        석유화학공업 종사자의 유전독성 위험성 평가

        In order to evaluate the genotoxic hazard among workers potentially exposed to low level petrochemical substances, the analyses of micronuclei(MN) and sister chromatid exchanges(SCEs) in lymphocytes were performed in 46 male workers(as exposed group) and 46 nonexposed subjects(as control group). Mean frequencies of MN and SCEs(respectively, 12.9/1000 cells and 6.5/cell) in exposed group were very significantly higher than those(10.2/1000 cells and 5.4/cell) in control group. And there were also significant differences in mean frequencies of MN and SCEs adjusted for age, employment duration, smoking, and drinking between two groups. Median frequencies of MN and SCEs in exposed group were very significantly higher than those in control group. Frequencies of SCEs were higher in smokers than in non-smoker. Frequencies of MN in smokers, however, were similiar to those of non-smoker. Interaction between exposure and smoking on MN and SCEs induction was not observed. The results suggest that there is genotoxic hazard in high risk group like workers handling carcinogens in petrochemical plants and the analyses of MN and SCEs are useful biomarkers for the exposure to hazard substances even at the level below the exposure limit.

      • 녹동균 세포외막 단백질 백신 CFC-1-101의 안정성 및 면역원성 검토 : 임상 제 Ⅰ/Ⅱa상 시험

        목적 : 제일제당에서는 녹농균의 세포외막 단밸질을 유효성분으로 하는 백신인 CFC-101을 개발하였으며, 동물시험에서 이 백신의 안전성과 유효성을 입증하였다. 본 연구에서는 이 녹농균 백신의 인체에 대한 안전성과 면역원성을 평가하는 동시에 인체 접종시의 최적 투여 용량을 결정하기 위하여 제 I/Ⅱa상 임상시험을 수행하였다. 방법 : 건강한 성인 남자를 피험자로 선별하여 각 용량군에 백신투여자 6명, 위약투여자 2명을 배정하였다. 백신 투여군은 0.25mg, 0.5mg 또는 1.0mg 용량의 녹농균 백신을 7일 간격으로 3회에 걸쳐 근육주사 하였으며, 위약 투여군에게는 세포외막 단백질을 제외한 동일한 성분을 투여하였다. 백신접종 후 국소적 또는 전신적인 반응의 발생여부를 관찰하고, 혈액시료를 체취하여 백신의 역가와 유효성을 검정하였다. 결과 : 녹농균 백신 CFC-101은 모든 접종자에서 양호한 내약성을 보였다. 또한 0.5mg 과 1.0mg 백신 투여군에서는 100%의 항체양전율을 나타내었다. 생성된 항체는 녹농균 세포외막단백질에 특이성을 보였고, 녹농균 감염에 대해 방어효능이 있었다. 결론 : 이와같은 결과로부터 이 녹농균 백신은 인체에 안전하게 투여할 수 있으며, 높은 항체 생성능으로 감염방어 효능을 보이고 0.5mg과 1.0mg이 최적용량인 것으로 판단되었다. Background : We developed a Pseudomonas aeruginosa outer membrane protein(OMP) vaccine CFC-101, and the prophylactic efficacy of which has been demonstrated in animal models. In order to evaluate the safety and immunogenicity of the P. aeruginosa vaccine, we carried out a phase I/Ⅱa clinical trial in healthy male volunteers. Methods : Groups of eight volunteers, including two placebo subjects, were vaccinated intramuscularly with three doses of 0.25, 0.5 or 1.0 mg of the vaccine at one week intervals. Sings of systemic and local reactions observed after vaccination were recorded for each vaccinee for 5 days. Physical examinations were performed on days 0, 1, 7, 8, 14, 15, 21, and 42, and clinical laboratory tests were done on days 0, 3, and 21. Blood samples for assay of serum antibody levels were obtained up to 42 days after the first vaccination. Results : The vaccine was generally well tolerated by all vaccinees, showing no significant side effects. In the three dosage groups, all vaccinees, except one receiving the 0.25 mg dose, showed significant elevation in serum IgG antibody titers against the vaccine proteins, indicating 100% seroconversion in 0.5 and 1.0 mg groups. The human antibodies induced by the vaccine were specific for P. aeruginosa OMPs, as confirmed by western blot analysis and immunoprecipitation assays. The capacity of the human antisera to enhance opsonophagocytic killing activity by polymorphonuclear leukocytes and to confer protection against P. aeruginosa infections indicates that the antibodies elicited by the vaccine have protective efficacy. Conclusion : We conclude that the P. aeruginosa OMP vaccine is safe and effective for human use and its optimal dose to be 0.5 or 1.0 mg.

        In a switched reluctance motor drive, it is important to synchronize the stator phase excitation with the rotor position, because the position of rotor is an essential information. In the high-speed region, switching angles are fluctuated back and forth out of the preset value, which is caused by the sampling period of the microprocessor. In this paper, a low cost analog encoder suitable for practical applications is proposed. The validity of the proposed analog encoder with a proper logic controller is verified from the experiments.

        재래종 콩 유전자원의 주요 형태적 특성과 변이

        본 실험은 한국, 중국 및 일본 재래종 콩 유전자 자원의 주요형질 변이를 조사하고 이들 자원간의 지리적 유연관계를 비교하고자 수행하였으며, 그 결과를 요약하면 다음과 같다. 1. 경장은 한국(66㎝)이나 일본자원(54.9㎝)보다 중국자원(71㎝)에서 더 컸으며, 또한 변이도 중국자원에서 가장 큰 것으로 나타났다. 2. 개화일수 및 생육일수는 한국자원(65.5일, 151.4일)이 가장 길었으며 중국(57일, 120일)과 일본자원(59.9일, 122.4일)은 같은 경향을 나타냈다. 또한 한국과 중국자원은 지역에 따른 차이를 나타냈으나 일본자원에서는 확인할 수 없었다. 3. 백립중은 한국(18.8g), 일본(17.2g) 및 중국(14.7g) 순으로 무거웠다. 특히 한국자원은 9~11g과 23~25g에서 정점을 나타냈고, 변이폭도 가장 컸다. 반면 일본과 중국자원은 13~17g사이에서 정점을 나타냈다. 4. 협수는 중국(46.9개)이나 일본자원(36.5개)보다 한국자원(75.5개)에서 가장 많았으며, 변이폭도 한국자원에서 가장 컸다. 내도복성은 일본자원에서 가장 강하였다. 5. 정준판별분석결과 can 1을 경계로 하여 오른쪽 상하에 한국재래종이 주로 분포하였고 왼쪽에는 중국과 일본 재래종이 주로 분포하여 각 나라별로 고유한 영역을 가지고 분리되었다. 전체적 경향이 중국과 일본자원이 한국자원보다 가까운 경향을 나타냈다. This experiment was carried out to compare the characters of 944 Korean soybean landraces, 716 Chinese, and 170 Japanese soybean landraces introduced from USDA soybean germplasm collection. Cannonical discriminant and cluster analyses were conducted by their origins. The population from China was taller in plant height than those from Korea and Japan, also its variation was largest in Chinese population. The population from Korea was langer than those from China and Japan in terms of days to flowering and maturity. Also, Korean and Chinese accessions appeared the difference of those traits according to latitude. One hundred seeds weight was in the order of Korean(18.8g), Japanese(17.2g) and Chinese populations(14.7g), especially Korean population was distinguished into two groups; the group including accessions between 10 and 15g, and the other group including accessions between 20 and 25g. However, others showed the peak in range of 13~17g. The number of pod per plant was more in Korean accession(75.5) with the largest variation than Chinese(46.9) and Japanese accession(36.5). The distribution of Korean, Chinese and Japanese accessions reflected the origin of accessions analyzed by cannonical discriminant analysis. The relationship between their geographical origins showed the population of China and Japan are losely related.

        치관보철용 글라스 세라믹 재료에서 조성비 변화에 따른 기계적 특성에 관한 연구

        Glass ceramics for dental crown prosthesis were prepared by heat-treatment of CaO-MaO-SiO₂-P₂O -TiO₂glasses. The crystallization behaviors of samples have been investigated as a function of heat-treated temperature, holding time and chemical composition with their mechanical properties. From the experiment, the following results were obtained: The nucleation rate of apatite increased with increasing CaO or P₂O and decreasing MgO or TiO₂. Vickers hardness and bending strength of samples increased with increasing MgO or TiO₂ and decreasing CaO or P₂O. Vickers hardness of samples increased due to the precipitation of apatite, whitlockite, β-wollastonite, magnesium titanate, and diopside crystal phases within glass matrix. The final crystalline phase assemblages and the microstructures of the samples were found to be dependent on glass compositions, heat-treatment temperature, and holding time.

        클럽 샤프트(Club Shaft) 특성에 따른 골프 스윙(Golf Swing)동작 분석

        본 연구는 클럽 샤프트의 대표적인 재질인 그라파이트(graphite)의 유연한(flexible) 클럽 샤프트(club shaft)의 특성에 따라 피험자가 운동학적(kinematics) 요인이 되는 관절의 각변위, 각속도, 각가속도, 클럽헤드의 속도와 가속도와 같은 변인들이 어떻게 적응하는지 비교 분석하여 보다 효율적인 드라이버 선택에 도움을 주며 샤프트 특성에 따른 신체관절의 움직임에 대한 자료를 제시하고자 하였다. 고속 카메라 2대의 속도는 500fps로 하였고 각 regylar,stiff, x-stiff, 자신의 클럽을 포함 각 4개의 클럽을 사용하여 각 클럽당 3번씩 촬영하였으며 목표방향에서 20m이상 벗어나는 경우의 촬영은 다시 촬영하였다. 본 연구에서는 디지타이징(digitizing)을 신체 9개 마커는 강체로 가정된 클럽과 신체분절 모델로 정의하였으며 2 대의 카메라(500fps)로부터 얻은 avi화일을 컴퓨터에 저장하고 자료로부터 Butterworth 6th order recursive digital filter를 사용하여 1차 자료를 smoothing 하고 DLT를 이용하여 3차원 좌표를 구성하도록 한다. 좌표값을 얻기 위하여 kwon3d v3.0을 이용하였다. 본 실험은 피험자 스스로 클럽의 특성에 따라 스윙의 속도를 달리 하기 때문에 스윙의 시간이 달라지며 어느 정도 클럽이 강성에 따라 스윙시간이 빨라지는 결과로 나타났다. 이것은 피험자가 샤프트가 강성(stiffness)에 따라 스윙 속도를 빨리 하게 되는 원인이 되는 것으로 생각된다. 어깨의 각변위는 클럽이 regular의 경우 임팩트에서 각속도를 계속 유지하고 있으며 stiff, x-stiff의 경우에는 어깨의 움직임이 임팩트에서 급격하게 감소되는 것을 알 수 있다. 이것은 팔의 동작과 클럽의 힘을 크게 하기 위한 동작으로 생각된다. 어깨 각속도는 클럽이 stiff할수록 각속도가 큰 감속하는 것으로 나타났다. 손목속도는 regular 클럽의 경우 손목의 감속이 늦게 되고 임팩트에서 손목의 감속이 적게 하는 것으로 나타났으며 stiff와 x-stiff의 클럽에서 임팩트 시에 순간적인 감가속으로 인해 클럽의 속도를 증가시키고 있다. 임팩트 시에 손목의 감가속은 클럽헤드의 임팩트 시 속도를 증가시키는 결과를 보였다. 클럽헤드는 regular 클럽이 임팩트전에는 속도 증가가 커지는 결과와 일치된 결과를 보이고 있다. The purpose of this study was to find the rational method to analyze golf swing with specific property of club shaft. Three subjects were filmed by two high speed digital cameras with 500fps. The phase analyzed was downswing of each subject. The three-dimensional coordinates of the anatomical landmarks were obtained with motion analysis system Kwon3d 3.0 version and smoothed by lowpass digital filter with cutoff frequency 6Hz. From these data, kinematic and kinetic variables were calculated using Matlab(ver 5.0) The variables for this study were angular velocity and accelerations, which were calculated and following conclusions have been made : 1) Golf swing time of stiff club is faster than that of regular club. 2) In shoulder joint motion of swing with the stiff club, x-stiff showed mort rapid negative acceleration than that of regular club. 3) In regular club, the velocity of club head would be more effective velocity, which was increasing, than those of other clubs before impact. 4) In wrist joint motion of swing with stiff club, x-stiff club showed faster than regular club in the downswing and impact more rapid negative acceleration.

      • 폐결핵 환자 혈청에서 수용성 Interleukin 2 수용체 및 Adenosine Deaminase 활성도에 관한 연구

        Interleukin-2 (IL-2) induces T cell proliferation in an autocrine manner and provides a means by which antigen triggered T cells can be clonally expanded in vitro. During the following activation, the activity of IL-2 is mediated by specific high affinity IL-2 binding membrane receptors which are expressed shortly after activation. In this process, a 42 KD-fragment (soluble IL-2R) is continuously cleaved off and circulates as a soluble marker of T lymphocyte activation. Elevated level of soluble IL-2R has been identified in the serum of patients with malignant autoimmune and allergic disorders, systemic parasitic infection, undergoing graft versus host disease, acute or chronic lymphocytic leukemia and HIV-infection. ADA (adenosine deaminase) completes the process of differentiation of T cell and is essential for progression of T cell maturation. Therefore level of ADA is to correlated with magnitude of T cell immune response. The fact that expression of sIL-2R and ADA activity increases in peripheral blood T-lymphocytes of patients with active pulmonary tuberculosis suggests that T cell activation might have a major role in the pathogenesis of pulmonary tuberculosis. In order to evaluate the T cell immune response in pulmonary tuberculosis, we measured the serum concentration of sIL-2R and ADA activity in 17 patients with current pulmonary tuberculosis, 10 chronic inactive pulmonary tuberculosis, 10 as normal controls. (1) Current pulmonary tuberculosis had significantly higher levels of sIL-2R (237.24±95.47)when compared with those of inactive tuberculosis (78.6±11.06). and the control (68.17±15.4) group. (2) ADA activity in current pulmonary tuberculosis was significantly increased (34.41±20.63) when compared with those of inactive tuberculosis (24.7±14.36) and control (17.65± 5.94) group. (3) There was good correlation between sIL-2R concentration and ADA activity in serum in current pulmonary tuberculosis group. (4) sIL-2R concentration and ADA activity was decreased significantly 6 months after anti-tuberculosis drug medication. In conclusion, sIL-2R concentration and ADA activity in serum in current pulmonary tuberculosis group was increased when compared with those of inactive pulmonary tuberculosis and the control group. By the way, T cell mediated immune response was enhanced in current pulmonary tuberculosis, but in inactive chronic pulmonary tuberculosis who had treated by antituberculosis drug medication, the concentration of sIL-2R and ADA activity was nearly normal.

      • 심한 척추후측만증 환자의 대퇴골절 복원술을 위한 부위마취 : 증례보고 A case report

        Kyphoscoliosis is a disease manifested as lateral curvature of spine and accompanying rotation of the vertebrae, resulting in distortion of rib cage. Restrictive lung disease and pulmonary hypertension progressing to cor pulmonale are the major cause of mortality in patients with kyphoscoliosis. We had experienced a successful spinal and caudal epidural anesthesia for the open reduction and internal fixation with plate of fractured femur shaft in two patients with severe kyphosoliosis. We had chosen regional anesthesia as an anesthetic method because of there poor pulmonary function and severe deformities of thoracolumbar spine. We could achieve the adequate anesthetic sensory dermatome level, T_(5) and T_(8), for operation without any significant deterioration of blood pressure and respiration. We recommended that regional anesthesia is a useful anesthetic method for a patients with severe kyphoscoliosis when the operation on hip or lower extremity is required.
