<P>The binding of SeqA protein to hemi-methylated GATC sequences (hemi-sites) regulates chromosome initiation and the segregation of replicated chromosome in <I>Escherichia coli</I>. We have used atomic force microscopy to examine the architecture of SeqA and the mode of binding of one molecule of SeqA to a pair of hemi-sites in aqueous solution. SeqA has a bipartite structure composed of a large and a small lobe. Upon binding of a SeqA molecule to a pair of hemi-sites, the larger lobe becomes visibly separated into two DNA binding domains, each of which binds to one hemi-site. The two DNA binding domains are held together by association between the two multimerization domains that make up the smaller lobe. The binding of each DNA binding domain to a hemi-site leads to bending of the bound DNA inwards toward the bound protein. In this way, SeqA adopts a dimeric configuration when bound to a pair of hemi-sites. Mutational analysis of the multimerization domain indicates that, in addition to multimerization of SeqA polypeptides, this domain contributes to the ability of SeqA to bind to a pair of hemi-sites and to its cooperative behavior.</P>
The Delay in Confirming COVID-19 Cases Linked to a Religious Group in Korea

Objectives: As of March 3, 2020, the Shincheonji religious group accounted for the majority of Korean cases of coronavirus disease 2019 (COVID-19). Nonetheless, the most likely cause of the broad spread of COVID-19 among members of the Shincheonji religious group remains largely unknown. Methods: We obtained data of laboratory-confirmed cases related to the Shincheonji religious group from press releases by Korean public health authorities and news reports. We measured the period from the date of illness onset to the date of COVID-19 confirmation. Results: We analysed data from 59 cases (median age, 30 years). The estimated median period between the date of symptom onset and the date of COVID-19 confirmation was 4 days (95% confidence interval, 1-12). Conclusions: There was a delay in COVID-19 confirmation from the date of illness onset among the cases linked to the Shincheonji religious group. This delay likely contributed to the occurrence of many cases of COVID-19 in the group.

최근 국내 반도체 DRAM 및 NAND flash 메모리 제조 기술 고도화로 인하여 wafer 상단에 수 나노 스케일 수준의 박막증착 및 패턴 제작이 이루어지고 있으며, 이에 공정라인에서 제작되는 시편을 검사/계측하기 위한 기술에 대한 필요성이 요구되는 추세이다. 반도체 생산 제조 공정라인의 검사/계측 기술로 활용되는 ellipsometry 기술은 산란을 이용한 비파괴성 광학적 측정 기술로, 시편에 의해 변화된 편광정보(Ψ, △)를 획득하고, 이를 분석하여 제작 시편의 물성 및 박막 두께, 나노 패턴의 3D 형상을 계측하는 기술이다. 현 반도체 산업계에서 제작되는 wafer의 균일도는 기업의 수율과 관계하므로 측정속도에 대한 솔루션의 필요성이 크게 증대되고 있는 상황이며, 이에 기존 ellipsometry 기술이 지닌 기계적 메커니즘 및 전기 변조 장치 등의 구동부로 인한 초 단위 이상의 측정속도 제한 문제를 극복하기 위하여 본 연구팀은 2016년 마하젠더 scheme의 분광편광타원계측 기술을 제안한바 있다. 이는 간섭편광변조모듈로부터 발생된 분광편광간섭신호를 시스템 알고리즘을 통해 분광편광위상정보(△)를 20Hz 이상 고속으로 획득 가능하다. 본 연구팀은 최근 간섭편광변조모듈을 일체형 마이켈슨 간섭계로 구성함에 따라, 측정 반복능을 ±1˚ 수준의 외란에 강인성을 지닌 시스템으로 성능을 크게 개선하였다. 아래 figure(a)의 Blue, Red line은 각각 reference(bare wafer), SiO2 30nm를 제안된 시스템으로 획득된 간섭spectrum이고, figure(b)의 Blue, red line은 각각 간섭계 기반 분광편광타원계와 상용 ellipsometer를 이용하여 측정된 분광편광위상정보(△)이다. SiO2 30nm 측정결과를 통해 간섭계 기반 분광편광타원계는 측정 반복능 뿐만 아니라, 측정에 사용된 전체파장에 대해서 상용장비 수준의 높은 정확도가 확보됨을 검증하였다. [그림 본문 참조]
Trigger Factor Interacts with DnaA Protein to Stimulate its Interaction with DnaA Box

While screening proteins that interact with DnaA protein, the initiator protein for Escherichia coil chromosomal DNA replication, we found a 52-kD sized protein which bound to DnaA protein in a salt-dependent manner. This protein was identified as trigger factor, a ribosome-associated peptidyl-prolyl- cisltrans isomerase with chaperone activity. Trigger factor was overproduced and purified to near homogeneity, and its effect on the function of DnaA protein was examined, Enhanced binding of DnaA protein to DnaA box with no apparent supershift in the gel-shift experiments suggested that trigger factor, by virtue of its chaperone activity, exerts a change on DnaA protein thus increasing its binding affinity for DnaA box.

This paper describes a dynamic spectroscopic ellipsometry based on dual-wavelength calibration. DSE provides ellipsometric parameters at rates above 20 Hz, but the interferometer's sensitivity to temperature makes it difficult for that proposed system to maintain stable 𝜟k over long periods of time. To solve this problem, we set up an additional path in the DSE to perform simulations of the polarization phase calibration method using dual wavelengths. Through simulation, we were able to eliminate most of the polarization phase error and maintain a stable 𝜟k in the long-term stability experiment for 10 hours. This is the result that the 𝜟k stability of the proposed system is improved tens of times compared to the existing system.

<P>Human TopBP1 with eight BRCA1 C terminus domains has been mainly reported to be involved in DNA damage response pathways. Here we show that TopBP1 is also required for G(1) to S progression in a normal cell cycle. TopBP1 deficiency inhibited cells from entering S phase by up-regulating p21 and p27, resulting in down-regulation of cyclin E/CDK2. Although co-depletion of p21 and p27 with TopBP1 restored the cyclin E/CDK2 kinase activity, however, cells remained arrested at the G(1)/S boundary, showing defective chromatin-loading of replication components. Based on these results, we suggest a dual role of TopBP1 necessary for the G(1)/S transition: one for activating cyclin E/CDK2 kinase and the other for loading replication components onto chromatin to initiate DNA synthesis.</P>

<P>Schlemm's canal (SC) is a specialized vascular structure in the eye that functions to drain aqueous humor from the intraocular chamber into systemic circulation. Dysfunction of SC has been proposed to underlie increased aqueous humor outflow (AHO) resistance, which leads to elevated ocular pressure, a factor for glaucoma development in humans. Here, using lymphatic and blood vasculature reporter mice, we determined that SC, which originates from blood vessels during the postnatal period, acquires lymphatic identity through upregulation of prospero homeobox protein 1 (PROX1), the master regulator of lymphatic development. SC expressed lymphatic valve markers FOXC2 and integrin α9 and exhibited continuous vascular endothelial-cadherin (VE-cadherin) junctions and basement membrane, similar to collecting lymphatics. SC notably lacked luminal valves and expression of the lymphatic endothelial cell markers podoplanin and lymphatic vessel endothelial hyaluronan receptor 1 (LYVE-1). Using an ocular puncture model, we determined that reduced AHO altered the fate of SC both during development and under pathologic conditions; however, alteration of VEGF-C/VEGFR3 signaling did not modulate SC integrity and identity. Intriguingly, PROX1 expression levels linearly correlated with SC functionality. For example, PROX1 expression was reduced or undetectable under pathogenic conditions and in deteriorated SCs. Collectively, our data indicate that PROX1 is an accurate and reliable biosensor of SC integrity and identity.</P>
