        Elevated extracellular calcium ions promote proliferation and migration of mesenchymal stem cells via increasing osteopontin expression

        Supplementation of mesenchymal stem cells (MSCs) at sites of bone resorption is required for bone homeostasis because of the non-proliferation and short lifespan properties of the osteoblasts. Calcium ions (Ca2+) are released from the bone surfaces during osteoclast-mediated bone resorption. However, how elevated extracellular Ca2+ concentrations would alter MSCs behavior in the proximal sites of bone resorption is largely unknown. In this study, we investigated the effect of extracellular Ca2+ on MSCs phenotype depending on Ca2+ concentrations. We found that the elevated extracellular Ca2+ promoted cell proliferation and matrix mineralization of MSCs. In addition, MSCs induced the expression and secretion of osteopontin (OPN), which enhanced MSCs migration under the elevated extracellular Ca2+ conditions. We developed in vitro osteoclast-mediated bone resorption conditions using mouse calvaria bone slices and demonstrated Ca2+ is released from bone resorption surfaces. We also showed that the MSCs phenotype, including cell proliferation and migration, changed when the cells were treated with a bone resorptionconditioned medium. These findings suggest that the dynamic changes in Ca2+ concentrations in the microenvironments of bone remodeling surfaces modulate MSCs phenotype and thereby contribute to bone regeneration.

        성인의 건강관심도 수준에 따른 식용곤충 인식도 및 식용곤충 활용제품 선택속성 분석

        This study aims to present theoretical-practical information for the vitalization of the food insect industry by identifying consumer awareness and selection attributes of edible insects according to the health concern level of edible insects, which are high-protein and widely known as eco-friendly foods. Subjects to the survey were male and female consumers in their 20s and under 70s nationwide, using structured questionnaires. Data from 412 respondents were used for analysis. The results were as follows: First, the results of a survey on perception levels of edible insects according to levels of health concerns showed that both ‘higher health concerns’ group and ‘lower health concerns’ group perceived edible insects as ‘edible insect menus and edible insect products are protein-rich foods(3.85 and 3.43 points, respectively; p<0.001) and ’edible insects are future alternative foods’(3.79 and 3.23 points, respectively; p<0.001). Second, according to the survey on the selection attributes when purchasing edible insect products, Selection attributes of ‘higher health concerns’ group was in the order of stability (4.67 points), hygiene (4.61 points), and health functional (4.25 points), while those of ‘lower health concerns’ group was in the order of hygiene (4.49 points), food stability (4.44 points) and sensual factors (4.11 points). When purchasing edible insect food, there were statistically significant differences between survey groups according to health concerns in all selection attributes, except for sensual factors and hygiene(p<0.01, p<0.001). At a time when a lot of researches is being conducted on edible insects, I think this article will contribute to popularizing them as a paper that has examined the perception of them according to the level of health concern.

        邱一馬 高速道路는 統通費用의 節減, 物的 統通過程의 合理化를 通한 流通構造의 效率性을 높여주고 있으며 各種財貨의 價格變動을 완화시켜주고 또한 價格의 地域的 隔差解消에 이바지하고 있다. 同時에 홈速道路의 開通되기 전에 不必要하게 介在되먼 中間商이 배제되므로서 流通 「마진」을 줄이며 市場組織의 合理化, 流通機能의 高度化및 物的 統通施設의 擴充이 急速히 推進되고 있다. 邱一馬 高速道路의 開通은 觀光의 供給側面에서는 有名觀光地를 有機的으로 連結시켜 觀光對象을 據大시켜 주고 있으며 需要個I面에서는 觀光에 대한 需要를 자극하여 원거리 觀光을 可能케 하여 주고 있다. 특히 邱一馬 高速道速는 過去 南海岸一帶를 觀光하려면 國道를 利用하거나 京釜高速道路와 南海高速道路를 거쳐야 하는 不便을 없애주며 時間의 短縮과 經濟的 負擔을 輕減시켜 觀光需要創出에 큰 效果를 가져 오며 有關工業團地를 순환적으로 連結시켜 産業觀光을 통한 觀光形態의 建全化에도 크게 기여 하고 있다.

        B형간염바이러스 표면항원과 C형간염바이러스 항체 헌혈혈액선별검사 숙련도시험 결과(2012∼2015년)

        Background: Donor screening test is one of the most important processes for blood safety management. Korea Center for Disease Control and Prevention (KCDC) has been conducting an annual proficiency test program that includes the distribution of specially manufactured panels for hepatitis B surface antigen (HBsAg) and antibody to hepatitis C virus (anti-HCV) to blood centers. Here, KCDC reports the results of these proficiency tests for HBsAg and anti-HCV blood donor screening for all licensed blood centers in Korea between 2012 and 2015. Methods: Panels for the proficiency tests were manufactured and distributed to blood centers by Chung-Ang University Hospital, which has been participating in the Korea Blood Safety Commission. Well-proven reactive sera and healthy donor’s sera acquired from the Human Serum Bank in Chung-Ang University were used to make the panels. To identify the S/CO ratio of the panel, three medical institutes triple-checked the results of each panel. Results: Most blood centers reported correct answers for the proficiency test with six panels. The average percentages (year) of correct answers were as follows: 98.7% (2012), 98.5% (2013), 99.1% (2014) and 99.6% (2015) for the HBsAg proficiency tests; and 97.7% (2012), 99.5% (2013), 99.1% (2014), and 99.8% (2015) for the anti-HCV proficiency tests. Conclusion: To improve the blood center’s ability for donor screening tests, KCDC will continue the proficiency test program by managing specialized panels for HBsAg and Anti-HCV tests. Furthermore, we will investigate the level of satisfaction to improve the quality of the program. 배경: 혈액원의 헌혈혈액 선별검사 능력은 혈액 안전성 확보를 위해 매우 중요하다. 질병관리본부는 혈액원을 대상으로 매년 항원성이 약하거나, 저농도의 항체를 가진 특화된 패널을 포함한B형간염바이러스 표면항원(Hepatitis B surface antigen, HBsAg) 및 C형간염바이러스 항체(anti-HCV) 검사의 숙련도시험을 실시하여 왔다. 이에 질병관리본부에서 2012년부터 2015년까지 모든 허가된 혈액원에서 실시한 헌혈혈액 선별검사인 HBsAg 과 anti-HCV 숙련도시험 결과에 대하여 보고하고자 한다. 방법: 질병관리본부는 HBsAg 및 Anti-HCV 검사의 숙련도시험 사업을 혈액안전사업단에 참여하고있는 중앙대학교병원에 위탁하여 특화된 패널을 제조하고 혈액원으로 발송하였다. 패널을제조하기 위하여, 중앙대학교 혈청검체은행으로부터 성상이 잘 밝혀져 있는 양성 검체와 음성 검체를 분양받아 사용하였고, 제조된 패널들의 설정값(S/CO) 확인을 위해 세 개의 의료기관에서 각패널마다 3회 반복 검사로 확인 후 사용하였다. 결과: HBsAg 및 Anti-HCV 검사의 숙련도시험을 실시한 혈액원은 6개 패널에 대하여 대부분정답을 보고하였다. 평균(연도) 정답률은 다음과같다: HBsAg 검사의 숙련도시험은 98.7% (2012), 98.5% (2013), 99.1% (2014) 및 99.6% (2015); anti- HCV 검사의 숙련도시험은 97.7% (2012), 99.5% (2013), 99.1% (2014), 그리고 99.8% (2015)였다. 결론: 혈액원의 선별검사 능력을 향상시키기위하여 질병관리본부는 HBsAg 및 Anti-HCV 검사에 대한 특화된 패널을 계속적으로 사용하는숙련도시험 위탁사업을 지속 운영하려한다. 또한, 프로그램의 질향상을 위하여 만족도 조사도실시할 것이다.

        한국 공공 제대혈은행의 보관 정책 개선: 장기 보관 및 단기 보관 제대혈의 품질 비교

        Background: The decreased use of cord blood units (CBU) due to improvements in haploidentical transplantation is a financial burden for public cord blood banks. Currently, there is no guidance regarding the length of cryopreservation of CBU in Korean public banks. The relative quality of long-term storage CB (LTCB) and short-term storage CB (STCB) needs to be evaluated to establish a storage policy. Methods: Thirty-four and thirty-one units of CB cryopreserved for less than one year and up to 14∼15.5 years, respectively, in the Busan Gyeongnam Public Cord Blood Bank were assessed. The total nucleated cells (TNCs), CD34+ cell counts, and colony-forming units-granulocyte monocyte (CFU-GM) were examined. The cell viabilities were evaluated by Eosin-Y exclusion staining and 7-aminoactinomycin D flow cytometry. The number of stored Korean public CB units from 2000 to 2016 was determined and categorized according to TNCs. Results: The post-thawing viability of the STCBs measured by flow cytometry was consistently higher than that of the LTCBs (TNCs, 62.5% vs 57.3%; MNCs, 93.1% vs 88.9%; CD34+ cells 95.7% vs 94.0%). The CD34+ cell viability was significantly higher in STCB (P=0.03). The CFU-GM after thawing was higher in STCBs (61.5±23.4 vs 49.9±22.8 [0.95 mm2 ] P=0.05). Of the 48,161 CB units stored until 2016, Dec, 9,493 (19.7%), which were stored until 2006, had been stored for more than 10 years. Conclusion: LTCB with a low number of cells (<0.7×109 cells) should be considered to exclude from storage for therapeutic purposes to improve the storage efficiency.         We investigated whether three flavonoids including daidzein, baicalein, hesperidin and ursolic acid, a triterpene acid, affect proliferation of MCF-7 human breast cancer cells stimulated by estrogenic compounds. Ursolic acid and baicalein inhibited proliferation of MCF-7 cells induced by PhIP, a food-derived carcinogen with estrogenic activity. Daidzein and hesperidin inhibited estradiol-induced proliferation of MCF-7 cells. These compounds should be further investigated for the possible involvement in signaling pathway after estrogen receptor binding in breast cancer cells.

        원주 칼국수 및 만두의 향토 음식 육성에 대한 소비자 의향 분석

        The purpose of this study was to analyze the consumer intentions on promote kalguksu (noodle soup) and dumplings as Wonju local food. The online survey was conducted for 527 Adults over 20 years old, and the all data were used for analysis. The data were analyzed using the SPSS program, and chi-care test, t-test, and frequency analysis were performed. The research results were as follows. The preferences of kalguksu (p<0.01) and dumplings (p<0.001) showed significant differences according to the region of residence and age. Kalguksu and dumplings were mainly eaten for lunch by visiting restaurants. Residents of other regions evaluated peach bulgogi (p<0.01), mulberry leaf rice (p<0.001), and jangkalguksu (noodle soup with sauces) (p<0.01) as local foods in Wonju more likely than residents of Wonju. The opinion prevailed that respondents were willing to purchase the possibility of developing kalguksu, dumplings, and kalmandu (3.53), foster professional restaurants (3.49), and inherit and develop them as representative foods in Wonju (3.41).
