        Studies on Lectins from Mushrooms

        Thirty-six species of higher fungi growing in the wild were collected and studied extensively for their lectin activities by using erythrocytes of human, rabbit and mouse blood. In total, 18 species demonstrated hemagglutination with some kinds of erythrocytes. Of thirty-six species, eight (Boletus edulis, B. Splendidus, Clavaria zollingeri, Lactarius subzonarius, L. volemus, Russula cutefracta, Pholiota squarrosa, and P. aspera) have shown lymphoagglutination with murine splenic lymphocytes. Protein contents were estimated from the curde lectin fraction. These mentioned eight species contained relatively high amounts of proteins than other mushrooms. Since these species had, coincidently, high hemagglutinating activity as well, we could define them as lectin-containing mushrooms. Some species also contained mitogenic lectins toward murine splenic lymphocytes. A lectin component was isolated and partially purified from agglutinins of Agaricus bisporus. Immunological experiments were performed with this lectin to investigate the properties and the similarities with other agglutinins.

        대퇴골절 수술 후 회복실에서 나타난 지방색전증후군

        The fat embolism syndrome is a serious and potentially life threatening complication of long bone trauma, blunt trauma and intramedullary manipulation. A 26-year-old woman underwent an intramedullary nailing of the femur under general anesthesia 45 hours after a traffic accident. The operation ended uneventfully and the patient was extubated in the operatirg room. About one hour after the patient arrived at the recovery room, she progressively developed tachycardia, tachypnea, cyanosis and drowsiness. The fat embolism syndrome was suspected because of the above clinical signs, and because pulmonary edema appeared on a simple chest A-P. During six days of intensive treatment in response to the fat embolism syndrome, the patient,s vital signs and ventilatory status progressively improved. The patient was uneventfully discharged from the hospital 19 days after her operation. (Korean J Anesthesiol 1999; 37: 731∼736)

      • 천연물자원과 향기(방향)요법

        색깔과 향기는 생명현상의 표현 그 자체중의 하나라 할 수 있다. 모든 생명체는 각각 감지되는 정도의 차이는 있을지언정 모두가 독특한 색과 냄새를 지닌 채 태어나고, 자라고, 노화하고, 죽음에 이르는 물질대사의 현상을 나타낸다. 그리고 사람들은 누구나 아름다운 색깔과 신선하고 감미로운 냄새를 좋아한다. 이렇게 좋은 냄새로 더 좋은 환경을 만들어 정신과 육체가 더 건강해 지고 그러므로 생활의 질이 향상되도록 하는 방법, 이것이 바로 향기(방향)요법(aromatherapy)이다. 여기에 이용되는 향의 원료는 천연물중 대부분이 식물의 생식기관에 해당되는 꽃, 열매, 과일, 종자 등에서 얻어진다. 이와 같은 현상은 고대에서부터 현대 문명에 이르기까지 그러했듯이, 미래에도 변함없이 인류는 자연의 산물중 하나인 방향성 식물을 이용할 것이다. 특히 앞으로는 과학기술의 발달과 더불어 열대 다우(多雨)지역 원시림 등 아직 사람들이 접근하지 못한 미개척지에서의 신품종 발견, 환경조절이 가능한 phytotron의 개발 이용, 새로운 생명공학기법의 도입 응용 등으로 더 많고 좋은 새로운 천연자원을 개발하게 될 것이다. 이런 꾸준하고 다양한 노력의 결과로 우리 인류의 생활의 질은 지속적으로 향상되리라 믿는다. 자연과학의 연구목표는 우리가 익혀 터득한 과학기술을 통해 인류를 더 행복하게 하려는데 있다. 이와 같은 자연과학의 바탕에는 문화예술이 있어야 그 과학은 더욱 건실하고 그런 사회와 국가는 번영함을 우리는 알아야한다. 꽃의 색과 향기를 찾아내고 이용하는 방안은 유사이래 문화예술임과 동시에 과학이었음을 강조하며, 다음과 같은 내용에 대하여 언급하고자 한다.

      • 별불가사리 렉틴의 특성 및 암 세포 성장저해 효과

        A new lectin was partially purified from starfish. Asterina pectinifera by means of physiological saline extraction, salt fractionation, ion exchange chromatography and hydroxyapatite chromatography, and it was named APL. The biochemical properties of the APL were characterized. In addition, its effects on lymphocyte mitogenicity and cancer cell agglutinability were tested. The APL agglutinated nonspecifically human erythrocytes and rabbit blood cells, Agglutinability was decreased to 30% of control activity below pH 5 and above pH 9 and was relatively unstable at increasing temperatures above 60℃. The activity was reduced by addition of two kinds of metal ions. Ba^(2+), Mn^(2+) and chelating agent. EDTA. APL was proved to be glycoproteins containing 9% sugars. For carbohydrate specificity, it was found that the activity of APL was inhibited by D(+)-glucosamine. D(+)-galactosamine, stachyose, N-acetyl-galactosamine and methyl-??-D-galactopyranoside among 35 sugars tested. In amino acid composition, the contents of acidic amino acids such as aspartic acid and glutamic acid were relatively high. This result suggest that the isoelectric point would be in a lower range. APL was found that in promotes the division of human lymphocytes. APL was proved to be a potent agglutinin for cancer cells such as HeLa, L929 and L1210 cells. Significant changes on the HeLa cell surfaces affected by APL were observed under the electron microscope.

        Observations of Infection Structures on the Leaves of Cucumber PlantsPre-treated with Arbuscular Mycorrhiza Glomus intraradices after Challenge Inoculation with Colletotrichum orbiculare

        Resistance inductions on the leaves of cucumber plant by an arbuscular mycorrhiza Glomus intraradices were investigated. In addition, the infection structures were observed at the penetration sites on the leaves of plant inoculated with Colletotrichum orbiculare using a fluorescence microscope. The severity of anthracnose disease caused by Colletotrichum orbiculare was significantly decreased on the leaves of cucumber plant colonized with G. intraradices compared with those of non-treated control plants. As a positive control, pretreatment with DL-3-aminobutyric acid (BABA) caused a remarkable reduction of the disease severity on the pathogen-inoculated leaves. There were no significant differences in the frequency of either germination or appressorium formation of the plant pathogen between mycorrhiza colonized and non-treated plants. It was also the same on the BABA pre-treated plants. However,the frequency of callose formation was significantly high on the leaves of G. intraradices colonized plants compared to those of non-treated control plants at 5 days after challenge inoculation. On the leaves of BABA treated plants callose formation was not significantly high than those of non-treated, although the disease severity was more strongly suppressed. It was suggested that the resistance induced by colonization with G.intraradices might be related to the enhancement of callose formation at the penetrate sites on the leaves invaded by the pathogen, whereas resistance by BABA did not

      • 사람 말초혈액 단핵세포에서 녹두 렉틴의 사이토카인 생성효과

        New lectins have been isolated and purified from mung bean(Phaseolus radiatus) through physiological saline extraction, ammonium sulfate salt fractionation and column chro-matographies. Ion exchanger were eluted by linear salt gradient and then further purified through gel filtration. Thus obtained lectin named as MBL. The gene expressions of 5 cytokines(IL-1, IL-2, IL-6, TNF-α and IFN-?) from human peripheral blood mononuclear cells(PBMC) stimulated with MBL were investigated by using reverse transcription polymerase chain reaction(RT-PCR). PBMC(1×106cells/㎖) isolated from healthy volunteers were stimulated with lectins(4㎎/㎖) for various time intervals(1 to 96 hrs). After each of the various stimulated times, total RNA was isolated and assessed for different cytokines mRNA by RT-PCR. The mRNA encoding IL-1, IL-2 were detected continuously from 1 to 20 hrs, and IL-6 was detected up to 24 hrs. But the mRNA encoding IFN-r and TNF-α were detected to 8 hours only and showed short time response com-pared with other cytokines. The significant expression of all cytokines mRNA were observed at 4 hrs. These results suggested that MBL, as inducer of cytokines could elicit detectable cytokine mRNA from PBMC.

      • 장수풍뎅이(Allomyrina dichotoma) 렉틴의 면역기능 증강효과

        A lectin was purified from Allomyrina dichotoma (ADL) by physiological saline extraction, ammonium solfate fractionation, anion exchange column chromatography on DEAE Sephadex A-50 and gel filtration column chromatography on Sephadex G-200. Several biochemical properties of ADL were characterized as follows: ADL from gel filtration column chromatography showed single band on SDS PAGE. ADL agglutinated the erythrocytes of rabbit and human A, B, O, AB. Agglutinability was relatively stable at basic pH, and was stable at temperature below 40℃. Agglutinability was not affected by metal ions and EDTA. This lectin was proved to be a glycoprotein which contains 0.47% of sugars. The molecular weight of ADL was estimated to be 97,000 dalton by SDS-PAGE By amino acid analysis, ADL exhibited high amounts of aspartic acid. The lectin's immunomodulating effect was measured as cytokine production. The productions of 5 cytokines (OL-1a, IL-2, IL-6, IFN?? and TNF-a) from peripheral blood mononuclear cells were measured by ELISA. The lectin induced the highest secretion of IL-2 at 8 hr. TNFa at 4 hr, and IFN?? at 24hr, respectively. These results suggest that ADL can elicit the production of detectable cytokines from PBMC.
