        The Effect of Topiramate on Hippocampal Neuronal Death and Expression of Glutamate Receptor in Kainate-induced Status Epilepticus Model

        신경흥분독성과 간질발작발현은 glutamate 수용체활성과 연관이 있다고 알려져 있다. a-amino-3-hydroxy-5-methyl-4-isoxazole-propionate (AMPA), kainate 수용체에 대한 glutamate 활성을 포함하는 다양한 기전을 가진 항전간제인 Topiramate는 신경보호작용을 가진다는 증거가 제시되어 Topiramate가 간질발작 후 해마의 glutamate 수용체 발현에 미치는 효과를 관찰하였다. 흰쥐에 kainate를 복강 내 주사하여 간질중첩증을 유발시킨 후 Topiramate를 1주일 주사하였다 Apop tag in situ detection kit를 이용하여 세포손상을 관찰한 결과 kainate 유발 간질중첩증 1주일 후 해마의 CA1, CA3에서 심각한 세포사를 보였으나, Topiramte 처리 군에서는 세포사가 현저히 감소하였다. 간질중첩증 이후 NMDA 수용체 아형 1,2a, 2b 발현이 현저히 증가했으나 Topiramate 처치에 의해 NMDA수용체의 발현에는 뚜렷한 변화가 없었다. AMPA수용체에서는 GluR1이 간질중첩증 이후 현저히 상향 조정되었고 GluR2는 현저히 하향조정 되었다 Topiramate 1주일 처리 시 간질중첩증으로 인해 변화된 CluR1과 GluR2의 발현이 역전되었다. 결론적으로 Topiramate는 간질중침증에 의한 CluR1/CluR2 발현비의 증가로 유발되는 흥분성 신경세포사를 억제시킴으로써 신경보호작용이 있는 것으로 보인다. Excitotoxicity and epileptogenesis have often been associated with glutamate receptor activation. Accumulating evidences indicates that topiramate (TPM), an antiepileptic drug with multiple mechanisms of action has neuroprotective activity. We explored the neuroprotective effect of TPM on the status epilepticus (SE)-induced hippocampal neuronal death. After development of SE by kainite injection (15 mg/Kg), rats were treated with TPM (10mg/kg) for 1 week. The neuronal death was detected by Apop tag in situ detection kit, and the expression levels of glutamate receptors were semi-quantitatively analyzed by immunoblot. Kainate-induced SE caused a significant neuronal death and cell loss in CAI and CA3 regions of hippocampus at 1 week. However, treatment of TPM for 1 week after SE markedly reduced hippocampal neuronal death. The expression of N-methyl-D-aspartate (NMDA) receptor subunit 1, was increased by SE, but was not affected by 1 week treatment of TPM. The expressions of NMDA receptor subunit 2a and 2b were not changed by either SE or TPM. As for ${\alpha}-amino-3-hydroxy-5-methyl-4-isoxazole-propionate$ (AMPA) glutamate receptors (GluR), kainate-induced SE markedly up-regulated GluR1 expression but down-regulated GluR2 expression, leading to increased formation of $Ca^{2+}$ permeable GluR2- lacking AMPA receptors. TPM administration for 1 week attenuated SE-induced expression of both the up-regulation of GluR1 and down-regulation of GluR2, reversing the ratio of GluR1/GluR2 to the control value. In conclusion, TPM protects neuronal cell death against glutamate induced excitotoxicity in kainate-induced SE model, supporting the potential of TPM as a neuroprotective agent.

      • 치과 기공사들에게서 나타나는 호흡기 증상과 이의 관련 요인

        This study was carried out to investigate subjective respiratory symptoms and their attributable factors in Korean dental laboratory technicians. A questionnaire was mailed to randomly selected 1,900 dental laboratory technicians. Among them, 1,344 dental technicians completed the questionnaires and returmned them. Validity and reliability tests on the questionnaires were carried out before the main survey. To investigate risk factors on respiratory subjective symptoms, we performed analysis on distribution of characteristics, and then investigated univariate analysis and multiple logistic regression analysis. The prevalence rate of wheezing, cough and phlegm, and dyspnea was 4.8%, 33.9%, 54.9%, respectively. Statistically significant factors inducing subjective respiratory symptoms by multiple logistic regression analysis were sex, smoking, exercise, health examination, work hours a day, investment gas, investment dust, metal fume, resin vapor, and asbestos dust. Considering the results above, it seems that most subjective respiratory symptoms are related with health habits and occupational environmental-related factors. The result, consideration in mind, suggest that health promotion programs for smoking cessation, regular health examination, and physical exercise in health habit, and improvement on casting and resin work in dental technological environment be necessary.

        혈관질환 정보관리 시스템

        As medical technology progresses rapidly, there is a rise in the average age along with the Korean dietary lifestyle becoming more westernized, which leads to an increase in the number of vascular disease patients in Korea. Thus, we need to manage the medical information of a disease systematically in order to diagnose and treat constructively. However, since there has been no standardized method of man agement to date, a great deal of information could not be properly utilized nor studied. Therefore, the departments of Cardiology, Radiology and Neurology of Samsung Seoul Hospital recently got together to develop an information management system called the Vascular Data System. This program was developed to be run on win98 O/S, upper Pentium Ⅲ, and upper 128 MB Memory, and its source code is Dephi 4.0. It was configured for the user to set the configurations as well as do a variety of search and analysis. If this program were to be updated continuously, it may be used extensively as well as in various parts of clinical research activities.

        <P><B>Abstract</B></P> <P>In this work, copper indium gallium selenide (Cu(In,Ga)Se<SUB>2</SUB>; CIGS) absorbers were grown on polyimide (PI)/molybdenum substrates by a three-stage co-evaporation process at various temperatures, film formation was systemically studied using various advanced characterization methods such as transmission electron microscopy, micro-Raman spectroscopy, Kelvin probe force microscopy, and atom probe tomography. The CIGS films on PI were found to exhibit considerable physical and electrical variations with respect to the process temperature of three-stage co-evaporation. In particular, when the process temperature reached 400 °C, the CIGS absorber on PI began to exhibit controlled microstructure and intergrain properties. By adjusting the microstructure and intergrain properties of the absorber films by means of the process temperature of three-stage co-evaporation, flexible CIGS solar cells on PI with an efficiency of 16.7% (with anti-refection coating) were achieved.</P> <P><B>Highlights</B></P> <P> <UL> <LI> CIGS absorber films were grown on flexible polyimide/molybdenum substrates. </LI> <LI> Low-temperature three-stage process (≤440 °C) with extrinsic Na addition was used to CIGS growth. </LI> <LI> CIGS film evolution was systemically observed using advanced material characterization techniques. </LI> <LI> Highly efficient CIGS cells on flexible polyimide substrates were realized while maintaining process manufacturability. </LI> </UL> </P> <P><B>Graphical abstract</B></P> <P>[DISPLAY OMISSION]</P>

        Bucillamine prevents cisplatin-induced ototoxicity through induction of glutathione and antioxidant genes

        <P>Bucillamine is used for the treatment of rheumatoid arthritis. This study investigated the protective effects of bucillamine against cisplatin-induced damage in auditory cells, the organ of Corti from postnatal rats (P2) and adult Balb/C mice. Cisplatin increases the catalytic activity of caspase-3 and caspase-8 proteases and the production of free radicals, which were significantly suppressed by pretreatment with bucillamine. Bucillamine induces the intranuclear translocation of Nrf2 and thereby increases the expression of γ-glutamylcysteine synthetase (γ-GCS) and glutathione synthetase (GSS), which further induces intracellular antioxidant glutathione (GSH), heme oxygenase 1 (HO-1) and superoxide dismutase 2 (SOD2). However, knockdown studies of HO-1 and SOD2 suggest that the protective effect of bucillamine against cisplatin is independent of the enzymatic activity of HO-1 and SOD. Furthermore, pretreatment with bucillamine protects sensory hair cells on organ of Corti explants from cisplatin-induced cytotoxicity concomitantly with inhibition of caspase-3 activation. The auditory-brainstem-evoked response of cisplatin-injected mice shows marked increases in hearing threshold shifts, which was markedly suppressed by pretreatment with bucillamine <I>in vivo</I>. Taken together, bucillamine protects sensory hair cells from cisplatin through a scavenging effect on itself, as well as the induction of intracellular GSH.</P>

        급성신부전을 동반한 알콜성 횡문근융해증 2례

        Rhabdomyolysis is a biochemical and clinical syndrome caused by lysis of skeletal muscle cell and release of muscle cell contents into the circulation. In addition to traumatic cause, various nontraumatic causes have been reported. Rhabdomyolysis has frequently been associated with seamy aspects of life and society: alcoholic abuse, drug addiction, sadistic drill exercise, war events and attempted suicide. The abuse of alcohol causes many hazards in many organs. One of these is acute alcoholic myopathy that may present as a fulminent syndrome of muscle pain and tenderness associated with rhabdomyolysis and acute renal failure. Because the prognosis of adequately treated rhabdomyolysis is excellent, early recognition and prompt management are essential in emergency situation. We experienced two typical cases of alcohol-induced rhabdomyolysis with acute renal failure and report these cases with literature review.

      • 光條件이 질경이(Plantago asiatica L.)의 生長에 미치는 影響

        被陰에 의한 光勾配實驗을 통하여 對照區와 相對照度 75,50,25% 處理區에서의 질경이(Plantago asiatica L.)의 生長狀態를 比較, 分析하였다. 相對照度가 높아질수록 植物體의 物質生産과 C/F ratio는 增加하였으나 T/R ratio는 減少하였다. 한편, 相對照度가 낮아질수록 뿌리와 花穗를 제외한 全 器官의 伸長成長이 增加되었고 水分含量 역시 높아지는 傾向이 있었으나, 花梗 및 잎의 數는 減少하였다. 乾物量에 의한 相對生長率은, 相對照度 75,50,25%區에서 對照區에 대해 각각 90,70,40%의 生長을 보여 照度低下에 따른 生長沮害를 觀察할 수 있었고, 花梗 및 花穗의 出現時期에 있어서는 相對照度가 가장 낮은 S₃區에서 다른 區에 비해 10日間의 差異로 늦어짐을 볼 수 있어 被陰이 질경이의 生長과 分布를 制限하는 要因의 하나임을 알 수 있었다. The light gradient of 100%, 75%, 50% and 25% in light intensity were established in order to investigate the difference of the growth of Plantago asiatica L. The amounts of matter production, water content, C/F ratio and numbers of leaf and scape tend to increase gradually with the increase of the relative light intensity. On the other hand, the elongations of all organs execpt root and spike and T/R ratio increase with the decrease of the relative light intensity. In the relative value of dry weight, the 75%, 50%, and 25% plots show the 90, 70 and 40% growth compared with the control group, respectively. Particulary, the emergence of spike and scape at 25% plot was delayed for 10 days compared with other plots. Judging from these results, shading is considered as a limiting factor in the growth and distribution of Plantago asiatica L.

      • 일부 출판업 VDT 작업자들의 근골격계질환 자각 증상 호소율과 관련요인에 관한 연구

        Objectives: This study was conducted to investigate the relationship between the self-reported musculoskeletal symptom prevalence and related factors among some VDT workers in publishing industries. Methods: The 201 study subjects consist of 130 VDT workers comprised of 59 edition workers and 71 data entry workers, and 71 Non-VDT workers. The study subjects were surveyed with structured self-reported questionnaire. Results: The prevalence of self-reported musculoskeletal symptom were 39.2% at neck, 36.2% at back and waist, 30.8% at shoulder, 25.4% at hand and wrist and 9.2% at arm on VDT workers. In logistic regression analysis. Type of VDT(data entry worker) work was related factor of musculoskeletal symptom prevalence at neck, shoulder, arm, back and waist, hand and wrist. VDT working time and working condition were related factors at back and waist. At neck, shoulder, hand and wrist, sex was one of the related factors. Conclusion: Our finding revealed that sex and type of VDT work were main related factors of musculoskeletal symptom prevalence in study subjects.
