        솔이끼로부터 플라보노이드 성분의 분리

        Four compounds, luteolin (1), quercetin (2) astragalin (3), rutin(4), were isolated from the methanol extract of the all part of Polytrichum commune. on the basis of chemical and spectroscopic evidence. These compounds have not been isolated from this plant.

        Contact stress of O-ring under uniform squeeze rate by photoelastic experimental hybrid method

        In this paper, photoelastic experimental hybrid methods using the external traction free boundary condition and that using the relative equation of two stress functions in contact problems are developed. The validities of these two methods are confirmed through experiments and discussions. Hertz’s contact theory and the two photoelastic experimental hybrid methods explained are applied to the analysis of the contact stress of an O-ring under 10% or 20% squeeze rate. The photoelastic experimental hybrid method using the relative equation of two stress functions in contact problems was found to be more effective. When the squeeze rates of an O-ring were 10% or 20%, the maximum of absolute te σ x was greater than the maximum of absolute σ y , but was almost equal. Maximums of absolute τ xy were 1 8 of absolute σ x and 1 5 of absolute σ x when the squeeze rates of the O-ring were 10% and 20%, respectively.

        A study on the development of photoelastic experimental hybrid method for colour isochromatics (II)

        Using the photoelastic experimental hybrid method, we can obtain stress intensity factors and separate the stress components near the crack tip of any material under various loading conditions from only isochromatics. However, monochromatic light has, until now been used in the photoelastic experimental hybrid method to produce black and white isochromatics. The existing photoelastic experimental hybrid method using black and white isochromatics requires high order fringes to obtain sufficient experimental data for photoelastic hybrid techniques. If a photoelastic experimental hybrid method for colour isochromatics is developed, isochromatics with a maximum fringe order of 2 will be adequate for gathering the necessary experimental data for stress analyses using photoelastic hybrid techniques. Therefore, in this work, photoelastic experimental hybrid method for colour isochromatics of a crack problem was developed, and its superiority demonstrated by comparing its results with the results from black and white isochromatics. The use of few fringe orders in photoelastic experimental hybrid method for colour isochromatics is an important feature of this method. Because of this feature, this method, specifically the reflection type photoelastic method where low fringe orders are produced compared to the transmission type photoelastic method, can be widely used in stress analysis of real components.

        A study on the development of a loading device using a photoelastic stress freezing method for the analysis of o-ring stress

        Typically, O-rings are used to prevent penetration of dust and alien substances from entering a cylinder during motion. Moreover, Orings are used to create an air tight seal around a stationary shaft. The stresses developed in O-rings depend on the squeeze rate, gap between the external diameter of the groove and internal diameter of the cylinder as well as internal pressure. In application, the stress distributions in O-rings can be very complicated and are almost always studied through experiment. Photoelastic experiment has been applied to the study of 3-dimensional stress distributions in O-rings. The loading device used in photoelastic experiment is important. Its function is to apply a uniform squeeze rate and internal pressure on the O-ring and to allow uniform squeeze rate to be controlled. In this research, a loading device was developed to perform these functions. The validity of this loading device was confirmed through the stress distribution, the configuration change and the contact length of the O-ring. When the squeeze rate was constant, the upper and lower contact lengths of the deformed O-ring were almost equal. When internal pressure was applied to the O-ring, while under a uniform squeeze rate the upper contact length increases slightly with increase in internal pressure, while the lower contact length of the O-ring is constant with an increase in internal pressure.

        털진득찰에서 분리한 Kirenol의 혈관 이완효과

        The purpose of this study is to investigate the vasodilatation effect of kirenol isolated from Sigesbeckia pubescens on the rabbit basilar artery. In this study, to determine the vasodilatation effect of kirenol on the rabbit basilar artery, arterial rings with intact or damaged endothelium were used for the experiment. And used an organ bath and force transducer were contracted by endothelin. Kirenol, major active constituents of S. pubescens, showed a moderate vasodilatation effect on the basilar arteries of rabbits. Therefore, treatment with kirenol may selectively accelerate cerebral blood flow through dilatation of the basilar artery. This result suggests a potential role of kirenol isolated from S. pubescens as a source of vasodilatation agent. 고혈압은 성인의 수축기 혈압이 140 ㎜Hg 이상이거나 이완기 혈압이 90 ㎜Hg 이상일 때를 의미하며 고혈압은 뇌졸중, 신부전 및 관상동맥질환 등 인체 전반에 걸쳐 다양한 합병증을 일으키며 고혈압 유병자의 생명과 건강을 직접적으로 위협한다(Kim and Kim, 2018). 그러나 고혈압 유병자들은 증상이 거의나타나지 않으므로 혈압을 측정하기 전까지는 진단되지 않는다. 진득찰 즉 한방에서 희렴은 고혈압을 치료하는 처방 제제로주로 사용됐기에 이에 착안하여 털진득찰을 이용하여 엔도델린(Endothelin)로 강제 수축 시킨 토끼 기저동맥의 이완 효과를평가하였다. 본 연구에서는 털진득찰(Sigesbeckia pubescens) 의 전초를 이용하여 키레놀 화합물을 분리 후 혈관 이완 효과를평가하였다. 인체 내 가장 강력한 혈관수축 물질로 알려진 엔도델린은 혈관 평활근 세포막에 존재하는 엔도델린 수용체 아형A (ETAR)와 혈관 내피세포막에 존재하는 수용체 아형 B2 (ETB2R)에 작용하여 혈관의 긴장도를 높이고, ETB1R에 작용하여 긴장도를 낮추는 조절자 임무를 수행한다(Lucchelli et al., 1999). 일반적으로 엔도델린 펩타이드의 경우 염증, 당뇨, 및 심혈관계 질환에서 혈중 농도가 증가하며, 엔도델린 체계의 비정상적 항진은 만성신부전 및 사구체 경화증과 같은 신질환, 특발성 폐섬유화증 및 만성폐쇄성 폐 질환 등의 호흡기계 질환, 대사질환의 중요한 관심거리가 되는 당뇨병성 신경병증 및 망막증, 수족괴사 등의 질환과 전립선 및 대장 등의 암질환 등을 초래한다(Lavoie et al., 1997; Pancrazio et al., 1998). 본 연구에서엔도델린 유도 뇌 기저동맥의 수축을 억제하는 키레놀의 농도(EC50)를 관찰한 결과 10 ㎍/mL의 농도에서 48% 이상의 유효한혈관 이완 효능이 관찰되었다. 따라서 키레놀을 이용하여 엔도델린 활성조절 신소재로의 구조적·기능적 도출 연구가 추가로진행된다면건강 기능성 식품 소재 또는 의약학 소재로의 개발이 가시화될 수 있을 것으로 사료된다.

        개소시랑개비 추출물의 RAW264.7대식세포에서 in vitro 항염효과

        Potentilla supina (Rosaceae) has traditionally been used to treat disorders of hemostasis, dysentery, malaria, bloody discharge and arthritis, and it has antinociceptive and anti-inflammatory properties. However, validity of the antiinflammatory activity has not been scientifically investigated so far. Therefore, the aim of this study was to investigate the anti-inflammatory potential of P. supina using the ethanolic extract of P. supina and its sub-fractions. To evaluate the anti-inflammatory effects of P. supina, we examined the inflammatory mediators such as nitric oxide (NO), inducible nitric oxide synthase (iNOS) and prostaglandin E2 (PGE2) in RAW 264.7 cells. Our results indicated that ethyl acetate fraction significantly inhibited LPS-induced NO, iNOS and PGE2 production in RAW 264.7 cells. This result showed that ethyl acetate fraction of P. supina is expected to be a good candidate for development into a source of anti-inflammatory agents.

        RAW 264.7 대식세포 염증반응에 대한 털연리초 에탄올 추출물의 항염증 효과

        Lathyrus palustris often used as a treatment for inflammation of the kidneys in Korean traditional medication. Generally, drugs for arthritis have anti-inflammatory and antinociceptive properties. However, the validity of the anti-inflammatory effect has not been scientifically investigated so far. Therefore, the purpose of the research was to investigate the latent anti-inflammatory ability of L. palustris using the ethanol extract. To evaluate the anti-inflammatory activities, we examined the inflammatory arbitrators such as a nitric oxide (NO) and prostaglandin E2 (PGE2) on RAW 264.7 cells. Our results indicated that ethanol extract significantly inhibited the lipopolysaccharide E (LPS) derived PGE2 production in RAW 264.7 cell. The inhibitory activity of ethanol extract for PGE2 tests with inhibition ratio showed in 40 ㎍ /mL. Overall, PGE2 tests had a higher inhibitory effect on inflammation than NO tests. This result anticipated that the ethanol extract from L. palustris is a good candidate for developing the origin of anti-inflammatory agents. 염증은 신체 특정 조직의 감염 및 손상에 관한 생체 반응이며, 매개하는 주요 대상은 면역세포이다. 염증은 급성과 만성 염증으로 나뉘며 신체 조직의 감염 및 손상부위의 규모에 따라 구분할 수 있다. 염증의 범위가 크게 발현되거나 급성염증 형태로 진행되지 않을 때 만성 염증으로 진행되며 대표적인 만성 염증 질환인 장 질환(Inflammatory bowel disease)의 일종인 크론병(Crohn’s disease)이나 관절질환인 류머티스성 관절염(Rheu�matoid arthritis)으로 나타난다. 낮은 수준이기는 하나 비만 역시 염증성 질환으로 분류할 수 있다. 연리초속 식물이 고래에 신장염을 치료하는 민간처방으로 주로 사용됐기에 이에 착안하여털연리초(Lathyrus palustris)를 이용하여 세포독성과 항염증활성 효과를 평가하였다. 대식세포인 RAW 264.7 세포에서 염증 유발 인자인 lipopolysaccharide (LPS)로 자극 후 NO와PGE2 같은 염증 매개 물질들의 억제 효과를 확인하였다. 털연리초 에탄올 추출물을 처리한 후 염증 매개 물질의 저해율(%)을 측정했을 때 NO 및 PGE2 생성을 농도 의존적으로 현저하게 억제하는 농도는 40 ㎍/mL이었으며 특이적으로 PGE2 발현을74% 이상 강력히 억제함을 확인하였다. 따라서 본 연구결과는 털연리초의 에탄올 추출물이 유의성 있는 항염증 효과를 나타내었고 이러한 생리활성 효과는 예방의학적 소재로서의 가능성을충분히 제시할 수 있기에 염증 질환의 예방 및 비만 억제를 위한기능성 건강식품의 개발로 이어질 것으로 기대된다. 또한 염증과 관련된 사이토카인 물질인 IL-4, IL-13 및 염증 지표 단백질인 iNOS, COX-2의 억제 메커니즘과 항염증 활성을 나타내는핵심 성분의 추가적인 연구가 차후 필요할 것으로 판단된다.

        지대 및 품종(재배종)별 콩사포닌그룹B 함량의 비교

        Contents of soyasaponin group B were compared according to two regions and ten varieties by HPLC. Those compounds were known to be beneficial for health. After soyasaponins were isolated and identified, those isolated compounds were used for HPLC analysis. The contents of soyasaponin were very different by regions but highest in the soybean with black seed coat. It was appeared that environmental difference for soybean growth could strongly change of soyasaponin contents. Soyasaponin group B함량의 재배지대간 비교에서는고랭지와 평난지에서 상이한 차이를 보였으며 품종 간에는대부분 자엽색이 녹색이고 종피색이 검정색인 콩의 soyasaponin group B함량이 높은 것으로 나타났다. 이상의 결과를종합해 볼 때 soyasaponin성분의 함량은 재배종의 종류즉 유전적인 요인뿐만 아니라 고랭지와 평난지처럼 환경적요인이라고 것을 확인할 수 있었으며 이는 식물이 환경적인 스트레스에 대한 생명유지 및 공격방어물질(Secondary matabolites,이차대사산물)이 식물내 생합성과정 중에서변이를 보이는 것으로 사료되어 진다.

        RAW264.7 대식세포에서 풀솜대 추출물의 nitric oxide 및prostaglandin E2생성 저해효과

        Smilacina japonica is a localized commonrhizomatous flowering plant, This plant is often used inKorean traditional systems of medicine as a remedy formigrain, diplegia, physical impurity, blood circulation,abscess and contusion. Generally drugs that are used forarthritis have antinociceptive and anti-inflammatory properties. However, validity of the anti-inflammatory activity has notbeen scientifically investigated so far. Therefore, the aim ofthis study was to investigate the anti-inflammatory potentialof S. japonica using the ethanolic extract and its subfractions. To evaluate the anti-inflammatory effects, weexamined the inflammatory mediators such as nitric oxide(NO) and prostaglandin E2 (PGE2) on RAW 264.7macrophages. Our results indicated that hexane fractionsignificantly inhibited the LPS induced NO and PGE2production in the cells. The hexane fractions inhibitoryactivity for NO tests with IC50 values showed in 53.3 μg/mland PGE2 tests with IC50 values showed at 32.5 μg/ml. Theseis result suggest a potential role of hexane fractionfrom S. japonica as source of anti-inflammatory agent.
