        오프라인 기반 가상공동체 구성원의 심리적 유형과 온라인 의사소통 매체에 대한 사회 연결망 분석

        오프라인 기반 가상공동체 활동을 위한 온라인 매체는 구성원간 사회적 관계형성을 위한 보완적 역할을 하고 있다. 온라인 의사소통 도구 중 전자우편과 메신저의 경우 개인간 뿐만 아니라 기업 내외적 활동에 있어서도 그 활용도가 중가하고 있다. 구성원간 사회적 관계형성과정은 심리적 성향 및 선호 커뮤니케이션 매체선택에 의해 영향을 받을 수 있음에도 불구하고 이에 대한 연구는 충분히 이루어지지 않았다. 따라서 심리적 성향과 온라인 매체 상에서의 네트워크 수준간의 관계에 대한 연구가 필요하다. 본 연구는 개인의 심리적 유형과 온라인 비동기 매체인 전자우편과 동기 매체인 메신저를 사용하는 구성원간의 사회연결망 관계를 연구하였다. 실증분석결과 내향사고형의 경우 메신저 상에서의 사회연결망 수준이 가장 높은 것으로 나타났으며, 전자우편의 경우는 외향사고형의 경우가 가장 높은 것으로 나타났다. 연구의 결과는 온라인상에서의 개인의 의사소통 방식에 대해 보다 이해의 폭을 넓힐 수 있으며, 심리적 성향에 따른 온라인 사회연계성 정도를 파악함으로써 온라인 마케팅활동과 관련한 시사점을 제공할 수 있을 것이다.

      • 뇌졸중 환자의 경상운동이 상지기능에 미치는 영향

        경상운동은 다른 쪽의 신체에서 일어나는 의도된 움직임의 경상 반전으로 발생하는 한 쪽 신체의 무의식적인 운동이다. 본 연구는 뇌졸중으로 인한 편마비 환자에게서 나타나는 경상운동이 상지기능에 미치는 영향에 대해 알아보고자 하였다. 65명의 뇌졸중 환자를 대상으로 연구한 결과, 경상운동이 나타나지 않은 환자군의 상지기능은 경상운동이 나타난 환자군의 상지기능과 통계학적으로 유의한 차이를 보였으며, 경상운동이 나타난 환자군에 비해 상지기능이 좋은 것으로 나타났다. 또한, 경상운동이 나타나지 않은 환자군과 경상운동이 경한 환자군, 경상운동이 중한 환자군의 상지기능을 비교한 결과 경사운동이 적게 나타날수록 상지기능이 좋은 것으로 나타났다. 연구의 결과를 종합하여 볼 때, 뇌졸중으로 인한 편마비 환자에게서 나타나는 경상운동은 상지기능을 감소시키는 요인 될 수 있는 것으로 나타났다. The purpose of this study was to compare upper extremity's motor function according to the mirror movements in hemiparetic patients with stroke. Mirror movements are involuntary movements in one side of the body that occur as mirror reversals of the intended movement on the other side of the body. The subjects of the study were 65 stroke patients who were admitted from May, 2004 to April, 2005 in the Y University medical center. This study used the Manual Function Test(MFT) and the Purdue pegboard test to evaluate upper extremity‘s motor function. The severity of mirror movements were assessed according to the criteria of Woods and Teuber. The results of the study was as follow; First, The upper extremity‘s motor function in hemiparetic patients without mirror movements was significantly different in hemiparetic patients with mirror movements(p< .05). Second, The upper extremity's motor function in hemiparetic patients with mild mirror movements was significantly different in hemiparetic patients with severe mirror movements(p< .05).       • 비·부비동 종양의 신생혈관 형성에서 COX-2 및 VEGF의 역할

        Background and Objectives : COX-2, the inducible form of cyclooxygenase(COX), is upregulated in transformed cells and in malignant cells, which have important roles in promotion of colorectal carcinogenesis, invasiveness and angiogenesis. Vascular endothelial growth factor(VEGF) may act as endothelial cell mitogen in various cancer tissues. It will increase tumor growth and angiogenesis. Aims of this study were to asses COX-2/VEGF expression and it's clinical correlation in sinonasal tumors and to investigate the effects on angiogenesis also. Materials and Method : The study samples were obtained from surgical removal of 27 cases of inverted papilloma(IPs), 5 cases of IP with malignant transformation(IP-MT), 16 cases of Squamous cell carcinoma(SCC) in sinonasal cavity COX-2 and VEGF expressions were assessed by immunohistochemical staining. Synchronism of COX-2/VEGF expression in one tumor cell was demonstrated with double immunofluorescence technique. Results : The expression rates of COX-2 were 37% in IP, 80% in IP-MT and 100% in SCC. The positive rates of VEGF were 29 6% in IP, 100% in IP-MT and SCC. COX-2 and VEGF expressions were increased as tumor progressed, and there is a positive correlation between COX-2 &EGF expressions. All clinical features which were compared are not associated with COX-2 &EGF expressions except malignant change in IP(p=0 028). Synchronism of COX-2/VEGF expressions was noted in all positive immunostaining samples. Conclusion : These findings suggest the role of COX-2 pathway could be involved in sinonasal tumor angiogenesis, which is modulated by VEGF COX-2 may act as autocrine factor in VEGF expression. There is a potential role for selective COX-2 inhibitors in the treatment of these lesions.

      • 후두 편평세포암종에서 Galectin-1 및 -7의 발현 의의

        Background and Objectives: The role of different Galectins in the pathogenesis of different types of malignancy is being profoundly investigated recently. In this study, the author investigated the level of Galectin-1 and -7 in the layngeal squamous cell carcinoma tissue to define its relationship to the tumor progression. Materials and Methods: Paraffin-embedded tissue specimens from 77 patients, who were diagnosed as laryngeal squamous cell carcinoma from 1993 to 2007, were immunohistochemically stained for Galectin-1 and -7. Results: Galectin-1 expression was positively correlated with tumor stage and nodal involvement. Galectin-7 expression was increased in better differentiated tumors. Conclusion: Expression of Galectin-1 and -7 can be used as a valuable indicator in laryngeal squamous cell carcinoma under certain limitations.

      • Association of Herpesvirus Saimiri Tip with Lipid Raft Is Essential for Downregulation of T-Cell Receptor and CD4 Coreceptor

        <B>ABSTRACT</B><P>Lipid rafts are membrane microdomains that are proposed to function as platforms for both receptor signaling and trafficking. Our previous studies have demonstrated that Tip of herpesvirus saimiri (HVS), which is a T-lymphotropic tumor virus, is constitutively targeted to lipid rafts and interacts with cellular Lck tyrosine kinase and p80 WD repeat-containing endosomal protein. Through the interactions with Lck and p80, HVS Tip modulates diverse T-cell functions, which leads to the downregulation of T-cell receptor (TCR) and CD4 coreceptor surface expression, the inhibition of TCR signal transduction, and the activation of STAT3 transcription factor. In this study, we investigated the functional significance of Tip association with lipid rafts. We found that Tip expression remarkably increased lipid raft fractions in human T cells by enhancing the recruitment of lipid raft-resident proteins. Genetic analysis showed that the carboxyl-terminal transmembrane, but not p80 and Lck interaction, of Tip was required for the lipid raft localization and that lipid raft localization of Tip was necessary for the efficient downregulation of TCR and CD4 surface expression. Correlated with this, treatment with Filipin III, a lipid raft-disrupting agent, effectively reversed the downregulation of CD3 and CD4 surface expression induced by Tip. On the other hand, Tip mutants that were no longer present in lipid rafts were still capable of inhibiting TCR signaling and activating STAT3 transcription factor activity as efficiently as wild-type (wt) Tip. These results indicate that the association of Tip with lipid rafts is essential for the downregulation of TCR and CD4 surface expression but not for the inhibition of TCR signal transduction and the activation of STAT3 transcription factor. These results also suggest that the signaling and targeting activities of HVS Tip rely on functionally and genetically separable mechanisms, which may independently modulate T-cell function for viral persistence or pathogenesis.</P>

        Apolipoproteins inhibit the innate immunity activated by necrotic cells or bacterial endotoxin

        <P>Summary</P><P>We suggested earlier that the <U>hy</U>dro<U>p</U>hobic <U>po</U>rtion<U>s</U> (Hyppos) of molecules, which are normally embedded in the membranes of cells or the core of molecular structures so as to be separated from the aqueous environment, might serve as evolutionarily ancient alarm signals of injury or stress to initiate innate immune responses when they are exposed on the surface. Under normal physiological conditions, the Hyppos released from endogenous or exogenous sources might be handled by ‘Hyppo-quenchers’<I>in vivo</I> to maintain the tissue homeostasis and immune modulation. To test this idea, we selected apolipoproteins, which have been known to transport blood lipids and play a role in a number of pathological inflammatory conditions. We examined their role as Hyppo-quenchers in early immune responses and found that apolipoproteins showed significant inhibition of the nuclear factor-&kgr;B-dependent gene expression in recombinant Chinese hamster ovary (CHO) cells and dendritic cells stimulated by necrotic cells or bacterial endotoxin. In addition, our results indicate that apolipoproteins could dramatically abrogate complement fixation on the surface of necrotic cells. These findings suggest that apolipoproteins, besides having known functions in lipid metabolism, also have a role in preventing the initiation of innate immunity, potentially through neutralizing Hyppos from injured cells or exogenous endotoxin.</P>

      • A multifunctional core-shell nanoparticle for dendritic cell-based cancer immunotherapy

        <P>Dendritic cell-based cancer immunotherapy requires tumour antigens to be delivered efficiently into dendritic cells and their migration to be monitored in vivo. Nanoparticles have been explored as carriers for antigen delivery, but applications have been limited by the toxicity of the solvents used to make nanoparticles, and by the need to use transfection agents to deliver nanoparticles into cells. Here we show that an iron oxide-zinc oxide core-shell nanoparticle can deliver carcinoembryonic antigen into dendritic cells while simultaneously acting as an imaging agent. The nanoparticle-antigen complex is efficiently taken up by dendritic cells within one hour and can be detected in vitro by confocal microscopy and in vivo by magnetic resonance imaging. Mice immunized with dendritic cells containing the nanoparticle-antigen complex showed enhanced tumour antigen specific T-cell responses, delayed tumour growth and better survival than controls.</P>
