        Synthesis and Solution Properties of Zwitterionic Copolymer of Acrylamide with 3-[(2-Acrylamido)dimethylammonio]propanesulfonate

        A novel zwitterionic monomer 3-[(2-acrylamido)dimethylammonio]propanesulfonate (DMADAS) was designed and synthesized in this study. Then it was polymerized with acrylamide (AM) by free radical polymerization in 0.5 mol/L NaCl solution with ammonium persulfate ((NH4)2S2O8) and sodium sulfate (NaHSO3) as initiator. The structure and composition of DMADAS and acrylamide-3-[(2-acrylamido)- dimethylammonio]propanesulfonate copolymer (P-AM-DMADAS) were characterized by FT-IR spectroscopy, 1H NMR and elemental analyses. Isoelectric point (IEP) of P-AM-DMADAS was tested by nanoparticle size and potential analyzer. Solution properties of copolymer were studied by reduced viscosity. Antipolyelectrolyte behavior was observed and was found to be enhanced with increasing DMADAS content in copolymer. The results showed that the viscosity of P-AM-DMADAS is 5.472 dl/g in pure water. Electrolyte was added, which weakened the mutual attraction between sulfonic acid group and quaternary ammonium group. The conformation became loose, which led to the increase of reduced viscosity. The ability of monovalent and divalent cation influencing the viscosity of zwitterionic copolymer obeyed the following sequence: Li+ < Na+ < K+, Mg2+ < Ca2+ < Ba2+, and that of anion is in the order: Cl− < Br− < I−, CO3 2− > SO3 2− ≈ SO4 2−.

      • Crocetin Induces Cytotoxicity in Colon Cancer Cells Via p53-independent Mechanisms

        Objective: Crocin has been proposed as a promising candidate for cancer chemoprevention. The purpose of this investigation was to investigate the chemopreventive action and the possible mechanisms of crocin against human colon cancer cells in vitro. Methods: Cell proliferation was examined using MTT assay and the cell cycle distribution fractions were analyzed using fow cytometric analysis after propidium iodide staining. Apoptosis was detected using theTUNEL Apoptosis Detection Kit with laser scanning confocal microscope. DNA damage was assessed using the alkaline single-cell gel electrophoresis assay, while expression levels of p53, cdk2, cyclinA and P21 were examined by Western blot analysis. Results: Treatment of SW480 cells with crocetin (0.2, 0.4, 0.8 mmol/L) for 48 h signifcantly inhibited their proliferation in a concentration-dependent manner. Crocetin (0.8 mmol/L) signifcantly induced cell cycle arrest through p53-independent mechanisms accompanied by P21 induction. Crocetin (0.8 mmol/L) caused cytotoxicity in the SW480 cells by enhancing apoptosis and decreasing DNA repair capacity in a time-dependent manner. Conclusions: This report provides evidence that crocetin is a potential anticancer agent, which may be used as a chemotherapeutic drug.

      • Efficacy of Prophylactic Entecavir for Hepatitis B Virus-Related Hepatocellular Carcinoma Receiving Transcatheter Arterial Chemoembolization

        Background and Aims: Hepatitis B virus (HBV) reactivation was reported to be induced by transcatheter arterial chemoembolization (TACE) in HBV-related hepatocellular carcinonma (HCC) patients with a high incidence. The effective strategy to reduce hepatitis flares due to HBV reactivation in this specific group of patients was limited to lamivudine. This retrospective study was aimed to investigate the efficacy of prophylactic entecavir in HCC patients receiving TACE. Methods: A consecutive series of 191 HBV-related HCC patients receiving TACE were analyzed including 44 patients received prophylactic entecavir. Virologic events, defined as an increase in serum HBV DNA level to more than 1 log10 copies/ml higher than nadir the level, and hepatitis flares due to HBV reactivation were the main endpoints. Results: Patients with or without prophylactic were similar in host factors and the majorities of characteristics regarding to tumor factors, HBV status, liver function and LMR. Notably, cycles of TACE were parallel between the groups. Ten (22.7%) patients receiving prophylactic entecavir reached virologic response. The patients receiving prophylactic entecavir presented significantly reduced virologic events (6.8% vs 54.4%, p=0.000) and hepatitis flares due to HBV reactivation (0.0% vs 11.6%, p=0.039) compared with patients without prophylaxis. Kaplan-Meier analysis illustrated that the patients in the entecavir group presented significantly improved virologic events free survival (p=0.000) and hepatitis flare free survival (p=0.017). Female and Eastern Cooperative Oncology Group (ECOG) performance status 2 was the only significant predictors for virological events in patients without prophylactic antiviral. Rescue antiviral therapy did not reduce the incidence of hepatitis flares due to HBV reactivation. Conclusion: Prophylactic entecavir presented promising efficacy in HBV-related cancer patients receiving TACE. Lower performance status and female gender might be the predictors for HBV reactivation in these patients.

        Ozone infusiblization and curing mechanism of polysilazane ceramic precursor fibers

        An appropriate infusible method plays a vital role as one of the key processes of the fabrication of polymer-derived ceramic fibers. In this work, ozone curing, a novel room temperature controllable curing strategy was used to prepare SiCN ceramic fibers. Some circular pores were observed on the surface of the obtained SiCN ceramic fibers. The tensile strength of the fibers was also investigated. To reveal the ozone-curing mechanisms of the PSZ fibers and the formation process of the circular pores, the FT-IR, SEM, OM, XRD and TG were characterized. The results showed that the Si–H bonds and Si-CH3 groups in the PSZ molecule were oxidized gradually to form oxygen-containing groups such as Si–OH, Si–O-Si and C = O in the ozone curing process, which contributed to the curing and weight gain of the PSZ fibers. Moreover, a hydrolysis reaction between Si-NH-Si and H2O might further accelerate the curing of the PSZ fibers. An unexpected result of the hydrolysis reaction is the formation of some circular pores on the surface layer of the PSZ fibers. This strategy provides a method to design, adjust and control the microstructure and composition of silicon-based ceramic fibers, and the obtained SiCN fibers may be used as catalyst support, bacterial culture and other fields.

      • Neutrophil Count and the Inflammation-based Glasgow Prognostic Score Predict Survival in Patients with Advanced Gastric Cancer Receiving First-line Chemotherapy

        Purpose: To explore the value of systemic inflammatory markers as independent prognostic factors and the extent these markers improve prognostic classification for patients with inoperable advanced or metastatic gastric cancer (GC) receiving palliative chemotherapy. Methods: We studied the prognostic value of systemic inflammatory factors such as circulating white blood cell count and its components as well as that combined to form inflammation-based prognostic scores (Glasgow Prognostic Score (GPS), Neutrophil-Lymphocyte Ratio (NLR), Platelet Lymphocyte Ratio (PLR), Prognostic Index (PI) and Prognostic Nutritional Index (PNI)) in 384 patients with inoperable advanced or metastatic gastric cancer (GC) receiving first-line chemotherapy. Univariate and multivariate analyses were performed to examine the impact of inflammatory markers on overall survival (OS). Results: Univariate analysis revealed that an elevated white blood cell, neutrophil and/or platelet count, a decreased lymphocyte count, a low serum albumin concentration, and high CRP concentration, as well as elevated NLR/PLR, GPS, PI, PNI were significant predictors of shorter OS. Multivariate analysis demonstrated that only elevated neutrophil count (HR 3.696, p=0.003) and higher GPS (HR 1.621, p=0.01) were independent predictors of poor OS. Conclusion: This study demonstrated elevated pretreatment neutrophil count and high GPS to be independent predictors of shorter OS in inoperable advanced or metastatic GC patients treated with first-line chemotherapy. Upon validation of these data in independent studies, stratification of patients using these markers in future clinical trials is recommended.

      • LANDUSE MONITORING BY RS, GIS AND GPS AT COUNTY LEVEL IN CHINA

        Using the synthetic technology of RS, GPS and GIS to monitor landuse at county level (scale 1:10,000) has characteristics of lower expense, high precision and efficiency. This method is suitable for China and has a great prospective. Landuse monitoring by 3S can be divided into five stages: ① finding out the possible changing area by RS, ②Capturing the spatial data of the changing area by GPS, ③Processing the GPS data, ④Editing GPS data in GIS and ⑤Analysis and output. In this paper, we discuss the techniqueflow and key techniques in each stage of landuse monitoring by 3S. Furthermore, we develop a prototype of a temporal landuse information system (TLIS) which built on a feature-based spatio-temporal data model. This system not only has the functions of traditional GIS but also has the sptio-temporal query and reasoning functions. It can play the process of changing feature instances which let us understand the nature of the landuse changing in spatio-temporal dimensions.

        BTXA Could Induce Fibroblast Apoptosis and Inhibit the Expression of α-SMA and Myosin II in Scar Tissue of Rabbit Ears

        Purpose This study aims to explore the effects of BTXA on the proteins that affect the contraction of fibroblasts. Moreover, the optimal dose of BTXA on hypertrophic scars has not yet been determined. The current study investigated the effect of different doses of BTXA on scar formation. Materials and Methods 18 rabbits’ ears of New Zealand rabbits were used to establish hypertrophic scar model. These wounds were divided into four groups as group B (treated with deferent dosage of BTXA), group T (treated with TAC), group S (not treated), and group C (normal skin). Fibroblast apoptosis was detected by the method of TUNEL. Collagen I, III, and TGF-β1 expression were examined by western blotting method. The immunohistochemical staining and computerized image analysis were used to measure the optical density (OD value) of positive staining of α-SMA and myosin II. Results With increasing BTXA dose, the apoptotic rates increased. Western blot showed that Collagen fibrils and TGF-β1 proteins were expressed lower with the increasing of dose of BTXA (P < 0.05). α-SMA and myosin II in Group B were significantly decreased than those in Group T (P < 0.05), moreover, more myosin II can be reduced with high concentrations of BTXA (2.0 IU). Conclusions It was shown that BTXA could induce fibroblast apoptosis and inhibit the expression of α-SMA and myosin II, which can inhibit FB contraction. It plays a positive role in the treatment of scar.

        Disease Course and Outcomes in Patients With the Limited Form of Neuromyelitis Optica Spectrum Disorders and Negative AQP4-IgG Serology at Disease Onset: A Prospective Cohort Study

        Background and Purpose Patients presenting with clinical characteristics that are strongly suggestive of neuromyelitis optica spectrum disorders (NMOSD) have a high risk of developing definite NMOSD in the future. Little is known about the clinical course, treatment, and prognosis of these patients with likely NMOSD at disease onset. Methods This study prospectively recruited and visited 24 patients with the limited form of NMOSD (LF-NMOSD) at disease onset from November 2012 to June 2021. Their demographics, clinical course, longitudinal aquaporin-4 immunoglobulin G (AQP4-IgG) serology, MRI, therapeutic management, and outcome data were collected and analyzed. Results The onset age of the cohort was 38.1±12.0 years (mean±standard deviation). The median disease duration was 73.5 months (interquartile range=44.3–117.0 months), and the follow-up period was 54.2±23.8 months. At the end of the last visit, the final diagnosis was categorized into AQP4-IgG-seronegative NMOSD (n=16, 66.7%), AQP4-IgG-seropositive NMOSD (n=7, 29.2%), or multiple sclerosis (n=1, 4.2%). Seven of the 24 patients (29.2%) experienced conversion to AQP4-IgG seropositivity, and the interval from onset to this serological conversion was 37.9±21.9 months. Isolated/mixed area postrema syndrome (APS) was the predominant onset phenotype (37.5%). The patients with isolated/mixed APS onset showed a predilection for conversion to AQP4-IgG seropositivity. All patients experienced a multiphasic disease course, with immunosuppressive therapy reducing the incidence rates of clinical relapse and residual functional disability. Conclusions Definite NMOSD may be preceded by LF-NMOSD, particularly isolated/ mixed APS. Intensive long-term follow-up and attack-prevention immunotherapeutic management is recommended in patients with LF-NMOSD.

      • Knockdown of GCF2/LRRFIP1 by RNAi Causes Cell Growth Inhibition and Increased Apoptosis in Human Hepatoma HepG2 Cells

        Background: GC-binding factor 2 (GCF2) is a transcriptional regulator that represses transcriptional activity of the epidermal growth factor receptor (EGFR) by binding to a specific GC-rich sequence in the EGFR gene promoter. In addition to this function, GCF2 has also been identified as a tumor-associated antigen and regarded as a potentially valuable serum biomarker for early human hepatocellular carcinoma (HCC) diagnosis. GCF2 is high expressed in most HCC tissues and cell lines including HepG2. This study focused on the influence of GCF2 on cell proliferation and apoptosis in HepG2 cells. Materials and Methods: GCF2 expression at both mRNA and protein levels in HepG2 cells was detected with reverse transcription (RT) PCR and Western blotting, respectively. RNA interference (RNAi) technology was used to knock down GCF2 mRNA and protein expression. Afterwards, cell viability was analyzed with a Cell Counting Kit-8 (CCK-8), and cell apoptosis and caspase 3 activity by flow cytometry and with a Caspase 3 Activity Kit, respectively. Results: Specific down-regulation of GCF2 expression caused cell growth inhibition, and increased apoptosis and caspase 3 activity in HepG2 cells. Conclusions: These primary results suggest that GCF2 may influence cell proliferation and apoptosis in HepG2 cells, and also provides a molecular basis for further investigation into the possible mechanism at proliferation and apoptosis in HCC.

        Synthesis of biscoumarin and dihydropyran derivatives as two novel classes of potential anti-bacterial derivatives

        A series of bisoumarin (1–4) and dihydropyran (5–8) derivatives were successfully synthesized as new antibacterial agents. The molecular structures of three representative compounds 1, 5 and 7 were confirmed by single crystal X-ray diffraction study. Among these compounds tested toward Staphylococcus aureus (S. aureus ATCC 29213), methicillin-resistant S. aureus (MRSA XJ 75302), vancomycin-intermediate S. aureus (Mu50 ATCC 700699), and USA 300 (Los Angeles County clone, LAC), compounds 1 and 2 displayed the most potent antibacterial activity. Additionally, the HB energy in biscoumarins 1–4 was calculated by density functional theory (DFT) [B3LYP/6-31G*] method.
