소 품종별 Melanocortin Receptor 1(MC1R) 유전자의 유전자형 빈도에 관한 연구

The melanocortin 1 receptor (MC1R) plays a central role in regulation of eumelanin(black/brown) and phaeomelanin (red / yellow) synthesis within the mammalian melanocyte and encoded by the classical Extension (E) coat color locus. The objectives of this study were carried out to observe the genotype frequencies of MC1R in 1,044 samples of imported beef and eight different cattle breeds including Hanwoo, Holstein, Angus, Brown-Swiss, Charolais, Limousin, Simmental and Hereford. The primers for the amplification of bovine MC1R gene were designed based on a bovine MC1R gene sequence (GenBank accession no. Y19103). A size of 350 by was amplified by polymerase chain reaction(PCR), digested with each or together of two different restriction enzyme, BsrFI and MspAII, and electrophoresed in 2.5% or 4% Metaphore agarose gel for determination of genotypes. One thousand and forty four samples including eight different cattle breeds and imported beef were determinated their genotypes by PCR-RFLP. Genotype frequencies of Hanwoo were 0.10 in E e and 0.90 in ee. Allele E^D was shown in all of Holstein and Angus breeds tested which have black coat color phenotypes. However, genotype frequencies of Limousin and Simmental breeds were 0.21 and 0.07 in E^DE^D, 0.00 and 0.03 in E^De^+ , 0.38 and 0.35 in E^De, respectively. These results of this study suggest that alleles of the bovine MC1R gene are associated strongly with coat color except for Simmental and Limousin breeds with dark brown, light yellow or mosaic coat color of red and brown. Therefore, it needs to be studied on relationship between A and E loci and coat color phenotypes. Furthermore, genotypes of the bovine MC1R gene can be used as a DNA marker for distinguishing. beef between Hanwoo and Holstein and Angus having black coat color phenotype.
p34 is a novel regulator of the oncogenic behavior of NEDD4-1 and PTEN

PTEN is one of the most frequently mutated or deleted tumor suppressors in human cancers. NEDD4-1 was recently identified as the E3 ubiquitin ligase for PTEN; however, a number of important questions remain regarding the role of ubiquitination in regulating PTEN function and the mechanisms by which PTEN ubiquitination is regulated. In the present study, we demonstrated that p34, which was identified as a binding partner of NEDD4-1, controls PTEN ubiquitination by regulating NEDD4-1 protein stability. p34 interacts with the WW1 domain of NEDD4-1, an interaction that enhances NEDD4-1 stability. Expression of p34 promotes PTEN poly-ubiquitination, leading to PTEN protein degradation, whereas p34 knockdown results in PTEN mono-ubiquitination. Notably, an inverse correlation between PTEN and p34/NEDD4-1 levels was confirmed in tumor samples from colon cancer patients. Thus, p34 acts as a key regulator of the oncogenic behavior of NEDD4-1 and PTEN.

The chemical composition of PM_(2.5) was measured at Gosan. Jeju for two weeks each in November 2001 and spring 2002. In the latter part of the measurement period of November 2001, designated as the polluted period in this work, secondarily formed ion components as well as primarily emitted elemental carbon were high, PM_(2.5) mass concentration was also high in this polluted period compared with the yellow sand period, in which daily average of PM_(10) peaked up to 520㎍/㎥. Increase of major components of anthropogenic origin in the polluted period was not correlated with the variation in sea salt components while increase of crustal components in the yellow sand period was highly correlated with the variation in sea salt components. Trace elements were generally higher in the yellow sand period: however, toxic heavy metals such as zinc, cadmium and lead were the highest in the polluted period.

Triclosan (TCS) and Triclocarban (TCC) are used as antimicrobial agents and have been widely dispersed and detected in the marine environment. However, the toxicities of TCS and TCC have been poorly investigated in marine invertebrates. In this study, the effects of TCS and TCC on mortality, population growth, lifespan, and fecundity were examined in the monogonont rotifer (Brachionus koreanus) using cellular ROS levels, GST enzymatic activity, and gene expression of defensomes. The median lethal concentration (LC50) of TCS (393.1μg/L) and TCC (388.1μg/L) was also determined in the same species. In TCS- and TCC-exposed B. koreanus, growth retardation and reduced fecundity were observed and were shown to have a potentially deleterious effect on the life cycle of B. koreanus. In addition, time-dependent increases in ROS content (%) and GST enzymatic activity were shown in response to TCS and TCC exposure. Additionally, transcript levels of detoxification proteins (e.g., CYPs), antioxidant proteins (e.g., GST-sigma, Cu/ZnSOD, CAT), and heat shock proteins (Hsps) were modulated in response to TCS and TCC exposure over a 24h period. Our results indicate that TCS and TCC induce oxidative stress and transcriptional regulation of detoxification, antioxidant, and heat shock proteins, resulting in changes in lifespan and fecundity.

The goal of this research was to develop recombinant Escherichia coli to improve fatty acid synthesis (FAS). Genes encoding acetyl-CoA carboxylase (accA, accB, accC), malonyl-CoA-[acyl-carrier-protein] transacylase (fabD), and acyl-acyl carrier protein thioesterase (EC 3.1.2.14 gene), which are all enzymes that catalyze key steps in the synthesis of fatty acids, were cloned and over-expressed in E. coli MG1655. The acetyl-CoA carboxylase (ACC) enzyme catalyzes the addition of CO<SUB>2</SUB> to acetyl-CoA to generate malonyl-CoA. The enzyme encoded by the fabD gene converts malonyl-CoA to malonyl-[acp], and the EC 3.1.2.14 gene converts fatty acyl-ACP chains to long chain fatty acids. All the genes except for the EC 3.1.2.14 gene were homologous to E. coli genes and were used to improve the enzymatic activities to over-express components of the FAS pathway through metabolic engineering. All recombinant E. coli MG1655 strains containing various gene combinations were developed using the pTrc99A expression vector. To observe changes in metabolism, the in vitro metabolites and fatty acids produced by the recombinants were analyzed. The fatty acids (C16) from recombinant strains were produced 1.23-2.41 times higher than that from the wild type.

This study was performed to investigate the behavioral characteristics of Hanwoo(Bos taurus coreanae) cows and their calves. Four cows and their calves were placed in pens. The behaviors of each cow and calf were recorded using a time lapse VCR from 07:00 h to 18:00 h for 6 days and were analyzed using the scan point sampling method. The cow's behaviors in order of frequency were LD(lying down, 25%), ST(standing, 23%), EA(eating, 13%), LR(lying down rumination, 10%) and SR(standing rumination, 7%). The cows spent less than 3% of their time PG(pairwise grooming) and SF(sniffing). The calves mainly showed LD(52%), ST(28%), LF(lying flat, 4%), EA(3%) and WA(walking, 3%). The calves spent less than 2% of their time SF(sniffing) and SUK(sucking). The behavioral patterns of cows and calves at 07:00 h, 12:00 h and 18:00 h were different, especially in ST and LD.

Valence states and electronic structures of Cr-doped Nd½A½MnO₃ (NAMO; A=Ca, Sr) manganites have been investigated using x-ray absorption spectroscopy (XAS) and high-resolution photoemission spectroscopy (PES). All the Cr-doped NAMO systems exhibit the clear metallic Fermi edges in the Mn e_g PES spectra near E_F. The spectral intensity at E_F is higher for Cr-doped Nd½Sr½MnO₃ (NSMO) than for Cr-doped Nd½Ca½MnO₃ (NCMO), reflecting the stronger metallic nature for NSMO than for NCMO. The measured Cr 2p XAS spectra are found to be very similar to that of Cr₂O₃, indicating that Cr ions in Cr-doped NAMO are in the trivalent Cr³+ states (t³_(2g)). The Cr 2p XAS data are consistent with the Cr 3d PES spectra located at ~1.3 eV below EF and having no emission near E_F.

Purpose: The purpose of this study was to explore the differences among social comparison orientation, depression, and self-esteem by patterns of using Facebook among Facebook users’ in their 20s. Methods: The participants in this descriptive study were 332 Facebook users in their 20s. Data collection took place between August 21 and 27, 2014. Questionnaires were the Iowa–Netherlands Comparison Orientation Measure (revised), Center for Epidemiologic Studies Depression Scale, and Rosenberg Self-Esteem Scale. Data were statistically analyzed using t-tests and ANOVAs. Result: Each mean score of social comparison, depression, and self-esteem was 26.04±6.82, 31.39±8.99 and 38.05±6.19. Significant differences emerged between the degree of social comparison and the following Facebook usage variables: average single-connection time to Facebook (F=3.570, p=.007) and self-evaluation of Facebook usage behavior (F=6.203, p<.001). Conclusion: The results of this study suggested that Facebook users in their 20s who access Facebook for long periods and assess themselves as using it more often had a high score of comparison orientation. This result can be used in Nursing by developing the theoretical basis for education and campaign related to Facebook usage. In-depth studies are needed for research on the effect social comparison has on Facebook users and relationships between social comparison and emotions.
