Bupivacaine과 ropivacaine이 Xenopus oocyte에 발현된 HERG 전류에 미치는 영향

Bupivacaine is an amide-type local anesthetic widely used for regional anesthesia. Ropivacaine is developed as a less cardiotoxic alternatives to bupivacaine. In the present study, we have analyzed the effects of bupivacaine and ropivacaine on HERG currents expressed in Xenopus oocytes. Bupivacaine and ropivacaine(3∼1,000μM) blocked HERG currents in a concentration dependent manner. EC_(50) was 26.1±3.1μM(n_(R) 0.65±0.04) and 43.5±7.9μM(n_(H) 0.99±0.13) in bupivacaine and ropivacaine, respectively. Bupivacaine and ropivacaine did not affect the activation and deactivation kinetics of HERG channels. However, the drugs decreased the slope conductance measured from fully activated current-voltage relationship curves. These results suggest that bupivacaine and ropivacaine have a similarinhibitory effect on HERG channels, which could be a possible cellular mechanism of LQT or ventricular arrythmia by the drugs.
요가 수리야-나마스카라 수련형태에 따른 자율신경계 변화 비교

The purpose of this study is to compare the effects of ashtanga surya-namaskar(ASN) emphasizing motion types on the autonomic nervous system during repeated execution of vini surya-namaskar(VSN) that emphasize mental movements. This study was conducted on 20 yoga instructors. Experiments were performed with all subjects repeatedly performing ASN and VSN at intervals of one week. Autonomic nervous system tests were performed before and after 12 consecutive runs of 12 ASN and VSN, respectively. Autonomic variables used heart rate variability. Heart rate variability variables were sandard deviation of the normal to normal interval(SDNN), the square root of the mean squared differences of successive NN intervals(RMSSD), very low frequency(VLF), low frequency(LF), high frequency(HF), ratio of LF per HF(LF/HF), and total power(TP). Statistical analysis was performed using repeated ANOVA, independent t-test, and dependent t-test. Statistical significant level was at .05. As a result, after 12 consecutive sessions, LF / HF increased ASN and decreased VSN, indicating that VSN is effective for parasympathetic activation.
요가 아사나(Yoga Asana)가 고관절,골반의 불균형에 따른 하지변형 교정

The goal of this study is to develop the healing program of recovering the body imbalance by means of yoga asana as well as to provide a fundamental data in domestic yoga therapeutics development, after revealing the effects of it. The subjects of this research were selected within the women members in their twenties to forties in Seoul M-yoga academy institute after conducting the survey in advance. The experiment was done 2 times a week for 8 weeks, and the vini healing yoga program(VHYP) was practiced distinguishing long legs, short legs, and O-shaped legs in accordance with physical traits, applying the principle of 2 to 1. X-ray photographing was done before the experiment began and after the experiment, in order to verify the effects which yoga asana had on the change in the iliac length(AS/PI), one in the iliac width(IN/EX), hip joint angle(internal rotation / external rotation), the change of the lower part of a leg, and the change of a pelvis circumference and that of the vini healing yoga program. In the O-shaped leg vini healing yoga program, there appeared some noticeable changes in the iliac width(L), in a hip joint angle(L), in the lower part of a leg(L), in a pelvis circumference in the second verification. In the O-shaped leg vini healing yoga program produced a more effectively. But there weren`t any noticeable changes in the pelvis length, in the pelvis width, and in the length of the lower part of the leg.
요가 수리야 나마스카라 수련형태에 따른 에너지소비량 및 호흡순환기능 비교

The purpose of this study was to provide a comprehensive reference base for designing exercise prescription program through a comparative analysis of Ashtanga Surya Namaskar (ASN) and Vini Surya Namaskar (VSN) by physiologically quantifying energy consumption and respiratory circulation response of 12 different postures of each yoga series in yoga instructors. Subjects were female yoga instructors (mean age, 35.5±7.0 years) with an experience of teaching for more than 3 years at least twice per week and 60 minutes per session. Subjects were asked to perform 12 poses of ASN and VSN as a chi-square test and their energy consumption and respiratory circulation response were measured while staying each stationary position for a minute. Based on resting heart rate and oxygen uptake, we minimized the effect of each position on a following position. Differences before and after each yoga pose and in energy consumption and respiratory circulation response between ASN and VSN were tested using two-way repeated m easures ANOV A. Moreover, t he d ata was analyzed u sing d ependent t -test to d etermine differences between the means of yoga practices. According to the results of this study, the mean energy consumption of 12 postures was 1.6 /min in both ASN and VSN. The energy ㎉ consumption of a series of 12 yoga poses was 3.6㎉/min in ASN and 3.2 ㎉/min in VSN. According to the analysis of exercise intensity when performing 12 postures of ASN and VSN, both yoga series required 1.7 METs, and HRmax was 48.2% in A SN and 50% in V SN. In addition, VO2max was measured 19.4% in ASN and 20.1% in VSN. The findings of this study are expected to provide quantitative physiological data necessary in choosing yoga postures and types, energy consumption patterns and exercise intensity appropriate for yoga trainees’ fitness levels.
고혈압백서 대동맥에서 Ca에 의한 수축에 미치는 Na/Ca교환기전의 영향

We investigated the contraction by sodium-calcium(Na-Ca) exchanges in the aortic strips of spontaneously hypertensive rats(SHR) and Wistar-Kyoto (WKY) rats. Resting tension of aortic strips in SHR was decreased by the removal of extracellular Ca and those were recovered by the addition of Ca. However, resting tone of aortic strips in WKY was not changed by Ca removal or Ca addition. Ca-induced contraction after exposure to Ca-free solution was suppressed by verapamil 10^-6M to 90% or more. In the presence of verapamil 10^-5M, phentolamine 10^-6M, caffeine 5mM and atropine 10^-6M, the reduction of extracellular sodium concentration([Na]o) from normal(158 mM) to 0 mM(replaced by choline chloride) caused the contraction(Ca entry by Na-Ca exchange). The amplitude of contraction was more greater in SHR than in WKY rats. In the presence of verapamil 10^-5M, phentolamine 10^-6M, caffeine 5mM and atropine 10^-6M, aortic strips were not contracted by Naand Ca-free solution, but was contracted by the addition of Ca. In the presence of verapamil 10^-5M and phentolamine 10^-6M and ouabain 10^-5M, Ca did not induce contraction after exposure to Ca-free Tyrode ' s solution, but in the presence of verapamil 10^-5M and phentolamine 10^-6M and monensin 10^-5M a sodium ionophore, Ca induced a little contraction in the aortic strips of SHR. These observations suggest that Na-Ca exchange does not contribute the development of Ca-induced contraction after exposure to Ca-free Tyrode ' s Solution in the presence of normal extracellular Na concentration. However, Na/Ca exchange in vascular smooth muscle is increased in SHR and may be involved in the mechanism for hypertension in SHR.
Redox Factor-1 Inhibits Cyclooxygenase-2 Expression via Inhibiting of p38 MAPK in the A549 Cells

In this study, we evaluated the role of apurinic/apyrimidinic endonuclease1/redox factor-1 (Ref-1) on the tumor necrosis factor-α (TNF-α) induced cyclooxygenase-2 (COX-2) expression using A549 lung adenocarcinoma cells. TNF-α induced the expression of COX-2 in A549 cells, but did not induce BEAS-2B expression. The expression of COX-2 in A549 cells was TNF-α dose-dependent (5∼100 ng/ml). TNF-α-stimulated A549 cells evidenced increased Ref-1 expression in a dose-dependent manner. The adenoviral transfection of cells with AdRef-1 inhibited TNF-α-induced COX-2 expression relative to that seen in the control cells (Adβgal). Pretreatment with 10μM of SB203580 suppressed TNF-α- induced COX-2 expression, thereby suggesting that p38 MAPK might be involved in COX-2 expression in A549 cells. The phosphorylation of p38 MAPK was increased significantly after 5 minutes of treatment with TNF-α, reaching a maximum level at 10 min which persisted for up to 60 min. However, p38MAPK phosphorylation was markedly suppressed in the Ref-1-overexpressed A549 cells. Taken together, our results appear to indicate that Ref-1 negatively regulates COX-2 expression in response to cytokine stimulation via the inhibition of p38 MAPK phosphorylation. In the lung cancer cell lines, Ref-1 may be involved as an important negative regulator of inflammatory gene expression.
