        수산물 중 에톡시퀸의 LC-MS/MS 정량분석법 개발

        Ethoxyquin (EQ, 1,2-dihydro-6-ethoxy-2,2,4-trimethyl-quinoline) is quinoline-based antioxidant used in the animal feed and food industry to protect the raw materials and final products against oxidation. In recent years the use of synthetic antioxidants in fishmeal ingredients carry-over to farmed fish fillets has received increasing attention in food safety. This study was conducted to develop an analytical method to determine EQ in aquatic products. The analytes were confirmed and quantified via liquid chromatography-tandem mass spectrometry (LC-MS/MS) in the positive ion mode using multiple reaction monitoring (MRM). The sample was extracted with 1 N HCl (in case of flatfish extracted with 1 N HCl containing 10% acetonitrile). Then, solid phase extraction (SPE) was used for the cleanup. Standard calibration curves presented linearity with the correlation coefficient (r2) > 0.99, analyzed at 0.005 - 0.2 mg/kg concentration. The developed method was validated according to the Codex Alimentarius Commission (CAC) guideline. The limits of quantitation for EQ were 0.01 mg/kg. Average recoveries ranged from 81.3% to 107%. The repeatability of measurements, expressed as the coefficient of variation (CV, %), was below 10%. The analytical method was characterized with high accuracy and acceptable sensitivity to meet CODEX guideline requirements and would be applicable to analyze the EQ residue in aquatic products.

        Gene expression profile of PM014 of immortalized mouse lung epithelial cells in response to the effect of PM014 on radiation‑induced fibrosis

        Radiotherapy is a major method for cancer treatment, but it frequently causes various side effects such as radiation-induced pneumonia and pulmonary fibrosis. Drugs to treat these side effects are urgently needed in the clinic, since there is no clearly defined medication for treating these symptoms. Previous studies demonstrated that the herbal formula, PM014, is effective for radiation-induced lung injury and fibrosis in mice. In this study, we investigated gene expression profiles to understand the mechanism of action behind the effects of PM014 on radiation induced damage in immortalized lung epithelial cells, MLE12. We performed QuantSeq 3′ mRNA-Seq analysis on the mRNA from radiation treated MLE-12 cells in the presence and absence of PM014. Transcriptome analysis found that 217 genes were significantly affected by PM014. Among them (217 genes, > twofold, p value < 0.05, 4 normalize), 77 genes were found to be upregulated, and 140 genes were downregulated in response to PM014 treatment in a dose dependent manner. Using the Kyoto Encyclopedia of Genes and Genomes analysis, we found that genes involved in cytokine–cytokine receptor interaction pathways were the most strongly affected by PM014. Based on these data, we selected 20 genes, and performed real-time PCR. Expression of 11 genes, including IL-18, IL-12a, Tnfrsf9, IL-17, CCR5, Blnk, Irf8, Nrros, TGF-β, Relt, and Cxcl2 was increased after irradiation, while PM014 treatment showed the reversed expression pattern of these genes. Therefore, PM014 may be useful for the treatment of radiation induced lung injury by modulating genes involved in cytokine–cytokine receptor interaction pathway.

        LC-MS/MS 및 GC-MS/MS를 활용한 수산물 중 디아제팜의 정량분석법 개발

        본 연구는 국내 생산 및 수입 양식 수산물에 대해 잔류할 수 있는 향정신성 의약품인 디아제팜 대한 안전관리강화기반을 위해 마련되었다. 중국인민공화국 국가 표준시험법(GB 29697-2013)을 기반으로 전처리 방법을 개선하여 GC-MS/MS 시험법을 확립하였으며, LC-MS/MS 방법과의 기기간 검증을 통해 확립된 시험법의 선택성, 정량한계 및 회수율에 대한 검증을 통해 디아제팜 시험법으로서의 유효성을 확인하였다. LC-MS/MS의 경우 아세토니트릴로 추출 후 PSA를 이용해 정제하였고, GC-MS/MS의 경우 아세토니트릴로 추출후 $C_{18}$카트리지를 이용해 정제하였다. 디아제팜은 표준용액을 정량한계를 포함한 농도에 따라 검량선을 작성한 결과 두 기기 모두 $r^2$> 0.99 이상의 직선성을 확인하였다. 본 실험에서의 검출한계와 정량한계는 LC-MS/MS 및 GC-MS/MS 모두 0.0004 mg/kg, 0.001 mg/kg 수준이었으며, 평균 회수율은 각각 99.8~106%, 109~124%이었다. 또한, 분석오차는 모두 15% 이하로 정확성 및 재현성이 우수하였으며, CODEX 가이드라인 규정에 만족하는 수준이었다. 따라서 개발된 시험법은 안전한 수산물의 국내 유통과 잔류실태조사를 위해 활용될 것으로 기대한다. The aim of this study was to develop an analytical method for the quantification of diazepam residues in fishery products, using liquid and gas chromatography-tandem mass spectrometry (LC-MS/MS and GC-MS/MS). The sample utilized in the study was extracted from the fish sample (crucian carp) using 0.1% formic acid in acetonitrile. For the utilization of the purification process, the dispersive solid phase extraction (dSPE) was used for LC-MS/MS, dSPE and SPE was used for GC-MS/MS, respectively. To be sure, the standard calibration curves showed a good linearity as the noted correlation coefficients, $r^2$ was > 0.99. The average recoveries for accuracy ranged in 99.8~124% for the samples which were fortified at three different levels (0.001, 0.002 and 0.010 mg/kg). The correlation coefficient for the precision effect was measured at a range of 4.01~11.8%. The limit of detection (LOD) for the diazepam analysis was 0.0004 mg/kg, and the limit of the quantification (LOQ) was 0.001 mg/kg. The proposed analytical method was characterized with a high accuracy and acceptable sensitivity to meet the established Codex Alimentarius Commission (CAC/GL71-2009) guideline requirements. We therefore established the optimal analysis method for the determination of diazepam in the fishery products using LC-MS/MS and GC-MS/MS. It would be applicable to analyze the diazepam residues in fishery products in further studies on this subject.

        LC-MS/MS를 이용한 수산물 중 콜리스틴 분석법 개발

        Colistin is a last resort antimicrobial agent against multi-drug resistant Gram-negative bacteria. This study was conducted to develop an analytical method to determine colistin in fish and shrimp. The analytes were confirmed and quantified via liquid chromatography-tandem mass spectrometry (LC-MS/MS) in the positive ion mode using multiple reaction monitoring (MRM). The sample was extracted with acidified 5% methanol (containing 0.5% formic acid). Then, solid phase extraction (SPE) was used for cleanup. Matrix-matched calibration curves were linear over the calibration ranges (0.05–1.2 mg/kg) for all the analytes into blank sample with r2>0.99. All the values fulfilled the criteria requested by the Codex guidelines. Average recoveries ranged from 85.9% to 107.9%. The repeatability of measurements, expressed as the coefficient of variation (CV, %), was less than 15%. The limit of detection (LOD) was 0.02 mg/kg, and the limit of quantitation (LOQ) was 0.05 mg/kg. This improved method showed higher accuracy and acceptable sensitivity to meet the CAC guideline requirements and is applicable for the analysis of residual colistin (A+B) in fish and shrimp.

        중주파 전기 근육자극을 활용한 가정용 미용기기의 안면 피부 개선 효과

        This study was aimed to investigate the effects of the EMS (Electrical Muscle Stimulation) at-home beauty device on the facial skin improvement. The EMS beauty device massages the skin surface and the muscle layer in the skin through a combination of medium frequency electrical stimulation and physical stimulation that are known to effective in skin rejuvenation. In this study, the EMS beauty device reproducing Petrissage massage method and functional cosmetic containing adenosine were used. For four weeks of study period, the EMS beauty device and the functional cosmetic were applied together to the test group with 11 women, whereas only the functional cosmetic was applied to the control group with 11 women. The EMS beauty device was applied for 10 minutes every night after cleansing face to the test group only, and the functional cosmetic was applied every morning and night after cleansing face to both groups. The skin improvements were evaluated after two and four weeks from the first visit (baseline) by mechanical measurements. As the evaluation results, significant (p<.05) improvements of skin elasticity (R2 and Mean CoR), skin sagging, and double chin lifting showed in the test group compared with the baseline, and compared with the control group. Therefore, the EMS beauty device is considered to be effective to improve skin features related to skin aging. And this study may expect to be useful data for global advancement and enforcement of market competitiveness of K-beauty industry and to be a foundation for further in-depth study.

        LC-MS/MS를 이용한 수산물 중 니트로빈의 정량분석법 개발 및 검증

        본 연구에서는 우리나라 식품공전에서 불검출 물질로 관리하고 있는 니트로빈(nitrovin)에 대해 고감도 정량 정성분석이 가능한 LC-MS/MS를 적용하여 적합한 분석법을 제시하고자 하였다. 수산물 시료는 아세토니트릴/물로 추출하고 아세토니트릴 포화 헥산으로 지방을 제거하여 고상추출 카트리지를 적용하여 정제하였다. 분석물질은 전기분무이온화방법의 positive mode에서 이온화하여 MRM 조건을 확립하여 분석하였다. 개선된 시험법은 CODEX CAC/GL-71 가이드라인에 따라서 정확성, 정밀성, 직선성, 정량한계에 대한 검증을 통하여 유효성을 확인하였다. 본 실험에서의 정량한계는 0.001 mg/kg 수준이며, 정량한계를 포함하는 표준시료에서 얻어진 검량선의 상관계수($r^2$)는 0.985 이상으로 시험법의 직선성이 유효함을 판단할 수 있었다. 또한, 수산물(넙치, 장어 및 새우) 시료에 대한 니트로빈의 평균 회수율과 변동 계수는 72.1~122%, 2.9~16.9%로 확인되어 정확성 및 정밀성이 CODEX가이드라인에 부합하였다. 따라서, 개선된 니트로빈 정량분석법은 수산물 중 니트로빈을 분석하는데 적합하며, 니트로빈에 대한 지속적인 잔류실태조사에 활용되어 수산물 중 니트로빈의 안전관리에 기여할 것으로 판단된다. The objective of this study was to develop a sensitive method for the identification and determination of nitrovin in fishery products by using a solid-phase extraction (SPE), as performed with a liquid chromatography-tandem mass spectrometry (LC-MS/MS). The samples were extracted with a mixture of acetonitrile and water, and were then defatted with acetonitrile saturated hexane, after which further clean-up was accomplished with SPE on the hydrophilic-lipophilic balance (HLB) cartridges. The analytes were subsequently ionized in the positive mode of an electrospray ionization (ESI), and where thereby detected in a process of multiple reaction monitoring (MRM). The linearity (expressed as correlation coefficients) of the matrix calibration curves was > 0.985. The limit of the quantification for the nitrovin was measured at 0.001 mg/kg. The accuracy (expressed as average recovery) was noted between 72.1 and 122%. The precision (expressed as coefficient variation) was noted from 2.9 to 16.9%. According to the CODEX CAC/GL-71 guideline accuracy, precision, linearity, and limit of detection were determined in three matrices (which were flatfish, eel and shrimp). The proposed method was suitable for analyzing the associated nitrovin residues. This application and result can also be a factor to contribute to the non-detection drugs management in fishery products.

        미세전류, 중주파 기능이 탑재된 가정용 미용기기가 안면 피부 탄력 및 리프팅에 미치는 영향

        At-home beauty devices used in facial skin are developed by combining various technologies such as electromagnetic waves, microcurrents, and lasers according to the main purpose of consumers. This study aimed to assess the effects of increasing of facial elasticity, improvement of eye wrinkles, and facial lifting in healthy Korean women to objectively prove the efficacy of an at-home beauty device equipped with microcurrent (MC) and medium-frequency electrical muscle stimulation (EMS). A total of 44 participants used the at-home beauty device and cosmetics together on the left side of the face (test area), and applied only cosmetics to the right side (control area). The results of the assessment, the improvement in the dermal density and eye wrinkles was clearly observed in the MC mode use group. In the EMS mode use group, the lifting effect of the cheek and decreasing facial swelling were clearly observed, and the lifting effect of the sagging double chin was also confirmed. Especially, EMS group had derived reproducible results like previous studies, and the skin improvement effects were proved again. MC and EMS, which are mainly used for therapeutic purposes, were developed into at-home beauty device, and this study confirmed that they are effective in facial elasticity, eye wrinkles, and facial lifting. Based on the research, we hope that various research for at-home beauty devices equipped with various functions are actively conducted.

        모발 염모제 삼원색의 혼합에 따른 헤어 컬러 변화에 관한 연구

        Hairstyles can be created by various treatments such as haircut, perm, coloring and bleaching. In particular, hair colors can vary depending on circumstance, place, season, feeling, mood and image. This study targeted to learn how to mix hair dyes to create wanted color with a better understanding of the mixture of hair colors and prove the acquisition of wanted color. For this, the followings were analyzed through a theoretical review: hair coloring, types and characteristics of hair dyes, characteristics of melanin pigments, color charts of ‘W’. In terms of an empirical study, color changes by ratio according to hair dye levels were derived. The acquired hair dyes were applied to white hair and black hair, and a total of 90 different colors were obtained. The test results found the followings: First, ‘45(R)’ and ‘88(B)’ were mixed in a 1:1 ratio, it was unable to create ‘6 (purple)’. When ‘88(B)’ and ‘/3(Y)’ were mixed, the secondary color of ‘/2(green)’ in the color chart wasn’t expressed. In fact, it was close to ‘/11(ash gray)’. When ‘/45(R)’ and ‘/3(Y)’ were blended, the secondary color of ‘/44(orange)’ in the color chart was ‘level 12’. When they were mixed in the ratio of 3 to 1, the color was closest to ‘12/44’. In other words, when hair dyes are mixed, color gets darker when hair dyes with strong color are used than when they are mixed at a certain ratio. In black hair, hair color does not change much by a mixing ratio. In fact, brightness can only be classified based on the existence of redness. It’s hoped that the study results would help hairdressers understand the standards of color changes by the mixture of hair dyes and establish the grounds for consulting hair coloring through the development of a color chart.
