      • 소 卵胞의 體外培養에 의한 卵子의 體外成熟과 體外受精에 관한 硏究

        This study was to investigate the effects of the follicular culture from which the oocytes origenate on their subsequent in vitro maturation. The follicular oocytes were cultured in TCM-199 medium containing FCS, hormones, BFF for 24-30 hrs in CO^2 incubator with 5% CO^2 in air at 38.5℃ and then matured oocytes were again 18-20 hrs with motile capacitated spermatozoa in the BO solution containing 100 ㎍/㎖ of heparin. The results obtained in these experiments were summarized as follows : 1.Ovarian follicles were isolated and cultured according to size 1-2 ㎜, 2-4 ㎜, 4-6 ㎜ and 6-8 ㎜ for 24-30 hrs. The rates of GVBD in each group were 80.0%(40/50), 84.4%(38/45), 77.8%(35/45) and 84.0%(42/50), but oocytes maturation were supressed at anaphaes-Ⅰ or telephase-Ⅰ stage. 2. When oocytes cultured for 4 hrs at 35℃ in medium with BFF and hormones after follicular culture for 24-30 hrs, the maturation completed to metphase-Ⅱ stage. However, rates of GVBD in oocytes from follicular culture were higher than oocytes cultured in medium. 3. When oocytes from follicle cultured for 4 hrs at various temperature and culture time were cultured for 24-30hrs the rates of oocytes maturation from follicle cultured at 20℃(43.6%, 24/55), 30℃(47.3%, 26/55) and 35℃(52.7%, 29/55) were significant higher than group cultured at 4℃(24.4%, 11/45). 4. When oocytes from follicles cultured for hrs at 35℃, the rates of oocytes maturation and fertilization rates were 67.3%(101/150), 29.3%(44/150) and oocytes cultured for 24-30hrs at 35℃, thr rates of oocytes maturation and in vitro fertilization were 86.5%(122/141), 53.9%(76/141), respectively.

      • HFC-134a와 대체냉매 HFC-152a에 CF_3I를 혼합한 자동차냉매 특성

        These days, an attention of environment has been increasing all over the world. to prevent the green house effect and destruction of an ozone layer, an ozone destruction potential(ODP) be zero and a refrigerant for low global warming potential(GWP) is needed. HFC-134a, in which hydrogen is mixed instead of chlorine is a refrigerant used for automobile conditioners and its destruction potential is ecologically zero. However, it is not considered as a perfect substitutive refrigerant as its GWP is high. It is studied refrigerant mixtures in which HFC-134a, HFC-152a and CF_3I with low GWP and zero ODP are mixed by experimentally and concluded as follows; 1. with the variation of speed of compressor outside temperature and flow rate, the heat of evaporator and compressor and coefficient of performance was varied, and influenced the air conditioner. 2. The pressure of evaporator was decreased with increasing the speed of compressor and that of the refrigerant HFC-134a was higher 21% than that of azotrope refrigerant mixed with CF_3I. The overall capacity of condensor and evaporator was increased and HGC-134a was higher than azotrope refrigerant mixed with CF_3I 3. The capacity of evaporator with the refrigerant HFC-134a was higher 6% than azotrope refrigerant mixed with CF31 and the required power with zaotrope refrigerant mixed with CF_3I was higher 11% than that of HFC-134a. 4. The COP of the refrigerant FHC-134a was higher 31% in 1000RPM. when outside temperature was increased, COP was decreased. 5. Performance of HFC-134a is better than azotrope refrigerant mixed with CF_3I but prefered zaotrope refrigerant mixed with CF_3I considering the environment.

        Involvement of ROS in Curcumin-induced Autophagic Cell Death

        Many anticancer agents as well as ionizing radiation have been shown to induce autophagy which is originally described as a protein recycling process and recently reported to play a crucial role in various disorders. In HCT116 human colon cancer cells, we found that curcumin, a polyphenolic phytochemical extracted from the plant <i>Curcuma longa</i>, markedly induced the conversion of microtubule-associated protein 1 light chain 3 (LC3)-I to LC3-II and degradation of sequestome-1 (SQSTM1) which is a marker of autophagosome degradation. Moreover, we found that curcumin caused GFP-LC3 formation puncta, a marker of autophagosome, and decrease of GFP-LC3 and SQSTM1 protein level in GFP-LC3 expressing HCT116 cells. It was further confirmed that treatment of cells with hydrogen peroxide induced increase of LC3 conversion and decrease of GFP-LC3 and SQSTM1 levels, but these changes by curcumin were almost completely blocked in the presence of antioxidant, N-acetylcystein (NAC), indicating that curcumin leads to reactive oxygen species (ROS) production, which results in autophagosome development and autolysosomal degradation. In parallel with NAC, SQSTM1 degradation was also diminished by bafilomycin A, a potent inhibitor of autophagosome- lysosome fusion, and cell viability assay was further confirmed that cucurmin-induced cell death was partially blocked by bafilomycin A as well as NAC. We also observed that NAC abolished curcumin- induced activation of extracelluar signal-regulated kinases (ERK) 1/2 and p38 mitogen-activated protein kinases (MAPK), but not Jun N-terminal kinase (JNK). However, the activation of ERK1/2 and p38 MAPK seemed to have no effect on the curcumin-induced autophagy, since both the conversion of LC3 protein and SQSTM1 degradation by curcumin was not changed in the presence of NAC. Taken together, our data suggest that curcumin induced ROS production, which resulted in autophagic activation and concomitant cell death in HCT116 human colon cancer cell. However, ROS-dependent activation of ERK1/2 and p38 MAPK, but not JNK, might not be involved in the curcumin-induced autophagy.

        Biocontrol of Anthracnose in Pepper Using Chitinase, β-1,3 Glucanase, and 2-Furancarboxaldehyde Produced by Streptomyces cavourensis SY224

        A strain of Streptomyces cavourensis subsp. cavourensis (coded as SY224) antagonistic to Colletotrichum gloeosporioides infecting pepper plants was isolated. SY224 produced lytic enzymes such as chitinase, β-1,3-glucanase, lipase, and protease in respective assays. To examine for antifungal activity, the treatments amended with the nonsterilized supernatant resulted in the highest growth inhibition rate of about 92.9% and 87.4% at concentrations of 30% and 10%, respectively. However, the sterilized treatments (autoclaved or chloroform treated) gave a lowered but significant inhibitory effect of about 63.4% and 62.6% for the 10% supernatant concentration, and 75.2% and 74.8% for the of 30% supernatant concentration in the PDA agar medium, respectively, indicative of the role of a nonprotein, heat stable compound on the overall effect. This antifungal compound, which inhibited spore germination and altered hyphal morphology, was extracted by EtOAc and purified by ODS, silica gel, Sephadex LH-20 column, and HPLC, where an active fraction was confirmed to be 2-furancarboxaldehyde by GS-CI MS techniques. These results suggested that SY224 had a high potential in the biocontrol of anthracnose in pepper, mainly due to a combined effect of lytic enzymes and a non-protein, heatstable antifungal compound, 2-furancarboxaldehyde.

        흡착튜브를 이용한 가스상물질 채취용 펌프의 유량성능 평가방법

        Objectives: Flow rate stability is very important for obtaining reliable measurements. However there is no easily used method for checking whether the flow rate set at the initial stage is sustained during sampling. The purpose of this study was to develop a method to evaluate the flow rate performance of gas sampling pumps with adsorbents commonly used to sample gases. Materials and methods: We tested the back pressure of gas sampling pumps commonly used in Korea with adsorbents such as charcoal and silica tubes and attempted to discover the combination conditions of adsorbents in accordance with back pressure required by ISO standard 13137. Results: We found the combination of sampling adsorbents to be applicable to the pressure drop required by the ISO standard for evaluating flow rate stability under increasing pressure drop and long term (eight-hour) performance. Conclusions: This evaluation method of using a sampling media matrix for checking flow rate stability as proposed by this study could be a highly useful tool for determining the reliability of the performance pumps before sampling.

        트레드밀 운동시 속도와 경사도에 따른 운동 역학적 변인의 특성 비교

        The purpose of this study is to define the mechanism by comparison analysis of mechanic movement by the inclination and the speed of the treadmill gait(walking and running) and to offer the basic data for the decision of the speed and the inclination to avoid injury. The selected subjects of this study were ten female college students, who had normal foot and no experience of the lower extremity injury. We used treadmill that set with forceplatform and Vicon 370 system for the three dimensional analysis. The treadmill speed was 1.25m/s for walking and 2m/s, 2.5m/s, 3m/s for running. The inclination were level(0%), up 5%, 10%, the speed and the order of the inclination selected random. We analyzed the walking value by the inclination and the speed and the angle of lower extremity, the vertical reaction force during the treadmill gait (walking and running). As follows are the result of the analysis, 1. The variation pattern of the inclination on treadmill has no effects during running(2/ms/, 2.5m/s, 3m/s), but change the movement pattern of lower extremity's on sagittal plane during walking(1.25m/s). 2. The difference by speed on the angle variation pattern of the lower extremity appear between walking(1.25m/s) and running(2m/s, 2.5m/s, 3m/s), and has effects on movement pattern of lower extremity's coronal plane and transverse plane. 3. The changing of the inclination to 5%, the DRM(dynamic range of motion) of the pelvic and ankle's on sagittal plane is increased but the case of the 10% the DRM(dynamic range of motion) of the knee's on sagittal plane and that of the pelvic's on transverse plane is increased. 4. The DRM(dynamic range of motion) by the speed in stance phase has no difference each running speed(2m/s, 2.5m/s, 3m/s), but differ with walking(1.25m/s) at pelvic tilt, pelvic rotation, hip rotation, knee flex/extension, knee rotation, foot dorsi/plantarflexion. So the DRM(dynamic range of motion) of most lower extremity in stance phase when it running is wider than walking, but has no difference each speed of running.
