        Dimethylnitrosamine 유발 급성 간 손상 흰쥐에서 ^(99m)Tc-Lactosylated Serum Albumin을 이용한 간 기능의 평가

        목적: ^(99m)Tc-lactosylated serum albumin (^(99m)Tc-LSA)은 간세포에 특이적으로 결합하는 간수용체 영상용 방사성의약품으로 새로이 합성되었다. 간섬유화를 유발하는 dimethylnitrosamine (DMN)을 투여한 간 손상 휜쥐 모델에서 ^(99m)Tc-LSa의 역동학적인 간섭취를 조사하고 간효소치의 변화와 조직학적 소견을 비교하여, LSA의 간섭취가 간기능의 변화를 반영하는지를 연구하였다. 대상 및 방법: SD계 흰쥐에 DMN를 27 mg/kg으로 복강 내 주사하여 급성 간손상을 유도하고 대조군과 비교하였다. DMN을 주사한 흰쥐를 3일(DMN-3), 8일(DMN-8), 21일(DMN-21)에 ^(99m)Tc=LSA (1,665 mg/kg) 29 MBq를 정맥 주사하여, 30분 동안 동적 영상을 획득하고 간과 신장부위에 관심영역을 설정하여 간과 심장부위의 시간방사능 곡선을 얻었다. 간기능 평가를 위해 시간방사능 곡선을 이용하여 간섭취지수와 혈중제거지수를 구하였고 곡선 최적화를 시행하였다. DMN 투여군과 대조군의 간효소치의 변화와 간조직의 광학현미경 소견을 비교하였다. 결과: 대조군에서는 ^(99m)Tc-LSA가 빠르게 간에 섭취되고 혈중에서 제거되었으나 DMN을 처리한 군에서는 간섭취가 낮았다. 간섭취지수의 비교에서 대조군에 비해 DMN 처리군에서 유의하게 간섭취지수가 낮았다(DMN-3: 0.842, DMN-8: 0.898, DMN-21: 0.91, 대조군: 0.96, p<0.05). 혈중제거지수의 비교에서도 대조군에 비해 DMN 처리군에서 혈중제거지수가 유의하게 높았다(DMN-3: 0.731, DMN-8: 0.654, DMN-21: 0.604, 대조군: 0.473, p<0.05). 비선형 회귀분석에서 R_(2) 값은 0.9이상으로 좋은 일치를 보였고, 대조군에ㅓ K값이 DMN처리군에 비해 크고(DMN-3: 0.28, DMN-8: 0.41, DMN-21: 0.46, 대조군: 0.97, p<0.05), T_(1/2)값은 작았다(DMN-3: 2.5, DMN-8: 1.7, DMN-21: 1.5, 대조군: 0.7, p<0.05). 간효소치의 변화는 DMN-3군에서는 대조군에 비해 상승하였으나 DMN-8, DMN-21군에서는 간효소치의 상승이 관찰되지 않았다. 간조직 소견의 경우 DMN-3군에서 중심정맥 주위에 괴사가 관찰되었으나 DMN-8군, DMN-21군에서는 미약한 정도의 염증세포 침윤만이 관찰되었다. 결론: ^(99m)Tc-LSA 간신티그래피의 간섭취 정도는 간손상과 반비례하였으며 간섭취의 변화는 조직학적 손상이 심한 정도와 간손상후 회복되는 과정을 반영하여 주었다. ^(99m)Tc-LSA 간신티그래피가 간손상을 평가하고 간손상후 회복되는 과정을 추적하는 간수용체 영상용 방사성 의약품으로 사용될 수 있을 것으로 생각된다. Objects: ^(99m)Tc-lactosylated human serum albumin(LSA) is a newly synthesized radiopharmaceutical that binds to asialoglycoprotein receptors, which are specifically presented on the hepatocyte membrane. Hepatic uptake and blood clearance of LSA were evaluated in rat with acute hepatic injury induced by dimethylnitrosamine(DMN) and results were compared with corresponding findings of liver enzyme profile and these of histologic changes. Materials and Methods: DMN (27 mg/kg) was injected intraperitoneally in Sprague-Dawley rat to induce acute hepatic injury. At 3(DMN-3), 8(DMN-8), and 21(DMN-21) days after injection of DMN, LSA injected intravenously, and dynamic images of the liver and heart were recorded for 30 minutes. Time-activity curves of the heart and liver were generated from regions of interest drawn over liver and heart area. Degree of hepatic uptake and blood clearance of LSA were evaluated with visual interpretation and semiquantitative analysis using parameters (receptor index : LHL3 and index of blood clearance : HH3), analysis of time-activity curve was also performed with curve fitting using Prism program. Results: Visual assessment of LSA images revealed decreased hepatic uptake in DMN treated rat, compared to control group. In semiquantitative analysis, LHL3 was significantly lower in DMN treated rat group than control rat group (DMN-3:0.842, DMN-8: 0.898, DMN-21: 0.91, Control: 0.96, p<0.05), whereas HH3 was significantly higher than control rat group (DMN-3: 0.731, DMN-8: 0.654, DMN-21: 0.604, Control: 0.473, p<0.05). AST and ALT were significantly higher in DMN-3 group than those of control group. Centrilobular necrosis and infiltration of inflammatory cells were most prominent in DMN-3 group, and were decreased over time. Conclusion: The degree of hepatic uptake of LSA was inversely correlated with liver transaminase and degree of histologic liver injury in rat with acute hepatic injury.

        <P>Solution‐processable reduced graphene oxide as a hole‐transporting layer for highly efficient and stable organic solar cells is reported on page 4923 by Dong‐Yu Kim, Seok‐In Na, and co‐workers. Introduction of a newly reduced graphene oxide by simple solution processing into solar cells dramatically raises the cell efficiency and cell life‐time. The results will allow full use of chemically reduced graphene and will advance the realization of carbon‐based printable optoelectronic devices. </P>

        경막 내 colistimethate 투여로 치료한 다제 내성 cinetobacter baumannii 뇌실염

        Acinetobacter species is a non-fermentative aerobic gram-negative coccobacillus that is an important pathogen found in nosocomial infections. Recently, multi-drug resistant Acinetobacter baumannii (MDR-AB) infections have been increasing and pose a serious problem. Most such infections present as bacteremia, pneumonia, or a wound infection; however, CNS infections are very rare. We herein present a case of ventriculitis caused by MDR-AB in a 37-year old man after a neurosurgical intervention. The patient was successfully treated with intrathecal colistimethate.

      • 구강점막 부착용 케토프로펜 고분자 필름의 제조 및 평가

        Abstract – The objective of this study was to prepare ketoprofen-loaded buccal adhesive patch. The adhesive patch was formulated by casting method using aqueous soluble polymer povidone K17 (PVP 17PF) as film-forming agent and hydroxypropylmethylcellulose (HPMC) as adhesive agent. To compare the effect of HPMC type, different molecular weight of K4M and K15M HPMC was used. The physicochemical properties of patches such as appearance, thickness, in vitro release, and adhesiveness were investigated. The concentration of ketoprofen was determined spectrophotometrically at a wavelength of 233 nm. The appearance of prepared patches was semi-transparent, light-yellow or almost colorless, and odorless. Thickness of each patches (n=6) was 0.895 ± 0.033 mm for K4M patch and 0.727 ± 0.036 mm for K15M patch. In vitro release test, both K4M and K15M patches showed over 20% release within 30 min. At 120 min, K4M and K15M patches demonstrated 95% and 67.5% release of ketoprofen, respectively, and up to 240 min, both patches released drug completely. Maximum adhesive force of K4M and K15M patches was 6.571 ± 2.703 gf and 2.735 ± 1.151 gf, respectively. Moreover, it took 28.29 ± 0.38 sec and 28.30 ± 0.34 sec for K4M and K15M patch to peel off them after adhesion, showing no significant difference. In conclusion, thickness, in vitro release, and maximum adhesive force could be modulated by alteration of polymer types.

      • 발생학습수업에 의한 국민학생들의 시각 개념의 변화

        The purpose of the study is to investigate the effect on the visual conceptual change in light with the application of generative learning model. 32 samples were selected in four elementary schools from Seoul, Kyung-gi areas. They were divided into 12 samples in grade 2, and 20 samples in grade 4, respectively. The results were shown as follows: 1. Visual concept on light was shown the receptive concept in the link of eye and object in lower grade, and the scientific concept from the source of light to object, and to eye in upper grade. 2. The direction of light vision was generally much more appeared after the application of generative learning strategy than before. 3. In lower grade light vision in the clock, candle, book context was represented with only one or two context of the three before the learning, however, after the learning they showed the dual representative model in the varieties of contexts. 4. Individual changes on light vision were shown much more scientific concepts than receptive concepts.

      • 국민학교 학생들의 과학에 관련된 태도와 과학 불안에 관한 연구

        The purpose of this study is to analyze the relationship between science anxiety and attitude related to the science of elementary students. To investigate the attitude, modified TOSRA and KEDI's attitude scale were used. To survey the science anxiety, modified science anxiety measurement scale was used. As the Cronbach α, the modified attitude instrument indicated 0.8 and modified anxiety instrument was 0.89. The 1,202 subjects were sampled by stratified cluster sampling method. The major findings of this study are as follows. 1. Fifth grade students have significantly higher attitude scores than sixth grade students. 2. Sixth grade students have significantly higher anxiety scores than fifth grade students. 3. There was no significant difference between male and female on science related attitude and science anxiety. 4. The rural area students has significantly higher anxiety scores than large city students. 5. The Pearson's correlation coefficient between anxiety and attitude was 0.57.

      • 댄스스포츠 경기판정 영향요인에 대한 인식차 비교분석

        The purpose of this study is to conduct an empirical analysis on differences in perspectives regarding background variables affecting dance-sport game decisions(including the referee's fairness, credibility, and possible misjudgement) and to provide fundamental information to help with fair game handling. The result of the analysis is as follows: First, all aspects such as fairness, partiality, and wrong decisions showed significant differences (p<.001) among groups after carrying out a comparative analysis on different perspectives regarding the influence of athlete and referee groups on the decision-making aspect. Second, fairness showed significant differences between male and female athletes (p<.01) after carrying out a comparative analysis on different perspectives regarding decision-making influences between different sex. Third, fairness (p<.05) and partiality (p<.01) showed significant differences among groups after carrying out a comparative analysis on different perspectives regarding the influence of different athletic divisions on the decision-making aspect. Fourth, fairness (p<.05) and partiality (p<.001) showed significant differences among groups after carrying out a comparative analysis on different perspectives regarding the influence of the athlete's different status (amateurs or professionals) as on the decision-making aspect. Fifth, fairness and partiality (p<.001 respectively) showed significant differences after conducting a comparative anlysis on different perspectives regarding the influence of the athlete's experience on the decision-making aspect.         만성 강박장애 환자의 정서처리 특성 : 정신분열병 환자 및 정상인과의 비교

        Objectives : Obsessive compulsive disorder (OCD) tends to take chronic course as schizophrenia. Researchers reported that OC patients had idiosyncratic sensitivity to threat-related information, while schizophrenic patients were reported to have affec-tive blunting. We hypothesized that there might be a difference in emotional response between OCD and SPR in cronic phase. Methods : Eight different emotional tasks were done by 25 healthy controls, 21 OCD patients and 25 SPR patients. Visual stimuli were made by pairing two words or pictures. Four kinds of emotional stimuli (positive, negative, combined and neutral)were presented to subjects through monitor and they were asked to report their subjective feelings by pushing mouse button, In the combined condition, a pair of positive words (or pictures) and negative words (or pictures) were presented simultaneously. The responses and response time were recorded and analyzed using SPSS 9.0 package. Results : In the negative condition, OCD group didn't show any deficit compared to healthy group. But SPR group showed significantly lower appropriate response rate than healthy group, In the combined condition, OCD and healthy control group Showed 'negativity bias' which SPR group didn't show. In the positive condition, however, OCD and SPR groups did show Significantly lower appropriate response rate than healthy controls. Conclusion : Our results suggested that OCD patients may not have deficits in the processing of negative emotion even in the chronic phase but to have a specific deficit in positive emotion. This result supports the 'threat-relatedness hypothesis' on attentional bias of OCD.

      • 모체 Thyroxine 투여가 태아알코올효과를 가진 흰쥐 대뇌겉질 및 해마에서 NPY함유 신경세포의 생후 발달에 미치는 영향

        Background and Objectives: Maternal alcohol abuse is considered to be one of the most prominent cause of neurobiological malformations in the postnatal and adult life of the offspring. In this study, we investigated the effects of maternal alcohol drinking on the postnatal development of NPY-containing neuron, and, the influence of thyroxine treatment on the cerebral cortex and hippocampus of pups of alcohol abused mother. Materials and Method: Time-pregnant rats were divided into three groups. Alcohol-fed group A received 35 calories of liquid alcohol diet daily from gestation day 6; control pair-fed group B was fed a liquid diet in dextrin replaced alcohol isocalorically: alcohol + T4 group C received 35 calories liquid alcohol diet and exogenous thyroxine subcutaneously. Results: Group C showed prominent NPY immunoreactivity in the cerebral cortex compared to group A and B at P7. In group C, NPY-containing neurons were widely distributed in the all layers of cerebral cortex after P14. Besides, numerical decrease of NPY-containing neuron as age increases was not found in group C. However, the decrease of NPY-containing neuron was clearly observed in group A compared to group C after P14. In hippocampus, group Band C were appeared similar patterns after P7. Additionally, in group C, NPY immunoreactivity was prominently appeared in CA2 and CA3 at P14 as compared to group B. Conclusions: The present results showed the increase of intensity and number of NPY-containing neurons in cerebral cortex and hippocampus of pups of exogenous T₄ supplemented alcohol-exposed dams as compared to control pair-fed and alcohol-exposed pups at P7. It presumably suggest that NPY-containing neurons might be regulated by the early postnatal growth stimulatory effect of the exogenously supplemented T₄. Therefore, the increase of NPY synthesis caused by maternal administration of exogenous thyroxine may ameliorate fetal alcohol effect, one of the ill effects as a result of the dysthyroid state following maternal alcohol abuse.
