        Adenosine triphosphate-based chemotherapy response assay-guided chemotherapy in unresectable colorectal liver metastasis

        <P><B>Background:</B></P><P>This study aims to evaluate the effectiveness of adenosine triphosphate-based chemotherapy response assay (ATP-CRA)-guided neoadjuvant chemotherapy for increasing resectability in patients with unresectable colorectal liver metastasis.</P><P><B>Patients and methods:</B></P><P>Patients were randomised into two groups: Group A was treated by conventional chemotherapy regimen and Group B was treated by chemotherapy regimen according to the ATP-CRA. Three chemotherapeutic agents (5-fluorouracil, oxaliplatin and irinotecan) were tested by ATP-CRA and more sensitive agents were selected. Either FOLFOX or FOLFIRI was administered. Between Group A and B, treatment response and resectability were compared.</P><P><B>Results:</B></P><P>Between November 2008 and October 2010, a total 63 patients were randomised to Group A (<I>N</I>=32) or Group B (<I>N</I>=31). FOLFOX was more preferred in Group A than in Group B (26 out of 32 (81.3%) <I>vs</I> 20 out of 31 (64.5%)). Group B showed better treatment response than Group A (48.4% <I>vs</I> 21.9%, <I>P</I>=0.027). The resectability of hepatic lesion was higher in Group B (35.5% <I>vs</I> 12.5%, <I>P</I>=0.032). Mean duration from chemotherapy onset to the time of liver resection was 11 cycles (range 4–12) in Group A and 8 cycles (range 8–16) in Group B.</P><P><B>Conclusion:</B></P><P>This study showed that tailored-chemotherapy based on ATP-CRA could improve the treatment response and resectability in initially unresectable colorectal liver metastasis.</P>

      • CD19 signalling improves the Epstein–Barr virus-induced immortalization of human B cell

        <P>Abstract. </P><P>Epstein–Barr virus (EBV) infection <I>in vitro</I> immortalizes primary B cells and generates B lymphoblastoid cell lines (LCLs). These EBV-LCLs have been used for several purposes in immunological and genetic studies, but some trials involving these transformations fail for unknown reasons, and several EBV-LCLs do not grow in normal culture. In this study, we improved the immortalization method by CD19 and B-cell receptor (BCR) co-ligation. This method shortens the time required for the immortalization and generation of EBV-LCLs but does not alter the cell phenotype of the LCLs nor the expression of the EBV genes. In particular, the CD19 and BCR co-ligation method was found to be the most effective method examined. EBV-infected B cells induced by CD19 and/or BCR ligation expressed the intracellular latent membrane protein LMP-1 earlier than EBV-infected B cells, and the expression of intracellular LMP-1 was found to be closely related to the time of immortalization. These results suggest that the modified method, using CD19 and/or BCR ligation, may efficiently generate EBV-LCLs, by expressing intracellular LMP-1 at an early stage.</P>

      • ASK1/JNK-mediated TAp63 activation controls the cell survival signal of baicalein-treated EBV-transformed B cells

        <P>Transcriptionally active p63 (TAp63) promotes cell cycle arrest, senescence, and apoptosis in several cancer cells. Migration inhibitory factor (MIF)/CD74 regulates B-cell survival through nuclear factor (NF)-kappa B-dependent TAp63 expression. In this study, we investigated how the level of TAp63 expression influences the induction of apoptosis in baicalein-treated EBV-transformed B cells. Baicalein induced the expression of TAp63 and apoptosis signal-regulating kinase 1 (ASK1), as well as cytotoxicity, by disrupting the mitochondrial membrane and inhibiting the activation of phosphoinositide 3-kinase (PI3K)/Akt and NF-kappa B. Genetic knockdown of TAp63 or ASK1 by small interfering RNA resulted in protection from apoptosis accompanied by the recovery of CD74, CD44, alpha 4 integrin, Bcl-2, and NF-kappa B activation. Baicalein-induced reactive oxygen species activated the ASK1/JNK pathway with subsequent expression of TAp63. Pre-engagement with MIF/CD74 maintained the expression of CD74, CD44, and alpha 4 integrin, as well as Syk/Src-mediated PI3K/Akt activation, in baicalein-treated EBV-transformed B cells. Meanwhile, ASK1/JNK-dependent TAp63 expression was efficiently suppressed after pre-treatment with MIF. Our results suggest that baicalein-mediated ASK1/JNK activation regulates the mitochondria-dependent apoptosis pathway through the up-regulation of TAp63 and downregulation of NF-kappa B and CD74/CD44 in B-cell malignancies.</P>

        포장방법과 저장조건이 돈육 등심의 저장성 및 보수력에 미치는 영향

        To investigate the effects of packaging method and storage condition on meat quality, a total of eight pork loins were selected from a commercial meat plant at 24 hr postmortem. The loins were cut to 2.5㎝ thick steaks and were randomly assigned to the five treatment combination; vacuum packaging and submersion in chilled salt water(SCSW)(T1), non-vacuum packaging and submersion in chilled salt water (T2), vacuum packaging and storage in conventional refrigerator(SCR)(T3), non-vacuum packaging and storage in conventional refrigerator(T4), and wrap packaging and storage in conventional refrigerator(TS). All samples were stored at 0℃ for 20 days to measure color of meat(CIE L*, a*. b*), pH, drip loss, purge loss %, lipid oxidation (TBARS; thiobarbituric acid reactive substances) and total plate counts (TPC). CIE L* and b* value of SCSW(T1, 2) samples were significantly(P$lt;0.05) lower than SCR samples when they were measured at storage 20 days. The sample of SCSW(T1, 2) showed significantly(P$lt;0.05) lower pH than those of SCR(T3, 4, 5) during storage. The sample of SCSW(T1, 2) showed significantly(P$lt;0.05) higher Drip loss than those of SCR(T3, 4, 5) during 5 days storage. T2 showed the lowest purge loss % whereas T3 remarked the highest purge loss %. The samples of SCSW showed significantly(P$lt;0.05) lower TBARS than those of SCR during storage. TPC was samples of SCSW(T1, 2) showed significantly(P$lt;0.05) lower than other treatments. These results suggested that NVP(non-vacuum packaging) and SCSW could reduce purge loss and discoloration, and extend shelf-life of pork loin.

        Purpose: The purpose of this study was to investigate clinical practice stress, academic self-efficacy, and career identity among undergraduate students in nursing practicum. Method: This study was a descriptive correlational design using the survey method. The study included 267 undergraduate students attending to 5 college in Seoul. The data was gathered from August 2014 to October 2014 and the data were analyzed using IBM SPSS statistics version 22.0 Program with descriptive statistics, t-test, analysis of variance with Scheffe test, and Pearson’s correlation coefficient. Results: Situational characteristics of Clinical Practice Stress(CPP) was significantly different with regards to task difficulties and efficacy self-confidence of academic self-efficacy(ASE) (r= .179, p= .003; r= .166, p= .006). Statistically significant positive relationship was founded between Personal characteristics of CPP and self-confidence of ASE (r= .239, p< .01). External regulators of CPP was significantly different according to self-confidence and task difficulties (r= .175, p= .003; r= .132, p= .028). The self-confidence and efficacy for self-confidence of ASE were a significantly correlated with Career Identity; The Career identity also was a significantly correlated with all sub-categories of CSS scale(r= -0.208, p< 0.01; r= -0.369, p< 0.01; r= -0.153, p< .011) Conclusion: This study showed positive statistical relationships associated with clinical practice stress, academic self-efficacy, and career identity as a preliminary evidence-based study for guiding education in nursing curriculum. Further research should be need for students’ future as a professional nurse.

      • Boro-Nitriding 처리한 FC-25계 편상흑연주철의 표면특성에 관한 연구

        Surface properties and oxidation behaviors were studied for FC-25 gray cast iron (GCI), plasma nitrided FC-25 GCI (P-N GCI), powder boronized FC-25 GCI (B GCI) and powder boronized N GCI (B-N GCI). A boride (Fe2B, FeB) layer with a tooth shape was formed by boronizing GCI at temperatures of about 650℃. The thickness and hardness of the layers were proportional to the boronizing temperature and time. The hardness was decreased by increasing boronizing depth. The activation energy for boride formation was 73.8kJ/mol. The thickness of the graphite layer formed by boronizing was increased by the temperature and time. A silicon layer insoluble in borides was accumulated under the boride layer. The major phases were Fe4N, Fe3N and FeN in a nitride layer formed by plasma. The thickness and hardness of the layer were increased with nitriding temperatures and time under the Fe4N decomposition temperature of 680℃. The hardness was maximum at the surface and decreased with nitrided depth on B-N GCI at 700℃, above the decomposition temperature of the Fe4N. Hardness of B-N GCI at the layer depth of 10 ㎛ was ≒1000Hv, which was lower than ≒1500Hv of the B GCI at the same condition. Also the layer thickness was 50 ㎛; deeper than that of B GCI by 10 ㎛ The reasons were that the nitrogen given off by the decomposition made the layer porous and facilitated the diffusion of oxygen and other reactants. The samples were oxidated at 800℃ for 48hrs under atmospheric air. Oxidation rate was in decreasing order of N GCI, GCI, B GCI and G-N GCI. Oxidation rates were greatly influenced by the diffusion of nitrogen formed by Fe4N decomposition over the decomposition temperature of 680℃ and by the oxidation barrier of the less corrosive boride layer.

      • Analysis of HOX Gene Expression Patterns in Human Breast Cancer

        HOX genes are highly conserved transcription factors that determine the identity of cells and tissues along the anterior-posterior body axis in developing embryos. Aberrations in HOX gene expression have been shown in various tumors. However, the correlation of HOX gene expression patterns with tumorigenesis and cancer progression has not been fully characterized. Here, to analyze putative candidate HOX genes involved in breast cancer tumorigenesis and progression, the expression patterns of 39 HOX genes were analyzed using breast cancer cell lines and patient-derived breast tissues. In vitro analysis revealed that HOXA and HOXB gene expression occurred in a subtype-specific manner in breast cancer cell lines, whereas most HOXC genes were strongly expressed in most cell lines. Among the 39 HOX genes analyzed, 25 were chosen for further analysis in malignant and non-malignant tissues. Fourteen genes, encoding HOXA6, A13, B2, B4, B5, B6, B7, B8, B9, C5, C9, C13, D1, and D8, out of 25 showed statistically significant differential expression patterns between non-malignant and malignant breast tissues and are putative candidates associated with the development and malignant progression of breast cancer. Our data provide a valuable resource for furthering our understanding of HOX gene expression in breast cancer and the possible involvement of HOX genes in tumor progression.

        Bioabsorbable bone plates enabled with local, sustained delivery of alendronate for bone regeneration

        <P>We prepared a bone plate enabled with the local, sustained release of alendronate, which is a drug known to inhibit osteoclast-mediated bone resorption and also expedite the bone-remodeling activity of osteoblasts. For this, we coated a bone plate already in clinical use (PLT-1031, Inion, Finland) with a blend of alendronate and a biocompatible polymer, azidobenzoic acid-modified chitosan (i.e., Az-CH) photo-crosslinked by UV irradiation. As we performed the in vitro drug release study, the drug was released from the coating at an average rate of 4.03 mu g/day for 63 days in a sustained manner. To examine the effect on bone regeneration, the plate was fixed on an 8 mm cranial critical size defect in living rats and the newly formed bone volume was quantitatively evaluated by micro-computed tomography (micro-CT) at scheduled times over 8 weeks. At week 8, the group implanted with the plate enabled with sustained delivery of alendronate showed a significantly higher volume of newly formed bone (52.78 +/- 6.84%) than the groups implanted with the plates without drug (23.6 +/- 3.81%) (p < 0.05). The plate enabled with alendronate delivery also exhibited good biocompatibility on H&E staining, which was comparable to the Inion plate already in clinical use. Therefore, we suggest that a bone plate enabled with local, sustained delivery of alendronate can be a promising system with the combined functionality of bone fixation and its expedited repair.         Quality characteristics of imitation crab sticks made from Alaska Pollack and spent laying hen meat

        Imitation crab stick (ICS) samples were divided into four treatments, a control (C) prepared with Alaska Pollack as a commercial ICS, T1, which consisted of Alaska Pollack with spent laying hen surimi collected by the pH adjust method, T2, which was composed of Alaska Pollack with spent laying hen surimi collected by the filter cake method, and T3, which consisted of Alaska Pollack with whole spent laying hen meat batter collected by the cutting method. The moisture content was significantly higher in T3 than in the other ICS samples; but, there was no significant difference in the crude protein, fat and ash content, regardless of the spent laying hen substitution methods. The lightness (L*) and whiteness (W) was higher in the control than in the other ICS samples at 0 days of storage, whereas the yellowness (b*) was significantly higher in T3 than in the other ICS samples. The level of polyunsaturated fatty acids was significantly higher in the control group than in the other ICS samples. Additionally, the pH increased with storage time in the spent laying hen substituted samples (T1, T2 and T3), with T1 showing a significantly higher pH during storage. The TBARS value increased with storage time in all ICS samples, with T2 showing a significantly lower TBARS value than the other ICS samples at the beginning and end of the storage periods. There was no significant difference in any sensory evaluation items among the ICS samples during storage. Thus, we assumed that T3 was better than other ICS samples, because T3 method (cutting) is much easier to collect spent laying hen surimi than T1 (pH adjust) and T2 (filter cake).
