      • A New Record of the Genus Toxares Haliday (Hymenoptera: Braconidae: Aphidiinae) from Korea

        The genus Toxares (Braconidae: Aphidiinae) is reported for the first time in Korea. Members of Toxares are known as solitary koinobiont endoparasitoids especially specific for aphidine aphids. In this study, Toxares deltiger (Haliday, 1833) is found as new to Korea, which has a wide host range for 31 aphid species within Aphidinae. Thus, this species would be useful as natural enemy for biological control in the future. Diagnosis and host information are included.

        도플러 심초음파도를 이용한 요독증 환자의 투석전후 좌심실 이완기능

        To investigate the acute effects of hemodialysis on the left ventricular function of dialysis patients, M-mode and Doppler echocardiographic studies were performed in 17 patients undergoing maintenance hemodialysis using acetate buffered dialysis solution. The results were as follows: 1) Blood urea nitrogen, creatinine, potassium, body weight, left ventricular end diastolic volume, left ventricular end systolic volume, sroke volume and cardiac index showed significant decreases after hemodialysis. 2) There was no change in sodium, heart rate, ejection fraction and fractional shortening. 3) Peak E velocity and peak A velocity were decreased significantly after hemodialysis and the E/A ratio was increased significantly. In conclusion, hemodialysis improves the left ventricular diastolic function in patients with chronic renal failure, and Doppler echocardiography is one of the useful tools for non-invasive and repeated evaluation of left ventricular diastolic function.

        Quercetin induces caspase-dependent extrinsic apoptosis through inhibition of signal transducer and activator of transcription 3 signaling in HER2-overexpressing BT-474 breast cancer cells

        <P>Flavonoids are assumed to exert beneficial effects in different types of cancers at high concentrations. Yet, their molecular mechanisms of action remain unknown. The present study aimed to examine the effect of quercetin on proliferation and apoptosis in HER2-expressing breast cancer cells. The anti-proliferative effects of quercetin were examined by proliferation, MTT and clonogenic survival assays. The effect of quercetin on expression of apoptotic molecules was determined by western blotting. Luciferase reporter assay was performed to measure signal transducer and activator of transcription 3 (STAT3) transcriptional activity. ELISA assay was performed to measure intracellular MMP-9 levels. Immunocytochemistry was performed to evaluate the nuclear STAT3 level. The results revealed that quercetin inhibited the proliferation of BT-474 cells in a dose- and time-dependent manner. Quercetin also inhibited clonogenic survival (anchorage-dependent and -independent) of BT-474 cells in a dose-dependent manner. These growth inhibitions were accompanied with an increase in sub-G<SUB>0</SUB>/G<SUB>1</SUB> apoptotic populations. Quercetin induced caspase-dependent extrinsic apoptosis upregulating the levels of cleaved caspase-8 and cleaved caspase-3, and inducing the cleavage of poly(ADP-ribose) polymerase (PARP). In contrast, quercetin did not induce apoptosis via intrinsic mitochondrial apoptosis pathway since this compound did not decrease the mitochondrial membrane potential and did not affect the levels of B-cell lymphoma 2 (Bcl-2) and Bcl-2-associated X protein (BAX). Quercetin reduced the expression of phospho-JAK1 and phospho-STAT3 and decreased STAT3-dependent luciferase reporter gene activity in the BT-474 cells. Quercetin inhibited MMP-9 secretion and decreased the nuclear translocation of STAT3. Our study indicates that quercetin induces apoptosis at concentrations >20 <I>µ</I>M through inhibition of STAT3 signaling and could serve as a useful compound to prevent or treat HER2-overexpressing breast cancer.</P>

        Anti-allergic effects of So-Cheong-Ryong-Tang in ovalbumin-induced allergic rhinitis model

        <P>Allergic rhinitis (AR) is an allergic inflammation of the nasal airways. The Korean herbal medicine, So-Cheong-Ryong-Tang (SCRT) has been typically used for the treatment of AR for hundreds of years. In the present study, we investigated whether SCRT suppresses the progression of AR in animal model. AR was induced by ovalbumin (OVA). Treatment with SCRT was assessed to study the effect of SCRT on AR in mice. Histological analysis, multiplex cytokine assay, blood analysis, cell viability assay, RT-PCR and Elisa assay were performed to verify inhibitory effect of SCRT on AR. SCRT reduced infiltration of inflammatory cells into nasal cavity. SCRT reduced infiltration of mast cells into nasal mucosa. SCRT reduced the levels of cytokines (IL-4 and LIF) in the serum. SCRT reduced the levels of leukocytes in the blood. SCRT decreased cell viability of HMC-1 cells and splenocyte. SCRT suppressed IL-4 level in HMC-1 cells and splenocyte cells in a dose-dependent manner. SCRT suppressed IL-6 level and TNF-alpha level in splenocyte. SCRT suppresses the progression of AR induced by OVA. SCRT might be a useful drug for the treatment of AR.</P>

        Estimation of centerline temperature of the waste form for the rare earth waste generated from pyrochemical process

        <P><B>Abstract</B></P> <P>Estimation of centerline temperature of nuclear glass waste form for each waste stream is very essential in the period of storage because the centerline temperature being over its glass transition temperature results in the increase of leaching rate of radioactive nuclides due to the devitrification of glass waste form. Here, to verify the effects of waste form diameter and transuranic element content in the rare earth waste on the centerline temperature of the waste form, the surrogate rare earth glass waste generated from pyrochemical process was immobilized with SiO<SUB>2</SUB> Al<SUB>2</SUB>O<SUB>3</SUB> B<SUB>2</SUB>O<SUB>3</SUB> glass frit system, and thermal properties of the rare earth glass waste form were determined by thermomechanical analysis and thermal conductivity analysis. The estimation of centerline temperature was carried out using the experimental thermal data and steady-state conduction equation in a long and solid cylinder type waste form. It was revealed that thermal stability of waste form in case of 0.3 m diameter was not affected by the TRU content even in the case of 80% TRU recovery ratio in the electrowinning process, meaning that the waste form of 0.3 m diameter is thermally stable due to the low centerline temperature relative to its glass transition temperature of the rare earth glass waste form.</P>

      • Cucurbitacin D induces cell cycle arrest and apoptosis by inhibiting STAT3 and NF-κB signaling in doxorubicin-resistant human breast carcinoma (MCF7/ADR) cells

        <P>Breast cancer is the most common cancer for women and is a major cause of mortality in women. Doxorubicin is a generally used chemotherapy drug for breast cancer. However, multidrug resistance of breast cancer interferes with the chemotherapy. We examined whether cucurbitacin D affects doxorubicin resistance of MCF7/ADR breast cancer cells. Cell viability was measured by MTT assay. Levels of p-STAT3, p-NF-κB, IκB, and caspases were measured by Western blot analysis. Nuclear staining of Stat3 and NF-κB was measured by immunocytochemistry. STAT3 and NF-κB transcriptional activity was detected by STAT3 and NF-κB luciferase reporter gene assays. Analysis of cell cycle arrest was performed by flow cytometry. Induction of apoptosis by cucurbitacin D was measured by Annexin V-FITC/propidium iodide assay. More than 90 % of MCF7/ADR cells lived upon treatment with doxorubicin for 24 h. However, upon treatment with cucurbitacin D, cell death was more than 60 %. Co-administration of cucurbitacin D and doxorubicin induced apoptosis, and G2/M cell cycle arrest, and inhibited upregulated Stat3 by doxorubicin on MCF7/ADR cells. Additionally, cucurbitacin D led to an increase in the IκBα level in the cytosol and a decrease in the p-NF-κB level in the nucleus. Finally, cucurbitacin D inhibited translocation of Stat3 and NF-κB and decreased transcriptional activity in the nucleus. Cucurbitacin D decreases cell proliferation and induces apoptosis by inhibiting Stat3 and NF-κB signaling in doxorubicin-resistant breast cancer cells. Cucurbitacin D could be used as a useful compound to treat adriamycin-resistant patients.</P>

      • S-411 Evaluating the value of BMD loss as a risk factor for fragility fractures in Koreans

        Summary: Baseline total hip BMD is important factor for estimating fragility fracture risk in Korea, while the rate of BMD losss at femur neck is important for predicting fragility fracture in non-osteoporotic men. Introduction: Whether BMD loss predicts the fragility fracture regardless of baseline BMD is not clear. We investigated that the accelerated loss of BMD increased the fragility fractures independent of baseline BMD in elderly Koreans. Methods: This is an ongoing prospective cohort study of BMD measurements at both 2009 and 2011 year at community-based Ansung cohort, which was originally recruited in 2001-2002. A total of 840 men and 1,005 postmenopausal women aged over 50 years from Ansung cohort were categorized according to the rate of BMD loss within 2009 and 2011. Primary outcome was the incident fragility fracture. Results: During a median follow-up of 5.8±0.2 years, incident fragility fractures occurred in 51 (6.1%) men and 96 (10.6%) women. Among baseline BMD measurements at lumbar spine, femur neck, and total hip, only total hip BMD was a significant risk factor for fragility fractures in both genders. Hazard ratio (HR) of baseline total hip BMD was 1.50 (95% confidential interval [CI], 1.03-2.07; p=.03) for men and 1.44 (95% CI, 1.09-1.90; p<.01) for women. In the subgroup analysis according to baseline BMD, non-osteoporotic men with accelerated loss of BMD at femur neck had a higher risk for fragility fractures (HR=3.55, 95% CI: 1.27-9.92; p<.02) after adjustment for covariates including baseline BMD. Conclusions: Measurement of baseline total hip BMD is the most important factor for estimating fragility fracture risk in Korean men and women. Rate of BMD loss in femur neck was an independent risk factor for fragility fractures in non-osteoporotic Korean men.

        조기 폐암과 동반된 내장형 Botryomycosis 1 예

        Botryomycosis is a rare, chronic and suppurative disease that is often mistaken clinically and histologically for a fungal infection, because the histologic feature shows a cluster of bacteria found within an eosinophilic matrix or capsule, giving the appearance of granules mimicking the sulfur granules of actinomycosis. Staphylococcus aureus is the most common organism cultured from lesions of botryomycosis, but other bacteria have also been isolated and implicated, including Psudomonas aeruginosa, E. coli, Proteus spp., Bacillus spp.. In some case, multiple pathogenic bacteria were found on culture. This disease has two patterns of organ involvement. The more common type is cutaneous type, which involves the skin, subcutaneous soft tissue, muscle and bone. The other is visceral type which involves the lungs, liver, tongue, orbit, bowel, brain, kidney or prostate. We report a case of brain and pulmonary botryomycosis in a 60 year-old man, who have early lung cancer. Botryomycosis was diagnosed by brain mass removal, PCNA and bronchoscopy, and lung cancer was detected incidentally by bronchoscope. The patient was treated with the Penicillin G after operation of brain mass, and right pneumonectomy was done.
