        성장생물학 : 체지방 감소 다클론 후보 항체가 비거세돈의 체중, 분 소화율 및 혈액 대사물질에 미치는 영향

        본 연구는 비거세돈 12두를 이용하여 돼지 복강 및 피하지방 감소 후보 항체를 생체 주사 처리 시 체중, 분 소화율 및 혈액 대사물질 등 영양생리대사에 미치는 영향을 조사하고자 실시하였다. Choi 등(2010)에서 개발된 돼지 복강 및 피하지방 감소 후보 항체를 피하 주사하였을 때 주사 후 4주까지 평균 체중은 수치적 증가가 발생했으나(P>0.05), 전 기간에 걸쳐 수치적 증가를 보여 항체의 처리로 인한 영향보다는 돼지의 일반적인 성장에 기인하는 것으로 판단된다. 항제 주사 처리 시 건물, 조단백질, 조지방 및 조섬유의 분 소화율은 항체 주사 전, 후 및 시험구 간의 통계적 유의성이 나타나지 않았다(P>0.05). 조회분 소화율의 경우 항체 주사 후 2주째 control에서 감소를 및 4주째 NAb에서 증가를 보였으나(P< 0.05), AAb 및 SAb에서는 유의적 변화가 없었다. 혈액 대사물질의 변화에서는 항체 주사 후 4주의 AAb 및 SAb에서 BUN 농도가 유의적으로 증가하는 것으로 나타났다(P<0.05). 하지만, 후보 항체 주사 전에서도 유사한 패턴으로 유의적 증가를 보여 항체 처리에 의한 차이보다는 개체간의 차이로 사료된다. 혈액 내 glucose 및 TG 농도는 후보 항체 처리에 의한 유의적 차이는 나타나지 않았다(P>0.05). 체지방 함량과 밀접한 관계에 있는 혈액 내 TC 농도에서는 control 대비 후보 항체 주사 후 4주차 AAb 및 SAb에서 유의적 감소가 나타나(P<0.05), 체지방 감소가 일어나고 있음을 간접적으로 추론할 수 있다. 이상의 결과를 볼 때, 본 연구에서 이용된 AAb 및 SAb는 in vivo 영양생리대사에 부정적 영향을 미치지 않는 안전한 항체로 판단된다. Twelve pigs were used to investigate the effects of polyclonal antibody candidate against abdominal (AAb) and subcutaneous adipocyte membrane proteins (SAb) on body weight, fecaldigestibility and blood metabolites. When AAb and SAb developed by Choi et al. (2010) were injected to pigs, the numerical increase in BW (body weight) occurred at 4 weeks post-treatment, but BW for an entire period was also increased, indicating that the BW increase may not be affected by the antibodies injection. Antibodies treatment did not affect (P>0.05) fecal digestibility of dry matter, crude protein, crude fat and crude fiber. Fecal digestibility of crude ash for control (no treatment) at 2 weeks decreased, and that for non-immunized serum treatmentgroup at 4 weeks post-treatment increased, respectively (P<0.05). However, fecal digestibility of crude ash for AAb and SAb groups did not significantly change. At 4 weeks after the antibodies treatment, blood urea N concentration for AAb and SAb groups was significantly increased (P<0.05). However, these increases may not be caused by the antibodies treatment because similar pattern in blood urea N concentration occurred before the antibodies treatment. Antibodies treatment did not affect concentration of plasma glucose and triglycerides (P<0.05). Compared with control, concentration of plasma total cholesterol for AAb and SAb groups at 4 weeks post-treatment was significantly (P<0.05) decreased. This may suggest that body fat reduction possibly occurs. In conclusion, the AAb and the SAb developed by Choi et al. (2010) may have safety in nutritional physiological metabolism in pigs. Further study on in vivo fat reduction of the antibodies against abdominal and subcutaneous adipocytes of pigs should be required for fat-reduced pork production.

        Study of Metabolic Profiling Changes in Colorectal Cancer Tissues Using 1D <sup>1</sup>H HR-MAS NMR Spectroscopy

        Metabolomics is a field that studies systematic dynamics and secretion of metabolites from cells to understand biological pathways based on metabolite changes. The metabolic profiling of intact human colorectal tissues was performed using high-resolution magic angle spinning (HR-MAS) NMR spectroscopy, which was unnecessary to extract metabolites from tissues. We used two different groups of samples, which were defined as normal and cancer, from 9 patients with colorectal cancer and investigated the samples in NMR experiments with a water suppression pulse sequence. We applied target profiling and multivariative statistical analysis to the analyzed 1D NMR spectra to identify the metabolites and discriminate between normal and cancer tissues. Cancer tissue showed higher levels of arginine, betaine, glutamate, lysine, taurine and lower levels of glutamine, hypoxanthine, isoleucine, lactate, methionine, pyruvate, tyrosine relative to normal tissue. In the OPLS-DA (orthogonal partial least square discriminant analysis), the score plot showed good separation between the normal and cancer groups. These results suggest that metabolic profiling of colorectal cancer could provide new biomarkers.

      • 위암세포주에서 Recombinant Human Interferon-r와 Adriamycin의 투여순서가 항암효과에 미치는 영향

        Numerous previous studies, both in vitro and in vivo, have demonstrated that the cytotoxicity can be enhanced by the combination of chemotherapeutic agent and interferons(IFNs) in various types of cancer cells. We have previously reported that combined treatment of MKN-45, human gastric adenocarcinoma cells, with adriamycin(ADM) and recombinant human interferon-r(rh-IFN-r) increased in the cytotoxicity. In this study, the effects of combination timing of rh-IFN-r and ADM on the cytotoxicity against MKN-45 were investigated using MTT assay. MKN-45 was treated with rh-IFN-r and ADM in vitro on three schedules : Treat A ; rh-IFN-r and ADM were treated simultaneously, Treat B ; rh-IFN-r was treated 24 hours after the treatment with ADM, Treat C ; rh-IFN-r was treated for 72 hours and followed by the treatment with ADM. The survival of MKN -45 was inhibited by ADM dose-dependently. 102 and 103U/ml of rh-IFN-r significantly inhibited the survival of MKN-45(% survival : 35.1 ±-1.2% and 34.4 ±1.1% in Treat A and 42.5 ± 2.1% and 45.9-±2.5% in Treat C, respectively). However no difference in the survival was observed between 102 and 103U/ml of rh-IFN-r. Combined treatment with rh-IFN-r and ADM significantly augmented the cytotoxicity at low concentrations of ADM. Combined effects of rh-IFN-r and ADM were evaluated using IC30(,ag/ml) to ADM. IC30s of MKN-45 in Treat A, B and C at 102 U/ml of rh -IFN-r _ were 0.019 -?- 0.003, 0.045 :I:0.001 and 0.054 ± 0.012, respectively, while IC30 of MKN-45 treated with ADM alone was 0.052±0.004. IC30s of MKN-45 in ADM alone group, Treat A, Treat B and Treat C at 103U/ml of rh-IFN-r were 0.047 ±0.003, 0.004 -±0.001, 0.031 ±0.004 and 0.056 0.008, respectively. These results indicate IC30s of Treat A and B were significantly lower than those of ADM alone(p<0.05) and IC30s of Treat A was significantly lower than those of Treat B(p <0.01). IC30s of Treat C, however, were not different from those of ADM alone. From these results demonstrating that cytotoxic effects were increased by the combination of rh-IFN-r and ADM in the order, Treat A > Treat B> Treat C, it can be concluded that the simultaneous administration of rh-IFN-r and ADM may be the most effective method to combine these two therapeutic modalties.

      • 유전자 증폭법(PCR)을 이용한 methicillin-registant Staphylococcus 균속의 신속동정에 관한 연구

        In order to indentify rapidly the methicillin-resistant genus Staphylococcus (MRS) which is known as a causative bacteria of nosocomial infection, we performed the polymerase chain reaction (PCR) method. In this study, we used 44 strains which were consist of 39 starins of Staphylococcus aureus, 2 strains of Staphylococcus epidermidis. 1 strain of Staphylococcus warneri, 1 strain of Staphylococcus cohnii isolated from clinical specimens and 1 type strain of Staphylococcus aureus KCTC 1928 for PCR experiments. DNA was extracted directly from the bacterial colony. 22 mer-oligonucleotides were used as primer which complements to the mecA region of structural gene coding low affinity binding pro-tein (PBP2a). A single band of 533 bp length from MRSA, MRSE and MRSW were confirmed. Also, an antibiotic susceptibility test by agar dilution method of NCCLS was performed for the strains isolat-ed from clinical specimens. The results obtained by this method were compared with the PCR results. In the result, most of the strains which have strong resistance to methicillin were amplified for the 533 bp fragment of the mecA gene in PCR experiment. However, 4 strains (MRA1-10, MRW2-3. MRE1-15. MRE1-16) that were classified as a sensitive group in the antibiotic susceptibility test also was showed mecA gene fragment in PCR. This suggested that the specificity of mecA gene is not always consistant with that of MRSA. In order to confirm the fragment in a region of mecA gene, a southern blot hybridization process was performed using the PCR products as probe. As a result, a single hybrid band was detected at about 4 Kb region of Hind III and Hae III-digested DNA fragment in MRS. Conclusively for the diagnosis of MRS, the confirmation of mecA gene by PCR seems to be more accurate and rapid than the antibiot-ic sensitivity test.

        The electrochemical reduction of 4-nitro-N-(triphenyl-phosphoranylidene) in nonaqueous media has been examined by polarography, cyclic voltammetry and controlled-potential coulometry. The reduction of 4-nitro-N-(triphenylphosphoranylidene) proceeds by a one electron-transfer to form anion radical which undergoes both protonation and a second one electron reduction followed by cleavage of a phosphorous-phenyl bond was also observed after reduction of triphenylphosphine oxide which is one of the major products of the chemical reaction which follow the primary electrode process.

      • 八公山의 山火跡地의 二次植生과 二次遷移

        This paper is the investigation of the secondary vegetation and succession in the plant community(pine forest) destroyed by the crown and surface fire(the early spring, 1969), and results are as follows: Soil gradually comes to be rich in humus and organic matters are accumulated. The vegetation of grassland tends to be substituted for shrubby vegetation. Compared with the change of relative humidity and saturating deficiency in the daytime, the west slope shows the larger change than the east slope. The floristic compositions are 25 families, 52 genera, 49 species, 6 varieties and 2 formae(57 kinds of vascular plants). Biological type succeeds in H-D_1-R_5-e type. Dominant species are Miscanthus sinensis var. purpurascens(79.10)-Lespedeza maximowiczii(61.38) and Carex bostrychostigma(60.45). Lespedeza maximowiczii is gradually prevailing as dominant species, Carex bostrychostigma and Miscanthus sinensis var. purpurascens are dominant species in the understory. The index of dominance(C) of east slope is higher than that of west slope. In the species diversity(H ̄) and evenness index(e), various species are distributed evenly throughout this area. The degree of succession of west slope is higher than that of east slope. The quotient of similarity between east slope and west slope is lower than that of same slope.

      • 구충감염에 의한 철결핍성 빈혈 1예

        Hookworm infection is one of the major parasitic diseases in warm moist climates, but its prevalence in Korea is very rare nowadays. Authors examined a case of iron deficiency anemia (IDA) associated with hookworm infection in a 46-year-old male. The peripheral blood examination showed markedly increased number of eosinophils, a low level of hemoglobin and decreased MCV. The iron study revealed that serum ferritin was 10.4 ng/ml, serum iron was 30㎍/dl. and total iron-binding capacity was 423㎍/dl. A large number of hookworm ova (7,925/g) were found in the stool sample. The G-I studies were all normal findings. We propose that we have to consider hookworm infection as one of the causes in a case of unknown IDA origin.

        This paper presents predicting an accelerated life test of glow starter for flourescent lamp by reliability tool ALTA(software). Failure mode/mechanism of glow starter can be classified into two groups. One is open failure due to glow tube cracking and capacitance changing. The other failure mode is short failure due to contact welding of bimetal in glow tube. In this study, main failure mechanism of glow starter was glow tube cracking due to on/off cycle in accordance with different test conditions of acceleration stresses(voltage).
