      • Alcohol extract of Geum Chung Cho alleviates alcohol-induced short-term memory impairment in gerbils

        Chronic ethanol consumption leads to memory impairment and also induces brain damage. The hippocampus has been particularly regarded as a possible target organ of alcoholic intoxication, since this structure plays a prominent function in memory processes. Cordyceps has been used as a traditional medicine in Asian countries for a long time. Different types of Cordyceps extract were reported to have various pharmacological activities including anti-tumor, anti-inflammatory, anti-viral, and immune modulatory effects. Guem Chung Cho is an artificial cultivated Cordyceps, and was supplied from GCordy (Seoul, Korea). Ethanol extract of Cordycep (50 g) was made by rotary evaporate, and resulting powder 8.29 g was obtained (yield 17%). In the present study, we investigated the effects of ethanol extract of Geum Chung Cho on short-term memory, cell proliferation and apoptosis in the hippocampus of ethanol-treated gerbils. The gerbils were divided into 5 groups (n = 10 in each group): Control group, alcohol-treated group, alcohol-treated and 0.001 mg/kg Geum Chung Cho-applied group, alcohol-treated and 0.01 mg/kg Geum Chung Cho-applied group, and alcohol-treated and 0.1 mg/kg Geum Chung Cho-applied group. Gerbils in the alcohol-treated groups were orally administered ethanol (1 g/kg) once a day for 2 weeks. Gerbils in the Guem Chung Cho-applied groups were orally administered Guem Chung Cho extract at each dosage 1 hour before ethanol consumption. Our results revealed that consumption of alcohol for 2 weeks deteriorated short-term memory, induced apoptosis, and inhibited cell proliferation, and reduced expressions of brain-derived neurotrophic (BDNF) and tyrosine kinase B (TrkB). However, Guem Chung Cho treatment alleviated alcohol-induced short-term memory impairment through enhancing cell proliferation and BDNF expression with inhibiting apoptosis in the hippocampus. Guem Chung Cho showed maximum effects at 0.1 mg/kg. Based on these results, alcohol extract of Guem Chung Cho may provide therapeutic value for the treatment of chronic alcohol-inducted brain injury.       • Changes in cAMP Response and Testosterone to 17β-Estradiol to FSH in Isolated Rat Sertoli Cells-Age and Dose Related Differences

        The Seroli cell was identified as> the main primary target site for the action of follicle stimulating hormone (FSH) in the testis. FSH has been known to bind specifically to Sertoli cell and to evoke a series of biochemical responses such as increased accumulation of cyclic AMP, production of androgen binding protein, and steroid metabolism. Recently age related changes in FSH stimulation of cAMP and aromatization of testosterone of 17β-estradiol were demonstrated. The purpose of this study was to investigate the changes in cAMP and testosterone to 17β-estradiol aromatization responses of FSH with dose differences in isolated rat Sertoli cells of different age. Methods for preparation of rat Sertoli cells were adapted from those used to prepare rat Sertoli cells. Homogenous preparations of primary Sertoli cell cultures were obtained from the testis of the rat of different age: prepubertal (14 days old rat), pubertal (18 days old rat) and adult (60 days old rat). For cAMP determinations, cells were stimulated for 30 min at 37?C with different dose of FSH in media containing 0.1 mg/m]l l-methyl-3-isobutylxanthine. cAMP assays were performed in duplicate using radioimmunoassay kit. FSH stimulated aromatization of testosterone of 17j3-estradiol was determined in cells stimulated with different dose of FSH in media containing 0.5 μmol/ml testosterone. 170-estradiol assays were performed in duplicate by means of specific radioimmunoassay. The results were as follows: 1. Sertoli cells from adult rats responded to FSH stimulation with over 30-fold rise at 5 min. At 30 min, cAMP is significantly lower and down to baseline level by 60 min. 2. There were age-related differences in Sertoli cell cAMP response after 30 min of FSH stimulation. Sertoli cells from pubertal rats responded most sensitively to FSH with a dose response relationship. 3. The heighest aromatase enzyme activity in converting testosterone to 17(3-estradiol was found in pubertal rat Sertoli cells. Sertoli cells from pubertal rats demonstrated a dose-response of FSH stimulation of testosterone to estradiol conversion. In conclusion, these results demonstrate that the age and dose-related differences in rat Sertoli cell response to FSH, and also provide direct evidence that the Sertoli cells represent a primary target site for FSH activity in the testes.

        Metal stain on monolithic zirconia restoration: A case report

        In restorative treatment using fixed dental prostheses, dentists should select appropriate restoration material among various types of dental materials. The strength, marginal fit, esthetics, wear resistance, biocompatibility, and cost are important factors in the choice of restoration materials. The present case showed a surface stain on a monolithic zirconia restoration that was due to wear between the monolithic zirconia restoration and the base metal alloy restoration. This phenomenon was confirmed by surface roughness measurement and electron probe micro-analysis.

        Production of Iron-Binding Peptides from Colostral Whey by Enzymatic Hydrolysis

        Colostral whey prepared from colostrum (pooled from first six post-partum milkings) was heated for 10 min at 100oC. Heated colostral whey was incubated with 1% enzymes (protein equivalent basis) for 15, 30, 60, 90, and 120 min at 50oC. Papain, pepsin, trypsin, and alcalase produced different degrees of hydrolysis (DH), 10.66%, 12.42%, 10.83%, and 25.31%, respectively, at an incubation time of 120 min. The SDS-PAGE reveals that significant amounts of bovine serum albumin (BSA), β-lactoglobulin (β-LG), and α-lactalbumin (α-LA) survived papain digestion. In contrast, pepsin completely removed BSA but not β-LG present in heated colostral whey. Alcalase completely eliminated BSA, β-LG, and α-LA. This differential hydrolysis was confirmed by reversed-phase HPLC analysis. Using ion-exchange chromatography, fraction-1 (F-1) was obtained from alcalase hydrolysate at a NaCl gradient concentration of 0.25 M. Reversed-phase HPLC chromatograms of alcalase F-1 showed numerous small peaks, which probably indicate that a variety of new peptides were produced. Iron content of alcalase F-1 was 28.94 ppm, which was the highest among all enzyme fractions, whereas iron content of colostral whey was 36.56 ppm. Main amino acids contained in alcalase F-1 were Thr (15.45%), Glu (14.12%), and Ser (10.39%). Therefore, alcalase can be used to generate good iron-binding peptides in heated colostral whey, and the resulting iron-binding peptides could be suitable as a value-added food ingredient for food supplements.

        Skin entrance dose for digital and film radiography in Korean dental schools

        Purpose : This study was aimed to compare skin entrance dose of digital radiography with that of film radiography and to show the dose reduction achievement with digital systems at 11 dental schools in Korea. Materials and Methods : Forty six intraoral radiographic systems in 11 dental schools were included in this study. Digital sensors were used in 33 systems and film was used in 13 systems. Researchers and the volunteer visited 11 dental schools in Korea. Researchers asked the radiologic technician (s) at each school to set the exposure parameters and aiming the x-ray tube for the periapical view of the mandibular molar of the volunteer. The skin entrance doses were measured at the same exposure parameters and distance by the technician for each system with a dosimeter (Multi-O-Meter : Unfors instruments, Billdal, Sweden). Results : The median dose was 491.2 μGy for digital radiography and 1,205.0 μGy for film radiography. The skin entrance dose digital radiography was significantly lower than that of film radiography (p<0.05). Conclusion : Fifty-nine percent skin entrance dose reduction with digital periapical radiography was achieved over the film radiography in Korean dental schools.

        Ethanol Production from Seaweed, Enteromorpha intestinalis, by Separate Hydrolysis and Fermentation (SHF) and Simultaneous Saccharification and Fermentation (SSF) with Saccharomyces cerevisiae

        Ethanol productions were performed by separate hydrolysis and fermentation (SHF) and simultaneous saccharification and fermentation (SSF) processes using seaweed, Enteromorpha intestinalis (sea lettuce). Pretreatment conditions were optimized by the performing thermal acid hydrolysis and enzymatic hydrolysis for the increase of ethanol yield. The pretreatment by thermal acid hydrolysis was carried out with different sulfuric acid concentrations in the range of 25 mM to 75 mM $H_2SO_4$, pretreatment time from 30 to 90 minutes and solid contents of seaweed powder in the range of 10~16% (w/v). Optimal pretreatment conditions were determined as 75 mM $H_2SO_4$ and 13% (w/v) slurry at $121^{\circ}C$ for 60 min. For the further saccharification, enzymatic hydrolysis was performed by the addition of commercial enzymes, Celluclast 1.5 L and Viscozyme L, after the neutralization. A maximum reducing sugar concentration of 40.4 g/L was obtained with 73% of theoretical yield from total carbohydrate. The ethanol concentration of 8.6 g/L of SHF process and 7.6 g/L of SSF process were obtained by the yeast, Saccharomyces cerevisiae KCTC 1126, with the inoculation cell density of 0.2 g dcw/L.

        Background/Aims: Endoscopic submucosal dissection (ESD) is an effective procedure for en-bloc curative resection of the colorectal tumor. As it requires high technical skills and experience in therapeutic endoscopy, it is important to understand learning curve of ESD technique. The aim of this study was to retrospectively describe the clinical results of ESD and to evaluate learning curve for the ESD of colorectal tumors. Methods: A total of 90 patients with 90 colorectal neoplasms, who had undergone ESD at a tertiary referral hospital from July 2009 to December 2012, were enrolled. The ESD was performed by a single endoscopist. All ESD cases were divided into three periods: first, cases 1-30; second, cases 31-60; and third, cases 61-90. Results: The en-bloc resection rates in third period (100%) was significantly higher than that of the first (93.3%) and second period (80%) (p=0.025). The perforation rate in third period (0%) also significantly decreased compared with that of the first (13.3%) and second period (20%) (p=0.032). To calibrate the difference of tumor size among periods, proficiency was calculated, as the procedure time per specimen area (min/cm2). The proficiency in third period (4.3) was significantly shorter than that of the first (16.8) and second period (10.2) (p=0.004). Conclusions: The learning curve of colorectal ESD in our study shows that at least 60 cases of ESD have to be conducted to acquire sufficient skill of degree without perforation.         Analysis of Lignans in Acanthopanax sessiliflorus Fruits and Their Fermented Wine by HPLC

        High performance liquid chromatography (HPLC) was used for the determination of lignans, eleutherosides B and E, in Acanthopanax sessiliflorus fruits and their fermented wine. The lignans were quantified by a reversed-phase system using a gradient of H2O and acetonitrile as a mobile phase within 20 min. The analysis was successfully carried out within 20 min. The contents of eleutherosides Band E as main active principles of Acanthopanax species were measured in A. sessiliflorus fruits (1.15 and 8.49 μg/mg, respectively), their fermented wine (0.45 and 1.33 μg/mg, respectively) and wine residues (no detection).
