A study on the determination of a Health based Guidance Value of Formaldehyde

Formaldehyde can be exposed through various routes, including the environment such as air and water, food and cosmetics. Therefore, a risk assessment is necessary to identifiy and manage human health effect. This study aims to identify the relationship between formaldehyde exposure and adverse effects as a hazard characterization step in the risk assessment process and to establish and propose a Health based Guidance Value. In order to carry out ther research, we used some online databases to find scientific papers, reports and opinions published by other institutions. Data for dose-response assessment were selected and classified, as properties of toxicity, and then the most appropriate study for establishing Health based Guidance Value was finally decided. Til et al (1989), 2-years carcinogenicity study with rats, was the optimal toxicity data. As the toxicity endpoint, the Health based Guidance Value TDI 0.15 mg/kg bw/day was proposed by applying an uncertainty factor of 100(interspecies difference 10, intraspeices difference 10) to NOAEL 15 mg/kg bw/day for chronic atrophic gastritis, focal ulceration and glandular hyperplasia in the landular forestomach.

Tofacitinib is an oral disease-modifying anti-rheumatic drug to selectively inhibit Janus kinases. Tofacitinib is a representative small molecule inhibitor that is used to treat many diseases including rheumatoid arthritis and various autoimmune conditions. Unlike biological agents, tofacitinib has several advantages, including the ability to be administered orally and a short half-life. This study aimed to evaluate the bioequivalence of the pharmacokinetics (PK) between tofacitinib aspartate 7.13 mg (test formulation) and tofacitinib citrate 8.08 mg (reference formulation; Xeljanz®) in healthy subjects. A randomized, open-label, single-dose, 2-sequence, 2-period, 2-treatment crossover trial was conducted in 41 healthy volunteers. A total of 5 mg of tofacitinib as the test or the reference formulation was administered, and serial blood samples were collected up to 14 hours after dosing for PK analyses. The plasma concentration of tofacitinib was determined by ultraperformance liquid chromatography-tandem mass spectrometry. A non-compartmental analysis was used to estimate the PK parameters. A total of 35 subjects completed the study and the study drug was well-tolerated. The mean maximum concentration (Cmax) and area under the concentration-time curve from time zero to the time of the last quantifiable concentration (AUClast) for the test formulation were 52.67 ng/mL and 133.86 ng∙h/mL, respectively, and 50.61 ng/mL and 133.49 h∙ng/mL for the reference formulation, respectively. The geometric mean ratios (90% confidence intervals) of the Cmax and AUClast between the 2 formulations were 1.041 (0.944–1.148) and 1.003 (0.968–1.039), respectively. Tofacitinib aspartate exhibited bioequivalent PK profiles to those of the reference formulation.

Previous studies reported that Bifidobacterium animalis ssp. lactis HY8002 (HY8002) improved intestinal integrity and had immunomodulatory effects. Lactobacillus plantarum HY7717 (HY7717) was screened in vitro from among 21 other lactic acid bacteria (LAB) and demonstrated nitric oxide (NO) production. The aims of this study were to investigate the individual and combined ex vivo and in vivo effects of LAB strains HY8002 and HY7717 at immunostimulating mice that have been challenged with an immunosuppressant drug. The combination of HY8002 and HY7717 increased the secretion of cytokines such as interferon (IFN)-γ, interleukin (IL)-12, and tumor necrosis factor (TNF)-α in splenocytes. In a cyclophosphamide (CTX)-induced immunosuppression model, administration of the foregoing LAB combination improved the splenic and hematological indices, activated natural killer (NK) cells, and up-regulated plasma immunoglobulins and cytokines. Moreover, this combination treatment increased Toll-like receptor 2 (TLR2) expression. The ability of the combination treatment to upregulate IFN-γ and TNF-α in the splenocytes was inhibited by anti-TLR2 antibody. Hence, the immune responses stimulated by the combination of HY8002 and HY7717 are associated with TLR2 activation. The preceding findings suggest that the combination of the HY8002 and HY7717 LAB strains could prove to be a beneficial and efficacious immunostimulant probiotic supplement. The combination of the two probiotic strains will be applied on the dairy foods including yogurt and cheese.

In this paper, a ROS-based autonomous driving framework designed by team ACCA from the School of Mechanical Engineering, Soongsil University, for the 2021 international college student creative car challenge held in K-city. The autonomous vehicle’s chassis used in this challenge is equipped with Velodyne 3D lidar, a Sick down-looking 2D lidar, Xsens MTi-30 AHRS, CCD camera, webcam, and PC-based controller. First, before the challenge in K-city, we evaluated the ROS-package-based SLAM such as LIO-SAM in a ring-shaped road environment on the campus of Soongsil University. After the successful SLAN and mapping process, the hdl_localization, which is a 3D lidar-based real-time 3D localization package, is used to estimate the global pose with respect to the global frame using NDT scan matching. For lane detection, traffic sign, and traffic signal recognition, the two well-known DNN models are utilized. Based on experimental results from both simulation and an actual autonomous vehicle platform, the Point Instance Network (PINet) for lane detection shows 88% of test accuracy, and the YOLO V4 for the traffic light and sign recognition offers 95% test accuracy.
