RBP2 induces stem-like cancer cells by promoting EMT and is a prognostic marker for renal cell carcinoma

        Dahai Zhou,Vinodh Kannappan,Xiang Chen,Jingqin Li,Xuefeng Leng,Jinping Zhang,Shiying Xuan 생화학분자생물학회 2016 Experimental and molecular medicine Vol.48 No.-

Renal cell carcinoma (RCC), one of the most common kidney cancers, has a poor prognosis. Epithelial to mesenchymal transition (EMT) is a hallmark of carcinoma invasion and metastasis. Several studies have examined the molecular regulation of EMT, but the relationship between histone demethylases and EMT is little understood. In this study, we investigated the role of retinoblastoma-binding protein-2 (RBP2), a histone demethylase that is highly expressed in RCC and is positively correlated with poor RCC prognosis in the regulation of EMT. We found that ectopic overexpression of RBP2 can induce cancer stem cell-like (CSC) phenotypes through EMT in RCC cells by converting them to a more mesenchymal phenotype. This results in increased resistance to apoptosis, which leads to enhanced tumor growth in xenograft models. Together, our data show that RBP2 is an epigenetic regulator that has an important role in the initiation of CSC phenotypes through EMT, leading to tumor progression. RBP2 is also a novel biomolecule for RCC diagnosis, and prognosis and may be a therapeutic target.

Pituitary P62 deficiency leads to female infertility by impairing luteinizing hormone production

        Li Xing,Zhou Ling,Peng Guiliang,Liao Mingyu,Zhang Linlin,Hu Hua,Long Ling,Tang Xuefeng,Qu Hua,Shao Jiaqing,Zheng Hongting,Long Min 생화학분자생물학회 2021 Experimental and molecular medicine Vol.53 No.-

P62 is a protein adaptor for various metabolic processes. Mice that lack p62 develop adult-onset obesity. However, investigations on p62 in reproductive dysfunction are rare. In the present study, we explored the effect of p62 on the reproductive system. P62 deficiency-induced reproductive dysfunction occurred at a young age (8 week old). Young systemic p62 knockout (p62 -/- ) and pituitary-specific p62 knockout (p62 flox/flox αGSU cre ) mice both presented a normal metabolic state, whereas they displayed infertility phenotypes (attenuated breeding success rates, impaired folliculogenesis and ovulation, etc.) with decreased luteinizing hormone (LH) expression and production. Consistently, in an infertility model of polycystic ovary syndrome (PCOS), pituitary p62 mRNA was positively correlated with LH levels. Mechanistically, p62 -/- pituitary RNA sequencing showed a significant downregulation of the mitochondrial oxidative phosphorylation (OXPHOS) pathway. In vitro experiments using the pituitary gonadotroph cell line LβT2 and siRNA/shRNA/plasmid confirmed that p62 modulated LH synthesis and secretion via mitochondrial OXPHOS function, especially Ndufa2, a component molecule of mitochondrial complex I, as verified by Seahorse and rescue tests. After screening OXPHOS markers, Ndufa2 was found to positively regulate LH production in LβT2 cells. Furthermore, the gonadotropin-releasing hormone (GnRH)-stimulating test in p62 flox/flox αGSU cre mice and LβT2 cells illustrated that p62 is a modulator of the GnRH-LH axis, which is dependent on intracellular calcium and ATP. These findings demonstrated that p62 deficiency in the pituitary impaired LH production via mitochondrial OXPHOS signaling and led to female infertility, thus providing the GnRH-p62-OXPHOS(Ndufa2)-Ca 2+ /ATP-LH pathway in gonadotropic cells as a new theoretical basis for investigating female reproductive dysfunction.

        Jingying Zhao,Xuefeng Wang,Yulong Cui,Sinuo Zhou 대한전기학회 2019 Journal of Electrical Engineering & Technology Vol.14 No.4

Magnetically-coupled resonant wireless power transfer (MCR-WPT) system with LCL-S structure is developing for higher transmission characteristics. This paper improves the resonance topology structure by adding a compensation capacitor in the transmitting side and proposes a parameter confi guration method with equivalent impedance. The transmission characteristics are analyzed fi rst based on simulation method to determine the parameters for the system requirements of 3 kW output power at working frequency of 85 kHz. Then the improved MCR-WPT system is developed. The realization methods of key modules such as electromagnetic coupling mechanism and drive circuit are given. The experiments are designed and are used to analyze the eff ect of diff erent frequency and diff erent distance on transmission characteristics. Output power and transmission effi ciency of the designed system can be 3141 W and 91% respectively. In addition, the inverter’s performance and the system electromagnetic radiation state are evaluated. The results show that the designed system can provide an eff ective means for 3 kW WPT application at 85 kHz.

The Role of Monoxide Hemoglobin in Color Improvement of Chicken Sausage

        Peng Xu,Xuefeng Zhu,Shengjiang Tan,Hao Qin,Cunliu Zhou 한국식품과학회 2016 Food Science and Biotechnology Vol.25 No.2

The role of monoxide hemoglobin (COHb) in improvement of chicken sausage color was investigated. COHb and NaNO2 synergistically increased a* values. Addition of 0.1% COHb decreased the residual nitrite content in the presence of 0.001% NaNO2. Compared with controls, the combined treatment resulted in significantly higher nitroso pigment contents while the single treatment resulted in slightly higher nitroso pigment contents. Visible spectrometry indicated that both nitrosohemochrome (NH) and hematin were the main ingredients of pigments extracted from chicken sausage treated with a combination of 0.006% NaNO2 and 0.6% COHb. Formation of NH and hematin caused an increase in a* values and a decrease in L* values, respectively. COHb showed potential for use in meat product formulations.

Optimization of Processing Conditions of Chinese Smoke-cured Bacon (Larou) with a New Natural Coating Solution during Storage Period

        Na Liu,Qiujin Zhu,Xuefeng Zeng,Bowen Yang,Meilian Liang,Li Deng,Laping He,Cai Liang,Ruping Zhang,Juan Zhou 한국축산식품학회 2018 한국축산식품학회지 Vol.38 No.3

The study aims to determine the optimum sterilization rate and water activity of Chinese traditional smoke-cured bacon product (Larou) in the preservation with natural coating solution. With the response surface methodology (RSM), we analyzed 3 factors of processing conditions (the concentration of lysozyme, concentration of sodium alginate, and concentration of chitosan) and 2 response factors (sterilization rate and water activity). Sterilization rate and water activity of Larou were largely affected by the concentration of lysozyme, concentration of sodium alginate, and concentration of chitosan. The final optimum concentrations of lysozyme, sodium alginate, and chitosan were 0.09, 1.40, and 1.60% and realized the high sterilization rate. Water activity of sliced Larou was significantly correlated with the sterilization rate. Low-field nuclear magnetic resonance analysis verified the optimum processing conditions. The coating resulted in 99.69% rate of reducing bacteria after 30-day storage. The data of the total number of colony, peroxidation value, moisture content, pH, and sensory evaluation provided the theoretical basis for extending the shelf life of Chinese traditional smoke-cured bacon product (Larou) with natural coating solution.

Reduction of the Retransmission Delay for Heterogeneous Devices in Dynamic Opportunistic Device-to-device Network

( Sixuan Chen ),( Weixia Zou ),( Xuefeng Liu ),( Yang Zhao ),( Zheng Zhou ) 한국인터넷정보학회 2018 KSII Transactions on Internet and Information Syst Vol.12 No.10

The dynamic opportunistic device-to-device (DO-D2D) network will frequently emerge in the fifth generation (5G) wireless communication due to high-density and fast-moving mobile devices. In order to improve the Quality of Experience (QoE) of users with different computing capacity devices in the DO-D2D network, in this paper, we focus on the study of how to reduce the packets retransmission delay and satisfy heterogeneous devices. To select as many devices as possible to transmit simultaneously without interference, the concurrent transmitters-selecting algorithm is firstly put forward. It jointly considers the number of packets successfully received by each device and the device’s connectivity. Then, to satisfy different devices’ demands while primarily ensuring the base-layer packets successfully received by all the devices, the layer-cooperation instantly decodable network coding is presented, which is used to select transmission packets combination for each transmitter. Simulation results illustrate that there is an appreciable retransmission delay gain especially in the poor channel quality network compared to the traditional base-station (BS) retransmission algorithm. In addition, our proposed algorithms perform well to satisfy the different demands of users with heterogeneous devices.

Shear-strain induced structural relaxation of Cu Σ3 [110](112) symmetric tilt grain boundary: The role of foreign atoms and temperature

        Li Yang,Xue Hongtao,Zhou Xin,Tang Fuling,Li Xiuyan,Ren Junqiang,Lu Xuefeng 한국물리학회 2021 Current Applied Physics Vol.28 No.-

Grain boundaries (GBs) relaxation is a promising and effective strategy to improving GB stability or stabilizing nanocrystalline metals. However, previous studies mainly focused on nanocrystalline pure metals and GB behaviors therein, without considering the role of foreign atoms such as impurity or alloying atoms in GB relaxation. In this work, the shear-strain induced structural relaxation of pure Cu Σ3 [110](112) symmetric tilt GBs (STGBs), and the effects of foreign elements (Fe and Ni) and temperature on the GB relaxation were investigated in detail by molecular dynamics method. The results show that shear strain can trigger the structural relaxation of pure, Fe- and Ni-containing Cu GBs by the emission of Shockley partial dislocations from Cu GBs. Both Fe and Ni have impediment effects on the shear-strain induced GB relaxation, though the content of Fe or Ni atom (0.00165 at.%) is quite low in the GB model. The temperature cannot trigger GB relaxation independently within the considered temperature range, but play a positive role in the shear-strain induced structural relaxations of pure, Fe- and Ni-containing Cu Σ3 [110](112) STGBs. Our work might gain new insights into the mechanically induced GB relaxation in nanocrystalline copper and could be beneficial for improving the stability of Cu GBs.

      • A Novel Algorithm for Green Citrus Detection based on the Reticulate Grayladder Feature

        Mingjun Wang,Jun Zhou,Weiyan Shang,Rufu Hu,Xuefeng Wang,Liang Gong 보안공학연구지원센터 2016 International Journal of Signal Processing, Image Vol.9 No.10

Immature green citrus fruit detection using conventional color images is a challenging task due to fruit color similarity with the background, partial occlusion, varying illumination and shape irregularity. Therefore, most existing green fruits detection algorithms, which use color as the main discriminant feature, have a low recognition rate and a high rate of false positives. In this manuscript, we developed a novel Green Citrus fruit Detection algorithm based on the proposed Reticulate Grayladder Feature (GCDRGF), which contained 4 major steps: First, an 8-graylevel image was generated by the preprocessing steps of median filtering, histogram-based equalization and 8-graylevel discretization of the input raw image. Secondly, reticulate grayladders were obtained by a multidirectional scanning on the 8-graylevel image, and rule-based pseudo-grayladder removal strategies were used to remove false positives of target grayladders. Thirdly, grayladder clustering and fruit location fitting were used to generate candidate regions for target fruits. Finally, majority voting was performed to determine the results of candidate regions based on the analysis of apparent features and recticulate grayladders within candidate regions. The experimental results proved the effectiveness of the proposed reticulate grayladder feature and the corresponding detection algorithm with respect to various illuminant and imaging conditions. Compared with the existed eigenfruit algorithm, our algorithm has a higher rate of successful recognition and a lower rate of false positives, which helps to greatly improve the productivity of robotic operations.

Subproteomic profiling from renal cortices in OLETF rats reveals mutations of multiple novel genes in diabetic nephropathy

        Li Zhiguo,Shen Hong,Liu Yeqiang,Zhou Xuefeng,Yan Meihua,He Hailan,Zhao Tingting,Zhang Haojun,Li Ping 한국유전학회 2022 Genes & Genomics Vol.44 No.1

Background: Ovarian cancer is a common gynecological malignancy among female patients and poses a serious threat to women's health. Although it has been established that Fos-like antigen 2 (FOSL2) is linked to ovarian cancer (OC), its exact role in the development of OC remains unknown. Objective: This article aims to investigate the role of FOSL2 in ovarian cancer development. Methods: FOSL2 expression in ovarian carcinoma and adjacent tissues was assessed using real-time fluorescent quantitative PCR and western blot. We constructed OE/sh-FOSL2 plasmids and Caspase-1 specific inhibitors (Yvad-CMK) and transfected A 2780 cells with them to identify the relevant cell functions. Furthermore, we used western blot assay to determine the changes in expression of apoptosis-associated speck-like protein containing a CARD (ASC), cysteine aspartate-specific proteasezymogen procaspase 1 (pro-caspase-1), cysteinyl aspartate-specific proteinase-1 (caspase-1), interleukin-1β precursor (pro-IL-1β), interleukin-1β (IL-1β), interleukin-18 precursor (pro-IL-18), and interleukin-18 (IL-18). In addition, we measured the concentration of IL-1β and IL-18 using an enzyme-linked immunosorbent assay (ELISA). Moreover, Tthe level of lactate dehydrogenase (LDH) in the cell supernatant was measured by LDH release assay kit. Results: The expression of FOSL2 was significantly higher compared with the surrounding tissues. The proliferation, migration, and invasion of A2780 cells were enhanced after transfection with OE-FOSL2 plasmids; however, the cell apoptosis was significantly decreased. When FOSL2 was overexpressed, the inflammasome-associated proteins such as ASC, caspase-1, IL-1β, and IL-18 were downregulated. Furthermore, FOSL2 induced apoptosis and activated the production of inflammasomes in A2780 cells. Co-therapy with Yvad-CMK and substantially inhibited apoptosis and activation of inflammasomes. Conclusions: Inhibition of FOSL2 promotes the apoptosis of OC cells by mediating the formation of an inflammasome.
