        Antioxidant and Biological Activity in the Leaves of Adzuki Bean (Vigna angularis L.)

        The adzuki bean (Vigna angularis L.) is a red-grained legume that has a number of essential nutrients and is used in traditional dishes in Asia. Adzuki bean industrial by-products are also a potential low-cost source of some unique bioactive polyphenols. Hence, here, the authors aimed to perform a comparative study of the phytochemical profiles of the leaves and seeds of the adzuki bean and compare their antioxidant, α-glucosidase inhibition, and tyrosinase inhibition activity. The authors assessed antioxidant activity by DPPH, ABTS, FRAP, PR, TPC, and SOD assays, which showed wide variation, respectively. From the relative antioxidant capacity index results, 10 adzuki bean landraces were selected to compare for phytochemicals and bioactivity using leaf and seed extracts. Antioxidant, α-glucosidase inhibition, and tyrosinase inhibition activity in the leaf extracts were higher than in the seed extracts, and there were more flavonols and isoflavones in the leaf extracts than in the seed extracts. This study demonstrated that adzuki bean leaf extracts could be a new natural antioxidant or antidiabetic agent and a skin whitener and can also be used in industrial applications.

        더덕과 일부 한약재 열수추출물의 혼합 비율에 따른 생리활성

        The aim of this study was to investigate the physiological functionalities of Codonopsis lanceolata, Glycyrrhiza uralensis, Chaenomeles sinensis, Crataegus pinnafida, and their mixtures. We also determined their antioxidative, fibrinolytic, and αglucosidase inhibitory activities. The antioxidative activities of Codonopsis lanceolata, Glycyrrhiza uralensis, Chaenomeles sinensis, and Crataegus pinnafida were 79%, 88.3%, 89.9%, and 89.3% respectively. Their fibrinolytic activities were 0.80plasmin unit/ml, 0.57 plasmin unit/ml, 0.52 plasmin unit/ml, and 0.53 plasmin unit/ml respectively. The αglucosidase inhibitory activity of Codonopsis lanceolata was 25%. The 10fold diluents of Glycyrrhiza uralensis, Chaenomeles sinensis, and Crataegus pinnafida showed αglucosidase inhibitory activities of 93.6%, 65.3%, and 61.3% respectively. In antioxidative activity tests of the medicinal plants mixtures at various ratios, the mixtures of Glycyrrhiza uralensis, Chaenomeles sinensis, and Crataegus pinnafida with Codonopsis lanceolata showed antioxidative activities of approximately 90%. In fibrinolytic activity tests mixtures(1:1) of Codonopsis lanceolata with Chaenomeles sinensis and Crataegus pinnafida exhibited increases of 23% and 24% in activity respectively. In αglucosidase inhibitory activity tests, a mixture (4:1) of Codonopsis lanceolata and 10fold diluted Glycyrrhiza uralensis showed an inhibitory activity of 98%, a mixture (3:1) of Codonopsis lanceolata and 10fold diluted Chaenomeles sinensis showed an inhibitory activity of 69.6%, and a mixture (1:1) of Codonopsis lanceolata and 10fold diluted Crataegus pinnafida showed an inhibitory activity of 50.2%. In conclusion, the mixtures of Glycyrrhiza uralensis, Chaenomeles sinensis, and Crataegus pinnafida with Codonopsis lanceolata will be used as a material for the development of biofunctional foods.

        Antioxidative and anti-inflammatory activities of Phaseolus aureus

        Phaseolus aureus (mung bean), Leguminosae, has been used as an antidote from the ancient time. Especially, it has been widely used for cleaning face and skin in oriental countries. Although several constituents such as fatty acids, phytoallexin and phaseol derivatives were reported in Paureus and related species including seedlings, there has been a few report to describe its biological activity. Therefore, in this investigation, the ethanol extract from R aureus was obtained and its biological activities including the antioxidative and anti-inflammatory activities were studied. The 70% ethanol extract from P aureus showed dose-dependent antioxidative activity (52.3% inhibition at 4 mg/ ml) against lipid peroxidation assay, while the extract did not show the inhibitory activity of superoxide radical formation. The extract also showed the topical anti-inflammatory activity against croton-oil and arachidonic acid induced mouse ear edema test (18-19% inhibition at 7.5 mg/ear) as well as mild inhibitory activity against picryl chloride induced delayed hypersensitivity in mouse. For investigating active principles, vitexin and isovitexin (apigenin C-glycoside) as flavonoids, and adenosine were isolated from the extract using silica gel chromatography. The actual contents of vitexin and isovitexin were found to be 3.7 and 2.4 mg/g extract, respectively. Vitexin and isovitexin showed the antioxidative activity. They showed the topical anti-inflammatory activity, although the activities were not potent compared to the reference compounds. These resuIts suggested that vitexin and isovitexin may be, at least in part, the compounds contributing the antioxidative activity in vitro and the topical anti-inflammatory activity of P.aureus in vivo. All results of present study might be one of the scientific rationale in using mung bean for skin care from the ancient time.

        Antioxidant Activity of Talinum portulacifolium (Forssk) Leaf Extracts

        The aim of the study is to investigate the antioxidant activity through, reducing power, 2, 2- diphenyl-1-picryl hydrazyl radical (DPPH), nitric oxide radical (NO), superoxide radical, hydrogen peroxide radical (H2O2) scavenging activity and the amount of total phenolic compounds of chloroform, ethyl acetate, methanol and aqueous extracts of the leaves of Talinum portulacifolium. Chloroform extract of leaves of T. portulacifolium showed highest antioxidant activity, with a direct relationship between activity and concentration of extracts (15 - 240 μg/mL). Among all the extracts, the highest amount of the total polyphenolic compounds was found in the chloroform extract. Chloroform extract of T. portulacifolium showed an important free radical scavenging activity towards the DPPH, NO, Superoxide and H2O2 radicals, with IC50 values of 133.26, 165.75, 156.34 and 135.29 μg/mL, respectively. In the lipid peroxidation assay, extracts of chloroform and ethyl acetate showed a remarkable inhibitory activity. The extracts showed significant activity in all the experiments but lower than the standard antioxidant, ascorbic acid.         식물유래 천연색소의 항산화 활성

        The objective of this study was to determine the effect of antioxidant enzyme activity and radical scavenging activities of thirteen kinds of natural plant pigments. The analytic method of antioxidant activities were measured by estimating DPPH free radical scavenging and nitrite scavenging ability activity. The free radical scavenging activity by method using stable free radical DPPH was the highest in the red cabbaged pigment. Addition of ethanol extract 1㎎/㎖ from onion peel pigment displayed remarkable effect on nitrite scavenging ability about 91.9%. Antioxidative enzyme activity was evaluated in terms of superoxide dismutase(SOD), catalase(CAT), and ascorbate peroxidase(APX) activity. The bitter melon pigment had the highest SOD activity of 87.3%. The activities of CAT and APX were higher in the mulberry leave pigment compared with other natural plant pigments. In contrast, CAT activity of plant pigment samples were unaffected. These results suggest that natural plant pigment had the potent biological activities such as antioxidant enzyme activities, and that their activities exhibited differently depending on each kind of pigments. 선발된 13종의 천연색소 추출물에 대하여 free radical 소거활성 및 아질산염 소거활성, 그리고 항산화효소 활성에 대하여 조사하였다. 천연색소 종류별 free radical 소거활성을 보면 적양배추(red cabbage), 양파껍질(onion peel), 레드비트(redbeet), 흑미(black rice), 오디(mulberry) 색소 등에서 상대적으로 높게 나타났고, 청치자(blue gardenia) 및 황치자(yellow gardenia), 완숙여주(mature bitter melon) 색소 등에서는 비교적 낮은 활성을 보였다 또한 아질산염에 대한 소거능을 조사한 결과, 반응용액 pH 1.2의 경우 양파껍질, 레드비트, 치자황, 지황(chinese foxglove), 치자청 색소에서 높은 소거 활성을 나타냈으며, 적양배추와 오디 색소에서는 상대적으로 낮은 소거활성을 보였다. 그리고 반응용액 pH 6.0에서는 대부분의 색소 추출물에서 아질산염 소거활성을 나타내지 않았다. 그리고 항산화효소 활성 결과를 보면, SOD 활성의 경우 여주, 흑미, 포도과피 등에서 높은 활성을 나타냈고, 자색고구마, 지황에서도 비교적 높은 활성을 보였다. 또한 APX 활성은 뽕잎, 오디, 지황의 순으로 높은 활성을 보였으며, CAT 활성도 APX 결과와 비슷한 양상으로 나타났다. 이는 천연색소에 있어 항산화 효소의 종류에 따라 그 활성에 차이가 있음을 알 수 있다. 본 연구결과를 통해 다양한 생리활성을 지니고 있는 천연식물색소에 대해 높은 항산화 기능성을 확인함으로서 향후 고부가가치의 향장소재 및 식품소재로서의 개발가능성이 매우 높음을 시사해 주고 있다.

        다류의 유형별 항산화 성분함량 및 항산화 활성 연구

        The purpose of this study was to investigate the contents of antioxidant compounds and antioxidant activities in teas. A total of 99 teas were tested for their antioxidant activities based on their ability to scavenge DPPH (1,1-diphenyl-2-picrylhydrazyl) free radical. Antioxidant activity was expressed as mg of ascorbic acid equivalents per 100 g tea sample (L-ascorbic acid equivalent antioxidant capacity, AEAC) and was expressed as mg per 1 serving size(free radical scavenging activity, FSC50). The total polyphenol contents of the extracts was quantified using the Folin-Ciocalteu method and the total flavonoid contents of the extracts was determined using a modified method of Davis. Vitamin C was analyzed by HPLC method. According to the AEAC value, mate tea, green tea, black tea, oriental raisin tea, chamomile tea and burdock tea showed relatively high antioxidant activities. Polyphenolic compounds were the major naturally occurring antioxidant compounds found in teas and the high concentrations of total polyphenol compounds were observed in black tea, green tea and mate tea. The high concentrations of total flavonoid compounds were observed in mate tea, black tea and yam tea and the amount of vitamin C contents were found to be high in citron tea and green tea. As a result, steeping tea was much higher in antioxidant activity than either powdered tea or liquefied tea. According to the study, a high correlation was demonstrated between the total polyphenol contents and antioxidant activities in teas(r=0.846) and correlations between the total flavonoid contents and antioxidant activities was statistically significant in teas(r=0.625). It was found that also the proportional relationship established among the total polyphenol content and antioxidant activities. That is, antioxidant activity of teas has been confirmed to have been caused by the total polyphenol.

        Antioxidant Activity of Some Yogurt Starter Cultures

        Several commercial yogurt starter cultures (L. acidophilus LA5, L. casei 01, L. acidophilus LA100, L. bulgaricus LB207 and L. rhamnosus GG744) were investigated for antioxidant activity by using in vitro assays. From the results of the present work, all strains tested showed varying degrees of antioxidant activity. L. bulgaricus LB207 showed the highest antioxidant activity, with a linoleic acid peroxidation inhibition of 81.3%. Hydroxy radical scavenging activity, ferrous iron chelating activity, reducing power and superoxide dismutase (SOD) activity were also studied. L. bulgaricus LB207 showed the highest hydroxy radical scavenging activity and L. casei 01 showed the highest chelating activity. L. bulgaricus LB207 and L. acidophilus LA100 showed good reducing power. All the strains in this study showed low SOD activity. The results of the present work suggest that antioxidant activity of L. bulgaricus LB207 due to its strong hydroxy radical scavenging activity and reducing power.

        천연복합물(애기달맞이꽃, 상황버섯, 감초) 에탄올 추출물의 항산화 활성 및 피부미백 효능

        The aim of this study was to assess the antioxidant activity and whitening efficacy of ethanolic extract of herb complex (Oenothera laciniata, Phellinus linteus and Glycyrrhiza uralensis Fischer). The antioxidant activity was evaluated by DPPH-radical inhibition activity, xanthine oxidase (XO) inhibition activity, superoxide dismutase (SOD)- like activity, and ABTS radical inhibition activity. Whitening efficacy was evaluated by mushroom tyrosinase inhibition activity. In the antioxidant activities of ethanolic extract of herb complex at 1,000 μg/mL concentration, DPPH radical inhibition activity was over 80% and similar to that of BHA, positive control. XO inhibition activity was over 90%. The SOD-like activity was over 30%. ABTS radical inhibition activity was over 95%. The ethanolic extract of herb complex reduced the mushroom tyrosinase activity in a dose-dependant manner, showing a inhibitory rate over 60% at 1,000 μg/mL concentration compared to the untreated control. The ethanolic extract of herb complex did not affect on cell growth at test concentrations (0~1,000 μg/mL). From these results, it was found that the ethanolic extract of herb complex had a good antioxidant activity and whitening efficacy. Hence, it could be used as cosmetic ingredients for skin whitening and/or skin anti-ageing.

        Change of Fibrinolytic and Antioxidative Activities of Codonopsis lanceolata According to Various Storage Conditions, and Heat or Salt Treatments

        We investigated the changes of fibrinolytic and antioxidative activities of wild or cultivated Codonopsis lanceolata, which were packed in woven polypropylene (WP) film or low density polyethylene (LDPE) film and stored for 0, 15, and 30 days at refrigerated (2~4℃) or room (18~20℃) temperature (So we have 16 samples, and wrote them as the abbreviated words such as W-WP-RE-15, W-WP-RO-15, W-LDPE-RE-15, W-LDPE-RO-15, W-WP-RE-30, W-WP-RO-30, W-LDPE-RE-30, W-LDPE-RO-30, C-WP-RE-15, C-WP-RO-15, C-LDPE-RE-15, C-LDPE-RO-15, C-WP-RE-30, C-WP-RO-30, C-LDPE-RE-30, C-LDPE-RO-30). Fibrinolytic activity of fresh Codonopsis lanceolata cultivated in the mountain or field were 0.8 unit (plasmin unit/ml) or 7.3 units, respectively. In descending order, the activities of wild Codonopsis lanceolata stored for 15 days were as followed ; LDPE-RE (0.70 unit), WP-RO (0.52 unit), WP-RE (0.45 unit), and LDPE-RO (0.30 unit). After 30 days, fibrinolytic activities of them decreased to 0.47 unit (LDPE-RE), 0.28 unit (WP-RO), 0.21 unit (WP-RE), and 0.30 unit (LDPE-RO). Considering from the point of fibrinolytic activity, the optimal storage condition of wild Codonopsis lanceolata was packing with LDPE film and storing at 4℃. The change of fibrinolytic activities of Codonopsis lanceolata cultivated in the field revealed the similar trend as wild samples, but the fibrinolytic activities of 30 days-stored samples were maintained better than the wild Codonopsis lanceolata. Fibinolytic activity of wild Codonopsis lanceolata was increased by heating for 5 min at 100℃ and decreased by addition of NaCl. Antioxidative activities of Codonopsis lanceolata were also compared from the electron donating activity. Fresh Codonopsis lanceolata had about 70% of electron donating activity. Independent of cultivation area, electron donating activity dropped to 19~74% (wild Codonopsis lanceolata) and 27~59% (cultivated Codonopsis lanceolata) during 15 days storage. But after 30 days storage, we obtained the unexpected results, which meant that the activities were higher activities than 15 days-stored samples or even though the fresh samples. In general, Codonopsis lanceolata could maintain antioxidative activities most strongly with LDPE film and chilled condition.

        Antioxidative Activity and Anti-inflammatory Effects on the Murine Macrophages of Methanol Extracts of Amphibians

        Oxidative stress has been reported to be one of causes of neuritis. This study examined antioxidative activities of methanol extracts of six amphibian species known to be medicinal animals (Rana catesbeiana, R. coreana, R. rugosa, R. dybowskii, R. nigromaculata, and Hyla japonica) and investigated their effects of inhibiting nitric oxide (NO) production and cytotoxicity on the murine macrophage RAW264.7 cells. As inflammation is closely associated with reactive oxygen species, assays on 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity, xanthine oxidase inhibitory activity, superoxide anion radical scavenging activity and NO scavenging activity of the extracts of the six species were performed to investigate their antioxidative activity. The results obtained were as follows; All extracts showed antioxidative activity, and the activity of R. dybowskii was the highest in comparison among those. Anti-inflammatory effects of the extracts were also examined, the five extracts except that of R. rugosa did not show cytotoxicity for RAW264.7 cells at the maximal concentration ($1,000{\mu}g\;mL^{-1}$). Selectivity index, meaning NO scavenging activity compared to cytotoxicity, showed the highest level in the extract of R. dybowskii. These results will be very useful basic data for future studies on prevention and treatment of human diseases to understand the biological roles of amphibian extracts throughout the antioxidative or anti-inflammatory pathways.
