      • Bupivacaine과 ropivacaine이 Xenopus oocyte에 발현된 HERG 전류에 미치는 영향

        Bupivacaine is an amide-type local anesthetic widely used for regional anesthesia. Ropivacaine is developed as a less cardiotoxic alternatives to bupivacaine. In the present study, we have analyzed the effects of bupivacaine and ropivacaine on HERG currents expressed in Xenopus oocytes. Bupivacaine and ropivacaine(3∼1,000μM) blocked HERG currents in a concentration dependent manner. EC_(50) was 26.1±3.1μM(n_(R) 0.65±0.04) and 43.5±7.9μM(n_(H) 0.99±0.13) in bupivacaine and ropivacaine, respectively. Bupivacaine and ropivacaine did not affect the activation and deactivation kinetics of HERG channels. However, the drugs decreased the slope conductance measured from fully activated current-voltage relationship curves. These results suggest that bupivacaine and ropivacaine have a similarinhibitory effect on HERG channels, which could be a possible cellular mechanism of LQT or ventricular arrythmia by the drugs.

      • 혈청 Fructosamine에 대한 임상적 유용성에 관한 연구

        Diabetes mellitus is a universal health problem and may occur at any age. The traditional biochemical measurements for initially detection patients with diabetes mellitus are random estimations of blood and urine glucose concentrations. Despite their common use both tests are fairly non-specific, being influenced by wide variety of condition. Measurement of the HbA_(1c) in a random blood specimen is a more simple screening test for diabetes, and widely used as an index of diabetic control. But unfortunately, it has difficulties with methodology. Fructosamine has been described as a new parameter for the estimation of glycosylated protein, It is based on the reduction potential of ketoamines in an alkaline medium and measures colorimethcally the concentration of all glycosylated semm proteins belonging to the class of fructosamine. It reflects the time-integrated glucose concentration over the previous 1 to 3 weeks and can be used for the follow up of diabetes mellitus. We measured serum fructosamine concentration and compared with any other items. The results are summehzed as follows : 1. Within-run and between-run imprecision were assessed by 20 analysis of 3 differential fructosamine concentration in control sera. The respective coefficients of variation were 2.9%, 6.6% in low concentration, 2.2%, 5.1% in medium concentration and 3.3%, 4.7% in high concentration. 2. The serum fructosamine concentration were 2.26±0.21 mmol/L in normal control subjects(n=29) and 3.35±0.65 mmol/L (n=62) in patients with diabetes mellitus (p<0.001). 3. In diabetic patients (n=62), the serum fructosamine concentrations correlated less significantly with glucose (Y(fructosamine) = 0.01×(glucose)+2.52, r=0.54, P<0.001) and more significantly with HbA_(1c) (Y(fructosamine) = 0.22×(HbA_(1c))+1.71 , r=0.63, P<0.001). 4. The concentrations of fructosamine in serum and EDTA plasma were not significently diferent (P<0.01). So, Plasma is as well suited to fructosamine determinations as serum(Y(serum) = 0.95×(Plasma)+0.25, r=0.96). 5. No interference was observed with lipemic sera (cholesterol) in detection of serum fructosamine(P=0.393).

      • 교과서 선정을 위한 중등학교 과학 교과서 평가틀 개발

        The major purpose of this thesis is to construct the evaluating framework of a science textbook with a high confidence and universal alidity, referring to a preceeding study and materials involved in research work, and in consideration of the educational conditions of the secondary school. The Framework developed in this study are composed of 7 categories: content, organization, representation, laboratory, illustrations, external form or editing system, and supplementary materals. Each category consists of detailed items. Each item in the check-list is to be given between one and five points and the total number of possible points in this evaluating framework is 290. As a result, these evaluating items enable quantitative analyses, afford an opportunity which we can look into many-sidedly the respects we should consider when a science textbook is adopted, and they can be estimated partly or synthetically as occasion demands. This study is expected to contribute to choose a good science textbook as fundamental data when selected in the future, and to help the lasting study or estimation of the textbook.

        수종 치과용 합금의 세포독성 평가에 관한 연구

        The restorations with dental alloys have toxic as well as mechanical stimulation to gingival tissue when they were contacted for long time. Cell culture method is one of the tests for evaluation of biocompatibility of the material or its components. In this study, the cytotoxicity of type Ⅲ dental crown and bridge casting alloys, dental amalgams, dental amalgam alloy containing metals, and copper-alluminum alloys was evaluated. L 929 cells were cultured in Eagle's MEM supplemented with 10% fetal bovine serum, 100 IU/㎖ of penicillin, and 100 ㎍/㎖ of streptomycin and seeded with ?? cells/㎖ in plastic dishes (100×15㎜). After the cell monolayer had formed, medium was removed and the medium containing agarose was added. The cells were stained with 0.01% neutral red solution and the specimens were placed on the agar. After incubation for 24 hours at 37℃ in the CO₂incubator, the cultures were examined macroscopically and microscopically under a microscope to determine the cytotoxicity. From the experiments, the following results were obtained : 1. Type Ⅲ dental crown and bridge casting alloys, dental ceramic alloys, dental Co-Cr alloys, pure silver, and pure tin showed mild cytotoxicity with zone index of 1.0-1.5 and lysis index of 1.0-2.0. 2. Copper-based alloys (NPG), after ethylene oxide gas sterilization, showed mild cytotoxicity with response index of 1.0/2.0 ; after autoclave sterilization, showed moderate cytotoxicity with response index of 2.0/3.0. 3. Dental high copper amalgam (HVA) showed moderate cytotoxicity with response index of 2.0/3.0. 4. Of the constituent metals of dental amalgam alloy, copper showed severe cytotoxicity with response index of 4.0/4.5, and zinc showed moderate cytotoxicity with response index of 3.0/3.3.

      • 적혈구침강속도와 급성기물질의 혈중변화에 따른 상관성과 임상적평가

        The authors determined erythrocyte sedimentation rate(ESR) and the level of acute phase reactants such as ceruloplasmin, α1-antitrypsin, haptoglobin and C-reactive protein and evaluated their correlation in 101 patients with active inflammation, chronic infection, collagen disease and neoplastic disease from Jan. 1990 to Aug. 1990 in Chosun university hospital, Shortly after the patients had been diagnosed, the tests was performed using their blood. ESR was measured by standardized Win trobe method. Ceruloplasmin, α1-antitrypsin, and haptoglobin were measured by radial immunodiffusion method. C-reactive protein was measured by turbidimethc immunoassay. The correlation of ESR and the levels of ceruloplasmin, α1-antitrypsin, haptoglobin and C-reactive protein was analysed in the control group and the disease group. The results were summarized as follows. 1. The ESR rose with each decade increase of age in most cases of the disease group. 2. The author judged that the ESR was increased when the ESR was upper than 20mm / hr. The 45 of 101 cases of the disease group showed increase of ESR. So the sensitivity of ESR to detect disease is 44.55%. 3. The difference of the level of ceruloplasmin and α1-antitrypsin of control group and that of the disease group showing normal ESR value was not statistically significant(P>0.05). 4. The difference of the level of C-reactive protein and haptoglobin of the control group and that of the disease group showing normal ESR value was statistically significant(P<0.05). 5. The difference of the level of ceruloplasmin, α1-antitrypsin and haptoglobin of the disease group showing normal ESR value and that of disease group showing high ESR value was not statistically significant (P>0.05). 6. The difference of the level of C-reactive protein of the disease group showing normal ESR value and that of the disease group showing high ESR values was statistically significant (P<0.05). 7. C-reactive protein played a major role in increasing ESR and haptoglobin played some influence also. But α1-antitrypsin and ceruloplasmin played little influence.

      • 165 rDNA PCR을 이용한 양수내 세균 검출

        Background and Objectives : Microbial invasion of the amniotic cavity is a significant clinical problem in obstetrics leading to infectious morbidity of both the mother and the newborn. So rapid and reliable test to detect bacteria in amniotic fluid is urgently needed. The purpose of this investigation was to determine the feasibility of using the 16S rDNA PCR to detect bacteria in amniotic fluid. Material and Method : Amniotic fluid samples from 15 pregnant women with hydramnios, preterm labor, or old age pregnancy were analysed with universal bacterial PCR. We used universal primers which amplify a DNA fragment of 16s rDNA from all known bacteria and sequenced the positive samples to identify the bacterial species, In some cases, routine bacterial culture, WBC, and CRP were studied also. Result : Three of 15 pregnant women were positive in the PCR. DNA sequencing of the PCR product detected Staphylococcus epidermidis in one case, and Streptococcus anginosus in two cases. Conclusion : The broad-spectrum bacterial 16S rDNA PCR assay, in properly controlled conditions, may provide a fast and reliable means for the detection of bacteria in amniotic fluid. 배경 및 목적 : 양수강의 미생물 침습은 산모와 신생아 양자에게 감염성 질환을 야기할 수 있어 산과학에 있어 중요한 임상적 문제이다. 따라서 양수에서 세균을 검출할 수 있는 신속하고 믿을 만한 검사가 절실히 필요하다. 이 연구의 목적은 16S rDNA PCR을 이용하여 양수에서 세균을 검출할 수 있는지 그 실행가능성을 검사하는데 있다. 대상 및 방법 : 양수과다증이나 조기 분만, 노산 등이 있는 임산부 15인의 양수 검체를 16S rDNA PCR로 분석하였다. 모든 알려진 세균의 16S rDNA 분절을 중폭할 수 있는 보편적인 시동물질을 사용하여 증폭하였고, 양성 검체를 염기순서분석하여 세균 균종을 동정하였다. 일부 환자들에서는 세균 배양검사와 WBC, CRP 검사도 함께 시행하였다. 결과 : 15명중 3명의 임산부가 PCR에 양성이었고 PCR 산물을 DNA 염기순서분석한 결과 한 환자에서 Staphylococcus edidermidis가 다른 두 환자에서 Streptococcus anginosus가 검출되었다. 결론 : 광범위 세균 16S rDNA PCR 분석은 적절히 관리된 조건 하에서 양수내 세균 검출을 위한 신속하고 믿을 만한 방법을 제공해 줄 수도 있다고 생각되었다.

        Micronucleus test를 이용한 수종 결합용레진과 상아질결합제의 돌연변이 유발에 관한 연구

        The purpose of this study was to evaluate the mutagenicity of the bonding resins of dental composites and the dentin bonding agents. Eight bonding resins, six dentin bonding agents and distilled water (negative control) were administered orally and cyclophosphamide (positive control) was administered intraperitoneally to the mice. The mice were killed by cervical dislocation at 24 hours after administration and the femurs were removed and dissected. The bone marrow cells were collected and smeared on glass slides and stained with haematoxylin-eosin staining method and were observed with light microscope(X450). The following results can be drawn: 1. The bonding resins of dental composites and the dentin bonding agents were tested in this study seemed to be non-mutagenic. 2. The numbers of micronucleated polychromatic erythrocytes were similar both in the bonding resins of dental composites and the dentin bonding agents. 3. In the case of some products(AP, SC' and AA), the number of micronucleated polychromatic erythrocs was high beyond other products.

      • 화학처리한 상아질 : 주사전자현미경 관찰 A comparision of conventional and environmental scanning electron microscopy findings

        목적 본 연구의 목적은 젖은 상태와 건조 상태에서 산 처리한 상아질의 구조를 비교하는 것이다. 방법 재래형과 조건형 주사 전자현미경을 이용하여 전처리한 상아질을 습윤상태, 공기건조 및 임계점건조 상태에서 보았다. 구연산, 말레산, 질산, 옥살산 및 인산 등을 포함한 6가지 서로 다른 처리를 제조사의 지시에 따라 실시하였다. 결과 간단한 공기건조와 조절 조건하에서의 건조 과정을 통하여 산처리한 상아질에서 수분을 상실함으로 섬유성, 콜라겐 구조의 형태변화를 가져왔다. 임계점 건조를 시행한 경우는 외부의 탈회층의 섬유구조를 유지하였다. 사용한 모든 산에서 같은 결과를 보였다. 의미 산처리를 통하여 상아질 콜라겐의 미세구조는 화학적으로는 안정하거나 거의 변하지 않으나 수분을 상실하는 동안 형태는 붕괴하였다. 임계점 건조만이 섬유경계 면에서 수분이 동과 상실을 수반하는 계면응력을 조절할 수 있다. 본 연구는 결합과정에서 레진 침투를 용이하기 위해서 콜라겐의 형태와 상아질의 습윤 상태를 유지하는 개념을 뒷받침하고 있다.
