      • 제대혈 CD34 양성세포를 이용한 수상돌기 세포의 배양

        Background: Dendritic cells have a distinctive morphology and immunostimulatory potential. These cells are thought to be the major APC involved in triggering primary T-cell responses and express little linage specific surface markers. We tried to establish the method for the culture of dendritic cells with CD34^(+) cells from cord blood. Methods: Cord blood was collected during normal full-term delivery CD34^(+) cells were purified with Magnetic Separation Column, and cultured over 14 days in the presence of GM-CSF and TNF-α. Flow-cytometric analysis was conducted before and after culture. CFU-GM and CFU-DL culture was done with cells, harvested from 14 days of culture. Results: 1) After 14 days of culture with CD34^(+) cells (5×10^(5)), total cell number was expanded upto 3.2±3.35×10^(7), about 63 fold, and about 20% of them were adherent cells with dendritic morphology of numerals long and thin cytoplasmic projections. 2) Flow-cytometric analysis of surface marker expression were examined before and after culture for 14 days. The expression of CD34 was 69.1±13.01 (%), initially, and was decreased to 0.5±0.41 (%) at 14th day. The expression of HLA-DR, CD14 and CD1a were increased to 82.4±11.47 (%), 39.8±30.00 (%) and 15.7±7.73 (%) from 72.9±14.79 (%), 3.4±0.25 (%) and 1.5 ±0.64 (%). 3) The ratio of CFU-GM (94.5±86.97) and CFU-DL (8±8.49) colonies were about 10 to 1. Conclusion: CD34^(+) enriched cord blood can be expanded upto 63 folds with GM-CSF and TNF-α. This study established the method for the culture and identification of dendritic cells morphologically and immunophenotypically. Clinical application of dendritic cells as an adoptive immunotherapy and therapeutic vaccination in infectious or malignant disease needs further investigations about the right time of exposure to antigens, the dosage of cells, and the root of administration of the dendritic cells.

        The Impact of COVID-19 on the Optimal Management of Osteoporosis

        Osteoporosis does not take a break while Coronavirus disease 2019 (COVID-19) stunned and overtook everyone’s lives. Medical resources were immediately shifted, self-isolation and telemedicine were expanded, ambulatory care services such as bone densitometry and osteoporosis-centered clinics came to a near halt. Progress with fracture prevention has been challenged because osteoporotic fracture with low energy injury is more prevalent even though restriction of people’s movement. Thus we must re-engage with chronic bone health concerns and fracture prevention. This review discusses challenges in management of osteoporosis during the COVID-19 pandemic and reinforces the need to implementing recommendations concerning the importance of bone fragility care with at least those patients who are already treated with antiosteoporotic drugs maintaining their adherence to treatments.

        Citation Analysis of the Journal of Bone Metabolism from Korean Citation Index, Web of Science, and Scopus

        Background: Nine years have passed since the Journal of Bone Metabolism (JBM) was launched as an English journal in 2012; it was finally included in Scopus in January 2019. Therefore, this study aimed to provide evidence of increased international recognition based on journal metrics and reflect on its efforts to be recognized as a top-notch journal. Methods: Databases, such as the Web of Science (WoS), Scopus, Korean Citation Index (KCI), and citation indicators, including the impact factor (IF) and SCImago journal rank (SJR) were reviewed and calculated according to years, and the results were drawn. Furthermore, country-wise contributions and top-cited articles were also investigated. Results: The JBM 2020 IF was 2.17 in the WoS. The 2020 SJR in Scopus was 0.334, with a ranking of 165/219 (75.3%) in the Endocrinology, Diabetes, and Metabolism category. The 2020 KCI was 0.42. Of 263 articles, 260 were citable (98.9%), and of 176 original articles, 15 (8.5%) were supported by research grants. The total citation of JBM has increased from 16 in 2014 to 141 in 2020; however, its KCI remained stationary from 0.29 in 2015 to 0.42 in 2020. Conclusions: Currently, JBM is increasingly cited by international researchers than Korean researchers, indicating that the journal’s content is valued at an international level. Its inclusion in PubMed Central appears to have increased its international relevance; however, publishing English-only articles may hinder its use domestically. Therefore, efforts should be made to increase citation rates and enhance domestic readership.

        Characterization of Differential Cesium Toxicity and Cesium-affected Transcription of K+ or Na+ Transporter Genes in Model Plants and Horticultural Crops

        Pollutant toxicity influences biomass increment and pollutant uptake in phytoremediation;therefore, tolerance to pollutant toxicity is important in determining applicability ofcandidate plants. This study investigated relative Cs toxicity in Arabidopsis, cabbage, lettuce, redpepper, rice, and tomato seedlings through growth, photosynthesis, and/or transcription assays. When treated with 100, 300, 800, or 1,500 μM CsCl, the six plant species showed substantialdifferences in changes in seedling weight, chlorophyll content, photosynthetic activity, ortranscription of K+ or Na+ transporter genes, and exhibited higher susceptibility to Cs toxicity inthe order of Arabidopsis<rice<red pepper<tomato <cabbage<lettuce. Co-treatment with NaClor mannitol demonstrated that salt or osmotic stress could increase Cs toxicity in Arabidopsisseedlings. Among the five K+ or Na+ transporter genes, NHX5, SOS1, KT1, NHD1, and HKT1, ahigh-affinity potassium transporter gene HKT1 and a sodium hydrogen antiporter gene NHD1showed Cs concentration-dependent decrease in transcription, associating more with Cs toxicitythan Cs uptake. In contrast, a low-dose gamma radiation of 8 Gy increased shoot growth slightlybut decreased Cs uptake in seedlings. Taken together, these results suggest that the differential Cstoxicity to these six plant species could be somewhat associated with the distinctive transcriptionof K+ or Na+ transporter genes and that salt or osmotic stress and gamma radiation couldindirectly modulate Cs toxicity or Cs uptake.

        Reduced gene expression at the branch point of chlorophyll and heme biosynthesis in Arctic Chlorella ArM0029B

        The gene expression at the branch point of chlorophyll and heme synthesis in the model microalga, Chlamydomonas reinhardtii, is different from that of higher plants. Another green alga, Arctic Chlorella, was recently isolated from Arctic sea ice and may be a promising candidate for a biofuel. To understand the chlorophyll metabolic pathway and relevant nuclear gene expression in Chlorella sp., we characterized chlorophyll-deficient mutants of the Arctic Chlorella sp. ArM0029B. First, we characterized the chlorophyll and heme biosynthetic pathways based on genes identified by bioinformatics analysis of the genome of Arctic Chlorella sp. ArM0029B. Then, we isolated and analyzed nine chlorophyll-deficient mutants that showed reduced expression of the ChlM gene, which encodes Mgprotoporphyrin methyltransferase. Expression of 5-amino levulinic acid dehydratase (encoded by ALAD) and glutamyl- tRNA reductase (encoded by HemA) was reduced in all nine independent mutants compared to wild type. These results indicated that Arctic Chlorella ArM0029B may have a regulatory mechanism of gene expression at earlier steps of the Mg-porphyrin branch that is more similar to higher plants than to the microalga C. reinhardtii. This study provides useful insight into the regulation of porphyrin precursor formation in Chlorella and related microalgae.

        Development of cyan fluorescent protein (CFP) reporter system in green alga Chlamydomonas reinhardtii and macroalgae Pyropia sp.

        Various fluorescent proteins have been developedfor in vivo reporter systems in diverse prokaryotesand eukaryotes. However, few in vivo imaging systemshave been reported for the model algae Chlamydomonasreinhardtii or Pyropia sp. In this study, an effectiveimaging system using cyan fluorescent protein (CFP) wasdeveloped for the green alga C. reinhardtii, and its applicationwas also successful in the red macroalgae Pyropiatenera and P. yezoensis. For optimization of CFP expressionin C. reinhardtii and Pyropia sp., we modified codonusage in the CFP gene (CFP), generating PtCrCFP (Pyropiatenera/Chlamydomonas reinhardtii CFP). PtCrCFPwas successfully expressed in PtCrCFP-expressingUVM11 transgenic lines, and high accumulation levels ofPtCrCFP were found by western blotting. Consistent withthese results, PtCrCFP fluorescence was clearly detectedwith a low level of chlorophyll background fluorescence inPtCrCFP-expressing UVM11 transgenic lines. In Pyropiasp. gametophytic cells, transient expression of PtCrCFPfluorescence was distinctly visualized. PtCrCFP fluorescencewas also observed during the regeneration ofmonospores and young gametophytes from PtCrCFPexpressingP. yezoensis gametophytic cells. These resultssuggest that PtCrCFP may be useful as an in vivo reporterin green algae due to the short emission wavelength ofCFP, which provides a low level of chlorophyll backgroundfluorescence. This study also presents the possibilityof PtCrCFP’s use as a visible selection marker for thegeneration of transgenic lines in the red algae Pyropia sp. Thus, PtCrCFP as an in vivo visualization tool may offernew opportunities for the functional analysis of geneticstudies in both green and red algae.

        대퇴골 원위부 골절에서 잠김 금속판 고정술과 역행성 골수정 삽입술의 결과 비교

        목적: 원위 대퇴골 골절에서 관혈적 정복 및 잠김 금속판 고정술(LCP군) 및 역행성 골수정 삽입술(RIN군)을 시행 후 두 군 간에 방사선학적, 임상적 결과 및 합병증에 대해 비교해 보고자 한다. 대상 및 방법: 2003년 10월부터 2020년 2월까지 52예(LCP군 31예, RIN군 21예)를 대상으로 하였으며, 평균 추시 기간은 19.1개월이었다. 수술시간, 실혈량, 입원기간을 비교하였고, 수술 후 불유합, 부정유합, 지연유합 및 금속실패의 발생을 평가하였으며, 수술 후 합병증의 발생을 비교 분석하였다. 결과: 수술시간은 두 군 간에 유의한 차이가 없었지만, 평균 실혈량은 LCP군에서 유의하게 많았다(LCP 683.5 ml vs RIN 134.9 ml; p=0.015). 52예 중 49예에서 추가적인 수술 없이 평균 6.8개월의 기간에 골유합을 얻었으며 두 군 간 유의한 차이는 없었다(LCP 6.9개월 vs RIN 6.6개월; p=0.512). 3예의 불유합(LCP 2예 vs RIN 1예; p=0.065)에서 추가 수술 후 완전한 골유합을 얻었으며, 각 군에서 1예씩의 부정유합 및 표재감염이 확인되었다. 결론: 두 군 모두 높은 골유합 및 낮은 합병증을 보여, 두 술기 모두 대퇴골 원위부 골절 시 유용한 수술적 치료법으로 생각된다. Purpose: We compared the radiological and clinical results of fixation for distal femoral fracture (DFF) using a locking compression plate (LCP) or a retrograde intramedullary nail (RIN). Materials and Methods: From October 2003 to February 2020, 52 cases of DFF with a minimum 1-year follow-up (with a mean follow-up of 19.1 months) were included: 31 were treated with LCP and 21 with RIN. The operation time, blood loss, and hospitalization period were compared, and the incidence of postoperative nonunion, malunion, delayed union and metal failure and other post-operative complications were evaluated and compared. Results: There was no significant difference in the operating time between the two groups, but the mean blood loss was significantly higher in the LCP group (LCP 683.5 ml vs RIN; 134.9 ml; p=0.015). In 49 out of 52 cases, bone union was achieved without additional surgery in an average of 6.8 months, and a complete union was achieved after additional surgery in three cases of nonunion (LCP 2 cases vs RIN 1 case; p=0.065). One case of malunion and superficial infection was confirmed in each group. Conclusion: Internal fixation using LCP and RIN give good outcomes with a low complication rate and can therefore be considered useful surgical treatments for DFF.
