before college and adjustment problems for college life were shared risk factors for both genders in the mild and moderate IA groups. Conclusion IA was a common phenomenon in college students with shared and unique precipitating and social risk factors in males and females. The gender-sensitive risk factors for IA warranted earlier and individualized intervention and prevention strategies for IA in this population.

        Cloning and Characterization of Filamentous Fungal S-Nitrosoglutathione Reductase from Aspergillus nidulans

        S-Nitrosoglutathione reductase (GSNOR) metabolizes S-nitrosoglutathione (GSNO) and has been shown to play important roles in regulating cellular signaling and formulating host defense by modulating intracellular nitric oxide levels. The enzyme has been found in bacterial, yeast, mushroom, plant, and mammalian cells. However, to date, there is still no evidence of its occurrence in filamentous fungi. In this study, we cloned and investigated a GSNOR-like enzyme from the filamentous fungus Aspergillus nidulans. The enzyme occurred in native form as a homodimer and exhibited low thermal stability. GSNO was an ideal substrate for the enzyme. The apparent Km and kcat values were 0.55 mM and 34,100 min-1, respectively. Substrate binding sites and catalytic center amino acid residues based on those from known GSNORs were conserved in this enzyme, and the corresponding roles were verified using site-directed mutagenesis. Therefore, we demonstrated the presence of GSNOR in a filamentous fungus for the first time.

        Hsa-let-7c controls the committed differentiation of IGF-1-treated mesenchymal stem cells derived from dental pulps by targeting IGF-1R via the MAPK pathways

        The putative tumor suppressor microRNA let-7c is extensively associated with the biological properties of cancer cells. However, the potential involvement of let-7c in the differentiation of mesenchymal stem cells has not been fully explored. In this study, we investigated the influence of hsa-let-7c (let-7c) on the proliferation and differentiation of human dental pulp-derived mesenchymal stem cells (DPMSCs) treated with insulin-like growth factor 1 (IGF-1) via flow cytometry, CCK-8 assays, alizarin red staining, real-time RT-PCR, and western blotting. In general, the proliferative capabilities and cell viability of DPMSCs were not significantly affected by the overexpression or deletion of let-7c. However, overexpression of let-7c significantly inhibited the expression of IGF-1 receptor (IGF-1R) and downregulated the osteo/odontogenic differentiation of DPMSCs, as indicated by decreased levels of several osteo/odontogenic markers (osteocalcin, osterix, runt-related transcription factor 2, dentin sialophosphoprotein, dentin sialoprotein, alkaline phosphatase, type 1 collagen, and dentin matrix protein 1) in IGF-1-treated DPMSCs. Inversely, deletion of let- 7c resulted in increased IGF-1R levels and enhanced osteo/odontogenic differentiation. Furthermore, the ERK, JNK, and P38 MAPK pathways were significantly inhibited following the overexpression of let-7c in DPMSCs. Deletion of let-7c promoted the activation of the JNK and P38 MAPK pathways. Our cumulative findings indicate that Let-7c can inhibit the osteo/odontogenic differentiation of IGF-1-treated DPMSCs by targeting IGF-1R via the JNK/P38 MAPK signaling pathways.

        Exponential Stabilization of Delayed Complex-valued Neural Networks with Aperiodic Sampling: A Free-matrix-based Time-dependent Lyapunov Functional Method

        In this paper, the exponential stabilization of delayed complex-valued neural networks (DCNNs) is addressed via sampled-data control. First, aperiodic sampled-data control aimed at further reducing the frequency of data transmission is adopted, which covers the periodic sampling as a special case. Then, a free-matrix-based timedependent Lyapunov functional is specially constructed for stability analysis of closed-loop DCNNs, in which two extra free matrices are introduced and the available information of system states at the sampling instants are fullyutilized. Accordingly, some less conservative stability conditions are established. By resorting to a matrix transformation, the design scheme for the feedback gains can be obtained. Meanwhile, the qualitative relationship between the decay rate and the upper bound of the variable sampling period is established and the maximum allowable value of the variable sampling period is determined. Finally, an illustrative example is provided to demonstrate the feasibility of the proposed stabilization criteria.

        As a new nanomaterial, HgWO4 nanofilms are prepared by the self-inventive technique of collodion nanoparticles-dispersal and film-formation. Their photoluminescent properties are enhanced and then controlled through nano-TiO2 doping. These approaches not only ensure film uniformity and fluorescence (FL) enhancement, but also realize a FL increase/decrease conversion effect. The relationships between HgWO4 nanofilms' PL intensities and TiO2 doped amounts are in good agreement with Gaussion function, and maximum PL intensities of HgWO4 nanofilms are located at 3.8% ratio of TiO2 nanoparticles.

        Analysis of delay compensation in real-time dynamic hybrid testing with large integration time-step

        With the sub-stepping technique, the numerical analysis in real-time dynamic hybrid testing is split into the response analysis and signal generation tasks. Two target computers that operate in real-time may be assigned to implement these two tasks, respectively, for fully extending the simulation scale of the numerical substructure. In this case, the integration time-step of solving the dynamic response of the numerical substructure can be dozens of times bigger than the sampling time-step of the controller. The time delay between the real and desired feedback forces becomes more striking, which challenges the well-developed delay compensation methods in real-time dynamic hybrid testing. This paper focuses on displacement prediction and force correction for delay compensation in the real-time dynamic hybrid testing with a large integration time-step. A new displacement prediction scheme is proposed based on recently-developed explicit integration algorithms and compared with several commonly-used prediction procedures. The evaluation of its prediction accuracy is carried out theoretically, numerically and experimentally. Results indicate that the accuracy and effectiveness of the proposed prediction method are of significance.

      • Convolutional neural network-based data anomaly detection considering class imbalance with limited data

        The raw data collected by structural health monitoring (SHM) systems may suffer multiple patterns of anomalies, which pose a significant barrier for an automatic and accurate structural condition assessment. Therefore, the detection and classification of these anomalies is an essential pre-processing step for SHM systems. However, the heterogeneous data patterns scarce anomalous samples and severe class imbalance make data anomaly detection difficult. In this regard, this study proposes a convolutional neural network-based data anomaly detection method. The time and frequency domains data are transferred as images and used as the input of the neural network for training. ResNet18 is adopted as the feature extractor to avoid training with massive labelled data. In addition, the focal loss function is adopted to soften the class imbalance-induced classification bias. The effectiveness of the proposed method is validated using acceleration data collected in a long-span cable-stayed bridge. The proposed approach detects and classifies data anomalies with high accuracy.

        Important Role of Immunological Responses to Environmental Exposure in the Development of Allergic Asthma

        Allergic asthma is a public health problem that affects human health and socioeconomic development. Studies have found that the prevalence of asthma has significantly increased in recent years, which has become particularly pronounced in developed countries. With rapid urbanization in China in the last 3 decades, the prevalence of asthma has increased significantly in urban areas. As changes in genetic backgrounds of human populations are limited, environmental exposure may be a major factor that is responsible for the increased prevalence of asthma. This review focuses on environmental components of farms and rural areas that may have protective effects in reducing the development of asthma. Farm and rural related microorganism- and pathogen-associated molecular patterns are considered to be important environmental factors that modulate host's innate and adaptive immune system to induce protection effects later in life. Environmental microbial-related immunotherapy will also be discussed as the future research direction for the prevention of allergic asthma.
