      • 總輸膽管 結紮術후 Hepatic Glycogen 變化에 관한 形態學的 硏究

        For the study on the changes of hepatic glycogen after common bile duct ligation, Sprague -Dawley rats (b.w. 250± 20gm, male) were used as experimental animal. In expermental group, at 3hrs, 6hrs, 9hrs, 12hrs, 24hrs, 48hrs, 72hrs, 96hrs, and 120hrs after ligation, the common bile duct at the point of about 1cm proximal portion from its duodenal opening, the liver specimens were collected under ether anesthesia. Simple laparotomy was performed in control rats. After fixation, the specimens were sectioned 5㎛ thickness and stained with Periodic Acid Schiff, and observed under light microscope. Also, transmission electron microscopic photos were obtained with H-7000(Hitachi Co.) TEM. The results were as followings: 1. PAS reactions were changed into negative 6hours after common bile duct ligation. 2. Glycogen depletion after common bile duct ligation was closely related with functional lobule, but not in simple laparotomized rats. 3. In the common bile duct ligated rats, PAS negativity continued in the zone I until 120hours after ligation.

        선박용 폐 FRP 수지의 재활용 공정 개발

        A recycling process for the waste FRP from boats was developed. The recycling process is composed of decomposition of waste FRP with propylene glycol and synthesis of recycled unsaturated polyester resin from the decomposed liquid material. Prior to the decomposition, waste FRP was cut into 2cm × 5cm segments and mechanical impact was applied by press roller to give gaps between cumulated laminates. Propylene glycol effectively decomposed the waste FRP segments and glass fibers were easily separated from decomposed liquid material. Recycled unsaturated polyester resin could be made from the decomposed liquid material by reaction with maleic anhydride and phthalic anhydride.

        The purpose of this study was to compare the verbal ability of institutional children with that of family children. We investigated the following question: ⑴ Is there a difference in verbal ability between institutional children and family children? ⑵ Are there age and sex differences in verbal ability between 4-year-olds and 6-year-olds? The sample of this study was 32 institutional children and 32 family children, with an equal number of 4-year-olds and 6-year-olds. The instrument for measuring children's verbal ability was the Children's Verbal Ability Test developed by Jang Young-ae(1981). The data were analyzed with a three-way ANOVA. The results of this study were the following: ⑴ There was a significant main effect of early care environment on children's verbal ability. That is, family children showed a higher verbal ability score that institutional children. There was a significant three-way interaction effect between early care environment, sex, and age. The size of the effects of early institutional care varied according to sex and age. ⑵ There was a significant main effect for age. That is, 6-year-olds showed higher verbal ability than 4-year-olds. The importance of an adequate verbal environment for institutionalized children was discussed.

        면접조사를 통한 초등학생의 영양교육 실태 및 실천도 조사

        This study investigated the experience and practice of elementary school students on nutrition education. The data were collected from 217 male and female students attending 5-6th grade elementary schools in Seoul and Kyunggi-Do from March to June 2007, interviewing face to face by a nutrition teacher and 3 interns of a nutrition teacher. The results were as follows: 86.5% of the subjects learned about ‘Table etiquette’, ‘Reasons for eating fruits and vegetables’ (78.7%), ‘Food waste and environment’ (72.3%), ‘Healthy snacks’ (55.7%), ‘Food sanitation’ (52.3%), ‘Food culture of foreign countries’ (48.1%). Nutrition education experience was significantly different by gender. A total of 43.5% boys responded that they never learned about ‘basic food preperation’ (p < 0.01). They had learned ‘Nutrients for body’ and ‘Food waste and environment’ in school, ‘Healthy weight loss’, ‘Food culture of foreign countries’, ‘Food circulation’ on television, Most content (‘Table etiquette’, ‘Simple cooking’, ‘Food sanitati n’, ‘Eating behaviors for health’, ‘Reasons for eating fruits and vegetables’, ‘Healthy snacks’) was learned from parents. The practice after nutrition education was higher in ‘Table etiquette’ (2.14), ‘Eating fruits and vegetables’ (2.07) than others compared with education experience. The most reason of non-practice on nutrition information was ‘Troublesome’. In ‘Nutrients for body’, a boy answered ‘Difficult for practice’ 20.0%, a girl answered ‘Difficult to understand’ 32.6%, showing a significant difference between the gender groups (p < 0.001). They remembered the ‘Nutrients for body’ (49.6%), ‘Food sanitation’ (44.5%) because of ‘important content’,‘Basic food preparation’ (40.6%), ‘Food culture of foreign countries’ (36.3%) because of ‘interesting content’, ‘Healthy weight loss’ (52.0%), ‘Eating behavior for health’ (44.5%) and ‘Healthy snacks’ (33.7%) because of ‘need for my health’.

      • 자가 말초혈액 조혈모세포 이식술의 임상적용

        Responses to chemotherapy correlate with the increased dose of chemotherapeutic agents in some cancers, e.g. breast cancer, Ewing's sarcoma, Hodgkin's disease(HD) and non-Hodgkin's lymphoma(NHL), then for the improvement of the chemotherapy responsiveness, high-dose chemotherapy(HDCT) was proposed. But, it's application was limited due to complications, especially bone marrow suppression. HDCT and succeeding autologous peripheral blood stem cell transplantation(APBSCT) was introduced to overcome this problem. This study was designed to determine the clinical pictures including clinical parameters and the responsiveness of HDCT followed by APBSCT in Hanyang University Hospital. Ten patients were enrolled. They were 8 adults (3 breast cancers, 3 multiple myelomas, 1 HD, 1 NHL) and 2 children (1 acute myelogenous leukemia (AML), 1 neuroblastoma). Mobilization chemocherapy(MCT) followed by Granulocyte Colony Stimulating Factor(G-CSF) was administered to mobilize stem cells from bone marrow to peripheral blood. The stem cells were collected by using CS-3000 and cryopreserved at -196℃ with programmed controlled rate freezer as the mixture with 10% dimethyl sulfoxide(DMSO). HDCT was administered and cryopreserved peripheral stem cells were infused on day 0 as planned. Patients' mean age was 41 years old. After induction chemotherapy three patients achieved complete response and 5 partial response. The median time for bone marrow recovery after MCT was 11.5 days for neutrophil(〉500/㎣) and 4.8 days for platelet(〉20,000/㎣). There were three cases of neutropenic fever and one case of gingivitis. After HDCT with APBSCT, one patient was converted partial response to complete response, and one patient achieved complete response after HDCT without induction chemotherapy. The median time for bone marrow recovery was 12.7 days for neutrophil(〉500/㎣) and 14.7 days for platelet(〉20,000/㎣). Eight patients developed neutropenic fever and required systemic antibiotics. Transfusion was required 1.6 pints for packed red cell and 23.4 pints for platelet concentrates. Toxicity consisted mainly of vomiting, diarrhea, and mucositis which were mild. HDCT with APBSCT can be performed safely with minimal complications. For the evaluation of the exact role of HDCT with APBSCT in malignant diseases, further studies are required as a large scale of patients and lung-term follow up.

        Characterizing affinity epitopes between prion protein and b-amyloid using an epitope mapping immunoassay

        Cellular prion protein, a membrane protein, is expressed in all mammals. Prion protein is also found in human blood as an anchorless protein, and this protein form is one of the many potential sources of misfolded prion protein replication during transmission. Many studies have suggested that b-amyloid1–42 oligomer causes neurotoxicity associated with Alzheimer’s disease, which is mediated by the prion protein that acts as a receptor and regulates the hippocampal potentiation. The prevention of the binding of these proteins has been proposed as a possible preventative treatment for Alzheimer’s disease;therefore, a greater understanding of the binding hot-spots between the two molecules is necessary. In this study, the epitope mapping immunoassay was employed to characterize binding epitopes within the prion protein and complementary epitopes in b-amyloid. Residues 23–39 and 93–119 in the prion protein were involved in binding to b-amyloid1–40 and 1–42, and monomers of this protein interacted with prion protein residues 93–113 and 123–166. Furthermore, b-amyloid antibodies against the C-terminus detected bound b-amyloid1–42 at residues 23–40, 104–122 and 159–175. b-Amyloid epitopes necessary for the interaction with prion protein were not determined. In conclusion, charged clusters and hydrophobic regions of the prion protein were involved in binding to b-amyloid1–40 and 1–42. The 3D structure appears to be necessary for b-amyloid to interact with prion protein. In the future, these binding sites may be utilized for 3D structure modeling, as well as for the pharmaceutical intervention of Alzheimer’s disease.

        과민성 장 증후군의 증상 경감을 위한 비약물적 접근 고찰

        There have been many trials of clinical efficacy of multicomponent and single component treatments for irritable bowel syndrome(IBS). We reviewed effects of non phamacological treatments in the IBS. Though the efficacy of multi-component approaches was unclear, several results suggest that cognitive behavioral therapy was effective in improving gastrointestinal symptoms of IBS. As a single component, cognitive therapy and relaxation with or without biofeedback could improve the symptoms and psychological health of IBS patients. Yoga, meditation, self-help information and hypnotherapy could be applicable to IBS.

        The purpose of this study was to identify the factors influencing the Exercise Performance of elderly patients with diabetes. The subjects were 153 elderly patients with diabetes who were selected from the public health center in Gwang ju. The data collected was analyzed using descriptive statistics, t-test, ANOVA, Pearson's correlation and stepwise multiple regression. This study found that approximately 52.9% of the subjects were exercising regulary. Exercise performance was significantly different according to education level, family income by month, and level of diabetes education. Significant factors influencing exercise performance were exercise self-efficacy, exercise social support and exercise benefits. The most powerful predictor of exercise performance was exercise self-efficacy(34.2%). This study suggests that nurses should emphasize exercise social support. and exercise benefits as well as reinforce exercise self-efficacy to improve exercise performance of the elderly patients with diabetes.

      • Effects of Pyrogallol on Growth and Cytotoxicity of Wild-Type and katG Mutant Strains of Vibrio vulnificus

        Vibrio vulnificus is a causative agent of fatal septicemia and necrotic wound infection and the pathogen infection became an important public health problem in many counties. Vibrio vulnificus causes RtxA1 toxin-induced acute cell death. We tried to identify natural products that inhibit the acute cytotoxicity of V. vulnificus using a lactate hydrogenase assay. A poly-phenol pyrogallol protected HeLa cells from V. vulnificus-induced cytotoxicity. Pyrogallol also decreased the growth of V. vulnificus; this inhibitory effect was more significant during log phase than stationary phase. To further elucidate the inhibitory mechanism, pyrogallol-induced toxicity was compared between a V. vulnificus catalase-peroxidase mutant (katG<sup>-</sup>) and the isogenic wild-type MO6-24/O strains. No growth was observed for the katG<sup>-</sup> mutant in the presence of pyrogallol (50 μg/mL) even after 24 h, whereas the wild-type strain dem-onstrated growth recovery following a prolonged lag phase. Pyrogallol-mediated growth inhibition of the katG<sup>-</sup> mutant strain was partially rescued by exogenous catalase treatment. These results indicate that the mechanism by which pyrogallol inhibits the growth and cyto-toxicity of V. vulnificus likely involves polyphenol-induced prooxidant damage. Taken together, these results suggest that pyrogallol has potential for development as a new para-digm drug treat infectious diseases.

      • Current Updates on the Genome Analysis of Leptotrombidium pallidum, a Major Vector Mite for Scrub Typhus

        Leptotrombidium pallidum is the major vector mite for Orientia tsutsugamushi, the causative agent of scrub typhus, in Asian countries, including Korea. The genome size of L. pallidum was previously estimated to be 191 ± 7 Mb (Kim et al., 2014). Genomic DNA (gDNA) was extracted from a single female from a 9-generation inbred L. pallidum colony and used for whole genome amplification (WGA). The resulting amplified gDNA was used for the construction of paired-end and mate-pair libraries and sequenced using Illumina platforms (HiSeq2000 and MiSeq). An unamplified gDNA sample extracted from 20 female mites was also used for sequencing in parallel. More than 45Gb sequence reads from both paired-end and mate-pair libraries of the WGA gDNA were trimmed and then de novo assembled using the CLC Asembly Cell v.4.0 for contig assembly and SSPACE for scaffolding. The assembly generated approximately 6,545 scaffolds with N50 value of 92,945 and total size of ~193Mb, which was in a good agreement with our previous estimation. Repeat analysis showed that about 30% of genome (~58Mb) was masked as repeats, most of which were unclassified novel elements. For gene predictions, generated were the PASA models based on genomic alignments of RNA-seq reads from 4 different chigger mite samples (i.e. male, female, larva, and protonymph) and the GeneWise models based on genomic alignments of protein sequences from 4 closely related species with chigger mite. Independently, ab initio gene predictions were performed with AUGUSTUS and FgeneSH with custom trained matrices optimized for L. pallidum and GENEID with pre-trained matrix for Acyrthopsiphon pisum. By combining all together, 15,842 genes were predicted finally. Manual curation is in progress for various groups of genes, including chemosensory receptor genes, immune-related genes, acaricide target genes, etc.
