        솔이끼로부터 플라보노이드 성분의 분리

        Four compounds, luteolin (1), quercetin (2) astragalin (3), rutin(4), were isolated from the methanol extract of the all part of Polytrichum commune. on the basis of chemical and spectroscopic evidence. These compounds have not been isolated from this plant.

        Isolation of a New Quercetin 3-O-trisaccharide from the Leavesof Rubus crataegifolius

        new quercetin 3-O-trisacharide was isolated from the leaves of Rubus crataegifolius (Rosaceae).The structure of this compound was determined as quercetin 3-O-α-L-arabinofuranosyl-(1. 2)-β-D-xylopyranosyl-(1. 2)-β-D-galactopyranoside on the basis of 2D-NMR spectroscopic data.         RAW264.7 대식세포에서 장대나물 추출물의 Nitric oxide 및 Prostaglandin E2생성 저해효과

        본 연구에서는 장대나물(Arabis glabra)의 전초를 이용하여세포독성 및 항염증 활성 효과를 평가하였다. 대식세포인 RAW264.7 cell에서 염증 매개 물질인 lipopo-lysacchride (LPS)로 염증을유발시켜 nitric oxide (NO)와 prostaglandi nE2 (PGE2) 같은 염증 유발인자들의 억제효과를 확인하였다. 장대나물의 hexane 과 chloroform 분획물의 염증 유발인자 억제 시 IC50 value를 측정하였을 때 nitric oxide 및 prostaglandin E2 생성을 농도의존적으로 현저하게 저해하는 농도는 각각 21.0과 18.0 ㎍/㎖였다. 따라서 본 연구 결과는 장대나물의 hexane, chloroform과 같은비극성용매 분획물들이 유의성 있는 항염증 효과를 나타내었으며, 이러한 효능은 예방의학적 가능성을 충분히 가지고 있기에염증성질환의 예방을 위한 건강 기능성식품의 개발 가능성을제시할 수 있을 것으로 기대된다. 또한 염증과 관련된 사이토카인 및 단백질 발현 메커니즘에 대한 추가적인 연구가 필요할 것으로 판단된다. Arabis glabra is a localized common rhizomatous flowering plant, This plant is often used in Korean traditional systems of medicine as a remedy for blood cleaning, detoxification, abscess, gastrospasm, arthritis, contraction and diarrhea. Generally drugs that are used for arthritis have antinociceptive and anti-inflammatory properties. However, validity of the anti-inflammatory activity has not been scientifically investigated so far. Therefore, the aim of this study was to investigate the anti-inflammatory potential of A. glabra using the ethanolic extract and its sub-fractions. To evaluate the anti-inflammatory effects, we examined the inflammatory mediators such as nitric oxide (NO) and prostaglandin E2 (PGE2) on RAW 264.7macrophages. Our results indicated that hexane and chloroform fraction significantly inhibited the LPSinduced NO and PGE2 production in the cells. The hexane fraction inhibitory activity for NO tests with IC50 values showed in 21.0 ㎍/㎖. The chloroform fraction inhibitory activity for PGE2 tests with IC50 values showed in 18.0 ㎍/㎖. These efficacy are expected to be able to present the potential for the development of health functional food for the prevention inflammatory diseases because it has sufficient preventive medical possibilities. Further, it is determined that it is necessary to further study the mechanism of cytokine and protein expression associated with inflammation.

        곧은 나무이끼(Climacium dendroides)의 식물 화학적 성분연구

        The chromatographic separation of organic solvent extracts of liverwort led to the isolation of six compounds. 2-Chromenone (1), 3, 4-dihydroxy-cinnamic acid (2), 3, 3', 4', 5, 7-pentahydroxy-2-phenylchromen- 4-one (3), kaemperol-3-O-β-D-glucopyranoside (4), 3-[[3-(3, 4-Dihydroxyphenyl)-1-oxo-2- propenyl]oxy]-1, 4, 5-trihydroxycyclohexane carboxylic acid (5) and quercetin-3-O-rutinoside (6) were isolated from the methanolic extracts of the all part of Climacium dendroides.         RAW264.7 대식세포에서 풀솜대 추출물의 nitric oxide 및prostaglandin E2생성 저해효과

        Smilacina japonica is a localized commonrhizomatous flowering plant, This plant is often used inKorean traditional systems of medicine as a remedy formigrain, diplegia, physical impurity, blood circulation,abscess and contusion. Generally drugs that are used forarthritis have antinociceptive and anti-inflammatory properties. However, validity of the anti-inflammatory activity has notbeen scientifically investigated so far. Therefore, the aim ofthis study was to investigate the anti-inflammatory potentialof S. japonica using the ethanolic extract and its subfractions. To evaluate the anti-inflammatory effects, weexamined the inflammatory mediators such as nitric oxide(NO) and prostaglandin E2 (PGE2) on RAW 264.7macrophages. Our results indicated that hexane fractionsignificantly inhibited the LPS induced NO and PGE2production in the cells. The hexane fractions inhibitoryactivity for NO tests with IC50 values showed in 53.3 μg/mland PGE2 tests with IC50 values showed at 32.5 μg/ml. Theseis result suggest a potential role of hexane fractionfrom S. japonica as source of anti-inflammatory agent.

        A study on the development of a loading device using a photoelastic stress freezing method for the analysis of o-ring stress

        Typically, O-rings are used to prevent penetration of dust and alien substances from entering a cylinder during motion. Moreover, Orings are used to create an air tight seal around a stationary shaft. The stresses developed in O-rings depend on the squeeze rate, gap between the external diameter of the groove and internal diameter of the cylinder as well as internal pressure. In application, the stress distributions in O-rings can be very complicated and are almost always studied through experiment. Photoelastic experiment has been applied to the study of 3-dimensional stress distributions in O-rings. The loading device used in photoelastic experiment is important. Its function is to apply a uniform squeeze rate and internal pressure on the O-ring and to allow uniform squeeze rate to be controlled. In this research, a loading device was developed to perform these functions. The validity of this loading device was confirmed through the stress distribution, the configuration change and the contact length of the O-ring. When the squeeze rate was constant, the upper and lower contact lengths of the deformed O-ring were almost equal. When internal pressure was applied to the O-ring, while under a uniform squeeze rate the upper contact length increases slightly with increase in internal pressure, while the lower contact length of the O-ring is constant with an increase in internal pressure.

        Contact stress of O-ring under uniform squeeze rate by photoelastic experimental hybrid method

        In this paper, photoelastic experimental hybrid methods using the external traction free boundary condition and that using the relative equation of two stress functions in contact problems are developed. The validities of these two methods are confirmed through experiments and discussions. Hertz’s contact theory and the two photoelastic experimental hybrid methods explained are applied to the analysis of the contact stress of an O-ring under 10% or 20% squeeze rate. The photoelastic experimental hybrid method using the relative equation of two stress functions in contact problems was found to be more effective. When the squeeze rates of an O-ring were 10% or 20%, the maximum of absolute te σ x was greater than the maximum of absolute σ y , but was almost equal. Maximums of absolute τ xy were 1 8 of absolute σ x and 1 5 of absolute σ x when the squeeze rates of the O-ring were 10% and 20%, respectively.

        갈퀴나물 에탄올 추출물의 RAW264.7 대식세포에서 LPS (Lipopolysaccharide)로 유도된 nitric oxide 및 prostaglandin E2 생성 저해효과

        Vicia amoena has traditionally been used to treat disease of rheumatism, arthralgia, muscular paralysis, abscess and eczema, and it has anti-inflammatory properties. However, validity of the anti-inflammatory activity has not been scientifically in vestige acted so far. Therefore, the aim of this study was to investigate the anti-inflammatory potential of V. amoena using the ethanolic extract. To evaluate the anti-inflammatory effects, we examined the inflammatory mediators such as nitric oxide (NO) and prostaglandin E2 (PGE2) on RAW264.7 cells. Our results indicated that ethanolic extract of V. amoena significantly inhibited the LPS-induced NO and PGE2 production in RAW264.7 cells. The ethanolic extract of V. amoena has inhibited the PGE2 production by 88.0±0.8 % at the concentration of 40μg/ml. This results showed that ethanol extract of V. amoena is expected to be a good candidate for development into source of inflammation inhibitor 본 연구에서는 갈퀴나물(Vicia amoena) 전초를 이용한 에탄올 추출물의 세포독성과 항염증 활성 효과를 평가하였다. RAW264.7 cell(대식세포)에서 염증 매개 물질인 lipopolysaccharide (LPS)로 염증을 유발시켜 nitric oxide (NO) 및 prostaglandin E2 (PGE2)같은 염증 유발 인자들의 생성 저해효과를 확인하였다. 갈퀴나물 에탄올 추출물의 염증 유발 인자 억제 시 저해효과를 측정하였을 때 nitric oxide 및 prostaglandin E2 생성을 농도 의존적으로 현저하게 저해하는 농도인 40 μg/㎖의 농도에서 LPS에 의해 유도된 NO를 36.0 ± 0.5 % 저해하였으며, 특히 PGE2에서는 88.0 ± 0.8 % 만큼 유의성 있는 저해효과를 나타내었다. 따라서 본 연구결과는 갈퀴나물의 에탄올 추출물이 유의성 있는 항염증 효과를 나타내며, 이러한 효능은 예방의학적 가능성을 충분히 가지고 있기에 염증성 질환의 예방을 위한 항염증 소재로의 개발 가능성을 제시할 수 있을 것으로 기대된다. 차후 에탄올 추출물의 보다 다양한 농도 및 유기용매 분획물에서 염증반응을 매개하는 iNOS․COX-2 등 의 다양한 효소의 발현과 Iκ-Bα의 분해 등 염증반응의 신호전달물질의 변화에 대한 추가적인 연구가 필요할 것으로 판단된다.

        지대 및 품종(재배종)별 콩사포닌그룹B 함량의 비교

        Contents of soyasaponin group B were compared according to two regions and ten varieties by HPLC. Those compounds were known to be beneficial for health. After soyasaponins were isolated and identified, those isolated compounds were used for HPLC analysis. The contents of soyasaponin were very different by regions but highest in the soybean with black seed coat. It was appeared that environmental difference for soybean growth could strongly change of soyasaponin contents. Soyasaponin group B함량의 재배지대간 비교에서는고랭지와 평난지에서 상이한 차이를 보였으며 품종 간에는대부분 자엽색이 녹색이고 종피색이 검정색인 콩의 soyasaponin group B함량이 높은 것으로 나타났다. 이상의 결과를종합해 볼 때 soyasaponin성분의 함량은 재배종의 종류즉 유전적인 요인뿐만 아니라 고랭지와 평난지처럼 환경적요인이라고 것을 확인할 수 있었으며 이는 식물이 환경적인 스트레스에 대한 생명유지 및 공격방어물질(Secondary matabolites,이차대사산물)이 식물내 생합성과정 중에서변이를 보이는 것으로 사료되어 진다.
