      • Quantitative Assay for the Binding of Jun-Fos Dimer and Activator Protein-1 Site

        The Jun and Fos families of eukaryotic transcription factors form heterodimers capable of binding to their cognate DNA enhancer elements. We are interested in searching for inhibitors or antagonists of the binding of the Jun-Fos heterodimer to the activator protein-1 (AP-1) site. The basic-region leucine zipper (bZIP) domain of c-Fos was expressed as a fusion protein with glutathione S-transferase, and allowed to form a heterodimer with the bZIP domain of c-Jun. The heterodimer was bound to glutathione-agarose, to which were added radiolabeled AP-1 nucleotides. After thorough washing, the gel-bound radioactivity was counted. The assay is faster than the coventional electrophoretic mobility shift assay because the gel electrophoresis step and the autoradiography step are eliminated. Moreover, the assay is very sensitive, allowing the detection of picomolar quantities of nucleotides, and is not affected by up to 50% dimethylsulfoxide, a solvent for hydrophobic inhibitors. Curcumin and dihydroguaiaretic acid, recently known inhibitors of Jun-Fos-DNA complex formation, were applied to this Jun-GST-fused Fos system and revealed to decrease the dimer-DNA binding.

        충남대학교 학술림 산책로 훼손실태 및 관리방안에 관한 연구

        This study concentrates an the trail of the CNU(Chungnam National University) forest. The amount of use and the pattern of damage are considered, as well as the extent of environmental damage. Finally, some managerial strategies are proposed in connection with sustainable maintenance. The trail of the university forest is composed of three sections; one is the neighborhood park and the Institute of Information Technology/Communications, the second is the Institute of Information Technology/Communications and the Dormitory, and the third section is the Dormitory and the Foreign prof's Apt. The total length of the trail is 2.6km. The daily amount of use is 1,269 visitors per day. The amount of using increased three times the amount of that in 1998. This study shows the pattern of damage of the trail in the order of root-exposure, widening, rock-exposure, deepening and the divergence in the frequency of occurrence of all these factors. Environmental damage is also graded thus: grade 3 is 27.4m, grade 4 is 0.8m, grade 5 is 1.5m, and grade 6 is 0.2m. Accordingly, some managerial strategies were proposed in connection with the sustainable trail maintenance.

        비응축성 기체 존재 시 수직관 외벽에서의 응축 열전달계수 상관식 개발

        본 연구에서는 비응축성 기체(공기)가 존재할 때, 10 mm의 직경을 갖는 수직 튜브 외벽에서의 응축 열전달계수를 측정하였다. 압력 2~5 bar, 공기 질량분율 10~75 %의 범위에서 실험을 수행하였다. 응축튜브 직경의 차이가 열전달계수에 미치는 영향을 평가하기 위해 측정 결과를 외경 40 mm의 응축튜브로부터 얻은 Lee의 실험 결과와 비교하였다. 본 연구의 측정결과와 Lee, 그리고 Dehbi의 실험결과를 이용하여 전열관의 직경에 의한 영향을 고려할 수 있는 실험적 상관식을 제안하였다. 제안된 상관식의 검증을 위해 Kawakubo의 실험결과와 예측값을 비교하였고, 평균 표준편차는 약 27 %로 나타났다. In this study, condensation heat transfer coefficients are measured in the presence of noncondensable gas (i.e., air) on a vertical tube with a 10-mm outer diameter. Experiments are conducted at pressures ranging from 2 to 5 bar and an air-mass fraction from 10 to 75 %. The experimental results are compared to the heat transfer coefficients obtained on a 40-mm O.D. tube, researched by Lee et al. Using the consolidated data from this study and Lee"s and Dehbi"s tests, a new empirical correlation for the condensation heat transfer coefficient of an air-steam mixture is developed. The proposed correlation is assessed against Kawakubo"s experimental results, and the mean standard deviation is ~27 %.

      • SD Rats를 이용 1-hexene의 흡입독성 연구

        The purpose of this study was to investigate the acute (4 hours) and repeated-dose (6 hours a day, 5 days a week, 4 weeks) toxic effects of I -hexene on Sprague- Dawley (SD) rats which were treated by inhalation. The results were as follows; I. The median lethal concentration (LC_(50)) was estimated 52,694 ppm (confidence limit 95%; 49,494~55,447 ppm) in acute inhalation. Abnormal clinical signs related to the l-Hexene were not observed with the acute inhalation dose. Gross findings of necropsy revealed on evidence of specific toxicity related to the 1-hexene, II. By repeated inhalation exposure the body weight of male were more or less reduced by the dose of 2,500 ppm and 5,000 ppm compared with control group. However there were no significant variation hematology and blood biochemistry for the exposed rats compared with the control rats. Abnormal clinical signs and gross findings of necropsy related to the 1-hexene were not shown. In conclusion when we exposed I-hexene to SD rats for 4 weeks, 5 days per week, 6 hours per day, the Lowest observed effect level (LOEL) was over 2,500 ppm and Non observed effect level (NOEL) was below 500 ppm.

      • Outsole 경도에 따른 피로도 분석에 관한 연구

        대형 할인업체간의 경쟁이 치열해지면서 계산원들의 업무가 복잡해졌다. 계산원의 업무 특성상 장시간 동안 한정된 장소에서 정적 기립자세를 유발하며 국소적 근육활동의 사용은 근육통과 피로누적 등 근골격계질환 발생 가능성을 가지고 있다. 본 연구에서는 이러한 문제점을 보완하기 위하여 근전도 측정을 통해 outsole의 경도에 따른 피로도를 비교분석하고자 하고자 한다. Outsole은 C형 경도계를 이용하여 경도(hardness)가 30~35인 경우를 Soft라 하고, 경도가 45~50을 Hard라 하였다. 계산원의 작업시간에 따른 변화를 알아보기 위해 작업시간은 0시간, 2시간, 4시간으로 구분하였고, 측정부위는 종아리(비복근, gastrocnemius), 허벅지(외측광근, vastus lateralis), 허리(척주기립근, erector spine)로 선정하였다. Outsole 경도에 따른 근전도 분석은 MF(Median Frequency), MPF(Mean Power Frequency), ZCR(Zero Crossing Rate) 3부류로 나누어 분석하였고 측정 부위별, outsole의 경도별 그리고 시간별 간의 통계적 유의성 분석을 실시하였다. MF, MPF, ZCR 분석 결과 세 측정 부위 중 종아리에서만 90% 유의 수준에서 유의성이 나타났으며 허벅지와 허리는 50%~80%정도의 유의 수준에서 유의한 차이를 보였다. Outsole의 경도별 분석은 Hard보다 Soft의 값이 현저히 낮게 나타났음을 알 수 있었고, 작업 시간간의 통계적 분석 결과는 20~30% 유의 수준에서 유의한 차이를 보였다. 이는 4시간 이내의 작업에서는 현 계산원들의 작업 시 허리 및 하지에 미치는 피로도 간의 차이가 크게 나타나지 않음을 알 수 있으며 허벅지와 허리보다 종아리에 있어서 피로도 감소 효과가 크게 나타남을 알 수 있다. 또한 45~50정도의 경도를 가진 outsole(hard)의 영향보다 30~35정도의 경도를 가지는 outsole(soft)이 피로감소에 따른 효과가 큼을 알 수 있다. 따라서 장시간 정적인 작업 자세에 있어서 단순 반복작업을 요하는 작업에서는 경도가 30~35정도의 soft type의 outsole 사용이 권장된다.

        우리 나라 종합병원 진료재료 구매와 재고관리 질 향상 방안에 관한 연구

        This study was conducted to investigate the current situation of medical supply purchasing and stock management at general hospitals having more than 150 beds in Korea and to find methods of effective purchasing and optimal stock management. Methods : Survey was done from staff at the purchasing departments of 299 general hospitals throughout Korea. Data collection was done using a structured questionnaire between January 3 to March 15, 2001. The survey form was returned from 88 hospitals (rate of return:38.4%). Results : Firstly, 13.6% of the hospitals did not carry the optimal stock of medical supplies, the lead time optimal stock was 3 weeks or longer in 64.4% of the hospitals. Secondly, since 69.8% of the hospitals showed passive attitude toward training on purchasing management and stock management techniques. Thirdly, as for the question on the presence or absence of a deliberation committee for purchasing of new medical supplies, 60% of the hospitals with less than 300 beds did not have one, and 9.4% of the hospitals opened the deliberation committee less than twice a year. Conclusion : At the time of purchasing new medical supplies, purchasing should be done according to the decision by the deliberation committee so that no deduction is made at the time of claiming insurance, and by setting a certain period of time, purchasing of those medical supplies that were not purchased during this period needs to be done according to the decision by the deliberation committee.

      • 균일 가열 수직 원형관에서의 R-12 임계열유속 예측 상관식 개발을 위한 파라미터의 경향성 분석

        The important parameters that have significant effects on the prediction of CHF (Critical Heat Flux) for R-12 (Dichlorodifluoromethane) were investigated. This was a preliminary study for a R-12 CHF correlation in uniformly heated vertical round tubes. Experimental data of Celata [1], Green and Stevens [2], Groenveld [3], Ilic [4], Katto [5], Merilo and Ahmad [6] and Stevens and Miles [7] were used for this analysis. Four representative CHF data sets at pressure condition of 9, 15, 19, and 27 bar were selected, analyzed, and compared to evaluate the effects other than the system pressure on the CHF were tube diameter (D), mass flux of water (G), and local true mass fraction of vapor (X_(t)). The results of this study have reaffirmed the feasibility that an advanced R-12 CHF correlation for uniformly heated vertical round tubes can be found.

      • Streptokinase와 Plasminogen의 활성 복합체에 대한 생화학적 특성

        Fig 1과 Fig 2의 실험 결과에 의하면 streptokinase에 의한 plasminogen의 활성화 기작은 먼저 streptokinase와 plasminogen 결합하여 복합체를 만들고 그 다음 다른 분자의 streptokinase에 의한 plasminogen을 streptokinase에 의한 plasmin으로 활성화 한다고 생각되었다.그리고 원래 효소활성이 없는 streptokinase량이 plasminogen보다 과량으로 존재하게 되면 전체 esterolytic 활성도는 plasminogen 의 량에 의존하게 될 것이다.Fig 3의 실험에서 streptokinase plasminogen 복합체의 정량적인 구성 비율을 조사하였더니 plasminogen 1㎍에 0.5㎍의 streptokinase가 결합하였다. 이는 보고된 plasminogen의 분자량을 87.000(10) 그리고 streptokinase분자량이 47.000(6)으로 계산하면 각각 한 분자씩 결합하고 있음을 알 수 있었다. 이 결과는 Fig. 1의 결과를 보충할 수 있는 data이다. streptokinase plasminogen 복합체의 생화학적 특성을 조사하기 위하여 복합체만의 활성을 assay 할 수 있는 방법을 고안하였다. 실험결과 soybean trysin inhibitor를 사용하면 plasminogen의 활성화에 의하여 생성된 plasmin의 esterolytic 활성은 완벽하게 저해할 수 있었으나 streptokinase plasminogen 복합체의 활성은 저해하지 못하였으므로 soybean trypsin inhibitor를 첨가하여plasmin활성을 제거하여 순수한 복합체만의 활성을 측정할 수 있었다.(Fig.5).그러나 pancreatic tyrpsin inhibitor는 Plasmin 뿐만 아니라 streptokinase plasminogen복합체의 활성을 모두 저해할 수 있었으므로 이 inhibitor 는 복합체의 활성을 측정하는데 사용하지 않았다. (Fig.4). 두가지 inhibitor의 복합체에 대한 상이한 저해 효과는 복합체의 활성화 자리 (active site)에 두 inhibitor 가 결합하고난 후 기질이 출입할 수 있는 공간의 넓이에 의한 효과인 것으로 생각되었다 (Table 1).따라서 soybean trypsin inhibitor을 이용한 assay는 원래의 생화확적 특성에 영향이 없는 것으로 생각되었다. 복합체의 esterolytic 반응은 빠른 첫 번째 반응 그리고 그 이후 늦은 두 번째 반응으로 진행되는 biphasic한 형태를 보여 주었다(Fig.7).이 반응 기작은 알려진 serine protease들이 acyl중간체을 이용한 biphasic 반응 기작과 유사한 형태이었다. 활성화 자리에 위치하고 있는 여러 작용기들을 확인하기 위한 기초 실험으로써 pH에 따른 esterolytic활성을 측정한 결과 최대 활성을 보이는 pH는 7.5이었다.(Fig.8). 이 근처의 pKa 값을 가지고 아미노산은 histidine 종류을 생각할 수있을 것이다. The kinetic data suggest that streptokinase interacts with plasminogen to become an active complex which in turn. activates another molecule of plasminogen to plasmin. This complex was fromed stoichiometrically at 1:1 molar ratio. A series of experiment were carried out to find the condition that the streptokinase plasminogen complex activities free from plasmin could be monitered. The soybean trypsin inhibitor wsas used to assay the activity of streptokinase-plasminogen complex. The lack of inhibiton of esterase activity by soybean trypsin inhibitor was thought to be the size of the protein in active site. The esterolytic reaction by streptokinase-plasminogen complex showed biphasic pattern. The pH profile showed typical bell-shape and optimal pH for streptokinas-plasminogen complex showed biphasic pattern. The pH profile showed a typical bell-shape and optimal pH for streptokinas-plasminogen complex was 7.5.

        Ziprasidone의 자연적 임상경험에 대한 후향적 조사 연구 : 3개 대학병원의 경험

        Objectives : This naturalistic retrospective study investigated the effectiveness of ziprasidone measured by mean improve-ments of the Cltnical Global Impression (CGI-I). Secondary objectives included prescribing patterns, tolerability and safety. Methods : From May to July 2006, 98 psychiatric patients who have been taking ziprasidone for at least 2 weeks were selected. Their current clinical statuses were assessed by the interview and laboratory tests including electrocardiogram (ECG). Data on DSM-IV diagnoses, dosing, concomitant psychotropic medications, clinical changes, adverse effects, and laboratory findings were collected retrospectively. Results : Ziprasidone was mostly prescribed for a switch from another antipsychotics for the treatment of schizophrenia. Two main reasons for a switch were unsatisfactory efficacy and poor tolerance, especially weight gain, of the preceding anti-psychotic medication, Participants as a whole showed mean 2.7±1.0 of the CGI-Improvement. Significant weight reduction after switching to ziprasidone occurred, which was related to the clinical improvement. No significant increase of the QTc interval except one case was observed. Conclusion : The present study confirmed previous tolerability and safety data of ziprasidone as well as the results of its effectiveness.
